        국내 이스라엘 잉어의 선발육종효과 평가

        1973년 이스라엘 잉어 (향어)가 한국에 양식을 위해 도입된 이후 현재까지 품종개량에 대한 연구가 전무한 실정이다. 본 연구는 지속적인 근친교배로 인해 낮아진 국내 이스라엘 잉어의 유전적 다양성을 회복하고, 성장이 빠르고 비늘 개선을 위해 유전적 기반 교잡육종 연구를 수행하였다. 본 연구는 한국의 이스라엘 잉어의 품종 개량을 위하여 국내 이스라엘 잉어와 중국의 송푸거울 잉어를 이용하여 4개의 교배구를 설정하여 F1을 생산하였다. 친어의 형태 및 유전학적 거리를 고려하여 교배지침을 설정하였다. 본 연구는 유전적 다양성과 친자분석을 위하여 microsatellite 마커와 유전형 데이터를 활용하였다. 그 결과, 국내 친어의 평균 대립유전자와 기대이형접합율은 8.3과 0.743이며, F1은 13.0과 0.764이었다. 국내 이스라엘 잉어와 중국 송푸거울 잉어의 품종 간 교배를 통하여 국내 이스라엘 잉어보다 F1의 유전적 다양성이 회복되었음을 나타내었다. 한국의 일반 이스라엘 잉어는 17개월에 1.7 kg이었고, 개량된 이스라엘 잉어는 2.2 kg에 도달하였다. 또한, KC (한국×중국) 교배그룹의 비늘수치는 2.52, 친어그룹의 비늘수치는 3.15로 나타나 F1은 친어보다 낮은 비늘수치 (0.63)를 나타내었다. 품종개량된 이스라엘 잉어 (F1; CK, KC)는 친어그룹 (F0)보다 비늘이 20% 개선되었으며, 일반 이스라엘 잉어에 비해 체중 (27%)과 비늘(25%)이 향상되었다. 유전적 데이터를 기반으로 개발된 이스라엘 잉어는 상업성이 좋아 국내 이스라엘 양식업에 크게 기여할 것으로 생각된다. Since the introduction of Israeli carp into Korea for farming in 1973, there are no breeding studies on developing Korea Israeli carp (domestic) so far. This study performed gene-based cross-breeding studies to restore genetic diversity of lowered Israeli carp through continuous inbreeding, and for rapid growth and better scales. This study produced four cross-breeding groups (F1) using Koean Israeli carp and Chinese Songpu mirror carp for the improvement of growth and scale of Israeli carp in Korea. And mating scheme for breeding groups was set in consideration of the morphological analysis and genetic distance of broodstock. In addition, this study used microsatellite markers and genotype data to analyze genetic diversity and parentage analysis. As a result, the average NA and HE values of Korean select broodstock are 8.3 and 0.743, and F1 is 13.0 and 0.764. This study shows that the genetic diversity of F1 has been recovered over Korean Israeli carp through breeding between Korean Israeli carp and Chinese Songpu mirror carp. Common Israeli carp in Korea reached 1.7 kg in 17 months, and improved Israeli carp reached to 2.2 kg. The KC (Korea×China, KC) group was 2.52 and broodstock group was 3.15. F1 showed lower scale score (0.63) than broodstock. The improved carp (F1; CK, KC) had 20% better scales than the parent group (F0), which improved 27% in weight and 25% in scales compared to common Israeli carp. The Israeli carp developed by the genetics-based breeding grew quicker and had improved genetic diversity and fewer scales, which will be of great value for Korean Israeli aquaculture industry due to good marketability.

