        하이드로젤 지지체 기반 3차원 환경에서 개 간엽줄기세포의 분화능 분석

        Mesenchymal stem cells (MSCs) are useful candidates for tissue engineering and cell therapy. Physiological cell environment not only connects cells to each other, but also connects cells to the extracellular matrix that provide mechanical support, thus exposing the entire cell surface and activating signaling pathways. Hydrogel is a polymeric material that swells in water and maintains a distinct 3-dimensional (3D) network structure by cross linking. In this study, we investigated the optimized cellular function for canine adipose tissue-derived MSCs (cAD-MSCs) using hydrogel. We observed that the expression levels of Ki67 and proliferating cell nuclear antigen, which are involved in cell proliferation and stemness, were increased in transwell-hydrogel (3D-TN) compared to the transwell-normal (TN). Also, transforming growth factor-${\beta}1$ and SOX9, which are typical bone morphogenesis-inducing factors, were increased in 3D-TN compared to the TN. Collagen type II alpha 1, which is a chondrocyte-specific marker, was increased in 3D-TN compared to the TN. Osteocalcin, which is a osteocyte-specific marker, was increased in 3D-TN compared to the TN. Collectively, preconditioning cAD-MSCs via 3D culture systems can enhance inherent secretory properties that may improve the potency and efficacy of MSCs-based therapies for bone regeneration process.

        Characteristics of mesenchymal stem cells derived from different tissues of goat

        Mesenchymal stem cells (MSCs) are multipotent cells capable of replicating as undifferentiated cells, and have the potential of differentiating into mesodermal lineages. Goats are commonly used as animal models for bone tissue engineering to test the potential of stem cells for bone regeneration. Goat MSCs isolated from bone marrow (BM) or adipose tissue (AD) should be evaluated using in vitro assays, prior to their application in a tissue engineering project. In this study, we compared the stem cell properties of MSCs derived from goat AD, BM and ear skin tissue (ESK). As results, BM and ESK-MSCs exhibited a spindle-shaped morphology comparable to that of AD-MSCs. Especially, BM-MSCs could be cultured for significantly longer periods and exhibited the greatest expansion capacity, whereas AD-MSCs had the shortest culture time and lowest growth rate. Also, we compared differentiation potentials of AD, BM and ESK-MSCs into adipogenic, chondrogenic, and osteogenic lineages through specific staining and quantitative real-time RT-PCR. Collectively, we successfully isolated ESK-MSCs from goat for the first time. This study suggests that adult skin tissue of goat could be used as a source of goat MSCs. Further studies are needed to show the more information for establishment and fully characterization of goat ESK-MSCs.

        Differentiation potential of mesenchymal stem cells through electrical stimulation

        Electrical stimulation (ES) is known to guide the development and regeneration of many tissues. Use of low-frequency ES for therapeutic purposes has been increasing during the last decades. Mesenchymal stem cells (MSCs) represent an appealing alternative cell source for cartilage repair. There are studies that induce differentiation into cartilage cells by treating the growth factors in stem cells or altering the properties of stem cells by genetic modification. In this study, we exposed equine adipose tissue-derived MSCs (eAD-MSCs) to ES and assessed changes in the chondrogenic differentiation potential. The cells obtained from equine adipose tissue attached to culture plates and expanded in vitro. Flow cytometric analysis at third passage indicated that the cells were strongly positive for CD44, CD90, and CD105, but negative for CD13, CD34, and CD45. Next, ES was applied to eAD-MSCs cultured under condition of high-density micromass under ES of 10 V/cm, with duration of 10 ms and a frequency of 2.0 Hz for three days. Gene expression of chondrogenic markers such as collagen type II, Aggrecan, and Sox9 was analyzed at three days of ES. As a result, we observed the differentiation potential of eAD-MSCs into chondrocytes by specific ES in absence of exogenous growth factors. We also found that ES upregulated the expression of heat shock protein 70, which affects cartilage formation. This study may contribute to the differentiation of MSCs into chondrogenic lineage under specific ES condition.

        Bacillus subtilis CH-10에 의한 폴리감마글루탐산의 생산에 관한 연구

        A bacterium that produced a large amount of poly-$\gamma$-glutamate (PGA) was isolated from the compost and designated as Bacillus subtilis CH-10. The optimum temperature and pH for PGA production were at $37^{\circ}C$ and 7.5, respectively. The maximum amount of PGA production (18.84 mg/ml) was obtained when it was grown in a medium containing 3% L-glutamate and 5% sucrose at $37^{\circ}C$ with shaking. The result that the L-glutamate significantly induced PGA production indicates that it produces a PGA by the glutamate dependent manner. Some properties of the PGA obtained at different times of cultivation were investigated by SDS-PAGE and ninhydrin analysis. The PGA production was elongated along with cultivation time and maximum amount was achieved at 96 h. Average molecular weight of PGA was estimated to be 1100 kDa by FDNB method. 폴리감마글루탐산(PGA)을 대량생산하는 균주를 축분퇴비에서 분리하고 생리 생화학적 특징 및 16s rRNA 염기서열을 분석한 결과, Bacillus subtilis로 동정하고 strain CH-10 으로 명명하였다. PGA 생산을 위한 배지의 최적 온도와 pH는 각각 $37^{\circ}C$, 7.5 이었다. 또한 배지성분 중에서 PGA 생산에 최적인 탄소원과 질소원은 각각 5% sucrose와 3% L-glutamate인 것으로 나타났다. 이 균주는 L-glutamate에 의해 PGA 생산이 대량으로 유도되었고 따라서 질소원으로 L-glutamate를 반드시 필요로 하는 glutamate 의존성 균주인 것으로 사료되었다. 배양시간에 따라 생산된 PGA의 생화학적 특정을 SDS-PAGE와 ninhydrin 분석을 통하여 검토하였다. PGA의 생산량은 배양시간에 따라 증가하였고 배양 후 96시간에서 최고 생산량을 나타내었다, 생산된 PGA의 평균분자량을 FDNB법으로 분석한 결과, 1,100 kDa인 것으로 나타났다.

