Mitochondrial dysfunction and calcium deregulation by the RanBP9-cofilin pathway

<P>Mitochondrial dysfunction and synaptic damage are important features of Alzheimer's disease (AD) associated with amyloid β (Aβ) and tau. We reported previously that the scaffolding protein RanBP9, which is overall increased in brains of patients with AD and in mutant APP transgenic mice, simultaneously promotes Aβ generation and focal adhesion disruption by accelerating the endocytosis of APP and β1-integrin, respectively. Moreover, RanBP9 induces neurodegeneration <I>in vitro</I> and <I>in vivo</I> and mediates Aβ-induced neurotoxicity. Here we show in primary hippocampal neurons that RanBP9 potentiates Aβ-induced reactive oxygen species (ROS) overproduction, apoptosis, and calcium deregulation. Analyses of calcium-handling measures demonstrate that RanBP9 selectively delays the clearance of cytosolic Ca2+ mediated by the mitochondrial calcium uniporter through a process involving the translocation of cofilin into mitochondria and oxidative mechanisms. Further, RanBP9 retards the anterograde axonal transport of mitochondria in primary neurons and decreases synaptic mitochondrial activity in brain. These data indicate that RanBP9, cofilin, and Aβ mimic and potentiate each other to produce mitochondrial dysfunction, ROS overproduction, and calcium deregulation, which leads to neurodegenerative changes reminiscent of those seen in AD.—Roh. <P>The generation of silver nanoparticle/bis(o-phenolpropyl)silicone composites have been facilitated by the addition of sodium tetrachloroaurate or gold(Ill) chloride (< 1 wt% of NaAuCl4 or AuCl3) to the reaction of silver nitrate (AgNO3) with bis(o-phenolpropyl)silicone [BPPS, (o-phenolpropyl)2(SiMe2O)n, n = 2,3,8,236]. TEM and FE-SEM data showed that the silver nanoparticles having the size of < 20 nm are well dispersed throughout the BPPS silicone matrix in the composites. XRD patterns are consistent with those for polycrystalline silver. The size of silver nanoparticles augmented with increasing the relative molar concentration of AgNO3 added with respect to BPPS. The addition of gold complexes (1-3 wt%) did not affect the size distribution of silver nanoparticles appreciably. In the absence of BPPS, the macroscopic precipitation of silver by agglomeration, indicating that BPPS is necessary to stabilize the silver nanoparticles surrounded by coordination.</P>
TWO DISTINCT RED GIANT BRANCHES IN THE GLOBULAR CLUSTER NGC 288

<P>We report the presence of two distinct red giant branches (RGBs) in the globular cluster NGC 288 from the narrowband calcium and Stromgren b and y photometry obtained at the CTIO 4 m Blanco telescope. The RGB of NGC 288 is clearly split into two in the hk [=(Ca - b) - (b - y)] index, while the split is not shown in the b - y color. Unlike other globular clusters with multiple populations reported thus far, the horizontal branch of NGC 288 is only mildly extended. Our stellar population models show that this and the presence of two distinct RGBs in NGC 288 can be reproduced if slightly metal-rich (Delta[m/H] approximate to 0.16) second generation stars are also enhanced in helium by small amount (Delta Y approximate to 0.03) and younger by similar to 1.5 Gyr. The RGB split in the hk index is most likely indicating that the second generation stars were affected by supernovae enrichment, together with the pollution of lighter elements by intermediate-mass asymptotic giant branch stars or fast-rotating massive stars. In order to confirm this, however, spectroscopy of stars in the two distinct RGB groups is urgently required.</P>

<B>ABSTRACT</B><P>During a previous study on the molecular interaction between commensal bacteria and host gut immunity, two novel bacterial strains, A911T and G707T, were isolated from the gut of <I>Drosophila melanogaster</I>. In this study, these strains were characterized in a polyphasic taxonomic study using phenotypic, genetic, and chemotaxonomic analyses. We show that the strains represent novel species in the family <I>Acetobacteraceae</I>. Strain G707T, a highly pathogenic organism, represents a new species in the genus <I>Gluconobacter</I>, “<I>Gluconobacter morbifer</I>” sp. nov. (type strain G707 = KCTC 22116T = JCM 15512T). Strain A911T, dominantly present in the normal <I>Drosphila</I> gut community, represents a novel genus and species, designated “<I>Commensalibacter intestini</I>” gen. nov., sp. nov. (type strain A911 = KCTC 22117T = JCM 15511T).</P>