      • Genetic Similarity and Difference between Common Carp and Israeli Carp (Cyprinus carpio) Based on Random Amplified Polymorphic DNAs Analyses

        Common carp (Cyprinus carpio) and its aquaculture breed Israeli carp samples were obtained from two separate aquaculture facilities under the similar raising conditions during two years in the Kunsan National University, Korea. Genomic DNA was isolated from the common carp and Israeli carp for identification of genetic characteristics and genomic polymorphisms by polymerase chain reaction amplification of DNA using arbitrary primers. The arbitrary primer No.21 (ACTTCGCCAC) yielded the highest number of fragments with the average of 15.0 among the primers used in Israeli carp. A tota1 of 294 polymorphic products in common carp and 336 in Israeli carp were observed by random primers. The average number of polymorphic products generated by random RAPD primer No. 2 (GTAGAC-CCGT) showed 8.0 in Israeli carp. On average, each random RAPD primer produced 5.4 amplified polymorphic products in common carp and 6.2 in Israeli carp. An average genetic similarity (BS value) was 0.44$\pm$0.05 within the common carp and 0.32$\pm$0.04 within the Israeli carp. The degree of similarity frequency (BS) between two carps was 0.67 as generated by the primer No. 19 (GACGGATCAG). The average level of bandsharing was 0.57$\pm$0.03 between the two carps. Accordingly, the two carp populations were genetically a little distant. The electrophoretic analysis of PCR-RAPD products showed middle levels of variation between the two carp populations. This result implies that the genetic diversity among intra-population may be higher when compared with that between the two carps. The RAPD polymorphism generated by these random primers might be used as a genetic marker for populations or lines identification in important aquacultural carp.

        Genetic variability comparison of cultured Israeli carp (Cyprinus carpio) from Korea using microsatellites

        In aquaculture, cultured fish often undergo continuous cross-fertilization without any inflow of new broodstock. This lowers genetic diversity, leading to increased disease rates and decreased survival rates. To improve the mass production and easy culture of Israeli carp, it is essential to investigate the population structure and genetic diversity of these fish. However, such a survey has not yet been performed on Korean Israeli carp. In this study, we used seven microsatellite markers to analyze the genetic diversity and association of cultured Israeli carp from Korea and China. The average numbers of alleles per locus (NA) for two Korean (KorA and KorB) and two Chinese (ChA and ChB) populations were as follows: KorA (10.42), KorB (14.43), ChA (20.57) and ChB (20.71). The expected heterozygosity (He) ranged from 0.672 to 0.897 and from 0.827 to 0.938 in the Korean sample and Chinese sample respectively. The genetic diversity of the Korean Israeli carp was about half that of the Chinese carp. The diversity of the Korean Israeli carp was very low, suggesting that the immunity of this population could be weak, and that diversity–recovery studies are urgently needed. Therefore, our results may therefore form the foundation for future research efforts towards genetic monitoring and selective breeding, continuous research needs to be conducted in order to recover the genetic diversity of the Korean Israeli carp.

        Growth Comparison of Israeli Carp (Cyprinus carpio) to Different Breeding Combination

        The purpose of this study was to investigate the improvement of growth in Israeli carp (Cyprinus carpio), and the cross experiment was carried out with two strains of Israeli carp. Four combinations of Israeli carp from Jeonbuk fisheries farm and Songpu mirror carp from Heilong Jiang, China (KK; Jeonbuk ♀ ${\times}$ Jeonbuk ♂, KC; Jeonbuk ♀ ${\times}$ China ♂, CC; China ♀ ${\times}$ China ♂ and CK; China ♀ ${\times}$ Jeonbuk ♂) were developed and reared. Body length, body weight and condition factor were determined at 20, 40, 60 and 170 days post-hatch (DPH). The results showed that there were differences in growth rate of the four groups. Body length of four groups were CK > CC > KC > KK and body weight were CC > CK > KC > KK at 170 DPH. The growth perfomance of four groups were statistically significant difference (P<0.05). During the rearing, CC group had longer length and higher weight at 170 DPH compared to other three groups and also condition factor was highest in the CC group, but there was no significant difference in a survival rate. These results indicated that the growth performance mainly depended upon brooder combination but survival rate could not significantly affect brooder.

