        Recombinant Human Granulocyte Colony-Stimulating Factor Promotes Preinvasive and Invasive Estrogen Receptor-Positive Tumor Development in MMTV-erbB2 Mice

        Purpose: We investigated whether recombinant human granulocyte colony-stimulating factor (rhG-CSF) could promote the development of preinvasive and invasive breast cancer in mouse mammary tumor virus (MMTV-erbB2) mice with estrogen receptor- positive tumors. Methods: MMTV-erbB2 mice were randomly divided into three experimental groups with 20 mice in each group. MMTV-erbB2 mice were treated with daily subcutaneous injections of vehicle or rhG-CSF (low-rhG-CSF group, rhG-CSF 0.125 μg; vehicle-rhG-CSF group, normal saline 0.25 μg; and high-rhG-CSF group, rhG-CSF 0.25 μg) at 3 months of age. Cellular and molecular mechanisms of G-CSF action in mammary glands were investigated via immunohistochemistry and reverse transcription polymerase chain reaction. Results: Low, but not high, rhG-CSF doses significantly accelerated mammary tumorigenesis in MMTV-erbB2 mice. Short-term treatment with rhG-CSF could significantly promote the development of preinvasive mammary lesions. The cancer prevention effect was associated with reduced expression of proliferating cell nuclear antigen, cluster of differentiation 34, and signal transducers and activators of transcription 3 in mammary glands by >80%. Conclusion: We found that G-CSF was regulated by rhG-CSF both in vitro and in vivo. Identification of G-CSF genes helped us further understand the mechanism by which G-CSF promotes cancer. Low doses of rhG-CSF could significantly increase tumor latency and increase tumor multiplicity and burden. Moreover, rhG-CSF effectively promotes development of both malignant and premalignant mammary lesions in MMTV-erbB2 mice.

      • Lobaplatin Combined Floxuridine/Pirarubicin-based Transcatheter Hepatic Arterial Chemoembolization for Unresectable Primary Hepatocellular Carcinoma

        Purpose: To assess the effect and safety of lobaplatin combinated floxuridine /pirarubicin in transcatheter hepatic arterial chemoembolization(TACE) of unresectable primary liver cancer. Patients and Methods: TACE combined with the chemotherapy regimen was used to treat 34 unresectable primary liver cancer patients. DSA/MRI/CT/blood routine examinations were used to evaluate short term activity and toxicity after 4-5 weeks, the process being repeated if necessary. Results: Among the 34 cases, 1 (2.9%) showed a complete response, 21 (61.7%) a partial response, 8 (23.5%) stable disease, and 4 progressive disease, with a total effective rate of 67.6%. The content of alpha fetoprotein dropped by over 50% in 20 cases (58.8%). The rate of recovery was hepatalgia (88.2%), ascites (47.1%), appetite (55.9%), Performance Status(30.4%). The median follow-up time (MFT) was 281 days (63-558 days), and median progression-free survival was 118.5 days (95%, CI:88.8-148.2days). Adverse reactions (III-IV grade) were not common, with only 4 cases of vomiting and 2 cases of thrombocytopenia (III grade). Conclusions: Lobaplatin-based TACE is an effective and safe treatment for primary liver cancer.

        Expression Patterns of Circular RNAs from Primary Kinase Transcripts in the Mammary Glands of Lactating Rats

        Purpose: Circular RNAs (circRNAs), a novel class of RNAs, perform important functions in biological processes. However, the role of circRNAs in the mammary gland remains unknown. The present study is aimed at identifying and characterizing the circRNAs expressed in the mammary gland of lactating rats. Methods: Deep sequencing of RNase R-enriched rat lactating mammary gland samples was performed and circRNAs were predicted using a previously reported computational pipeline. Gene ontology terms of circRNA-producing genes were also analyzed. Results: A total of 6,824 and 4,523 circRNAs were identified from rat mammary glands at two different lactation stages. Numerous circRNAs were specifically expressed at different lactation stages, and only 1,314 circRNAs were detected at both lactation stages. The majority of the candidate circRNAs map to noncoding intronic and intergenic regions. The results demonstrate a circular preference or specificity of some genes. DAVID analysis revealed an enrichment of protein kinases and related proteins among the set of genes encoding circRNAs. Interestingly, four protein-coding genes (Rev3l, IGSF11, MAML2, and LPP) that also transcribe high levels of circRNAs have been reported to be involved in cancer. Conclusion: Our findings provide the basis for comparison between breast cancer profiles and for selecting representative circRNA candidates for future functional characterization in breast development and breast cancer.

