Effect of Developmental Lead Exposure on the Expression of Hippocampal NMDA Receptor Subunit mRNA

목적: in vivo 및 vitro에서 해마 신경세포의 발생단계별 NMDA 수용체 아단위 mRNA 발현에 대한 연 폭로 영향을 알아보고자 하였다. 방법: 흰쥐 해마 신경세포의 발생단계별 NMDA 수용체 NR2A, NR2B 아단위 mRNA 발현에 대한 연의 영향은 정상군과 연 폭로군의 출생 후 7일, 14일, 22일 흰쥐의 해마에서 in situ hybridization으로 mRNA 발현 정도를 densitometer로 측정하여 비교하였고, 연과 NMDA 의 세포독성은 해마 신경세포 일차배양 후 도립현미경을 이용한 형태학적인 관찰과 LDH 활성도를 이용하여 측정하였다. 결과: 연 과 NMDA 에 의한 세포독성에 대한 in vitro 실험에서 형태학적 소견과 LDH 활성도에서 해마 미성숙 신경세포와 성숙 신경세포사이의 차이가 있었으므로, 연과 NMDA 독성효과는 해마 신경세포의 발달 단계에 따라 차이가 있다. 정상군의 해마에서의 NR2A mRNA 발현은 출생 후 연령이 증가함에 따라 점진적으로 증가하였으나, NR2B mRNA 발현은 출생 후 연령이 증가함에 따라 점진적으로 증가하였으나 NR2B mRNA 발현은 연령의 증가에 따른 변화가 없었다. 연 폭로에 희한 NR2A mRNA 발현은 유의하게 감소하였으나(p<0.05), NR2B mRNA 발현은 변화가 나타나지 않았다. 만성적 연 폭로는 NR2A를 포함하는 NMDA 수용체를 감소시킬 수 있음을 알 수 있다. 결론: 연은 해마신경세포의 발생단계에서 NMDA 수용체 아단위 특히 NR2A mRNA 발현의 변화를 야기하여 시냅스 신호 전달에 영향을 나타냄을 알 수 있었다. The up-to-date small and medium-sized enterprises (SMEs) in Korea have tried to respond flexibly and rapidly to dynamic business environment and to establish efficient production management system based on information technologies. However, most of SMEs have faced with low applicability of the production management system resulting from high costs of introduction and maintenance. In this paper, a production planning and control system, that is S-PMS (production management system for SMEs), is proposed to solve the problem of low applicability and limited human resources. S-PMS enables production managers to efficiently collect and manage master data with the actual target production systems and explores the bottleneck process by means of simulation techniques to improve productivity. Furthermore, it implements rescheduling mechanism in terms of a variety of process routes. In essence, intuitive dispatching rules and integrated data management of S-PMS improve field applicability of production management system. Consequently, S-PMS is expected to be used as an efficient production management system of SMEs in Korea.
S-573 : Depression in patients with SLE and their association with serum BDNF levels

Introduction: Patients with systemic lupus erythematosus (SLE) are vulnerable to depression because of the chronic nature and neuropsychiatric involvement of the disease. Serum brain-derived neurotrophic factor (BDNF) was reported to be decreased in depression. The aim of the study was to investigate the prevalence of depression and its related factors including BDNF in patients with SLE. Material and Methods: One hundred and eighty patients were enrolled at the Rheumatology Clinic from January to March in 2012. The prevalence of depression was assessed using the center for epidemiologic studies depression (CES-D) scale. We evaluated disease activity by physician`s global assessment (PhyGA), patient`s global assessment (PGA), SLE disease activity index (SLEDAI) and disease related organ damage. The EuroQol-5 dimensions (EQ-5D), sociodemographic features and laboratory tests were also surveyed. Serum BDNF was measured using enzyme-linked immunosorbent assay (ELISA). Patients having a CES-D score of 24 or more were considered to have depression. Results: The prevalence of depression in SLE was 22.8% (n=41). In univariate analysis, unmarried/disrupted marital status (patients with single/divorced/estrangement/bereavement status than those with married status), unemployment, low annual income, higher PGA, higher PhyGA, lower EQ-5D index score and severe pain/discomfort were significantly associated with depression. In multivariate analysis, unmarried/disrupted marital status, higher PGA and severe pain/discomfort were significantly associated with depression. Serum BDNF levels were not associated with depression (p=0.62) but negatively correlated with disease activity in patients with SLE (r=-0.188, p=0.021). Conclusion: Depression is prevalent in patients with SLE and is associated with unmarried/disrupted marital status, severe pain/discomfort and higher PGA. Serum levels of BDNF were not associated with depression but were negatively correlated with disease activity. Treatment of depression may be beneficial in patients with severe pain/discomfort or high PGA.

