        Effects of epigallocatechin gallate on CoCl_(2)-induced apoptosis in PC12 cells

        Neuronal apoptotic events, consequently resulting in neuronal cell death, are occurred in hypoxic/ischemic condition. This cell death has been shown to be accompanied with the production of reactive oxygen species (ROS), which can attack cellular components such as nucleic acids, proteins and phospholipid. However, the underlying mechanisms of apoptosis induced in hypoxic/ischemic condition and its treatment methods are unsettled. Cobalt chloride (CoCl_(2)) has been known to mimic hypoxic condition including the production of ROS. Epigallocatechin gallate (EGCG). a green tea polyphenol, has diverse pharmacologial activities in cell growth and death. This study was aimed to investigate the apoptotic mechanism by CoCl_(2) and effects of EGCG on CoCl_(2)-induced apoptosis in PC12 cells. Administration of CoCl_(2) decreased cell survival in dose- and time-dependent manners and induced genomic DNA fragmentation. Treatment with 100 µM EGCG for 30 min before PC12 cells were exposed to 150 µM CoCl_(2), being resulted in the cell viability and DNA fragmentation being rescued. CoCl_(2) caused morphologic changes such as cell swelling and condensed nuclei, whereas EGCG attenuated morphologic changes by CoCl_(2). EGCG suppressed the apoptotic peak and a loss of Δψ_(m) induced by CoCl_(2). CoCl_(2) decreased Bcl-2 expression but Bax expression was not changed in CoCl_(2)-treated cells. EGCG attenuated the Bcl-2 underexpression by CoCl_(2). CoCl_(2) augumented the cytochrome c release from mitochondria into cytoplasm and increased caspase-8, -9 and caspase-3 activity, a marker of the apoptotic executing stage. EGCG ameliorated the incruement in caspase-8, -9 and -3 activity, and cytochrome c release by CoCl_(2). NAC (N-acetyl-cysteine), a scavenger of ROS, attenuated CoCl_(2)-induced apoptosis in consistent with those of EGCG. These results suggest, that CoCl_(2) induces apoptotic cell death through both mitochondria- and death receptor-dependent pathway and EGCG has neuroprotective effects against CoCl_(2)-induced apoptosis in PC12 cells. 신경세포자멸사는 저산소 및 허혈환경에서 일어나며 이러한 세포죽은 reactive oxidant species (ROS) 생성을 동반함이 알려져있다. 그러나, 저산소 및 허혈환경에서 일어나는 세포자멸사의 기전 및 그 치료방법은 아직 정립되어 있지 않다. CoCl_(2)는 ROS를 생성하는 등 저산소환경과 유사한 조건을 초래하는 것으로 알려져 있다. Epigallocatechin gallate (EGCG)는 녹차의 polyphenol 성분으로서 세포성장과 죽음에 다양한 약리학적 효과를 나타냄이 알려져 있다. 본 연구는 PC12 세포에서 CoCl_(2)에 의한 세포자멸사기전을 밝히고 이에 미치는 EGCG의 효과를 조사하는데 목적이 있다. Cell viability는 MTT 측정으로 조사되었고, DNA fragmentation은 DNA laddering으로 조사되었다. Bcl-2와 Bax발현 정도는 RT-PCR로, caspase-3와 -9의 활성은 spectrophotometer, caspase-8의 활성은 flow cytometry에 의해 측정되었다. 미토콘드리아에서 세포질로 분비된 cytochrome c는 western blot으로, 분해된 DNA 양과 미토콘드리아 세포막전위 (Δψ_(m))는 FACScan으로 조사되었다. CoCl_(2)투여로 PC12 세포수는 용량 및 시간 의존형태로 감소하였고, genomic DNA fragmentation이 발생하였다. CoCl_(2)투여로 야기된 cell viability의 감소와 DNA fragmentation은 EGCG 전처치에 의해 억제되었다. CoCl_(2)은 세포용적팽창과 condensed nuclei 같은 형태적 변화를 일으켰으며, apoptotic peak, Δψ_(m)감소 및 cytochrome c 유리를 야기하였다. EGCG는 CoCl_(2)에 의한 세포형태변화, apoptotic peak, Δψ_(m)소실 및 cytochrome c 유리를 억제시켰다. CoCl_(2)는 Bcl-2 발현을 감소시켰지만, Bax 발현에는 영향을 미치지 않았다. EGCG는 CoCl_(2)에 의해 야기된 Bcl-2 발현 감소를 억제시켰다. CoCl_(2)는 caspase-3, -8, 그리고 -9의 활성을 증가시켰으며, EGCG는 CoCl_(2)에 의한 세포자멸사를 억제시켰다. 본 실험결과는 PC12 세포에서 CoCl_(2)가 미토콘드리아 의존 및 death receptor의존 기전으로 세포자멸사를 일으키며, EGCG는 세포자멸사기전을 억세지킴으로 신경보호기능을 가짐을 시사하였다.

