        Antioxidant, Anti-Melanogenic and Anti-Wrinkle Effects of Phellinus vaninii

        In this study, the antioxidant, anti-xanthine oxidase, anti-melanogenic and anti-wrinkle effects of methanol (ME) and hot water (HE) extracts from the fruiting bodies of Phellinus vaninii were investigated. The 1,1-diphenyl-2-picryl-hydrazyl free radical scavenging activity of 2.0mg/mL HE (95.38%) was comparable to that of butylated hydroxytoluene (96.97%), the reference standard. The hydroxyl radical scavenging activities of ME (98.19%) and HE (97.55%) were higher than that of butylated hydroxytoluene (92.66%) at 2.0mg/mL. Neither ME nor HE was cytotoxic to murine melanoma B16-F10 cells at 25-750 mg/mL. Although the xanthine oxidase (XO) inhibitory effects of ME and HE were significantly lower than that of allopurinol, the values were higher than 84 percent. The in vitro tyrosinase inhibitory activities of ME and HE were comparable to kojic acid at 2.0mg/mL. The cellular tyrosinase and melanin synthetic activities of ME and HE on B16-F10 melanoma cells at 500 mg/mL were higher than arbutin, indicating that the inhibitory effects of arbutin on the tyrosinase and melanin synthesis were higher than those of ME and HE. The collagenase inhibitory activity of HE was comparable to EGCG at 2.0mg/mL, however, the elastase inhibitory activity of ME and HE was lower than EGCG at the concentration tested. The study results demonstrated that the fruiting bodies of Ph. vaninii possessed good antioxidant, anti-xanthine oxidase, cell-free anti-tyrosinase, cellular anti-tyrosinase, anti-collagenase, and moderate anti-elastase activities, which might be used for the development of novel anti-gout, skin-whitening, and skin anti-wrinkle agents.

        연근과 우절 에탄올 추출물의 향장효능 검증

        본 연구는 화장품 소재로서 연근과 우절의 가능성을 확인하기 위한 것이다. 이를 위해 우리는 연근과 우절 에탄올 추출물을 사용하여 항산화, 항염증 및 항주름에 대한 생물학적 활성 평가를 수행하였다. 연근과 우절을 95% 에탄올로 추출한 다음 항산화 평가를 위해 샘플을 농도 (100, 500, 1000) μg/mL 에 따라 처리하여 1,1-diphenyl-2-picrylhydrazyl (DPPH) 라디칼 소거능과 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) 양이온 라디칼 소거능을 확인하였다. 또한 항주름 효과를 평가하기 위해 Elastase 저해 활성 평가를 수행 하였다. 항염증 효과를 평가하기 위해서 대식세포 (Raw 264.7 cells)를 이용해 MTT assay를 통한 샘플의 독성평가와 nitric oxide 생성 저해 활성을 평가하였다. 그 결과 연근과 우절의 1000 μg/mL 농도에서 DPPH 라디칼 소거능 활성이 각 66.7%, 99.5% 로 ABTS+ 라디칼 소거능 활성은 동일농도에서 각 51.2%, 98.3%로 나타났다. elastase저해 활성 결과 우절이 연근에 비해 높은 항주름 효능을 나타내었다. 우절은 1000 μg/mL 농도에서 대조군 EGCG 보다 높은 활성을 나타내었다. Nitric oxide 저해 활성 결과에 따르면 연근은 55.8%의 효과를 나타내었고 우절은 66.6%의 효과를 각각 나타내어 우절 추출물이 연근보다 항염증 효과가 더 우수함을 나타내었다. 결과적으로 연근 및 우절 추출물은 안전한 항산화 항염증 및 항주름 천연화장품소재로 사용될 수 있음을 시사한다. This study is for checking the possibility of Lotus Rhizome and node of Lotus Rhizome as cosmetic materials. For this we carried out biological active evaluation about anti-oxidant, anti-inflammatory and anti-wrinkle by using ethanol extract of Lotus Rhizome and node of Lotus Rhizome. We extracted Lotus Rhizome and node of Lotus Rhizome with 95% ethanol and then in order to evaluate anti-oxidant activity we treated samples by concentrations (100, 500, 1000) μg/mL and carried out 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and The activity of 2, 2’-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) cation radical scavenging. Also, in order to evaluate effect of anti-wrinkle we carried out evaluation of Elastase inhibitory activity. To evaluate effect of anti-inflammatory we evaluated toxicity of samples through MTT assay with a macrophage (Raw 264.7 cells) and measured nitric oxide production inhibitory activity. As a result, DPPH radical scavenging activity of Lotus Rhizome and node of Lotus Rhizome at 1000 μg/mL was 66.7% and 99.5%, respectively and ABTS + radical scavenging activity was 51.2% and 98.3% at the same concentration, respectively. Elastase inhibitory activity results showed that the nodes of the Lotus Rhizom extract excellent anti-wrinkle efficacy than Lotus Rhizom. Node of Lotus Rhizome showed higher anti-wrinkle activity than the positive the control group BHT at 1000 μg/mL concentration. According to the result of nitric oxide production inhibitory activity, Lotus Rhizom showed 55.8% effect and nodes of the Lotus Rhizom showed 66.6% effect respectively. This showed that effect of anti-inflammatory was greater in nodes of the Lotus Rhizom extracts. As a result it suggests that Lotus Rhizome and node of Lotus Rhizome extracts can be used as natural substance of cosmetics which are safe in antioxidant, anti-inflammatory, and anti-wrinkle.

