        정신지체 특수학교 전공과 및 전환교육의 운영실태와 개선방향

        본 연구의 목적은 정신지체 특수학교의 전공과 및 전환교육의 운영실태와 개선방향을 탐색하는데 주안점을 두었다. 전공과 및 전환교육의 운영실태는 16개 영역으로 조사하고 문제점과 개선방향은 10개 영역을 중심으로 세부적인 내용을 조사 분석하였다. 여기서 조사분석된 내용은 정신지체 직업 · 전환교육에 좋은 지침이 될 수 있을 것이며 타장애영역 학생들의 직업진로 및 전환교육에 실질적인 도움을 줄 수 있을 것이다. The aims of this study are finding out the reality of the operation of the majored subjects and problems and finding out the improved methods about them. And 1 surveyed special teachers who are attending 17 special schools in Korea in which have the majored subjects about the reality of operating them and improved methods about them. First, about surveying the reality of operation, as to the majored subjects, they are mainly teaching students in the form of work training, guiding job, preparing job and daily Iiving skills through classes. The selection of majored subjects conceming jobs are made through school's direction and the cooperation among teachers, students and parents, and teaching jobs are now changed or will be changed because of the maladjustment to student' s aptitude, the assessment on their job aptitude is not being made now. There are many qualified teachers who have the certification in special school and they think that their roles as directors, guiders are important. The majoring students don’t have enough practicing places and factories. Even though they are received training about job in school, they get jobs unrelated with their received training. And they have verγ low employment as a whole. Though the transitional education program whose aims are selection in jobs, employment and enjoying jobs, it is started especially in the middle and high school courses. And it is thought that official and individualized education program is very important, also needed the cooperation among related authorities and it’s major program is used as the programs on job training. programs on job training. Next, as to the problems and improved methods, the problems about the majored subjects are the unrelatedness with job training and unreality and only one line of selection in job training. The problem about the transitional education program is the shortage in social skills, in the basement of job training and in carrγing out ability. The professional ability which is required in job guiding teacher is job training guide, many methods, the ability about assessing job aptitude, and testing their ability. The construction of the curriculum is not applying the guided curriculum rather than transforming the curriculum according to the atmosphere, and their school year should not be stuck to any one standard but be varied according to their ability and improvement. In the operation of majored subjects, it is necessarγ for them to provide real career education, the opportunity to receive various job training and the supported employments in which they can be included in their society actively.

        Identification of a Pathogen-Induced Glycine max Transcription Factor GmWRKY1

        On screening pathogen-resistant soybean, we identified a WRKY type transcription factor named a Glycine max WRKY1 (GmWRKY1). Expression of GmWRKY1 gene was induced in the soybean sprout by Pseudomonas infection. The GmWRKY1 was expressed in all of the tissues with high levels in stems, leaves and developing seeds. The protein GmWRKY1 contains highly conserved two WRKY DNA-binding domains having two C2-H2 zinc-finger motif (C-X4-5-C-X22-23-H-XH) in its N-terminal and C-terminal amino acid sequences. In electrophoresis mobility shift assay, the GmWRKY1 protein bound specifically to W-box elements in the promoters of defense related genes. These results demonstrated that GmWRKY1 is one of the soybean WRKY family genes and the plant-specific transcription factors for defense processes.

        호밀 염색체 도입 밀 계통 선발을 위한 RAPD, GISH 및 FISH분석

        Rye (Secale cereale L.) is known as an important source of useful genes for the improvement of resistance to rust, powdery mildew and tolerance to drought and cold on wheat (Triticum aestivum L.). We describe the identification and utilization of rye genome- specific DNA markers, OPH20-1494 bp and OPC10-1012 bp, by RAPD analysis. These markers were used to detect rye chromatin among 467 BC1F6 lines derived from the backcross of the F1 hybrid of wheat cv. Olmil and rye cv. Paldanghomill. Thirty-two lines out of 467 BC1F6 population possessed the part of rye genome. Moreover GISH and FISH analysis confirmed the presence of rye chromosome fragment on wheat background in these lines. Therefore these rye-specific markers can be used effectively in the breeding program for introgression of rye chromatin into wheat.

        팔당호밀의 단부염색체가 부가된 올밀계통의 선발

        Many progenies were produced from the backcross of Triticum aestivum cv. Olmil×Secale cereale cv. Paldanghamil hybrid using Olmil as the male parent. Nineteen ditelosomic rye chromosome addition lines (1RL) were selected in the BC_1F_6 generation by cytogenetic and genomic in situ hybridization (GISH) analysis. Progenies of BC_1, which were derived from one seed, were advanced to BC_1F_6 generation. The percentage of seed set in backcross derivatives of T. aestivum cv. Olmil×S. cereale cv. Paldanghomil was enhanced in progression, after each generation. GISH technique was applied to select the plants having the rye chromosome in BC_1F_6 generation. The presence of rye chromatin was identified in 32 out of 467 plants in 77 BC_1F_6 families. GISH analysis showed that the modes of rye-chromatin in BC_1F_6 plants were almost telocentric addition lines including nineteen ditelocentric and ninety two monotelocentric rye chromosomes. The added ditelocentric chromosomes were found as pairs and underwent normal meiotic pairing and segregation, while monotelocentric chromosomes showed meiotic irregularities.

        Identifications of a Sprout-Rot Pathogen Pseudomonas Species SN239 and Selection Resistant Soybean Line

        콩나물은 우리나라에서 오래 전부터 재배하여 온 채소로서 그 기호성이 매우 높으며 영양학적으로 우수하나 일부 열악한 재배환경으로 콩나물의 부패문제가 자주 발생해 왔다. 따라서 본 연구는 시중의 부패된 콩나물로부터 다양한 병원균을 분리함과 동시에 재래콩 유전자원으로부터 시중의 콩나물 부패병에 강한 품종을 탐색하고 선발된 내병성 계통의 생육특성을 조사하였다. 분리된 콩나물 부패균들 중 병원성이 강한 콩나물 부패균인 Pseudomonas sp. SN239을 분리하고 16S rRNA 염기서열을 동정한 결과 P. putita, P. plecoglossicida, P. monteilii 및 P. mevalonii와 근연관계를 보였으나 완전히 일치하지는 않았으므로 Pseudomonas sp. SN239는 새로이 동정된 콩나물 부패균으로 여겨진다. 또한 재래콩 194계통에 콩나물 부패병균 Pseudomonas sp. SN239을 접종하여 저항성을 검정한 결과, 이병성 계통은 심하게 부패되었으나 한국 고유계통 YNPCS3-19는 병원성이 없었으며 또한 지속적으로 생육하였다. 그러므로 부패균 저항성 계통 YNPCS3-19는 부패균 저항성 품종 육성에도 활용 가치가 크다고 판단된다. Control microbial contamination in pathogens to soy sprouts has always been highly concerned in soybean sprout industries because the soybean sprouts are consumed largely as a nutritious fresh vegetable around the world. However, pathogens in soy sprouts are little known. Here, we isolated a strain of Pseudomonas sp. SN239 that caused severer symptoms in sprouts of many soybean cultivars. In phylogenetic relationships using 16S ribosomal RNA sequences of the Pseudomonas species, the identified Pseudomonas sp. SN239 was grouped with P. putita, P. plecoglossicida, P. monteilii and P. mevalonii. Thus, the bacterial strain SN239 might be a newly identified Pseudomonas species which closely related to P. putida. Furthermore, we found that a Korean indigenous soybean (Glycine max) cultivar YNPCSS3-19 has strong resistance against the Pseudomonas sp. SN239.