        8년간(1992~1999) 산업안전보건연구원에 의뢰된 직업병 심의 사례 분석

        Objectives : Pneumoconisis and noise-induced hearing loss (NIHL) have been reported as main occupational diseases by the Special Health Examination. The Industrial Accident Compensation Insurance has reported various work-related diseases, however, these two diseases occupied almost a half of compensated cases. Therefore, it was not well known about the status of occupational diseases other than pneumoconiosis, NIHL, and cardio-cerebrovascular accident (CVA). This study was conducted to analyze claimed cases as an occupational disease, that was requested to the Korea Industrial Safety and Health Agency (KOSHA). Methods : The local office of the Korea Labor Welfare Corporation (KLWC) has asked the KOSHA for confirmation of claimed cases as an occupational disease. We analyzed 379 cases requested from KLWC, the Ministry of Labor, employers, unions and occupational health agencies from 1992 to 1999. Results : Male was 80.7 % of the requested cases. Their mean age was 42 years old and 75.5 % of them were more than 35 years old. The requested cases were increased rapidly from 25 cases in 1992 to 108 cases in 1999 and the accept rate was 50.7 %. The majority of requested cases were respiratory diseases(22.4%), cancers(18.5%), Neuropsychiatric problems(14.5%), and musculoskeletal problems(13.5%). The accept rate was high in reproductive, respiratory, musculoskeletal and digestive disorders and low in neuropsychiatric, renal and otologic problems and occupational cancers. 73.6% of them were caused by chemical agents, especially 28.5 % were by organic solvents. 67% of them were clinically confirmed at university hospitals. A half of the cases were from KyongIn area, even the request came from the whole country. Conclusions : A claim was common in workers whose age was over 35 years old and exposure history was over 10 years. The respiratory diseases and neuropsychiatric disorders were still main problems in occupational health and occupational cancers was increasing even though its accept rate was not high yet.