        Anti-inflammatory and anti-wrinkle effect of Sophora flavescens Aiton extracts treated with protein hydrolytic enzyme

        This study was conducted to investigate anti-inflammatory and wrinkle improvement effects of Sophora flavescens Aiton water extracts (SWE) treated with proteolytic enzyme. The antioxidant activity of proteolyzed Sophora flavescens Aiton water extracts (SWE-E) showed increased total polyphenol content, total flavonoid content, electron-donating ability and ABTS⁺ free radical scavenging activity compared with SWE. To investigate the anti-inflammatory effects, the inhibition of NO production was assessed in RAW 264.7 cells induced by LPS. The SWE-E showed an increased anti-inflammatory effect compared with SWE at the same concentration. The anti-wrinkle effect was evaluated by the rate of collagenase and elastase inhibitory activity, which was determined by the MMP-1 mRNA measured in human dermal fibroblast (HDF) by quantitative polymerase chain reaction (qPCR). Collagenase and elastase are important enzymes that play roles in wrinkle formation, and SWE-E showed a significantly higher collagenase inhibition rate than non-treated extracts. The MMP-1 mRNA in HDF cells decreased in a dose-dependent manner. Moreover, SWE was shown to be a non-irritant in the BCOP assay, which is an alternative method to in vivo eye irritation test. Taken together, these results suggest that proteolytic enzyme could enhance the antioxidant activity, as well as the anti-inflammatory and anti-wrinkle effects of SWE, and that SWE-E could be used as a cosmeceutical ingredient.

        국내산 개똥쑥 소수성 분획물의 항산화 활성 및 항주름 효과 탐색

        Artemisia annua belongs to the plant family of Asteraceae and is an annual short-day plant. This plant is known for its various effects such as its anti-cancer, anti-antioxidant, anti-inflammatory, and cardiovascular disease therapies. However, the anti-wrinkle effect of the hydrophobic fraction extract has not been reported yet. In this study, the antioxidant activity and anti-wrinkle activity of the number of hydrophobic fractions of domestic Artemisia annua L is investigated. Three solvent extracts (hot water, 70% ethanol, and 100% methanol) of Artemisia annua L are prepared. In the evaluation results of Artemisia annua L. leaf, antioxidant and anti-wrinkle activities are uncovered. The 100% methanol extract shows the highest antioxidant activities and total phenolic and total flavonoid contents among them. Collagenase inhibition, ellstase inhibition, and hyaluronidase inhibition IC50 rate are 124, 200, and 324 ug/mL, respectively. Therefore, this plant would be a useful anti-wrinkle cosmetic material.

