        Hemipteleae davidii(Ulmaceae) is a tree which is widely distributed in Korea and China. It has been used as treatment of cancer, diuretic, tuberculosis, anti-inflammatory agent in folk remedies. Lipid peroxide is associated with aging, diabetes, mutation and cancer. Therefore acute toxicity, carrageenin edema, anti-lipid peroxidation effect are examined. MeOH Ex. has none toxicity and show significant anticarrageenin action by dose dependence. Chloroform and ether fractions show potent inhibition of lipid peroxidation in vitro and in vivo. Butanol fraction in vitro, water fraction in vivo by does dependence had significant inhibition.

      • 배(梨)의 메탄올 추출물이 마우스 복강대식세포의 nitric oxide와 tumor necrosis factor-a 생성에 미치는 영향

        This study was conducted to investigate the effects of pear methanol extracts (P-M) on mouse peritoneal macrophages. Peritoneal macrophages were isolated from mice and cultured with or without LPS in the presence of P-M (0.16, 0.31, 0.63, 1.25, 2.50mg/ml) for 48hrs (TNF-a) or 96hrs (NO). NO levels in culture supernatant were determined by measuring the amount of nitrite, the stable oxidation product of NO, and TNF-a levels in culture supernatant were assayed by a sandwich ELISA. P-M significantly stimulated NO and TNF-a production from peritoneal macrophages at all doses. However, LPS-induced NO production was inhibited by P-M in a dose dependent manner and LPS-induced TNF-a production was not altered by P-M.

      • Prostaglandin E₂가 파골세포 형성에 미치는 영향

        Osteoclasts, the multinucleated giant cells that resorb bone, develop from hemopoietic cells of monocyte-macrophage lineage. Differentiation of osteoclasts is strictly regulated by osteoblasts, which deposit bone. In the present study, prostaglandin E₂ (PGE₂) induced osteoclast formation in co-cultures of mouse bone marrow cells and calvarial osteoblasts. This activity was not observed in the absence of calvarial osteoblasts, suggesting that calvarial osteoblasts are likely target cells of PGE₂. Northern blot analysis revealed that the effect of PGE₂ is associated with increased expression of the osteoclast differentiation factor TNF-related activation-induced cytokine (TRANCE, identical to RANKL, ODF, and OPGL) in mouse calvarial osteoblasts. Induction of TRANCE expression by PGE₂ was partially suppressed by the inhibitors of ERK and p38 MAPK, suggesting that activation of ERK and p38 MAPK is involved in TRANCE expression by PGE₂.

      • 양강의 nitric oxide synthase 저해활성

        Nitric Oxide (NO), derived from L-arginine, is produced by two types of nitric oxide synthase (constitutive NOS: cNOS, inducible NOS: iNOS). The NO produced in large amounts by the iNOS is known to be responsible for the vasodilation and hypotension observed in septic shock and inflammation. The inhibitors of iNOS, thus, may be useful candidates for the treatment of inflammatory diseases accompanied by the overproduction of NO. The activity guided column chromatography of the chloroform soluble fraction obtained from the methanol extract of Alpinia officinarum gave an inhibitory active compound of NO synthesis in LPS-activated macrophages (IC_(50) value; 0.6 μM). The structure of active compound was elucidated as 7-(4-hydoxy-3-methoxyphenyl)-1-phenylhept-4-en-3-one by spectral analysis. This compound can be an useful candidate for the development of new drug to treat endotoxemia and inflammation accompanied by the overproduction of NO.

      • 파골세포 분화에 관여하는 Phosphodiesterase Isozyme 의 규명

        Phosphodiesterases (PDEs) are enzymes that degrade intracellular cAMP and cGMP. In the present study, inhibition of PDE4 was demonstrated to influence osteoclast formation in co-cultures of mouse bone marrow cells and calvarial osteoblasts. Furthermore, PDE4 was shown to modulate mRNA expression of EP4, TRANCE, and COX-2 in calvarial osteoblasts. These results suggest that PDE4 is a key regulator of EP4, TRANCE, and COX-2 expression in osteoblasts, which in turn controls osteoclast formation.