      • 중탄산나트륨(NaHCO₃) 투여가 1500m 런닝기록과 심박수, 혈중 pH, HCO₃??, 젖산농도에 미치는 영향

        본 연구에서는 고등학교에 재학중인 학생 8명을 대상으로 중탄나트륨(NAHCO??)투여에 대한 다음과 같은 연구가설을 검증하는 것이었다. 1)중탄산나트륨(300mg/kg.wt)의 섭취는 염기성증(alkalosis)의 유발을 촉진시키는가? 2)중탄산나트륨(300mg/kg.wt)의 섭취는 1500m 런닝기록에 유의한 효과를 나타내는가? 3)중탄산나트륨(300mg/kg.wt)의 섭취는 1500m 런닝수행시 심박수 및 혈중 pH, HCO????, 젖산농도에 유의한 효고를 나타내는가? 중탄산나트륨 투여 전,후간 심박수 및 1500m 런닝기록은 유의한 효과를 나타내지 못하였으나, 혈중 pH는 중탄산나트륨 투여후 1500m 런닝직후에서 유의한 효과(P<0.01)를 나타내었으며 HCO????농도의 경우에 있어서도 투여 60분후(pre-running, P<0.0001)동일한 효과를 나타내는 것이 증명 되었다. 또한 중탄산나트륨 투여후 혈중 젖산농도는 투여전에 비해 투여후에 더욱 크게 증가된 수준(P<0.01)을 나타내었다. 결론적으로, Ergogenic aids로서 중탄산나트륨(300mg/kg.wt)의 투여는 생체내 알카리성 유발을 촉진시키며 혈중 pH, HCO???? 및 젖산농도에 유의한 생리적 효과를 나타내었으나 1500m 런닝기록에 단축효과는 나타내지 못하였다. 이러한 결과가 시사하는바는 운동수행능력의 향상을 위한 NaHCO?? 섭취는 무산소성 운동능력 및 NaHCO??의 투여량과 밀접한 관련성을 나타낸다는 것이다. The purpose of this study was to examine the hypothesis as follows : Firstly, Does the ingestion of sodium bicarbonate(NaHCO??) result. in induce of alkalosis? Secondly, Does the ingestion of sodium bicarbonate result in improving 1500m running records? Lastly, Does the ingestion of sodium bicarbonate result significant effet during 1500rn running in the variables such as heart rate, venous pH, HCO???? and blood lactate concentration? Record of 1500m running and heart rate before and after the sodium bicarbonate ingestion was not shown a significant effect, but it was proved a significant effect, in case after sodium bicarbonate ingestion of venous pH and HC0?? Also, blood lactate concentration was displayed a significantly increased ievels after sodium bicarbonate treatment, compared with sodium bicarbonate treatment before. Conclusively, Treatment of sodium bicarbonate as ergogenic aids results in increasing a alklosis inducing, and a physiological effect of venous pH, HCO???? and blood lactate concentration, but it was not improved a record of 1500m running. We concluded that NaHCO?? ingestion for improvement, of performance was closely related to factors such as total dosage of NaHCO?? and exercise protocol.

        Activator 와 Anterior high pull headgear를 이용한 골격성Ⅱ급 부정교합의 치험례

        저자는 전남대학병원 소아치과에 내원한 혼합 치열기 Ⅱ급 부정교합환아를 Activator와 Anterior high pull headgear로 치료하여 다음과 같은 결론을 얻었다. 1. 상악골의 전,하방 성장을 억제하였다. 2. 하악골의 전방 전위 및 반시계방향 회전을 유도하였다. 3. Deep overbite와 large overjet이 개선되었다. This article describes the use of an activator with anterior high pull headgear to treat a skeletal Class Ⅱ malocclusion in children in the mixed-dentition phase. A combination of headgearactivator appliance can inhibit forward and downward growth of the maxillary complex while stimulating mandibular growth. The correction of Class Ⅱ malocclusion can be achieved by careful case selection of a motivated patient with a favorable growth pattern. The patients who have skeletal Class Ⅱ malocclusions were treated by means of activator with anterior high pull geadgear and the followintg results were observed; 1. Forword and downward growth of the maxillary complex were inhibited 2. Mandibualr growth was stimulated and counterclockwise rotation of the mandible was observed. 3. Large overjet and deep overbite were corrected.

        데페록사민 전처치가 토끼 심근경색 크기의 감소에 미치는 효과

        Background: Reperfusion of ischemic myocardium has been postulated to result in a specific oxygen radical mediated tissue injury. Iron may liberate during ischemia and we hypothesized that administration of the iron chelator, deferoxamine during ischemia would result in improved recovery after postischemic reperfusion. Purpose: To test whether iron-catalyzed processes contribute to myocardial necrosis during ischemia and reperfusion, deferoxamine was administered to block iron catalyzed hydroxyl radical formation in rabbits. Methods: Eleven rabbits were divided into two groups : control group (n=5) and deferoxamine pretreatment group (n=6). The left circumflex coronay artery was ligated for 30 minutes and reperfused for 180 minutes. Area at risk (AR) was measured by non-stained area with methylene blue injection into left atrium after left circumflex coronary artery ligation. Infarct size was measured by weighing after triphenyltetrazolium chloride staining. Heart rate was measured using electrocardiographic recording and systemic blood pressure was monitored by pressure transducer connected to the catheter in the left ventricle. Results: 1. There was no significant difference of heart rate and blood pressure in deferoxamine pretreatment group compared with control group. 2. There was significant decrease of serum iron concentration after continuous infusion of deferoxamine compared with serum iron concentration before ligation of coronary artery(P<0.05). 3. There was no significant difference of area at risk between control and deferoxamine pretreatment group. 4. Area at necrosis to area at risk was significantly reduced in deferoxamine pretreatment group compared with control group(P<0.05). The results suggest that deferoxamine infusion prior to coronary artery occlusion has a significant benefit in reducing infarct size in this model.