        제주 자생 우슬(Achyranthes japonica Nakai)의 항산화, 항염증과 항주름 평가

        본 연구는 화장품 소재로서 우슬(Achyranthes japonica Nakai)의 가능성을 확인하기 위한 것이다. 우슬 추출물이 가지고 있는 항산화, 항염증, 항주름 효과를 측정하였다. 각각의 식물 재료는 70% 에탄올을 이용하여 우슬 뿌리(Achyranthes japonica Nakai roots, AJNR)와 우슬 줄기 (Achyranthes japonica Nakai stalks, AJNS)로부터 추출하였다. RAW 264.7 세포를 배양하여 추출물의 Nitric oxide assay를 진행하였고, 섬유아세포 CCD-986sk를 배양하여 추출물의 MMP-1 assay, Type I procollagen synthesis assay를 실시하였다. 이 연구의 결과, 항산화 활성은 우슬 뿌리와 우슬 줄기 모두 우수하였고, 뿌리는 함염증 효과가 월등하게 우수하였으며 줄기는 뿌리에 비해 MMP-1 저해 활성과 Type I procollagen 합성 효과가 조금 더 높았다. 이 연구의 결과, 우슬 뿌리와 우슬 줄기의 항산화 활성은 유사한 수준으로 우수하였고, 뿌리의 함염 활성은 줄기보다 월등하게 높았다. MMP-1 저해 활성과 Type I procollagen 합성 효과는 대체적으로 우수하였는데 줄기가 뿌리에 비해 가 조금 더 뛰어났다. 그러므로 우슬은 항산화, 항염증, 항주름의 활성을 갖는 기능성 화장품 소재로서 활용이 가능할 것으로 판단된다. The aim of this study was to assure the possiblility of Achyranthes japonica Nakai for a cosmetic material. For this we examined the Anti-oxidation, Anti-inflammation, Anti-wrinkle effects of Achyranthes japonica Nakai extract. Each material is extracted from Achyranthes japonica Nakai roots(AJNR) and Achyranthes japonica Nakai stalks(AJNS) using 70% ethanol. We carried out Nitric oxide assay of the extract with cultured RAW 264.7 macrophage cells, And we conducted MMP-1 assay, Type I procollagen synthesis assay of the extract with cultured CCD-986sk fibroblast cells. After we measured ABTS+ assay and SOD assay of AJNR and AJNS, we knew that effect of anti-oxidant increased as their concentration get higher. The result of this study was that the antioxidant activity of AJNR and AJNS was similarly excellent and the anti-inflammatory activity of AJNR was significantly higher than AJNS. The MMP-1 inhibitory activity and Type I procollagen synthesis of AJNR and AJNS was generally excellent, AJNS had slightly better than AJNR. Therefore I thought it has the chance to play an imfortant role as a functional cosmetic material for anti-oxidant, anti-inflammation and wrinkle reduction effects.

