<P>In spite of severe side effects, chemotherapy is widely used as a major anticancer treatment in non-small cell lung cancer (NSCLC). In order to enhance the therapeutic properties and reduce side effects, enormous efforts have been devoted to direct anticancer agents specifically to tumor tissues by the use of nanoparticles, or cancer cell marker attached drugs. However, cell-specific chemotherapy is still in its infancy and is not applicable to all types of cancers due to the complexity of the cancer occurrence and progress. In this study, we demonstrate that hyaluronan (HA)-conjugated cisplatin has highly selective and sensitive anticancer effects in NSCLC cells that overexpress the trans-membrane receptor, CD44, as HA is a specific ligand for CD44. In proliferation experiments, HA-conjugated cisplatin showed dramatic cell viability decreases (from 100% to 42.31%) in H1299 cells, which overexpress CD44, whereas no such change was observed in A549 and HFL1, which have little to no expression of CD44. More importantly, conjugation with HA decreased the dosage concentration of cisplatin by 50-fold for both cytotoxic and anti-metastatic effects. In addition, HA-cisplatin conjugate treatment selectively decreased migration (from 100% to 7.8%) and invasiveness (from 100% to 21.4%, respectively) of H1299. Based on our experimental results, we strongly believe that HA-cisplatin conjugate is a potential anticancer chemo-agent, which target CD44 overexpression in NSCLC, with minimal side effects and high therapeutic properties.</P>

<P>Conclusions: We successfully created an ischemic animal model of the gastric conduit. Using this animal model, we evaluated the sensitivity and applicability of the fluorescent imaging system for observation and identification of ischemic areas during an esophagectomy.</P>
Taguchi Approach for Anti-heat Stress Prescription Compatibility in Mice Spleen Lymphocytes In Vitro

Heat stress (HS) may induce immunosuppression as well as inhibit the proliferation of lymphocytes. This study evaluated the effects on immune function of our prescription on splenic lymphocytes under HS as well as its compatibility. The effects of four herbal extracts from Agastache rugosa, Atractylodes lancea, Cortex Phellodendri, and Gypsum Fibrosum on heat treated splenic lymphocytes were investigated and the compatibility of the prescription was also explored by using the Taguchi method. This study revealed changes in proliferation by traditional Chinese medicines of splenic lymphocytes after HS. Proliferation in the HS group was significantly lower than the control group. Under HS, the effects of higher concentrations of Agastache rugosa (100 and 200 μg/mL), Atractylodes lancea (100 and 200 μg/mL), Cortex Phellodendri (50 and 100 μg/mL) and Gypsum Fibrosum (100 and 200 μg/mL) caused a significant increase on ConA/LPS-induced proliferation of lymphocytes than lower concentrations. We, therefore, conclude that the prescription of traditional Chinese medicines may recover splenic lymphocytes from the immunosuppression induced by HS. The Taguchi design, which allows rapid and high efficiency for the selection of the best conditions for our prescription on HS-treated splenic lymphocytes, demonstrated that Agastache rugosa (200 μg/mL), Atractylodes lancea (200 μg/mL), Cortex Phellodendri (100 μg/mL) and Gypsum Fibrosum (100 μg/mL)were the optimal conditions for the prescription. The validation experiment confirmed that our composition in optimum extraction conditions enhanced effects on ConA or LPS-stimulated lymphocytes under HS. The results showed that the Taguchi optimization approach is a suitable method for optimization of the composition of prescription.

The effect of cooling rate on the microstructure evolution of liquid Cu nanoparticles during their solidification process is investigated by using a molecular dynamics simulation based on the embedded atom method (EAM) potential developed by Foiles et al.. The potential energy analysis,the pair distribution function and the common neighbor analysis have been used. The results show that the solidification point increases with decreasing cooling rate and that the solidification of the microstructure of Cu nanoparticles varies with the cooling rate. The microstructure consists of fcc,hcp and bcc crystals or mixtures, though the fcc structure dominates, except in the amorphous state. An amorphous structure was obtained when the cooling rate reached 1.0 × 1013 K/s or higher while crystallization degree increased with decreasing cooling rate, and the total content of crystal structures reached to 95% when the cooling rate dropped to 4.0 × 1011 K/s, which was nearly a perfect crystal structure. The results also indicate that a single-crystal nanoparticle will not be obtained by quenching the liquid metal under various cooling rates.
Structure and property of a vertical cutting Ca0.4Sr0.6Bi4Ti4O15 ferroelectric ceramic

Ca0.4Sr0.6Bi4Ti4O15 ceramics with a-axis preferred orientation were prepared by a solid-state reaction method. The cylindrical Ca0.4Sr0.6Bi4Ti4O15 ceramics were cut into two types of plates: one was along the cylinder axis (A), and the other was perpendicular to the direction of the cylinder axis. The effects of sintering temperature on the structure and electrical properties of the Ca0.4Sr0.6Bi4Ti4O15 ceramics were investigated. When the sintering temperature was 1180 oC, sample A gave the highest a-axis (200) orientation with a value of about 27.1%. At this sintering temperature, sample A showed better dielectric properties and ferroelectric properties, the relative dielectric constant (εr), dielectric loss (tan δ), remnant polarization (Pr), and coercive field (Ec) were 217.6, 0.0027, 14.2 μC/cm2 and 49.6 kV/cm, respectively.

Ascaris suum eggs are inactivated by composting conditions; however, it is difficult to find functional changes in heat-treated A. suum eggs. Here, unembryonated A. suum eggs were incubated at 20˚C, 50˚C, and 70˚C in vitro, and the gene expression levels related to viability, such as eukaryotic translation initiation factor 4E (IF4E), phosphofructokinase 1 (PFK1), and thioredoxin 1 (TRX1), and to apoptosis, such as apoptosis-inducing factor 1 (AIF1) and cell death protein 6 (CDP6), were evaluated by real-time quantitative RT-PCR. No prominent morphological alterations were noted in the eggs at 20˚C until day 10. In contrast, the eggs developed rapidly, and embryonated eggs and hatched larvae began to die, starting on day 2 at 50˚C and day 1 at 70˚C. At 20˚C, IF4E, PFK1, and TRX1 mRNA expression was significantly increased from days 2-4; however, AIF1 and CDP6 mRNA expression was not changed significantly. IF4E, PFK1, and TRX1 mRNA expression was markedly decreased from day 2 at 50˚C and 70˚C, whereas AIF1 and CDP6 mRNA expression was significantly increased. The expressions of HSP70 and HSP90 were detected for 9-10 days at 20˚C, for 3-5 days at 50˚C, and for 2 days at 70˚C. Taken together, incremental heat increases were associated with the rapid development of A. suum eggs, decreased expression of genes related to viability, and earlier expression of apoptosis-related genes, and finally these changes of viability- and apoptosis-related genes of A. suum eggs were associated with survival of the eggs under temperature stress.