        정상교합자와 골격성 Ⅲ급 부정교합자의 경추골성숙도에 관한 연구

        정상교합자와 골격성 Ⅲ급 부정교합자간의 성장 정도에 차이가 있는지를 평가하기 위해 8-12세 여자를 대상으로 하여 진단 석고 모형과 측모두부방사선사진을 기준으로 정상교합자 172명 , 골격성 Ⅲ급 부정교합자 191명 총 363명을 분류하여 연구를 시행하였다. 경추의 하연의 만곡도와 수직·수평 길이의 비를 측정해서 연령이 증가함에 따른 변화를 분석하여 다음과 같은 결론을 얻었다. 1. 연령이 증가함에 따라 제2~6경추의 하연의 만곡도는 정상교합자와 골격성 Ⅲ급 부정교합자 모두에서 일정하게 증가하였다. 2. 연령이 증가함에 따라 제3~6경추의 수직·수평 길이의 비도 정상교합자와 골격성 Ⅲ급 부정교합자 모두에서 일정하게 증가하였다. 3. 제2~6경추의 하연의 만곡도와 제3~6경추의 수직·수평 길이의 비에서 정상교합자와 골격성 Ⅲ급 부정교합자간의 경추골성숙도 차이를 조사한 결과 통계적으로 유의한 차이를 보이지 않았다. 이상의 결과에서 정상교합자와 골격성 Ⅲ급 부정교합자간의 경추골성숙도를 비교했을 때 유의한 차이를 보이지 않았으므로 성장에 차이가 없음을 시사하였다. This study was to evaluate and compare differences of the cervical vertebral skeletal maturity of normal occlusion and skeletal Class Ⅲ malocclusion. Normal occlusion (172 girls) and skeletal Class Ⅲ malocclusion(191 girls) were classified according to diagnosis stone model and lateral cephalogram of Korean girls aging from 8 to 12 years. The concavity of inferior border, vertico-horizontal ratio of cervical vertebrae were observed and measured according to age. Differences of the cervical vertebral skeletal maturity were evaluated. The results were as follows : 1. The concavity of inferior border of the 2nd to 6th vertebrae of normal occlusion and skeletal Class Ⅲ had uniformly increased with age. 2. The vertico-horizontal ratio of the 3rd to 6th vertebrae of girls with normal occlusion and skeletal Class Ⅲ had uniformly increased with age. 3. There was no significant difference in cervical vertebral skeletal maturity between normal occlusion and skeletal Class Ⅲ malocclusion in the concavity of inferior border of the 2nd to 6th vertebrae and in the vertico-horizontal ratio of the 3rd to 6th vertebrae. The results in the study indicate that there is no significant difference of cervical vertebral skeletal maturity between girls with normal occlusion and skeletal Class Ⅲ malocclusion.

        Air Rotor Stripping with Essix Anterior Anchor를 이용한 교정치료

        Air turbine handpiece를 사용하여 치간 법량질을 삭제하는 Air-rotor stripping 술식은 악궁확장이나 발치 술식에 대한 대체방안으로써 arch length discrepancy를 해결할 수 있는 효과적인 접근법으로 시료된다. Conventional interproximal stripping using abrasive strips is normally limited to the anterior teeth. The strips must be forced between the contact points of the teeth creating patient discomfort and the risk of cutting gingival tissue. Air-rotor stripping (ARS) with Essix anterior anchor enables the clinician to remove a precise amount of interproximal enamel to create space, primarily in the buccal quadrants, for aligning or retracting teeth. In selected cases, ARS can resolve significant differences in ratios of tooth size to arch length, and the technique can become an alternative to extraction or expansion. ARS can create substantially more space than that is usually obtained by conventional interproximal stripping, and it can be done at any time during treatment without discomfort to the patient and without adversely affecting the function of the dentition, interocclusal relations, or tooth form.