        Original Article : Optimal Dietary Protein and Lipid Levels for Growth of Juvenile Israeli Carp Cyprinus carpio

        A feeding trial of four dietary protein levels (20, 30, 40, and 50%) and two lipid levels (7 and 14%) with a factorial design was conducted to determine the optimal dietary protein and lipid levels for juvenile Israeli carp Cyprinus carpio. Triplicate groups of fish (average body weight, 1.3 ± 0.02 g) were fed the experimental diets for 9 weeks. Survival of fish was not affected by either dietary protein or dietary lipid level. Weight gain and feed efficiency increased as dietary protein levels increased up to 40 and 50%, respectively. Weight gain was higher in fish fed the high-lipid diets with 20 and 40% protein content. Feeding efficiency increased as the dietary lipid level increased for the 30, 40, and 50% protein diets. Daily feed intake decreased with increasing protein level and the minimum feed consumption was observed in fish fed the 50% protein diet with 14% lipid content. Moisture and lipid contents of the whole body were affected by both dietary protein and lipid levels. The crude lipid content of fish fed the 14% lipid diet was higher than that of fish fed the 7% lipid diet at each protein level. The results of this study indicate that a diet containing 40% protein with 14% lipid content is optimal for the growth and effective protein utilization of juvenile Israeli carp.

        향어(Cyprinus carpio) 함유 쿠키의 영양특성

        This study was conducted to compare the nutritional properties of cookies made with Israeli carp Cyprinus carpio (C-IC) to those made without Israeli carp (control). The proximate composition of C-IC per 100 g was 4.1 g moisture, 9.7 g protein, 29.2 g lipid, 1.4 g ash, and 55.6 g carbohydrates. Moisture, protein and ash contents were significantly higher and the carbohydrate content was significantly lower (P< 0.05) in C-IC than control, but there was no difference in lipid content (P >0.05). The total amino acid content of C-IC per 100 g was 9.46 g and the major amino acid was glutamic acid (2.49 g). The mineral contents of C-IC per 100 g were 216.6 mg calcium, 193.2 mg phosphorus, 170.9 mg potassium, and 18.2 mg magnesium, which were all significantly higher than the contents of the control (P<0.05). The major fatty acids of C-IC were 16:0, 18:1n-9, and 18:2n-6. The digestibility of C-IC in the small intestine was 51.3%, which was higher than the digestibility of the control. These results suggest that C-IC have better nutritional properties than the control.

        향어(Cyprinus carpio) 함유 야채죽의 제조공정 최적화

        This study optimized the vegetable rice porridge preparation process with Israeli carp Cyprinus carpio (VRP-IC). The optimum heating period for Israeli carp paste (IC-P) in a retort was 120 min as per yield results. Response surface methodology was performed with IC-P/[rice (glutinous rice: non-glutinous rice=6:4) (Rice, RI)+vegetable (carrot: zucchini=1:1) (vegetable, VE)] (X1) and RI/VE (X2) as independent variables; viscosity (Y1), hardness (Y2), amino nitrogen (Y3), and overall sensory acceptance (Y4) as dependent variables. Optimum rates of IC-P, RI, and VE were 47.37%, 24.49%, and 28.15%, respectively; predicted multiple response optimum values for the dependent variables were 20,150 mPa·s, 2,002.9 N/m2, 193.1 mg/100 g, and a 6.4 score for Y1, Y2, Y3, and Y4, respectively. Under optimum conditions, experimental values for Y1, Y2, Y3, and Y4 were 20,359.8±191.9 mPa·s, 2,015.0±9.6 N/ m2, 188.1±4.9 mg/100 g, and a 6.5±0.2 score, respectively, which were not significantly different from the predicted values (P < 0.05). Results of F0 value, viscosity, hardness, volatile odor intensity, and browning suggested that the optimum sterilization period was 25 min. VRP-IC prepared under the optimum conditions was superior to commercial vegetable rice porridges in sensory evaluations.