<P>The limited localization and penetration of monoclonal antibodies (mAb) into solid tumors restricts their antitumor efficacy. Here, we describe a solid tumor–targeting antibody with enhanced tumor penetration activity. We designed a 22-residue peptide (A22p), which was extracted from the C-terminal basic region of semaphorin 3A (Sema3A) but modified to have higher affinity with neuropilin receptors (NRP), and genetically fused it to the C-terminus of Fc of human immunoglobulin G1 via a 15-residue (G<SUB>4</SUB>S)<SUB>3</SUB> linker, generating Fc-A22p, for the bivalent binding to NRPs. In contrast to Fc or the monovalent A22p peptide alone, Fc-A22p homed to tumor vessels and induced vascular permeability through VE-cadherin downregulation and penetrated tumor tissues by interacting with NRPs in mice bearing human tumor xenografts. We extended the Fc-A22p platform by generating mAb-A22p antibodies of two clinically approved solid tumor–targeting mAbs, the anti-EGF receptor mAb cetuximab (erbitux), and the anti-Her2 mAb trastuzumab (herceptin). The mAb-A22p antibodies retained the intrinsic antigen binding, natural Fc-like biophysical properties, and productivity in mammalian cell cultures, comparable with those of the parent mAbs. In mouse xenograft tumor models, the mAb-A22p antibodies more efficiently homed to tumor vessels and spread into the extravascular tumor parenchyma, which significantly enhanced antitumor efficacy compared with the parent mAbs. Genetic Diversity of Entomopathogenic fungi Beauveria and Metarhizium from Soils in Korea

In agricultural fields, the entomopathogenic fungal species have been investigated for their potential as the biological control agents due to their role of natural enemies for insects. To address the requirements of a potential South Korea based biocontrol effort using entomopathogenic fungi, we investigated the occurrence of various entomopathogenic fungi in 1080 soil samples representing from various area and locations in South Korea. Entomopathogenic fungi were isolated from soils using semiselective medium SDA-D50 contained saboraund dextrose agar, 50 ug/ml dodine, 100 ug/ml chloramphenicol and 50 ug/ml streptomycin. The isolated putative fungi were identified by the determination of internal transcribed spacer (ITS) region sequences of the nuclear ribosomal analysis. As a result, entomopathogenic fungi were found to occur in 30.8% of the soil samples studied. The most abundant species were Beauveria bassiana (Bals.) Vuill. and Metarhizium anisopliae (Metschn.) Sorok. Isolates of B. brongniartii, Cordyceps sp., Lecanicillium sp., Isaria sp. and Tolypocladium cylindrosporum were also found. The occurrence of entomopathogenic fungi was analyzed by the area and soil types. These positive entomopathogenic fungi may have potential against variety pests in agriculture and forest

Effect of Terminalia chebula fruit on anaphylaxis by anal therapy

The effect of aqueous extract of Terminalia chebula fruit (Combretaceae) (TCAE) by anal administration on mast cell-dependent immediate-type anaphylactic reactions was investigated. TCAE (0.005 to 1 g/kg) inhibited systemic anaphylaxis induced by compound 48/80 in mice. When TCAE was pretreated at the same concentrations with systemic anaphylaxis, the plasma histamine levels were reduced in a dose-dependent manner. TCAE (0.1 and 1 g/kg) also significantly inhibited local anaphylaxis activated by anti-DNP IgE. TCAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-DNP IgE. Moreover, TCAE (0.01 and 0.1 mg/ml) had a significant inhibitory effect on anti-DNP IgE-mediated tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$ production from RPMC. These results provide evidence that anal therapy of TCAE may be beneficial in the treatment of systemic and local mast cell-dependent anaphylaxis.