      • 이유백서에서 교근절제후 하악과두의 면역조직화학적 연구

        The purpose of this study was to determine whether there was any correlation between temporomandibular joint dysfunction and structure of the mandibular condyle. Weanling rats had their masseter muscles resected and immunohistochemical findings were observed a light microscope. The results obtained were as follows : 1. The condylar cartilage region was divided into articular, proliferating, cartilage cell and hypertrophic cell layers according to cell morphology. 2. In light microscopic views, the proliferating and cartilage cell layers of the experimental group decreased gradually and at the 8the week significantly. 3. In immunohistochemical staining for type Ⅰ and Ⅱ collagen, a reaction was detected in the lower part of proliferating cell and cartilage cell layers. In the acrtilage cell layers, a stronger cellular reaction was present. Immunohistochemical staining for type Ⅱ collagen reacted more strongly than that of type Ⅰ collagen. 4. In immunohistochemical staining for proteoglycan, the staining of the experimental group resembled the control group and gradually showed a weak reaction. The proliferating and cartilage cell layers reacted more strongly than the hypertrophic cell layer. 5. In immunohistochemical staining for proliferating cell nuclear antigen(PCNA), the strong reaction was detected in the nucleus of the proliferating cell layer both in control and experimental groups. But the thickness of the proliferating layer decreased in experimental group, consequently the reaction of the experimental group was reduced more than that of the control group.

        Er:YAG 레이저를 이용한 치아삭제 효과

        Er:YAG 레이저는 파장이 물의 주흡수 정점과 일치하는 2.94㎛의 방출파장을 가지기 때문에, 물분사를 하면서 사용했을 때, 다른 레이저보다 열손상은 더 작으면서 치아경조직을 보다 효과적으로 삭제한다고 알려져 있다. Er:YAG 레이저의 상아질 삭제 효과를 평가하고자 발거된 소구치와 대구치를 대상으로 상아질 시편을 제작하고 2.94㎛의 pulsed Er:YAG 레이저 (SDL-300EN, 삼성전자, 한국)를 handpiece형의 전달계를 이용한 비접촉식 방법으로, 물에 담궈진 시간과 레이저 조사시간에 따른 삭제량과 형태를 조사, 분석하여 다음과 같은 결과를 얻었다. 1. 건전 상아질에 대한 조사 결과 물에 담군 시간이 증가할수록 삭제율이 증가 하였다. 2. 건전 상아질과 우식 상아질에 150mJ, 10Hz의 조건으로, 30초간 레이저를 조사했을 때, 우식상아질에 비해 건전 상아질에서 surface irregularity가 증가하고 상아세관이 열리는 경향이 있었으며 crack이 관찰되었다. 또한 우식 상아질에 비해 많은 탄조각(ash flecks), crack이 관찰되었고 상아세관의 불연속성(inconsistency)이 관찰 되었다. 3. 우식 상아질에서 30초로 조사한 경우에서는 dark zone이 와동에 국한되어 나타났으며 작은 fleck들이 관찰되었고, 1분간 조사한 경우에서는 30초의 경우보다도 와동이 더 넓었으며, crack이 상아세관 주행방향과 직각으로 형성된 것을 관찰 할 수 있었다. 2분간 조사한 경우는 와동이 가장 넓었으며 crack이 더 많이 형성된 것을 볼 수 있었다. The purpose of this study was to evaluate the effects of Er:YAG laser on cutting efficacy and its histologic findings. Dentin specimens of human premolars and molars were used and irradiated by Er:YAG laser with noncontact handpiece type delivery system under different treatment condition of irradiation tiem. Cavity pattern and volume were evaluated to determine the cutting efficacy and following results were obtained. 1. Cutting volume of sound dentin was getting larger with time immersed in water increase 2. With the condition of irradiation (150mJ, 10Hz, 30sec), surface irregularity was more increased in sound dentin comparing to carious dentin. For the light microscopic examination, dentinal tubules were opened and ash flecks and cracks were noted with inconsistence of dentinal tubules. 3. In case of 30 sec, irradiation in carious dentin, dark zone was limited to small focus whereas 1 min, irradiation, more wider, and cracks were noted in the perpendicular to direction of dentinal tubules. For the 2 min, irradiation, cavity was the widest and more cracks were found.

