        Wind loads on fixed-roof cylindrical tanks with very low aspect ratio

        Wind tunnel tests are conducted to investigate the wind loads on vertical fixed-roof cylindrical tanks with a very low aspect ratio of 0.275, which is a typical ratio for practical tanks with a volume of 100,000 . Both the flat-roof tank and the dome-roof tank are investigated in present study. The first four moments of the measured wind pressure, including the mean and normalized deviation pressure, kurtosis and skewness of the pressure signal, are obtained to study the feature of the wind loads. It is shown that the wind loads are closely related to the behavior of flow around the structure. For either tank, the mean wind pressures on the cylinder are positive on the windward area and negative on the sides and the wake area, and the mean wind pressures on the whole roof are negative. The roof configurations have no considerable influence on the mean pressure distributions of cylindrical wall in general. Highly non-Gaussian feature is found in either tank. Conditional sampling technique, envelope method, and the proper orthogonal decomposition (POD) analysis are employed to investigate the characteristics of wind loads on the cylinder in more detail. It is shown that the patterns of wind pressure obtained from conditional sampling are similar to the mean pressure patterns.An instantaneous pressure coefficient can present a wide range from the maximum value to the minimum value. The quasi-steady assumption is not valid for structures considered in this paper according to the POD analysis.

        An improved horizontally reversible plow design based on virtual assembly semantics and constraint

        There exit model errors in the constructed Horizontally reversible plough (HRP) structure through Interference check technique (ICT). This is basically due to missing parts and geometric interference and, hence, has significantly adverse effects in improving HRP. In thispaper an improved design to refine the three dimensional (3D) model of HRP is implemented by using Virtual assembly technology(VAT). To achieve this, the authors propose a combined virtual assembly semantics and constraint for assembly planning and simulationof HRP in the commercial software, e.g., SolidWorks. First, the assembly planning of HRP was captured with a semantics-based model,including spatial position, assembly orientation, type and parameters of the semantic entities; secondly, the assembly simulation of HRPwas performed by using constraint-based VAT, e.g. component grasping, moving and releasing; finally, the obtained HRP model wasverified through ICT again. The results demonstrate that not any model error exists in the refined 3D HRP model any longer and thatsemantics and constrain based VAT can support the interactive operation more effectively and accurately than those with geometric constraints. This technique was previously used for HRP model. Based on the refined 3D HRP model, the design improvement of Remotecylinder base (RCB), an important component of HRP, will be forthcoming in a future paper.

      • An Arabidopsis SUMO E3 Ligase, SIZ1, Negatively Regulates Photomorphogenesis by Promoting COP1 Activity

        <P>COP1 (CONSTITUTIVE PHOTOMORPHOGENIC 1), a ubiquitin E3 ligase, is a central negative regulator of photomorphogenesis. However, how COP1 activity is regulated by post-translational modifications remains largely unknown. Here we show that SUMO (small ubiquitin-like modifier) modification enhances COP1 activity. Loss-of-function siz1 mutant seedlings exhibit a weak constitutive photomorphogenic phenotype. SIZ1 physically interacts with COP1 and mediates the sumoylation of COP1. A K193R substitution in COP1 blocks its SUMO modification and reduces COP1 activity in vitro and in planta. Consistently, COP1 activity is reduced in siz1 and the level of HY5, a COP1 target protein, is increased in siz1. Sumoylated COP1 may exhibits higher transubiquitination activity than does non-sumoylated COP1, but SIZ1-mediated SUMO modification does not affect COP1 dimerization, COP1-HY5 interaction, and nuclear accumulation of COP1. Interestingly, prolonged light exposure reduces the sumoylation level of COP1, and COP1 mediates the ubiquitination and degradation of SIZ1. These regulatory mechanisms may maintain the homeostasis of COP1 activity, ensuing proper photomorphogenic development in changing light environment. Our genetic and biochemical studies identify a function for SIZ1 in photomorphogenesis and reveal a novel SUMO-regulated ubiquitin ligase, COP1, in plants.</P>