      • Pre-operative Predictive Factors for Intra-operative Pathological Lymph Node Metastasis in Rectal Cancers

        Background: A number of clinicopathologic factors have been found to be associated with pathological lymph node metastasis (pLNM) in rectal cancer; however, most of them can only be identified by expensive high resolution imaging or obtained after surgical treatment. Just like the Child-Turcotte-Pugh (CTP) and the model for end-stage liver disease (MELD) scores which have been widely used in clinical practice, our study was designed to assess the pre-operative factors which could be obtained easily to predict intra-operative pLNM in rectal cancer. Methods: A cohort of 469 patients who were treated at our hospital in the period from January 2003 to June 2011, and with a pathologically hospital discharge diagnosis of rectal cancer, were included. Clinical, laboratory and pathologic parameters were analyzed. A multivariate unconditional logistic regression model, areas under the curve (AUC), the Kaplan-Meier method (log-rank test) and the Cox regression model were used. Results: Of the 469 patients, 231 were diagnosed with pLNM (49.3%). Four variables were associated with pLNM by multivariate logistic analysis, age<60 yr (OR=1.819; 95% CI, 1.231-2.687; P=0.003), presence of abdominal pain or discomfort (OR=1.637; 95% CI, 1.052-2.547; P=0.029), absence of allergic history (OR=1.879; 95% CI, 1.041-3.392; P=0.036), and direct $bilirubin{\geq}2.60{\mu}mol/L$ (OR=1.540; 95% CI, 1.054-2.250; P=0.026). The combination of all 4 variables had the highest sensitivity (98.7%) for diagnostic performance. In addition, age<60 yr and direct $bilirubin{\geq}2.60{\mu}mol/L$ were found to be associated with prognosis. Conclusion: Age, abdominal pain or discomfort, allergic history and direct bilirubin were associated with pLNM, which may be helpful for preoperative selection.

        Effect of Different Nanofillers on Non-Isothermal Crystallization Kinetics and Electric Conductivity of Dynamically-Vulcanized PP-EPDM Blends

        In this study, nanofillers composed of hydroxylated carbon nanotubes (h-CNT), carbon nanotubes (CNT) and graphene (GR) were separately added into the dynamically-vulcanized polypropylene (PP)/ethylene-propylenediene monomer (EPDM) blend. Differential scanning calorimetry (DSC), wide-angle X-ray diffraction (WAXD) and electrical resistivity measurements were employed to study the effect of nanofillers on the melt behavior, non-isothermal crystallization behavior and electrical conductivity of the prepared composites. WAXD results showed that h-CNT had a better induction effect of β-PP in the nanocomposites. The sequence of the activity in inducing the formation of β-PP was h-CNT>GR>CNT. However, the total crystallinity of the nanocomposites nearly remained constant. Non-isothermal crystallization kinetic analysis indicated that the presence of nanofillers improved the crystallization rate of the nanocomposites. The consequence of nucleation activity was as follows: CNT>GR>h-CNT. Although EPDM hindered the macromolecular motion of PP, interestingly it could increase the crystallization rate to an extent. Besides, the influence of nanofillers on enhancing the conductive property of the nanocomposites can be ranked as follow: CNT>GR>h-CNT.

        Microstructure Formation Mechanism and Properties of AZ61 Alloy Processed by Melt Treatment with Vibrating Cooling Slope and Semisolid Rolling

        A melt treatment with a vibrating cooling slope and a semisolid rolling process to produce an AZ61 alloy strip was proposed. The microstructure formation mechanism and the properties of the AZ61 alloy produced by the proposed process were investigated. Due to the high cooling rate and stirring action caused by the vibration cooling slope, the nucleation rate was greatly improved, which caused the formation of fine spherical or rosette primary grains. During the rolling process, the solid fraction increased from the entrance to the exit of the roll gap, and under the shearing action of the roller, the distribution of solute in the melt was homogenous, and the primary grains grew further. When the casting temperature was 680 °C,a strip with a cross section of 4 mm×160 mm was produced and a homogeneous microstructure was obtained. The ultimate tensile strength of the AZ61 alloy strip produced by the proposed method reached 242 MPa, and the corresponding elongation to failure was 4%, which were better than those achieved in previous similar studies.

      • Garcinol, an Acetyltransferase Inhibitor, Suppresses Proliferation of Breast Cancer Cell Line MCF-7 Promoted by 17β-Estradiol

