        Some Factors Affecting Freezing of Boar Semen in 5 ml Maxi-straws

        A series of experiments were conducted to determine the suitable freezing and thawing temperatures for the freezing of boar semen in 5 ml maxi-straws. The ultrastructure, in vitro fertilization (IVF) and artificial insemination (AI) of frozen-thawed semen were also be evaluated. The 5 cm freezing height gave the best results not only in post-thaw motility rate (54.00%), but also in normal acrosome morphology rate (NAR) (80.23%). There was no significant difference in the post-thaw motility between different thawing temperatures and corresponding thawing times (p>0.05); the group of $52^{\circ}C$ and 25 s gave the highest motility rate (45.00%). As a whole, not only from the motility but also the NAR, thawing at $42^{\circ}C$ was better than the other two treatments. In the freezing packages, 5 ml maxi-straw gave a little lower mobility (40%), viability rate (49.58%), plasma membrane integrity rate (53.91%) and NAR (52.65%) than the 0.25 ml straw, but there was no significant difference between the two straw volumes (p>0.05). The IVF capacity of frozen-thawed semen in this experiment was similar to fresh semen. From ultrastructure observation, the main damage to boar spermatozoa after freezing was seen in the acrosome, such as swelling and formation of vesicles. After AI in recipient Shanghai White sows, frozen-thawed semen from 5 ml maxi-straws and pellets produced 72.2% and 80% conception rate and 7.8 and 8 litter sizes, respectively, and there was no significant difference between the 5 ml maxi-straw and the pellet (p>0.05).

        R&D Status of High-current Accelerators at IFP

        High-current accelerators have many important applications in Z-pinches, high-power microwaves, and free electron lasers, imploding liners and radiography and so on. Research activities on Zpinches, imploding liners, radiography at the Institute of Fluid Physics (IFP) are introduced. Several main high-current accelerators developed and being developed at IFP are described, such as the Linear Induction Accelerator X-Ray Facility Upgrade (LIAXFU, 12 MeV, 2.5 kA, 90 ns), the Dragon-I linear induction accelerator (20 MeV, 2.5 kA, 60 ns), and the Primary Test Stand for Z-pinch (PTS, 10 MA, 120 ns). The design of Dragon-II linear induction accelerator (20 MeV, 2.5 kA, 3 × 60 ns) to be built will be presented briefly.

      • Synthesis, characterization of the phenylquinoline-based on iridium(III) complexes for solution processable phosphorescent organic light-emitting diodes

        A new series of highly efficient Ir(III) complexes, (DPQ)<SUB>2</SUB>Ir(pic-N-O), (F<SUB>4</SUB>PPQ)<SUB>2</SUB>Ir(pic-N-O), (FPQ)<SUB>2</SUB>Ir(pic-N-O), and (CPQ)<SUB>2</SUB>Ir(pic-N-O) were synthesized for phosphorescent organic light-emitting diodes (PhOLEDs), and their photophysical, electrochemical, and electroluminescent (EL) properties were investigated. The Ir(III) complexes, including picolinic acid N-oxide (pic-N-O) ancillary ligand, are comprised with the various main ligands such as 2,4-diphenylquinoline (DPQ), 4-phenyl-2-(2,3,4,5-tetrafluorophenyl)quinoline (F<SUB>4</SUB>PPQ), 2-(9,9-diethyl-9H-fluoren-2-yl)-4-phenylquinoline (FPQ) and 9-ethyl-3-(4-phenylquinolin-2-yl)-9H-carbazole. Remarkably, high performance PhOLEDs using a solution-processable (DPQ)<SUB>2</SUB>Ir(pic-N-O) doped CBP host emission layer were fabricated to give a high luminance efficiency (LE) of 26.9cd/A, equivalent to an external quantum efficiency (EQE) of 14.2%. The calculated HOMO-LUMO energy gaps for (DPQ)<SUB>2</SUB>Ir(pic-N-O), (F<SUB>4</SUB>PPQ)<SUB>2</SUB>Ir(pic-N-O), (FPQ)<SUB>2</SUB>Ir(pic-N-O) and (CPQ)<SUB>2</SUB>Ir(pic-N-O) were in good agreement with the experimental results.

      • Understanding of the capacity contribution of carbon in phosphorus-carbon composites for high-performance anodes in lithium ion batteries

        <P>Phosphorus has recently received extensive attention as a promising anode for lithium ion batteries (LIBs) due to its high theoretical capacity of 2,596 mAh.g(-1). To develop high-performance phosphorus anodes for LIBs, carbon materials have been hybridized with phosphorus (P-C) to improve dispersion and conductivity. However, the specific capacity, rate capability, and cycling stability of P-C anodes are still less than satisfactory for practical applications. Furthermore, the exact effects of the carbon support on the electrochemical performance of the P-C anodes are not fully understood. Herein, a series of xP-yC anode materials for LIBs were prepared by a simple and efficient ball-milling method. 6P-4C and 3P-7C were found to be optimum mass ratios of x/ y, and delivered initial discharge capacities of 1,803.5 and 1,585.3.mAh.g(-1), respectively, at 0.1 C in the voltage range 0.02-2 V, with an initial capacity retention of 68.3% over 200 cycles (more than 4 months cycling life) and 40.8% over 450 cycles. The excellent electrochemical performance of the 6P-4C and 3P-7C samples was attributed to a synergistic effect from both the adsorbed P and carbon.</P>

      • Hydrate nucleation in quiescent and dynamic conditions

        Gas hydrate nucleation is the spontaneous formation of an ordered crystalline lattice from a disordered phase. This inherently random process often involves long induction times particularly in quiescent conditions. An experimental study was conducted to explore the kinetics of hydrate formation in the presence of mineral grains and when subjected to mechanical agitation. Results show that tetrahydrofuran THF hydrate nucleation is facilitated in the presence of most minerals, and induction times are a function of mineralogy and surface characteristics. While mechanical vibration does not suppress the inherent stochastic nature of nucleation, mechanical agitation triggers nucleation when the imposed acceleration exceeds ~10m/s<SUP>2</SUP>.