        노화 및 주름개선화장품 선호브랜드의 성분현황과 만족도

        생활수준의 향상, 소득수준 증가, 여성들의 사회진출 증가로 미적욕구의 표출의 하나인 화장품 역시 많은 발전을 거듭해왔다. 화장품에 대한 기대심리가 높아지고 보습과 보호의 단순개념에서 다기능을 포함한 효능과 효과를 중요시하는 기능성 화장품의 수요가 증가하는 경향을 보이고 있는 것이 현실이다. 따라서 본 연구는 피부 관리실에서 노화 및 주름 개선을 목적으로 사용되는 화장품의 성분현황과 만족도를 조사하여 사용성분의 기초자료를 제시하는데 목적이 있다. 연구대상은 광주지역 3인 이상 근무하는 피부 관리실의 피부관리사를 대상으로 연구방법은 1차 설문지와를 통하여노화 및 주름개선을 목적으로 관리실에서 사용되고 있는 화장품 브랜드의 선호도 및 제품선택 시 중점요소와 효과를 조사한다. 둘째, 설문에서 밝혀진 브랜드 선호도를 조사한 후 상위 10개의 제품 중 에센스, 크림, 팩 제품의 전 성분 현황과 노화 및 주름개선 화장품에 사용되고 있는 유효성분과 추출물, 방부제, 계면활성제, 보습제 및 컨디셔닝제, 오일 또는 알콜류, 기타 등으로 구분하여 사용되고 있는 성분을 조사하고자 한다. 셋째, 노화 및 주름개선 화장품의 성분과 지식에 따른 관리 만족도와의 관련성을 밝히고자 한다. 수집된 자료는 윈도우용 SPSS 11.0 통계 패키지를 이용해서 기술 통계 분석, 교차분석(cross-tabulation analy sis), T-TEST, 다중회귀분석을 실시, 연구결과는 다음과 같다. 첫째, 노화 및 주름개선 화장품을 선택할 때 중요하게 생각하는 요소로 화장품의 사용감이 26명(42.6%), 유효성분은 24명(39.3%)으로 나타났고 효과가 나타나는 시기는 또 4개월에서 6개월 사이가 26명(44.1%)로 나타났다. 둘째, 노화 및 주름개선을 위한 관리용 화장품의 선호 브랜드를 조사해본 결과 우선순위 10개 브랜드의 성분을 조사한 결과 에센스 21.7 ± 7.9개, 크림은 24.3 ± 8.9, 팩은 23.7 ± 10.3개의 성분이 포함되어 있는 것으로 나타났다. 셋째, 유효성분과 추출물은 브랜드별로 다양하게 나타났고 방부제는 메틸파라벤(Methyl Paraben), 계면활성제는 글리세릴스테아레이트(Glyceryl Stearate), 보습제와 컨디셔닝제는 글리세린(Glycerine), 유연화제로는 싸이클로펜타실록산(Cyclopentasiloxane)이 화장품에 가장 많이 사용된 것으로 나타났다. 넷째, 기타성분으로는 오일이나 알콜류로는 비즈왁스(Beeswax), 실리콘유에는 디메치콘(Dimethicone), 점증제로는 카보머(Carbomer), 금속이온 봉쇄제로는 디소디움이디티에이(Disodium EDTA), 산화방지제로는 메에이치티(BHT)가 가장 많이 사용되고 있음을 보였다. 다섯째, 노화 및 주름개선 관리를 위한 화장품의 만족도는 피부 관리 화장품에 대한 이해도(β=.013, t=.079, p>.05)에는 유의적 영향을 미치지 않는 것으로 나타났으나 피부 관리 화장품 성분에 대한 지식(β=.318, t=2.002, p<.01)과 만족도는 유의한 영향을 미치는 것으로 나타났다. 결론적으로 노화 주름관리에 사용되는 화장품들은 특성상 보습제가 포함되어있는 화장품을 선택, 사용되고 있었으며 성분의 지식이나 이해도가 높을수록 만족도가 높아지는 것을 알 수 있었다. As living quality is improving, people earn more income and more women work outside, cosmetics has rapidly developed for expression of their aesthetic desires. Expectancy on cosmetics becomes higher, and the demand of cosmetics is increasing with a focus on its efficacy and effect instead of simple concepts such as refreshing and protection. Therefore, this study aims to analyse components of cosmetics which is used against ageing and wrinkles in skin care shops and satisfaction in it and present basic data on components used. Therefore this study analysed components of cosmetics used for anti-aging and anti-wrinkles in skin care shops and satisfaction, aiming to present basic data on components used. The subjects of the study were skin care experts who are working for skin care shops with more than three employees in Gwangju. For the purpose, the study analysed preference of cosmetic brands used for anti-aging and anti-wrinkles in skin care shops and considerations in selection of cosmetics and effects using a questionnaire. Second, the study examined preferences of cosmetic brands revealed in the questionnaire and analysed components of essence, cream, and pack of top ten brands, and effective components, extracts, preservatives, surfactant, humectant, conditioner, oil, alcohol and others used for anti-ageing and anti-wrinkles. Third, the study identified the relations between components of anti-aging and anti-wrinkles, and care satisfaction. The data collected had the following analyses using SPSS 11.0: a descriptive statistics, a cross-tabulation analysis, a t-test and a multi-regression analysis and the results are presented as follows: First, 26 respondents(42.6%) considered texture of cosmetics important in selecting cosmetics and 24 ones (39.3%) considered effective components important. 26 subjects (44.1%) answered that the period that effectiveness appears is between four and six months. Second, as a result of surveying components of 10 preferred cosmetic brands of anti-ageing and wrinkle-free cosmetics, it was discovered that essence included components of 21.7 ± 7.9, cream included those of 24.3 ± 8.9, and pack included those of 23.7 ± 10.3. Third, each brand had different effective components and extracts, and mainly used methyl paraben as a preservative, glycery istearate as a surface-active agent, glycerine as a humectan and conditioning agent, and cyclopentasiloxane as a softening agent Fourth, it mainly used other components as follows: bees wax for oil and alcohol, dimethicone for silicon oil, and carbom as a thickener. disodium EDTA for sequestering agent, and BHT for antioxidant. Fifth, it was discovered that satisfaction in anti-ageing and wrinkle-free effect did not have a significant effect on understanding of skin care cosmetics (β=.013, t=.079, p>.05), but it had a significant effect on knowledge on components of skin care cosmetics (β=.318, t=2.002, p<.01). In conclusion, it was discovered that cosmetics with humectant was preferred for anti-ageing and wrinkle-free effect and the more knowledge and understanding on components they had, the higher satisfaction they had in cosmetics.

