Steel Bridge Construction of Hong Kong–Zhuhai–Macao Bridge

The 55-km-long Hong Kong–Zhuhai–Macao Bridge (HZMB) is the world’s longest sea-crossing bridge, connecting Hong Kong with Zhuhai and Macao at the mouth of the Pearl River Estuary in China, comprising 22.9-km-long steel bridges. HZMB is the leading steel bridge in China, with top-level manufacturing and installation technology. This paper outlines the steel bridge construction experiences of HZMB to provide comparisons for the construction of other long sea-crossing steel bridges at home or abroad. The main considerations of construction constraints, scheme selection, structural and aesthetic design of HZMB are presented, and the following points related to new strategies in the steel bridge construction of HZMB are elaborated: (1) construction quality assurance, (2) automatic manufacturing technology, (3) large segment off shore installation, (4) eco-friendly paint (content limitation of volatile organic compounds) and new multifunctional inspection gantry, and (5) Guss Mastic Asphalt steel deck pavement system. The successful implementation of those strategies shows that the steel bridge construction of HZMB promotes improvement in the overall construction and management level of the Chinese bridge industry. The advanced experience of HZMB has opened up broad prospects for the design and construction of off shore bridge engineering in China.
Bacterial Fruit Rot of Apricot Caused by Burkholderia cepacia in China

An unreported disease of apricot was observed in orchards in Zhejiang province, China. Symptoms started as water soaked lesions on the fruit surface. Later, water-soaked areas developed and spread to the entire fruit, resulting in soft rot of the whole fruit. The causal organism isolated from symptomatic fruits was identified as Burkholderia cepacia based on its biochemical and physiological characteristics and confirmed by the cellular fatty acid composition and Biolog data as well as 16S rRNA gene sequence analysis. The bacterial isolates caused similar symptoms when inoculated onto fruits of apricot. In addition, European plum, Japanese plum, nectarine and kiwifruit were susceptible to the B. cepacia pathogen. However, the B. cepacia pathogen failed to cause any visible symptoms when it was inoculated onto 16 other fruits. This is the first report of a bacterial disease of apricot caused by B. cepacia in China.

MicroRNAs (miRNAs), a class of small non-coding RNAs, mediate gene expression by either cleaving target mRNAs or inhibiting their translation. They have key roles in the tumorigenesis of several cancers, including non-small cell lung cancer (NSCLC). The aim of this study was to investigate the clinical significance of miR-638 in the evaluation of NSCLC patient prognosis in response to chemotherapy. First, we detected miR-638 expression levels in vitro in the culture supernatants of the NSCLC cell line SPC-A1 treated with cisplatin, as well as the apoptosis rates of SPC-A1. Second, serum miR-638 expression levels were detected in vivo by using nude mice xenograft models bearing SPC-A1 with and without cisplatin treatment. In the clinic, the serum miR-638 levels of 200 cases of NSCLC patients before and after chemotherapy were determined by quantitative real-time PCR, and the associations of clinicopathological features with miR-638 expression patterns after chemotherapy were analyzed. Our data helped in demonstrating that cisplatin induced apoptosis of the SPC-A1 cells in a dose- and time-dependent manner accompanied by increased miR-638 expression levels in the culture supernatants. In vivo data further revealed that cisplatin induced miR-638 upregulation in the serum derived from mice xenograft models, and in NSCLC patient sera, miR-638 expression patterns after chemotherapy significantly correlated with lymph node metastasis. Moreover, survival analyses revealed that patients who had increased miR-638 levels after chemotherapy showed significantly longer survival time than those who had decreased miR-638 levels. Our findings suggest that serum miR-638 levels are associated with the survival of NSCLC patients and may be considered a potential independent predictor for NSCLC prognosis.