        복합레진은 치질과 우수한 결합력을 가지며 심미적인 이유로 치과의 대표적 수복재로 자리잡고 있다. 그러나 저작력에 따른 마모와 구강내 환경에서의 수분흡수에 따른 가수분해가 복합레진의 강도약화를 야기하고 있다. 실험에 사용된 복합레진은 현재 임상에서 많이 사용되고 있는 Point4, Tetric flow, Heliomolar flow, FiItek supreme 이다. 각 제품의 화학적 분해와 마모도를 평가하기 위해 각각 10개의 시편을 제작하여 그 중 5개는 37℃의 수중하에 1Kg의 하중을 가하여 10,000 cycle동안의 마모도를 측정하였다. 나머지 5개 시편은 무게 측정을 한 후, 0.1N NaOH 용액, 3ml에 저장하여 60℃에서 보관한 후 2주 후 제거하여 1.23% HCI로 2시간 동안 중화하고 증류수로 세척한 후 60℃에서 건조하였다. 무게 손실, 분해층 깊이 및 시편내의 Si의 농도 변화, 최대 마모 깊이를 근거로 평가하여 다음과 같은 결과를 얻었다. 1. 각 제품의 무게 손실량은 Heliomolar flow, Filtek supreme, Point4, Tetric flow 순이었으며 Heliomolar flow, Filtek supreme을 제외한 다른 제품간에는 유의한 차이를 보였다(P<0.05). 2. 각 제품의 표면하 분해층 깊이는 Filtek supreme, Heliomolar flow, Tetric flow, Point4 순이었으며 Heliomolar flow, Tetric flow을 제외한 다른 제품간에는 유의한 차이를 보였다(P<0.05). 3. 각 제품으로부터 용출된 Si은 Filtek supreme, Heliomolar flow, Point4, Tetric flow 순이었으며 Point4, Tetric flow을 제외한 다른 제품간에는 유의한 차이를 보였다(P<0.05). 4. 최대마모깊이 측정시 Heliomolar flow, Point4, Filtek supreme, Tetric flow 순이며 0.1 N NaOH 용액에 보관한 레진의 마모깊이가 물에 보관한 레진에 비해 증가한 양상을 보였다. 5. 주사전자현미경 관찰시 NaOH 용액에 보관한 후 레진 기질과 충전제 사이의 결합의 파괴를 관찰 수 있었으며, 공촛점 레이저 현미경 관찰시 NaOH 용액에 보관한 후 수복재의 기질과 충전제 사이의 파괴양상인 분해층 깊이를 관찰할 수 있었다. The aim of this study was to evaluate the resistance to degradation and to compare the wear resistance characteristics of four composite resins in an alkaline solution. The resistance to degradation was evaluated on the basis of mass loss(%), degradation depth(㎛). Si loss(ppm) and wear depth. The brands studied were Heliomolar flow, Filtek supreme. Point4, Tetric flow. The results were as follows: 1. The sequence of the mass loss was in descending order by Heliomolar flow, Filtek supreme, Point4, Tetric flow. There was significant differences among the materials except Heliomolar flow and Filtek supreme. 2. The sequence of the degree of degradation layer depth was in descending order by Filtek supreme, Heliomolar flow, Tetric flow, Point4. There were significant differences among the materials except Heliomolar flow and Tetric flow. 3. The sequence of Si loss was in descending order by Filtek supreme, Heliomolar flow, Point4, Tetric flow. There were significant differences among the materials except Point 4 and Tetric flow. 4. The sequence of maximum wear depth was in descending order by Heliomolar flow, Point4, Filtek supreme, Tetric flow and there was increasing wear depth on soaking in 0.1N NaOH solution. 5. When observed with SEM, destruction of bonding between matrix and filler was observed and when observed with CLSM, the depth of degradation layer of specimen surface was observed. There results indicate that wear and hydrolytic degradation could be considered to be evaluation factors of composite resins.
