A Novel Bromoindole Alkaloid from a Korean Colonial Tunicate Didemnum sp.

Chemical investigation on a colonial marine tunicate, Didemnum sp. led to the isolation of a series of indole alkaloids including a new (1) and two known metabolites (2-3). Based on the spectroscopic analysis including 1D and 2D NMR along with MS spectra, the structure of 1 (16-epi-18-acetyl herdmanine D) was elucidated as a new amino acid derivative. The absolute configuration of 1 was determined by comparison of specific rotation with the known compound. The structures of compounds 2 and 3 were also identified as bromoindole containing compounds N-(6-bromo-IH-indole- 3-carbonyl)-L-arginine and (6-bromo-IH-indol- 3-yl) oxoacetamide, respectively, based on <sup>1</sup>H and <sup>13</sup>C NMR data, MS data and specific rotation value. Their pharmacological potentials as antibacterial agents and FXR antagonists were investigated, but no significant activity was found. However, the structural similarity of compound 1 to compound 4 suggested the anti-inflammatory potential of compound 1.

Three new structurally related depsipeptides, halieylindramides F-H (1-3), and two known halieylindra-mides were isolated from a Petrosia sp. marine sponge collected off the shore of Youngdeok-Gun, East Sea, Republic of Korea. Their planar structures were elucidated by extensive spectroscopic data analyses including ID and 2D NMR data as well as MS data. The absolute configurations of halicylin-drarnides F-H (1-3) were determined by Marfey``s method in combination with Edman degradation. The absolute config-urations at C-4 of the dioxyindolyl alanine (Dioia) residues of halieylindramides G (2) and H (3) were determined as 4S and 4R, respectively, based on ECD spectroscopy. The C-2 configurations of Dioia in 2 and 3 were speculated to both be 2R based on the shared biogenesis of the halicylindramides. Halieylindrarnides F (1), A (4), and C (5) showed human farnesoid X receptor (hFXR) antagonistic activities, but did not bind directly to hFXR.

Three novel scalarane sesterterpenes were isolated from a Korean marine sponge, Psammocinia sp., along with four known derivatives. Their structures were elucidated on the basis of NMR, MS and IR spectroscopic data. The three new compounds are 12-deacetoxy-23-hydroxyscalaradial (1), 12-dehydroxy-23-hydroxyhyrtiolide (2) and 12-O-acetyl-16-deacetoxy-23-acetoxyscalarafuran (3), respectively, and the four known compounds are 12-deacetoxy-23-hydroxyheteronemin (4), 12-deacetoxy-23-acetoxy-19-O-acetylscalarin (5), 12-deacetoxy-23-O-acetoxyheteronemin (6) and 12-deacetoxyscalaradial (7). They exhibited cytotoxicity against intractable human cancer cell lines A498, ACHN, MIA-paca and PANC-1, with an IC50 range of 0.4-48 μM. ⓒ2013 Elsevier Ltd. All rights reserves.
Safety and Lung Damage Reduction Activity of Morinda citrifolia (noni) seed

Noni used in food and folk remedies for over 2000 years. Recently, the consumption of noni products has increased, as there is a growing interest in noni as various bioactivity has been revealed. But, anthraquinone, one of the components of noni, causes side effects like hepatotoxicity in some papers. It was present in the husk and seeds. Accordingly, it is important to explore bioactive components while excluding anthraquinone. Meanwhile, it is considered that fine dust is the cause of respiratory disease. It consists of minerals, nitrogen oxide, sulfur oxide, and volatile organic compounds. 1-Nitropropane (1-NP), constituting fine dust, have toxicity to lung cells. In this study, Noni seed was extracted with dichloromethane: methanol (1: 1) solution and then fractionated hexane, ethyl acetate, butanol, and water layer. The ethyl acetate layer was subdivided by size exclusion chromatography, and some components were separated using preparative HPLC. 1H-NMR and 13C-NMR were used for components structure elucidation isolated from noni seed. 1-NP had been tested into lung cells with lignans from noni seed, and cell viability was analyzed compared to those not treated.
Quantification of Allantoin in Yams (Dioscorea sp.) Using a <sup>1</sup>H NMR Spectroscopic Method

Allantoin is an abundant component of yams and has been known as a skin protectant due to its pharmacological activities. In previous methods for allantoin determination using high-performance liquid chromatography (HPLC), the separation was unsatisfactory. We herein developed a 1H quantitative nuclear magnetic resonance (qNMR) method for quantification of allantoin in the flesh and peel of yams. The method was carried out based on the relative ratio of signals integration of allantoin to a certain amount of the internal standard dimethyl sulfone (DMSO<sub>2</sub>) and validated in terms of specificity, linearity (range 62.5-2000 μg/ml), sensitivity (limit of detection (LOD) and quantification (LOQ) 4.63 and 14.03 μg/ml, respectively), precision (RSD% 0.02-0.26), and recovery (86.35-92.11%). The method was then applied for the evaluation of allantoin in flesh and peel extracts of four different yams cultivated in Korea.

Phenanthrenes are representative compounds found in the genus Dioscorea and have many bioactivities. The recent study confirmed that phenanthrenes were present in higher amounts in the peel than the flesh of Dioscorea. Dioscorea is not highly storable, so most of them are peeled and then vacuum-packed or dried and then powdered before distribution. In this process, many peels are produced as a byproduct, discarded because there is no use. This study was conducted to develop an optimized extraction method for peels for other industries such as cosmetics. As standard for evaluation of the extraction method, we used three compounds isolated from Dioscorea batatas. A quantitative analysis method using HPLC was developed to quantify the phenanthrenes in the extract, and method validation was performed. Quantitative analysis was performed in 1 to 10 μg/mL concentration of the standard compounds and confirmed acceptable linearity, accuracy, and precision. Four different conditions extracted the peels, and quantitative analysis was performed. As a result, it showed the highest extraction efficiency in steaming pretreatment, high ethanol concentration, and high extraction temperature.