        Otaku means the people who are addicted to manga, animation, game, PC, SF. trick shooting, figure and the others, the namely Japanese subculture, which is deeply related to one another. The generation of Otaku culture is more connected with inherent collective tendency of social inevitability in Japan. Otaku appeared on that stage in the era of high economic growth and development, which means that the formation of Otaku stands for the appearance of the generation who has experienced the division of reality and fantasy through the full-color combination of the animation which dominates the public perspective and the monochrome of Construction State represented by air pollution and cement during the period of growth. In other words, although they have material prosperity in reality, they failed to relieve it but just satisfied with the visual fetish through popular fantasy in extremely individualized status. This visual fetish has become an important social and cultural current in Japan since they have played major role in the society. Murakami Takashi and Nara Yoshitomo, the very artists leading ‘NiPPJl1 Pop’ flooded in 1990s’, are the representative artists in Japan recently. The post-war contemporary art in Japan has enclosed itself in the name of ‘Art’ and has produced recursive meaning constantly, while the art of the Otaku generation has been accomplished in the process of breaking from ‘art’ area and redefining of themselves. It is caused by mixing the basement of the so-called subculture with the rigid highbrow, but one can say that it is the reflection of will for flatting the prevalent cultural hierarchy rather than just transplanting lowbrow into the frame of highbrow or just pastiche it. Accordingly, contemporary art in Japan, which has developed with Otaku culture is still ongoing project. However, Otaku culture that constantly reproduces a sharing point in extremely individualized taste refers to the way of identifying the individual who live in the giant Japanese society, and also the way that could be extended only in so far as it continually evolves and dissolves in development.

      • 비평과 전시의 새로운 관계 - 스페이스 애프터를 중심으로

        In Korean art criticism, there are functions and possibilities of criticism that have not yet been practised. However, art criticism is systematically fixed, old negations and controversies surrounding criticism, such as the crisis of criticism, inactivity of criticism, and disappearance of criticism, are repeating the same trajectory. On the other hand, both the art market and art history are firmly established when the conventional framework of criticism is converted into precedency, and various art projects are followed through this. It is that criticism is an active 'leading' existence between academics and the art scene. And the role of this traction is explored through the new art gallery Space Aefter. 한국의 미술비평에는 아직 검토되지 않은 비평의 기능과 가능성이 존재한다. 그러나 비평에 대한 고정된 소비가 제도적으로 고착되어 비평의 위기, 비평의 부진, 비평의 소멸 등 비평을 둘러싼 해묵은 논쟁과 회의는 늘 같은 궤도만 을 반복하며 공회전 하고 있다. 이에 비평의 후행성이라는 관습적인 틀을 선행성으로 전환하고, 이를 통해 가능한 여 러 가지 적극적 기획이 뒤따를 때, 미술시장과 미술사 모두 견고하게 구축된다. 그리고 이것은 비평이 학문과 현장 사 이에 ‘끼인’ 존재가 아니라 능동적인 ‘견인’의 존재임을 전제로 한다. 그리고 이러한 견인의 역할을 신생 전시 공 간 스페이스 애프터를 통해 타진한다.

      • 코로나 팬데믹 이후, MZ 세대와 미술 시장의 변화

        현재 미술시장에서 나타나는 변화는 MZ 세대가 아니라 XY 세대가 주도하는 것이다. 그러나 20대 초반의 참여도 확인할 수 있으므로, 더 넓게 XYZ 세대로 확장해서 살펴볼 것을 제안 할 수 있다. 또한 정보화와 문화화의 면에서 보 자면 코로나19 사태를 계기로 촉진된 미술시장의 변화는 일시적인 것이 아니라 구조적인 것이다. 이 변화는 첫째 기술 적으로 정보기술이 더욱 적극적으로 활용되는 것이고, 둘째 경제적으로 미술 작품에 대한 투자가 더욱 성장하는 것이 며, 셋째 문화적으로 미술 작품에 대한 관심이 더욱 확산되는 것이다. 세계와 국내의 미술시장에서 젊은 세대의 참가 확대는 명확히 확인할 수 있는 새로운 현상이다. 현재 그 실체는 MZ 세대가 아니라 XY 세대이나 Z 세대도 이 대열에 곧 합류하게 될 것이다. 현재 한류와 같은 문화 향유의 면에서 보자면 Z 세대의 주도가 매우 뚜렷이 나타나고 있다. 그리고 이들 세대에는 디지털 원주민으로서의 정보화와 이를 통해 확산되는 세대의 문화가 강력히 연관되어 있다. 정보기술을 사용하는 것은 사실 문화 활동을 하는 것이며, 이를 통해 미술 작품을 향유하고 거래하는 것 역시 경제 활동 이다. The changes in the current art market are not led by the MZ generation but by the XM(Y) generation. However, since participation in the early 20s can be confirmed, it can be suggested to expand to XYZ generation more widely. In terms of information and culturalization, the change of art market promoted by the phandemic is not temporary but structural. This change is summarized in three meanings. first, information technology is more actively used technically. Second, the investment in art works is growing more economically, Third, interest in art works is spreading more culturally. The expansion of participation of younger generations in the world and domestic art market is a new phenomenon that can be clearly confirmed. The reality at the moment is not the MZ generation, but the XM(Y) generation or the Z generation will soon join this ranks. And these generations are strongly related to the informatization as digital natives and the culture of generations spreading through them.
