        Authentication of an Endangered Herb Changium smyrnioides from Different Producing Areas Based on rDNA ITS Sequences and Allele- Specific PCR

        The rDNA ITS region of 18 samples of Changium smyrnioides from 7 areas and of 2 samples of Chuanminshen violaceum were sequenced and analyzed. The amplified ITS region of the samples, including a partial sequence of ITS1 and complete sequences of 5.8S and ITS2, had a total length of 555 bp. After complete alignment, there were 49 variable sites, of which 45 were informative,when gaps were treated as missing data. Samples of C. smyrnioides from different locations could be identified exactly based on the variable sites. The maximum parsimony (MP) and neighbor joining (NJ) tree constructed from the ITS sequences based on Kumar’s two-parameter model showed that the genetic distances of the C. smyrnioides samples from different locations were not always related to their geographical distances. A specific primer set for Allele-specific PCR authentication of C. violaceum from Jurong of Jiangsu was designed based on the SNP in the ITS sequence alignment. C. violaceum from the major genuine producing area in Jurong of Jiangsu could be identified exactly and quickly by Allele-specific PCR.

        Design of Smart City Considering Carbon Emissions under The Background of Industry 5.0

        Industry 5.0 puts forward higher requirements for smart cities, including low-carbon, sustainable, and people-oriented, which pose challenges to the design of smart cities. In response to the above challenges, this study introduces the cyber-physical-social system (CPSS) and parallel system theory into the design of smart cities, and constructs a smart city framework based on parallel system theory. On this basis, in order to enhance the security of smart cities, a sustainable patrol subsystem for smart cities has been established. The intelligent patrol system uses a drone platform, and the trajectory planning of the drone is a key problem that needs to be solved. Therefore, a mathematical model was established that considers various objectives, including minimizing carbon emissions, minimizing noise impact, and maximizing coverage area, while also taking into account the flight performance constraints of drones. In addition, an improved metaheuristic algorithm based on ant colony optimization (ACO) algorithm was designed for trajectory planning of patrol drones. Finally, a digital environmental map was established based on real urban scenes and simulation experiments were conducted. The results show that compared with the other three metaheuristic algorithms, the algorithm designed in this study has the best performance.

        Ultrasonic/microwave synergistic synthesis of well-dispersed hierarchical zeolite Y with improved alkylation catalytic activity

        Well-dispersed hierarchical zeolite Y, structure-directed by ionic liquid 1-methyl-3-[3'-(trimethoxysilyl) propyl] imidazolium chloride, was synthesized for the first time using a novel ultrasonic/microwave synergistic synthesis (UMSS). The time required for the synthesis of zeolite Y by UMSS method was remarkably reduced to 6 h instead of 24 h under conventional hydrothermal conditions. The structures of all samples were characterized by XRD, BET, FT-IR, SEM and TEM. The results clearly demonstrated that the UMSS is a promising strategy to achieve zeolite Y with improved crystallinity, large BET surface area, bulky mesopore volume, well-dispersed morphology, small zeolite nanoparticle. The catalytic activity of hierarchically micro-mesoporous zeolite Y was evaluated by using the alkylation of o-xylene with styrene. Mesoporous zeolite Y synthesized by UMSS method showed significantly higher catalytic activity, stability and reusability, compared with conventional hydrothermal synthesis. It might be ascribed to its large mesoporous volume, small crystal nanoparticle and high surface area, minimizing the diffusion length as the reactant molecules diffuse into the pores, and correspondingly the products diffuse out of zeolites. This study suggests that UMSS method is a good alternative for the synthesis of micro-mesoporous zeolite Y, which may be of remarkable benefit for industrial applications.

        Effects of gear eccentricity on time-varying mesh stiffness and dynamic behavior of a two-stage gear system

        Gear eccentricity, one of the most common defects of gear systems, affects not only dynamic behavior but also mesh stiffness. Accurate mesh stiffness under defect conditions is vital for the dynamic simulation and fault diagnosis of gear systems. Hence, the timevarying mesh stiffness affected by gear eccentricity, which is commonly neglected in most studies, is incorporated in the proposed twostage spur gear dynamic model and calculated by a novel gear mesh model with an improved potential energy method. The effects of mesh stiffness with and without gear eccentricity on the gear system are compared on the basis of the constructed dynamic model. The influences of gear eccentricity on dynamic transverse and torsional responses, as well as dynamic transmission errors, are also quantitatively studied. Some helpful analytical results are then presented.

        Effective adsorption of phenols using nitrogen-containing porous activated carbon prepared from sunflower plates

        Nitrogen-containing porous carbons, the 800SP-NH3, were synthesized using sunflower plates as the major carbon source carbonized at 800 oC and activated with concentrated aqueous ammonia at the same temperature. The porous carbons were characterized by nitrogen physical adsorption-desorption, surface area analyzer, FT-IR, and SEM. The adsorption properties of the porous carbons towards phenols were also investigated by batch methods. The test results show that the average pore diameter of porous carbon is smaller than 2 nm, and nitrogen-containing chemical groups are formed on its surface. The adsorption capacity for phenol, 4-chlorophenol, and p-nitrophenol is 316.5mg/g, 330.24mg/g and 387.62mg/g due to its developed pore structure and nitrogen-containing chemical groups. The adsorption isotherm data greatly obey the Langmuir model.