<P>The purpose of this study is to assess greenhouse gas (GHG) emission reduction performance among certification criteria constituting “materials and resources” category, as a part of study that configures weighted values of categories for the Korea Green Building Scheme (G-SEED). For this purpose, the subjects of assessment were classified into “reference apartment house” newly constructed without application of certification criteria and “assessed apartment house” newly constructed with reflection of certification criteria. In addition, direct and indirect GHG emission reduction performance of the assessed apartment house was assessed from life cycle perspective in comparison to the reference apartment house, reflecting the purpose of assessment according to certification criteria within the “materials and resources” category. As a result, GHG emission reduction performance of “displaying carbon emission of material” and “recycling” were deduced to be extremely excellent. GHG emission reduction performance of “flexibility” and “reducing food waste” ware analyzed to be relatively satisfactory. On one hand, GHG emission reduction performance of “appropriateness of measure to reduce furniture materials” and “use of eco-friendly products” ware assessed to be relatively insufficient.</P>

Aims: Laparoscopic liver resection has evolved and widened in its scope. While open liver resections are currently being performed safely in our hospital, laparoscopic liver resections are being implemented in fewer cases. We review our early experience in laparoscopic liver resection to assess the early outcomes of the procedure. Methods: A retrospective chart review was conducted of 25 patients who underwent laparoscopic liver resections for various indications between January 2014 and July 2016 by single surgeon, who had performed 124 open liver resections (≥3 segments/< 3 segments: 91/33) at the same time. Results: There were 25 laparoscopic liver resections preformed. Male/female ratio: 15/10. Mean age: 64. 8 cases were wedge resections, left lateral sectionectomy: 7, left hepatectomy: 6, Right hepatectomy: 4. Pathology included Hepatocellular carcinoma: 12, cholangiocarcinoma: 2, Intrahepatic duct stones:5, metastatic liver carcinoma: 5, primary neuroendocrine tumor of liver:1. The mean operation time was 168.9min (40-410 minutes). Mean blood loss was 90.9 mL (10-180mL). There were no intraoperative and postoperative complications except for the following case. Severe portal vein stenosis in laparoscopic right hepatectomy occurred postoperatively, the patient underwent reoperation (portal vein resection and anastomosis, stenting). The mean hospital stay was 8.71 days (3-44). Conclusions: Even though our experience in laparoscopic liver resection is not enough, our results are comparable to those in other studies. Therefore, an experienced surgeon in open liver resection will be able to perform the laparoscopic liver resection safely.

Background/Aims: Recent data from Western populations have suggested that patients with sporadic duodenal adenomas are at a higher risk for the development of colorectal neoplasia. In this study, we compared the frequency of colorectal neoplasia in patients with sporadic duodenal adenomas to healthy control subjects. Methods: This retrospective case-control study used the databases of 3 teaching hospitals in Gyeonggi-do Province, South Korea. The colonoscopy fi ndings of patients with sporadic duodenal adenomas were compared with those of age- and gender-matched healthy individuals who had undergone gastroduodenoscopies and colonoscopies during general screening examinations. Results: Between 2001 and 2008, 45 patients were diagnosed endoscopically with sporadic duodenal adenomas; 26 (58%) of these patients received colonoscopies. Colorectal neoplasia (42% vs 21%; odds ratio [OR], 2.8; 95% confi dence interval [CI], 1.1 to 7.4) and advanced colorectal adenoma (19% vs 3%; OR, 9.0; 95% CI, 1.6 to 50.0) were signifi cantly more common in patients with sporadic duodenal adenomas than in healthy control subjects. Conclusions: Compared with healthy individuals, patients with sporadic duodenal adenomas were at a signifi cantly higher risk for developing colorectal neoplasia. Such at-risk patients should undergo routine screening colonoscopies. (Gut Liver 2011;5:432-436)