      • Molecular and structural characteristics of multidrug resistance-associated protein 7 in Chinese liver fluke Clonorchis sinensis

        <P>Multidrug resistance-associated protein 7 (MRP7, ABCC10) is a C subfamily member of the ATP-binding cassette (ABC) superfamily. MRP7 is a lipophilic anion transporter that pumps endogenous and xenobiotic substrates from the cytoplasm to the extracellular milieu. Here, we cloned and characterized CsMRP7 as a novel ABC transporter from the Chinese liver fluke, Clonorchis sinensis. Full-length cDNA of CsMRP7 was 5174 nt, encoded 1636 amino acids (aa), and harbored a 147-bp 5'-untranslated region (5'-UTR) and 116-bp 3'-UTR. Phylogenetic analysis confirmed that CsMRP7 was closer to the ABCC subfamily than the ABCB subfamily. Tertiary structures of the N-terminal region (1-322 aa) and core region (323-1621 aa) of CsMRP7 were generated by homology modeling using glucagon receptor (PDB ID: 5ee7_A) and P-glycoprotein (PDB ID: 4f4c_A) as templates, respectively. CsMRP7 nucleotide-binding domain 2 (NBD2) was conserved more than NBD1, which was the sites of ATP binding and hydrolysis. Like typical long MRPs, CsMRP7 has an additional membrane-spanning domain 0 (MSD0) and cytoplasmic loop, along with a common structural fold consisting of MSD1-NBD1-MSD2-NBD2 as a single polypeptide assembly. MSD0, MSD1, and MSD2 consisted of TM1-7, TM8-13, and TM14-19, respectively. The CsMRP7 transcript was more abundant in the metacercariae than in the adult worms. Truncated NBD1 (39 kDa) and NBD2 (44 kDa) were produced in bacteria and mouse immune sera were raised. CsMRP7 was localized in the apical side of the intestinal epithelium, sperm in the testes and seminal receptacle, receptacle membrane, and mesenchymal tissue around intestine in the adult worm. These results provide molecular information and insights into structural and functional characteristics of CsMRP7 and homologs of flukes.</P>

        A simple method for the parallel quantification of nicotine and major solvent components in electronic cigarette liquids and vaped aerosols

        <P>An analytical method was developed for the parallel quantitation of nicotine (Nic) and two key solvents (propylene glycol (PG) and vegetable glycerin (VG)) in e-cigarette (EC) liquids before vaping and from aerosols after vaping. For analysis of the EC refill solutions, the samples were diluted by a factor of about 100 in methanol. The aerosol samples generated by a modified international puffing protocol were initially collected on Cambridge filter pads and extracted with methanol. Both types of samples were analyzed by a gas chromatography-flame ionization detector (GC-FID) together with the mass change tracking (MCT) procedure introduced in our earlier study. The recovery of all three target components (Nic/PG/VG) in both EC liquid and aerosol samples was assessed after spiking Nic at four different concentrations (2, 5,10, and 20 mg g(-1)) in the e-solutions (prepared in the laboratory at three different PG:VG mass ratios of 10:0, 5:5, and 0:10). The method recoveries of Nic, PG, and VG in the e-liquid samples were 96.0 +/- 1.0, 96.0 +/- 1.2, and 101.4 +/- 6.9%, respectively, while those in the aerosol samples were slightly lower at 94.7 +/- 5.6, 85.5 +/- 3.0, and 91.4 +/- 15.7%, respectively. The amounts of VG and Nic in the e-liquid had a significant influence on the emission factors of PG, VG, and Nic. The detection limit values (ng) were 036 (Nic), 0.72 (PG), and 8.15 (VG) for the liquid samples and 0.51 (Nic), 0.96 (PG), and 3.99 (VG) for the aerosol samples.         New Evidence of Alleles (V199I and G52S) at the PRKAG3 (RN) Locus Affecting Pork Meat Quality

        The porcine PRKAG3 (RN) gene encodes the regulatory gamma subunit of adenosine monophosphate-activated protein kinase (AMPK), which is a good candidate gene affecting meat quality. In this study, the effects of two missense mutations A595G (Ile199Val) and G154A (Gly52Ser) in porcine PRKAG3 gene on meat quality traits were studied in M. Longissimus dorsi (LD), M. Semispinalis capitis (SC) and M. Biceps femoris (BF) from different populations of 326 pigs. The PRKAG3 alleles 199I, 199IV, 52S and 52G were identified with PCR-RFLPs and all genotypes - 199I/199I, 199I/199V, 199V/199V, 52S/52S, 52S/52G and 52G/52G - were found. The frequency of V allele was larger than that of I allele in all populations. I allele frequency was zero in Chinese Meishan pigs (population D) especially. G allele frequency was larger than that of S allele in all populations except Large White (population A). Both variations at the PRKAG3 locus significantly affected these meat quality traits. The pork meat quality has not previously been established in Meishan or crosses thereof. The results suggested that generally pH of LD, SC and BF was higher in Meishan pigs than that in other populations. Moreover, Meishan pigs showed higher water-holding capacity and intramuscular fat (IMF), lower water content and water loss percentage compared to other populations in terms of the two variations. The results present here supply new evidence that alleles V199I and G52S at the PRKAG3 locus affect pork meat quality and provide useful information on pork production.