      • 한국인의 고지혈증과 CETP 및 APOE 유전자다형

        Since 1980s, hyperlipidemia in Korea has been rapidly increased and its related cardiovascular diseases are one of the main causes of death in Korea. Thus, for effective prevention of hyperlipidemia, clarification of etiology of the hyperlipidemia in Koreans is required. In this study, we focused on polymorphisms in genes, which are involved in cholesterol-transfer, and -binding, e.g. cholesteryl ester transfer protein (CETP) and apolipoprotein E (APOE). Study subjects were 257 Koreans (cases who had hyperlipidemia, LDL≥130, N=47; controls, N=210; Men, 62%). Genetic polymorphisms were determined by PCR-RFLP, and single base primer extension assays. As results, allele frequency of the CETP -Tag IB1 was significantly higher in cases than that in controls (p<0.05). When age and BMI were controled for statistics, there was 2.3 fold increase of OR (95%CI, 1.1-5.1) for hyperlipidemia in the Bl/Bl homozygotes compated to the B1/B2 heterozygotes. However, 2 SNPs, CI 12R and R158C of the APOE, or 5 haplotypes of the 2 SNPs were not associated with hyperlipidemia risk. Therefore, presence of the CETP-B1 allele has a potential of risk for Korean hyperlipidemia.

      • 의약품 분석에 대한 정도관리에서의 z-스코어에 의한 평가에 대한 대안

        Proficiency test is an essential tool for ensuring analytical ability of analytical chemists and analytical institutes. Usually, the standard protocol for proficiency test is based on mean values or any other reference values and standard deviation estimated by actual analysis or Horwitz's method. In the cases of data set with a certain amount of outliers, which moves means toward them and.inflate standard deviation, the proficiency test often fails in producing satisfactory results. It is because the existence of outliers results in reduced z-score calculated from these mean and standard deviation and in turn, the process fails in detecting unsatisfactory analytical values. In addition to this, Horwitz's estimation of standard deviation accounts solely for the mass-to-mass concentration level, which is not the only factor determining standard deviation in real-world analysis of pharmaceutical formulations. To deal with this kind of problem, various methods for the calculation of z-scores were evaluated and the results were compared with those obtained from median values and median absolute deviation(MAD) introduced by Huber. Sample data were collected from quality control departments of pharmaceutical companies in Kyungln province in Korea and these data were used to evaluate conventional and new methods for proficiency test. As a result, a new method using median values and MAD gave more reliable decisions in proficiency test as applied to the data studied for this experiment.

      • 다양한 다변량 거리측정법에 따른 군집분석에 의한 황금 원산지 구분

        NIR(Near-Infrared) spectroscopy is a nondestructive technique and gives fingerprint-type information. Its scanning speed is much fasten compared to other analytical techniques, and usually, minimum sample preparation is required. Particularly, it has been well studied in agricultural and food industry because of fast analysis. Recently, it became important to discriminate the geological origins of natural products for pharmaceutical uses. In this paper, a method of pattern recognition basically based on a form of cluster analysis is described for the discrimination of geological origin of Scutellaria Radix. When 2nd derivative of NIR spectra were used for the calculations of multivariate distances, the results of geological discrimination of the crude drug was successful without any misclassification. The result of successful and perfect discrimination of geological origins of the samples was achieved using several multivariate distance measures, such as Euclidean distance(2-norm), Minkowski distance with norms of 0.5, 1.5 or 3, cosine distance, city block distance(l-norm), or Chebychev distance(oo-norm). With correlation distance, there were a few cases of wrong classification results. On the contrary, with reflectance spectra or 1st derivative spectra, the discrimination was not successful and some distance measures such as Mahalanobis distance or Standardized Euclidean distance were not appropriate for this kind of cluster analysis.

      • RANKL 유도성 파골세포 분화에 있어서 LTB4의 역할 규명

        Leukotriene B4 (LTB4) is a potent lipid mediator of inflammation formed by the 5-lipoxygenase(5-LO)-catalyzed oxidation of arachidonic acid. LTB4 mediates diverse biological activities through two distinct highly conserved G protein-coupled receptors BL T1 and BL T2. In this study, we investigated the role of LTB4 in receptor activator of nuclear factor-kappaB ligand (RANKL)-induced osteoclast formation. 5-lipoxygenase inhibitor and antagonists for BLT1 or BLT2 were shown to suppress the RANKL-induced osteoclast formation in mouse bone marrow-dereived macrophage (BMM) culture system. Furthermore, RT-PCR analysis demonstrated that rnRNA expression levels for 5-LO, BLT1, and BLT2 on BMM cells were significantly increased by the treatment of RANKL. Taken together, these data suggest the possible role of LTB4 on RANKL-induced osteoclast formation.