        산유자 잎 에탄올 추출물의 미백, 주름억제, 항염증 및 항산화 효능

        본 연구는 제주도 해변의 벌판에서 드물게 자라는 산유자(Xylosma congesta) 잎 에탄올 추출물의 화장품 기능성 소재로서의 활용 가능성을 알아보기 위하여 cell culture model 및 invitro assay system을 이용하여 미백, 주름개선, 항염증 및 항산화 활성을 분석하였다. 그 결과 산유자 잎 에탄올 추출물은 B16F10 세포에 세포독성 없이 효과적으로 α-MSH에 의해 유도된 melanin 합성을 억제함으로써 높은 미백 활성을 가지는 것이 관찰되었다. CCD-986SK 세포를 이용하여 산유자 잎 에탄올 추출물의 주름개선 활성 능력을 procollagen 합성시험을 통해 분석한 결과, 양성 대조군으로 사용한 TGF-β 만큼은 아니지만 농도별로 각각 120, 122%의 procollagen 합성을 증가시키는 것을 통하여 산유자 잎 에탄올 추출물의 주름개선 활성을 확인하였다. 또한 RAW 264.7 murine macrophage를 이용하여 NO 생성 억제능을 분석함으로써 산유자 잎 에탄올 추출물의 항염증 활성 정도를 측정한 결과, LPS에 의해 유도된 NO 생성이 산유자 잎 에탄올 추출물의 농도 의존적으로 감소하는 결과를 얻을 수 있었다. DPPH 법, ABTS 법, ORAC 법을 이용하여 항산화 활성을 분석한 결과, 50 μg/mL의 농도에서 DPPH및 ABTS 라디칼 소거능이 각각 75.2, 99.1% 증가하는 것이 확인 되었다. ORAC activity assay kit를 이용하여 항산화 활성을 측정한 결과 산유자 잎 에탄올 추출물의 농도에 따라 높은 항산화 활성이 관찰 되었고, DPPH와ABTS 라디칼 소거능 결과와 유사하게 50 μg/mL의 농도에서 가장 높은 항산화 활성(573.74±0.79 μM TE/g)을 나타내었다. 이러한 결과를 통하여 산유자 잎 에탄올 추출물이 높은 미백과 주름개선, 항염증 및 항산화 활성을 나타내는 것을 확인 할 수 있었으며, 기능성 화장품 소재로서의 활용가능성을 제시 할 수 있었다. In the present study, we investigated the biological activities of Xylosma congesta leaf ethanol extract (XCO) using a variety of in vitro and cell culture model systems for antimelanogenic, anti-wrinkle, anti-inflammatory and anti-oxidant activities. First, XCO markedly inhibited α-melanocyte stimulating hormone-stimulated melanin synthesis in B16F10 cells. Secondly, XCO marginally induced procollagen synthesis in CCD-986SK cells. Thirdly, XCO dose-dependently suppressed lipopolysaccharideinduced nitric oxide (NO) production in RAW 264.7 cells. XCO did not affect cell viability at different concentrations used in this study, indicating that XCO-mediated inhibition of melanin,procollagen and NO synthesis is not mediated by cytotoxicity. Finally, XCO was found to exert anti-oxidant effect. Taken together, these findings demonstrate for the first time that XCO possesses anti-melanogenic, anti-wrinkle, anti-inflammatory andanti-oxidant activities, and suggest further evaluation and development of XCO as a functional supplement or cosmetic that may be useful for whitening skin, reducing wrinkles and treating inflammatory responses.