        Multi-objective optimum design of fast tool servo based on improved differential evolution algorithm

        The flexure-based mechanism is a promising realization of fast tool servo (FTS), and the optimum determination of flexure hinge parameters is one of the most important elements in the FTS design. This paper presents a multi-objective optimization approach to optimizing the dimension and position parameters of the flexure-based mechanism, which is based on the improved differential evolution algorithm embedding chaos and nonlinear simulated anneal algorithm. The results of optimum design show that the proposed algorithm has excellent performance and a well-balanced compromise is made between two conflicting objectives, the stroke and natural frequency of the FTS mechanism. The validation tests based on finite element analysis (FEA) show good agreement with the results obtained by using the proposed theoretical algorithm of this paper. Finally, a series of experimental tests are conducted to validate the design process and assess the performance of the FTS mechanism. The designed FTS reaches up to a stroke of 10.25 μm with at least 2 kHz bandwidth. Both of the FEA and experimental results demonstrate that the parameters of the flexure-based mechanism determined by the proposed approaches can achieve the specified performance and the proposed approach is suitable for the optimum design of FTS mechanism and of excellent performances.

        Piperine increases striatal levels of DA and TH and decreases α-syn and Aβ42 deposition in PDD mice by regulting autophagy: downexpression Beclin-1 and LC3B and upexpression p62

        Piperine, the major pharmacological ingredient of pepper, can delay the procession of neuropharmacological effects, but its effects and mechanisms on Parkinson’s disease dementia (PDD) mice is still unclear, we investigated whether piperine could help treating PDD mice. Here, PDD mice were randomly divided into eight groups (n = 12/group): a normal control group, a PDD model group, a madopar group, an autophagy inhibitor group, an autophagy activator group, and groups receiving low, medium or high doses of piperine respectively. The normal control and PDD model mice were injected with saline. Treatments were administered to the mice once per day continuously for 30 days. The behavioral tests were assessed. Dopamine (DA), Monoamine Oxidase-B (MAO-B), DOPA decarboxylase (DDC), β-secretase, acetylcholinesterase (AChE), amyloid β42 (Aβ42), tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) levels were detected. α-synuclein (α-syn), tyrosine hydroxylase (TH), HSP90, Beclin-1, LC3B, p62 mRNA levels and miRNA-99a-5p expression were determined. Neuronal histology was observed. The behavior of PDD mice improved significantly after peperine treatment compared with the PDD model mice. In addition, our results also showed that peperine treatment increased DA, TH, DDC and p62 levels, decreased MAOB, β-secretase, AChE, Aβ42, TNF-α, IL-6, Beclin-1 and LC3B levels, and down-regulated α-syn, HSP90, Beclin-1, LC3B mRNA levels and miR-99a-5p expression. These findings suggest that piperine may reduce the expression of mmu-miR-99a-5p and autophagy-related factors (HSP90, Beclin-1, LC3B and p62) to alleviate the neurological impairment of PDD mice, which is shown to slow down the process of DA metabolism and Aβ production and resist neuroinflammation.         RPTOR methylation in the peripheral blood and breast cancer in the Chinese population

        Background: Altered regulatory-associated protein of mTOR, complex 1 (RPTOR) methylation levels in peripheral blood was originally discovered as breast cancer (BC)-associated risk factor in Caucasians. Objective: To explore the relationship between RPTOR methylation and BC in the Chinese population, we conducted two independent case-control studies. Methods: Peripheral blood samples were collected from a total of 333 sporadic BC cases and 378 healthy female controls for the DNA extraction and bisulfite-specific PCR amplification. Mass spectrometry was applied to quantitatively measure the levels of methylation. The logistic regression, Spearman's rank correlation, and Non-parametric tests were used for the statistical analyses. Results: In our study, we found an association between BC and RPTOR_CpG_4 hypomethylation in the general population (per-10% of methylation, OR 1.29, P = 0.012), and a weak association between BC and RPTOR_CpG_8 hypomethylation in the women with older age (per-10% of methylation, OR 2.34, P = 0.006). We also identified age as a confounder for the change of RPTOR methylation patterns, especially at RPTOR_CpG_4, which represented differential methylation comparing age groups especially in the BC cases (age < 50 years vs age ≥ 50 years by Mann-Whitney U test, P < 0.0001 for BC cases and P = 0.079 for controls). Conclusion: Our study validated the association between hypomethylation of RPTOR and BC risk in the Chinese population also with weak effect and mostly for postmenopausal women. In addition, our findings provided novel insight for the regulation of DNA methylation upon aging or the change of hormone levels.

        Non-resonant 3D Elliptical Vibration Cutting Induced Submicron Grating Coloring

        Surface Submicron grating manufacturing has broad application prospects, which has also been the subject of intensive research and development by scholars with many eff orts. In this paper, a grating coloring manufacturing method is proposed based on three-dimensional elliptical vibration cutting and non-resonant piezoelectric actuator. Based on the mechanism of elliptical motion, an effective cutting depth model was established. Nonlinear interaction of machining parameters (overlap rate, nominal cutting speed, vibration frequency) on the geometry of the grating were analyzed and controllable modulation model of grating geometry was established. The geometrical dimensions of the grating were predicted and grooved experiments were performed on brass and aluminum surfaces. A set of angle-adjustable detection device was designed independently to check the diffraction quality of the grating. The quantitative experimental results show that grating distances of copper and aluminum rods are 752 nm and 684 nm, respectively. The average width of the aluminum rod overall grating is 764 nm, the accuracy error is 9.25%. In addition, aluminum rod gratings can diffractred light at non-specific angles, which is more effective at viewing angle of 70°. The experimental results are in agreement with the theory which have positive significance for expanding the functional surface.