        The acetyltransferase inhibitor garcinol, a polyisoprenylated benzophenone, is extracted from the rind of the fruit of Garcinia indica, a plant found extensively in tropical regions. Anti-cancer activity has been suggested but there is no report on its action via inhibiting acetylation against cell proliferation, cell cycle progression, and apoptosis-inhibtion induced by estradiol ($E_2$) in human breast cancer MCF-7 cells. The main purposes of this study were to investigate the effects of the acetyltransferase inhibitor garcinol on cell proliferation, cell cycle progression and apoptosis inhibition in human breast cancer MCF-7 cells treated with estrogen, and to explore the significance of changes in acetylation levels in this process. We used a variety of techniques such as CCK-8 analysis of cell proliferation, FCM analysis of cell cycling and apoptosis, immunofluorescence analysis of NF-${\kappa}B$/p65 localization, and RT-PCR and Western blotting analysis of ac-H3, ac-H4, ac-p65, cyclin D1, Bcl-2 and Bcl-xl. We found that on treatment with garcinol in MCF-7 cells, $E_2$-induced proliferation was inhibited, cell cycle progression was arrested at G0/G1 phase, and the cell apoptosis rate was increased. Expression of ac-H3, ac-H4 and NF-${\kappa}B$/ac-p65 proteins in $E_2$-treated MCF-7 cells was increased, this being inhibited by garcinol but not ac-H4.The nuclear translocation of NF-${\kappa}B$/p65 in $E_2$-treated MCF-7 cells was also inhibited, along with cyclin D1, Bcl-2 and Bcl-xl in mRNA and protein expression levels. These results suggest that the effect of $E_2$ on promoting proliferation and inhibiting apoptosis is linked to hyperacetylation levels of histones and nonhistone NF-${\kappa}B$/p65 in MCF-7 cells. The acetyltransferase inhibitor garcinol plays an inhibitive role in MCF-7 cell proliferation promoted by $E_2$. Mechanisms are probably associated with decreasing ac-p65 protein expression level in the NF-${\kappa}B$ pathway, thus down-regulating the expression of cyclin D1, Bcl-2 and Bcl-xl.

        Effect of Glutinous Rice Slurry on the Unconfined Compressive Strength of Lime-Treated Seasonal Permafrost Subjected to Freeze-Thaw Cycles

        It is important to account for the poor ecological environment of weak seasonal permafrost regions while developing a suitable ground improvement technique. As an organic non-polluting material, glutinous rice slurry improves the unconfined compressive strength of the soil through induction and synergy with lime. The Qinghai-Tibet Plateau has cold weather, large temperature differences between day and night, and a fragile ecological environment. Once damaged, it is extremely difficult to recover. This study investigates the effects of glutinous rice slurry on the unconfined compressive strength (UCS), failure mode, and microstructure of lime-treated seasonally frozen soil subjected to freeze–thaw cycles. Organic glutinous rice slurry with concentrations of 0, 2, 4, 6, and 8% was added to soil specimens that were treated with 10% lime and subjected to 0, 1, 3, 6, and 10 freeze–thaw cycles. In tests, the addition of glutinous rice pulp could increase the UCS of the soil by up to 40%. The first freeze-thaw cycle has the greatest impact on the UCS of the soil sample, with a decrease of 6 – 15%. Glutinous rice slurry can reduce the degradation effects of freeze-thaw cycles by increasing the contact area between soil particles. In a comprehensive evaluation under test conditions, a glutinous rice slurry concentration of 4% led to optimal results. It revealed the change characteristics of the microstructure under the action of freeze-thaw cycles, which has a good corresponding relationship with the macroscopic mechanical properties. The results can provide reference value for the application of glutinous rice slurry in the modification of subgrade soil in seasonal permafrost areas.

        Antiblurry Dejitter Image Stabilization Method of Fuzzy Video for Driving Recorders

        Video images captured by vehicle cameras often contain blurry or dithering frames due to inadvertent motion from bumps in the road or by insufficient illumination during the morning or evening, which greatly reduces the perception of objects expression and recognition from the records. Therefore, a real-time electronic stabilization method to correct fuzzy video from driving recorders has been proposed. In the first stage of feature detection, a coarse-to-fine inspection policy and a scale nonlinear diffusion filter are proposed to provide more accurate keypoints. Second, a new antiblurry binary descriptor and a feature point selection strategy for unintentional estimation are proposed, which brought more discriminative power. In addition, a new evaluation criterion for affine region detectors is presented based on the percentage interval of repeatability. The experiments show that the proposed method exhibits improvement in detecting blurry corner points. Moreover, it improves the performance of the algorithm and guarantees high processing speed at the same time.

      • Predictive Value of the Surface-Enhanced Resonance Raman Scattering-Based MTT Assay: A Rapid and Ultrasensitive Method for Cell Viability in Situ

        <P>SERRS (surface-enhanced resonance Raman scattering) has been used to develop and optimize a novel and quantitative MTT assay for living cell viability. This highly sensitive method derives from two factors for formazan signal enhancing: the addition of Au nanoparticles and the resonance effect by 632.8 nm of excitation. The results show that the background elements, such as excessive MTT residues, serum, and the drug, did not interfere with the detection of formazan. Moreover, the detection limit of formazan is as low as 1 ng/mL. With the use of this method to quantify metabolically viable cells, dose–response curves of treated and untreated cells with the drug were constructed on the human lung cancer cell A549. The results also show that the Raman signal generated is dependent on the degree of activation of the cells. In comparison to the traditional method, the main advantages of this method are its rapidity (30 min), high-selectivity, high-precision, and cost-effectiveness (0.1 mg/mL MTT) without time-consuming steps and any modifying or labeling procedure.
