        Upregulation of PITX2 Promotes Letrozole Resistance Via Transcriptional Activation of IFITM1 Signaling in Breast Cancer Cells

        Purpose Although the interferon  (IFN) signaling and the paired-like homeodomain transcription factor 2 (PITX2) have both been implicated in the progression of breast cancer (BCa), it remains obscure whether these two pathways act in a coordinated manner. We therefore aimed to elucidate the expression and function of PITX2 during the pathogenesis of endocrine resistance in BCa. Materials and Methods PITX2 expression was assessed in BCa tissues using quantitative reverse transcription polymerase chain reaction (RT-qPCR) and immunohistochemistry and in experimentally induced letrozole-resistant BCa cells using RT-qPCR and immunoblotting. Effects of PITX2 deregulation on BCa progression was determined by assessing MTT, apoptosis and xenograft model. Finally, using multiple assays, the transcriptional regulation of interferon-inducible transmembrane protein 1 (IFITM1) by PITX2 was studied at both molecular and functional levels. Results PITX2 expression was induced in letrozole-resistant BCa tissues and cells, and PITX2 induction by IFN signaling powerfully protected BCa cells against letrozole insult and potentiated letrozole-resistance. Mechanistically, PITX2 enhanced IFN-induced AKT activation by transactivating the transcription of IFITM1, thus rendering BCa cells unresponsive to letrozoleelicited cell death. Additionally, ablation of IFITM1 expression using siRNA substantially abolished IFN-elicited AKT phosphorylation, even in the presence of PITX2 overexpression, thus sensitizing BCa cells to letrozole treatment. Conclusion These results demonstrate that constitutive upregulation of PITX2/IFITM1 cascade is an intrinsic adaptive mechanism during the pathogenesis of letrozole-resistance, and modulation of PITX2/IFITM1 level using different genetic and pharmacological means would thus have a novel therapeutic potential against letrozole resistance in BCa.

        A new species of Aphis Linnaeus from Guangxi, China (Hemiptera, Aphididae), based on morphological characters and DNA sequences

        A new species of the genus Aphis with stridulatory apparatus, Aphis gnetuma Qiao sp. n., feeding on Gnetum parvifolium which is a new host plant record for aphids from Guangxi, China is described. The new species is distinguished by a well-developed median frontal tubercle and body bearing white wax powder in life. The taxonomic position of this new species is supported by analysis results of COI and gnd genes. A key to Aphis species with stridulatory spines on the hind tibiae of apterous viviparous females is provided. The type specimens studied are deposited in the National Zoological Museum of China, Institute of Zoology, Chinese Academy of Sciences, Beijing, China.

      • Knocking Down Nucleolin Expression Enhances the Radiosensitivity of Non-Small Cell Lung Cancer by Influencing DNA-PKcs Activity

        Nucleolin (C23) is an important anti-apoptotic protein that is ubiquitously expressed in exponentially growing eukaryotic cells. In order to understand the impact of C23 in radiation therapy, we attempted to investigate the relationship of C23 expression with the radiosensitivity of human non-small cell lung cancer (NSCLC) cells. We investigated the role of C23 in activating the catalytic subunit of DNA-dependent protein kinase (DNA-PKcs), which is a critical protein for DNA double-strand breaks (DSBs) repair. As a result, we found that the expression of C23 was negatively correlated with the radiosensitivity of NSCLC cell lines. In vitro clonogenic survival assays revealed that C23 knockdown increased the radiosensitivity of a human lung adenocarcinoma cell line, potentially through the promotion of radiation-induced apoptosis and adjusting the cell cycle to a more radiosensitive stage. Immunofluorescence data revealed an increasing quantity of ${gamma}$-H2AX foci and decreasing radiation-induced DNA damage repair following knockdown of C23. To further clarify the mechanism of C23 in DNA DSBs repair, we detected the expression of DNA-PKcs and C23 proteins in NSCLC cell lines. C23 might participate in DNA DSBs repair for the reason that the expression of DNA-PKcs decreased at 30, 60, 120 and 360 minutes after irradiation in C23 knockdown cells. Especially, the activity of DNA-PKcs phosphorylation sites at the S2056 and T2609 was significantly suppressed. Therefore we concluded that C23 knockdown can inhibit DNA-PKcs phosphorylation activity at the S2056 and T2609 sites, thus reducing the radiation damage repair and increasing the radiosensitivity of NSCLC cells. Taken together, the inhibition of C23 expression was shown to increase the radiosensitivity of NSCLC cells, as implied by the relevance to the notably decreased DNA-PKcs phosphorylation activity at the S2056 and T2609 clusters. Further research on targeted C23 treatment may promote effectiveness of radiotherapy and provide new targets for NSCLC patients.

        An Adaptation Method in Noise Mismatch Conditions for DNN-based Speech Enhancement

        The deep learning based speech enhancement has shown considerable success. However, it still suffers performance degradation under mismatch conditions. In this paper, an adaptation method is proposed to improve the performance under noise mismatch conditions. Firstly, we advise a noise aware training by supplying identity vectors (i-vectors) as parallel input features to adapt deep neural network (DNN) acoustic models with the target noise. Secondly, given a small amount of adaptation data, the noise-dependent DNN is obtained by using L2 regularization from a noise-independent DNN, and forcing the estimated masks to be close to the unadapted condition. Finally, experiments were carried out on different noise and SNR conditions, and the proposed method has achieved significantly 0.1%-9.6% benefits of STOI, and provided consistent improvement in PESQ and segSNR against the baseline systems.