        본 연구는 에탄올 농도별(0, 30, 50, 70, 100 %)로 추출한 사매추출물의 항산화 효과를 Oxygen Radical Absorbance Capacity(ORAC)로 평가한 결과 30 % 에탄올 추출물에서 가장 우수하였다. 따라서 30 % 에탄올 추출물의 주름완화에 관련한 효능 및 효과를 확인하기 위하여 마우스 유래 섬유아세포에 대한 세포 독성, MMP-8 collagenase 억제활성 검사를 실시하였다. 그 결과 30 % 에탄올 사매추출물을 섬유아세포에 처리한 결과 세포 독성이 없었고, 20 %의 MMP-8 collagenase 억제 활성을 보였다. 사매 추출물 1 %가 함유된 Di-Wrinkle Free Cream (DWFC)으로부터 35 ~ 50세의 21명 여성을 대상으로 하루에 2번 도포하여 8주간 인체 안전성과 주름개선 임상평가를 수행하였다. 주름 평가 방법은 3차원 입체 측정기(PRIMOS)를 이용하여 평가를 하였다. DWFC에 대한 안전성 평가는 피험자 방문 시 마다 이상반응, 부종, 인설, 가려움, 자통, 작열감, 뻣뻣함, 따끔거림이나 다른 이상반응이 발생하는지 육안평가와 문진으로 평가하였다. PRIMOS(GFM, Germany)를 이용하여 눈 주변의 주름을 분석한 결과 시험제품 사용 8주 후 통계적으로 유의하게 주름이 13 % 감소하였으며, 시험기간 동안 특별한 피부 이상반응은 관찰되지 않았다. 이와 같은 결과로 보아 사매추출물은 인체에 부작용이 없는 주름개선에 효과 있는 화장품으로 개발이 가능할 것으로 사료된다. In this study, we investigated anti-oxidative effects of Duchesnea indica extracts by using Oxygen Radical Absorbance Capacity (ORAC). The extracts were prepared with 0 %, 30 %, 50 %, 70 % and 100 % aqueous ethanol respectively. The 30 % EtOH D. indica extract showed higher ORAC activity than the other extracts. Therefore, we performed in vitro studies on cytotoxicity of NIH-3T3 cells and MMP-8 collagenase inhibition using by the 30 % EtOH extract. The 30 % EtOH extract showed no cytotoxicity and significant inhibition on MMP-8 collagenase. And we performed clinical studies for the anti-wrinkle effect of the Di-Wrinkle Free Cream. The cream formula was prepared with 2 % arbutin and 1 % D. indica extract. Twenty one healthy women volunteers, ages of 35 and 50, applied the cream on their faces twice a day for 8 weeks. The skin was evaluated with PRIMOS (phaseshift rapid in vivo measuring of human skin) system and analyzed by the student's paired t-test. The wrinkles on the eye region were reduced by 13 % based on the PRIMOS system after 8 weeks. In the safety study of the Di-Wrinkle Free Cream, no symptoms were observed such as erythema, edema, scaling, itching, stinging, burning, tightness and prickling by visual observation and medical examination of volunteers for 8 weeks. Moreover, there was no noticeable skin disorder during experience period. These results suggested that D. indica extracts could be applied as cosmeceuticals effective for anti-wrinkle.