Alu Tandem Sequences Inhibit GFP Gene Expression by Triggering Chromatin Wrapping

Alu elements belonging to the short interspersed nuclear elements (SINE) of repetitive elements are present in more than one million copies which altogether represent 10% of the whole human genome. In this study, the roles of Alu tandem sequences in the process of GFP gene (GFP) expression and packing into chromatin of its DNA were studied. To detect the effect of Alu repeats on gene expression, different copies of Alus were inserted GFP downstream respectively in pEGFP-C1 vector. We found that Alu sequences decreased the amount of GFP transcription, the percentage of GFP positive cells and the accessibility to DNase I in length-dependent manner. Inserting Alu caused the production of higher-molecular-mass RNA, indicating Alu sequence did not induce premature transcriptional termination. Tight packing chromatins keep silent and resist to DNase I digestion, which is a general phenomenon. We suggested that head and tail tandem Alu sequences suppressed GFP expression in length dependent manner by triggering chromatin packing.

In this paper, an adaptive controller with structurally dynamic wavelet network is developed for a harmonic drive subject to parameter varying friction. The control architecture integrates a proportional controller, a feedback adaptive component and sliding component to adaptively compensate for the friction to achieve accurate position tracking. Global asymptotic stability of the algorithm is proved by using Lyapunov function. In parallel to the adaptive controller, a fuzzy reconfiguration scheme is devised to change the structure of the network along with weights updating to improve the system tracking performance and robustness. Experimental tests on a harmonic drive manipulator verify the effectiveness of the proposed control method.
Visual Servoing Using an Optimized Trajectory Planning Technique for a 4 DOFs Robotic Manipulator

Although visual servoing has been considered as a solution to increase dexterity and intelligence ofthe robotic systems specially in unstructured environments, some prominent deficiencies are preventing it frompractical employment. Trajectory planning is a solution to overcome the shortcomings of visual servoing andmakes it practical for industrial applications. In this paper, a new trajectory planning technique is developed toperform image-based visual servoing (IBVS) tasks for a 4 DOFs robotic manipulator system. In this method, thecamera’s velocity screw is parameterized using time-based profiles. The parameters of the velocity profile are thendetermined such that the velocity profile takes the robot to its desired position. This is done by minimizing theerrors between the initial and desired features. A depth estimation technique is proposed to provide the trajectoryplanning algorithm with an accurate initial depth. This algorithm is tested and validated via the experiment on a4 DOFs Denso robot in an eye-in-hand configuration. Experimental results demonstrate that the proposed methodprovides with a reliable visual servoing algorithm by overcoming the IBVS drawbacks such as surpassing the systemlimits and causing instability of the system in fulfilling the tasks which require a 180o rotation of the camera aboutits center.
Image-based Visual Servoing using Improved Image Moments in 6-DOF Robot Systems

This paper addresses the challenges of choosing proper image features for planar symmetric shape objects and designing visual servoing controller to enhance the tracking performance in image-based visual servoing (IBVS). Six image moments are chosen as the image features and the analytical image interaction matrix related to the image features are derived. A controller is designed to efficiently increase the robustness of the visual servoing system. Experimental results on a 6-DOF robot visual servoing system are provided to illustrate the effectiveness of the proposed method.
Adaptive Switch Image-based Visual Servoing for Industrial Robots

In this paper, an adaptive switch image-based visual servoing (IBVS) controller for industrial robots is presented. The proposed control algorithm decouples the rotational and translational camera motions and decomposes the IBVS control into three separate stages with different gains. This method can increase the system response speed and improve the tracking performance of IBVS while the proposed adaptive law deals with the uncertainties of the monocular camera in eye-in-hand configuration. The stability of the designed controller is proved using Lyapunov method. Experimental results on a 6 degree of freedom (DOF) robot show the significant enhancement of the control performance over other IBVS methods, in terms of the response time and tracking performance. Also the designed visual servoing controller demonstrates its capability to overcome some of the inherent drawbacks of IBVS, such its inability to perform a 180◦ camera rotation around its center.