        Doxorubicin-Loaded CuS Nanoparticles Conjugated with GFLG: A Novel Drug Delivery System for Lymphoma Treatment

        Doxorubicin (DOX) plays an important part in lymphoma treatment. However, various side effects on normal tissues restrict its clinical use. Nanocarriers connected by Gly–Phe–Leu–Gly (GFLG) can be equipped with the advantages of nanoparticles (NPs), their enhanced permeability and retention (EPR) effect, and surface modifiability. Nanocarriers can also be specifically enzymatically hydrolyzed by cathepsin (Cath) B, a kind of enzyme highly expressed in tumor cells. In this work, we proposed a novel drug delivery system comprising GFLG conjugated with copper sulfide (CuS) NPs loaded with DOX. The system, designated as CuS-GFLG-DOX, could be used for NP-based targeted combination chemotherapy. Results showed that the drug delivery system had an appropriate diameter, good dispersibility, high encapsulation efficiency and high drug loading. The system also exhibited an excellent targeting of lymphoma cells and an enhanced antitumor activity. The possible pathway to induce cytotoxic effects was Bcl-2/caspase-mediated apoptosis pathway. In conclusion, CuS-GFLG-DOX could precisely deliver drugs to lymphoma cells and could be a novel and promising therapeutic option for lymphoma.

      • <i>Sinomicrobium oceani</i> gen. nov., sp. nov., a member of the family <i>Flavobacteriaceae</i> isolated from marine sediment

        <P>A marine bacterium, designated SCSIO 03483<SUP>T</SUP>, was isolated from a marine sediment sample collected from the Nansha Islands in the South China Sea. The strain produced roundish colonies with diffusible yellow-coloured pigment on nutrient agar medium or marine agar 2216. Optimal growth occurred in the presence of 0–4 % (w/v) NaCl, at pH 7.0 and a temperature range of 28–37 °C. 16S rRNA gene sequence analysis indicated that the isolate belonged to the family <I>Flavobacteriaceae</I> and showed relatively high sequence similarity with <I>Imtechella halotolerans</I> K1<SUP>T</SUP> (92.7 %). Phylogenetic analysis based on nearly complete 16S rRNA gene sequences revealed that the isolate shared a lineage with members of the genera <I>Imtechella</I>, <I>Joostella</I> and <I>Zhouia</I>. Phospholipids were phosphatidylethanolamine, two unidentified aminolipids and three unknown polar lipids. The major respiratory quinone was MK-6 and the major fatty acids were iso-C<SUB>15 : 0</SUB>, iso-C<SUB>17 : 0</SUB> 3-OH and summed feature 3 (C<SUB>16 : 1</SUB>ω6<I>c</I>/C<SUB>16 : 1</SUB>ω7<I>c</I>). The DNA G+C content of strain SCSIO 03483<SUP>T</SUP> was 38.4 mol%. On the basis of phenotypic, chemotaxonomic and molecular data, strain SCSIO 03483<SUP>T</SUP> represents a novel species in a new genus in the family <I>Flavobacteriaceae</I>, for which the name <I>Sinomicrobium oceani</I> gen. nov., sp. nov. is proposed. The type strain of <I>Sinobacterium oceani</I> is SCSIO 03483<SUP>T</SUP> ( = KCTC 23994<SUP>T</SUP> = CGMCC 1.12145<SUP>T</SUP>).</P>

        The Reflux Symptoms Before and After Peroral Endoscopic Myotomy of Chinese Patients With Achalasia

        Background/Aims Achalasia is a rare disease, but the incidence is increasing recently. Peroral esophageal myotomy (POEM) is an effective treatment. Regurgitation is a common symptom before and after POEM. Our aim is to investigate the factors related to preoperative and postoperative reflux symptoms. Methods Our study was retrospective. The achalasia patients diagnosed by high-resolution manometry and gastroscopy were divided into reflux group and non-reflux group before and after POEM, respectively. General information, symptoms, POEM information, and manometric results were compared. Results (1) Ninety-six of 130 patients had reflux symptoms before POEM. The lower esophageal sphincter pressure (LESP) in the reflux group was significantly higher than the non-reflux group (P = 0.023), while integrated relaxation pressure (IRP) was similar. The reflux group had longer esophagus than the non-reflux group (P = 0.006). Reflux symptoms were not related to subtypes of achalasia. (2) Twenty-five of 84 patients had reflux symptoms after POEM. Postoperative Eckardt scores, LESP, and 4-second IRP (4sIRP) were significantly lower than the preoperative values (P < 0.001). The preoperative values and POEM information were similar between the postoperative 2 groups and there was no significant difference in the presence of preoperative reflux symptoms between 2 groups. The postoperative LESP and 4sIRP were similar between the 2 groups, however, the postoperative UESP was significantly higher in the reflux group than the non-reflux group (P = 0.042). The non-reflux group had more declines in Eckardt scores and LESP than the reflux group. Conclusions The reflux symptoms of achalasia patients without treatment were mainly due to food retention. The postoperative reflux symptoms were not the sign of the excessive relaxation of lower esophageal sphincter. (J Neurogastroenterol Motil 2021;27:377-389)

      • Research on the Optimization of the Performance of CTL Loop Discriminator

        Chip Tracking Loop (CTL) is the key to ensure the accuracy of regenerative pseudo-noise ranging in the deep space measurement and control communication system. However, due to the impact of the code imbalance, clock attenuation rate and noise, the loop locking time of CTL is prolonged and the ranging accuracy is degraded. In this paper, we proposed the discriminator optimization algorithm to address this problem. This algorithm is based on the loop discrimination performance of clock code and composite code, and the impact of the noise on the discrimination performance. In order to improve the tracking accuracy and shorten the loop locking time, this algorithm corrected the curve of discriminator’s gain by compensating discriminator’s gain. Both the theory analysis and simulation results illustrate that the algorithm can correct the discriminator’s gain curve, reduce the loop locking time and ensure the significant tracking accuracy in low SNR.