        Microstructure and Magnetic Property of Ferromagnetic Fe-Pd-Rh Alloys

        This study shows the appearance of an intermediate martensitic structure between the fcc → L10 martensitic transformation in aged Fe-30Pd-4Rh alloys. The intermediate phase (L1m) has a monoclinic structure with lattice parameters of a = 3.193 A, b = 3.684 A, c = 3.141 A and β = 92.042˚, as confirmed by transmission electron microscopy (TEM) and X-ray diffraction. The crystal orientation between L1m and L10 can be demonstrated as [101]L10//[100]L1m. This observation suggests that the observed intermediate L1m monoclinic phase may be an adaptive martensite. The magnetization (M) versus magnetic field (H) M-H curve measured at temperatures of 50, 200 and 350 K of the alloys, which were first solution-treated (ST) and then thermally aged at 450 ℃ for 100 hours, revealed an abrupt drop at the saturation remanence (Mr). This result indicates that two phases exist in the aged alloys and that the saturation remanences of these two phases differ. The two phases, i.e., the adaptive L1m monoclinic phase and the ordered L10 martensitic structure, were conrmed by TEM and X-ray studies.       • Scutellaria Extract Decreases the Proportion of Side Population Cells in a Myeloma Cell Line by Down-regulating the Expression of ABCG2 Protein

        Background and Aims: Scutellaria is one of the most popular traditional Chinese herbal remedies against various human diseases, including cancer. In this study, we examined the active effects of Scutellaria extract and its main flavonoid constituents on the proportion of side population cells within human multiple myeloma cell line RPMI8226 in vitro and explored the potential molecular mechanisms involved. Materials and Methods: The contents of flavonoids in ethanolic extract of Scutellaria baicalensis Georgi were determined using high performance liquid chromatography. The antiproliferative effect of the ethanolic extract on RPMI-8226 was determined by CCK assay. Apoptosis was measured by annexin combining with propidium iodide in a flow cytometer. Cell cycle analysis was performed by propidium iodide staining in combination with flow cytometry analysis. Hoechst 33342 exclusion assay was used for the identification of side population within RPMI8226 cells. The expression of ABCG2 protein was assessed by Western blotting assay. Results: The content of major flavonoids constitutents of Scutellaria extract was baicalin (10.2%), wogonoside (2.50%), baicalein (2.29%), and wogonin (0.99%), respectively. The crude Scutellaria extract did not show significant anti-proliferative effect, apoptosis induction and cell cycle arrest in RPMI-8226 within the concentrations of $1-75{\mu}g/mL$. However, the ethanolic extract, baicalein, wogonin and baicalin reduced the side population cells in RPMI-8226, and data showed that baicalein and wogonin had stronger inhibitory effects. Correspondingly, they also exhibited significant effects on decreasing the expression level of ABCG2 protein in RPMI-8226 in vitro. Conclusions: Our results for the first time demonstrated a novel mechanism of action for Scutellaria extract and its main active flavonoids, namely targeting SP cells by modulating the expression of ABCG2 protein. This study provides an insight for new therapeutic strategies targeting cancer stem cells of multiple myeloma.

        A Convergence Study on the Factors Influencing E-Learning Intentions of English Knowledge Short Videos Users

        Against the backdrop of rapid digitalization in education and media, English knowledge short videos, as an emerging online learning tool, are transforming traditional educational models. This mode of learning is not only universally significant for scientists across disciplines but also offers deeper insights for experts in educational technology and online learning. This study aims to explore the factors influencing the E-learning intentions of users of this learning tool, a critical issue for the current educational environment. Based on the Technology Acceptance Model (TAM), the study conducted a questionnaire survey on 388 valid sample users and analyzed the data using a structural equation model. The results indicate that perceived interactivity and technical support significantly enhance users perception of platform ease of use, while cognitive style has a less pronounced impact on this perception. Moreover, these factors also positively influence users perceived usefulness of English knowledge short videos. Perceived interactivity and technical support significantly impact E-learning intentions through perceived ease of use and perceived usefulness. These findings provide new theoretical perspectives in the field of online learning, particularly in understanding and enhancing E-learning intentions. This study offers practical guidance for the innovative design of online educational resources and the development of educational technology. Furthermore, the findings underscore the importance of considering users interactive experience and technical support in the design and implementation of online learning platforms, providing valuable insights for educational practices and policy-making in online learning environments.