        Functional Properties of Newly-Bred ‘Summer King’ Apples

        In this study, we investigated the various physiological properties of ‘Summer King’ apples (Malus domestica Borkh) to highlight this fruit as a high value functional food source. The peels of newlybred ‘Summer King’ apple were extracted using water and ethanol, and each extract demonstrated relatively high levels of phenolic compounds at 4.41 ㎎·g<SUP>-1</SUP> and 7.39 ㎎·g<SUP>-1</SUP>, respectively. The water and ethanol extracts also showed antioxidant protection factor (PF) levels of 3.37 PF and 2.65 PF, respectively, and the antioxidation effects towards thiobarbituric acid reactive substances were 38% and 30% at 100 ㎍·mL<SUP>-1</SUP> phenolic concentrations. Therefore, extracts of ‘Summer King’ apple possess substantial properties for anti-aging. The anti-inflammatory effect (hyaluronidase inhibition) of the extracts were 4.97% for the water extract and 20.40% for the ethanol extract, both at a 200 ㎍·mL<SUP>-1</SUP> phenolic concentration. Both the water and ethanol extracts showed low α-amylase inhibition effects, but each demonstrated 93.59% and 98.80% α-glucosidase inhibition at a 200 ㎍·mL<SUP>-1</SUP> phenolic concentration, respectively. In an anti-wrinkle assay, the water extract showed 14.55% and 67.73% inhibition of elastase and collagenase, respectively, and the ethanol extract had 56.50% and 77.81% inhibition, respectively. These results support the high potential for the use of these extracts in functional food and cosmetics, and the ‘Summer King’ apple peel was determined to have various functions including anti-oxidation, anti-inflammation, anti-wrinkle, and anti-diabetic effects.

        Anti-Oxidative, Anti-Inflammatory, and Anti-Wrinkling Activities of Sambucus williamsii Hance Methanol Extracts

        Sambucus williamsii Hance traditionally used in oriental herbal medicine to treat bone disease. However, its anti-aging effects as a skin care ingredient has not been fully evaluated. Therefore, we investigated the antioxidant, anti-inflammation, and anti-wrinkling activities of S. williamsii methanol extract in vitro using human keratinocytes, HaCaT cells. The total polyphenol contents of the leaf (SWL), tree (SWT), and seed (SWS) extracts were 48.605, 84.132, and 17.750 mgGAE/g, respectively. The DPPH and ABTS radical scavenging activities of S. williamsii methanol extracts showed dose-dependent anti-oxidant activity. Nitrite production and pro-inflammatory cytokine, interleukin (IL)-6 were reduced by after treatment with SWL extract. In addition, extract caused reduction in nitric oxide by affecting expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 in LPS-activated RAW264.7 murine macrophages. S. williamsii tree extract decreased matrix metalloproteinase (MMP)-1 and increased hydration related gene, AQP-3 expression in TNF-a stimulated HaCaT cells. Furthermore, SWT increased the hyaluronan synthase (HAS)-1, 2, and 3 mRNA levels. From these results, we conclude that S. williamsii methanol extract has excellent antioxidative, anti-inflammatory, and even better anti-wrinkle effects. These results suggest that S. williamsii could be used in functional cosmetics for the prevention or alleviation of skin aging.
