        Titania nanopores with dual micro-/nano-topography for selective cellular bioactivity

        <P><B>Abstract</B></P> <P>This letter describes a simple surface modification strategy based on a single-step electrochemical anodization towards generating dual micro- and nano-rough horizontally-aligned TiO<SUB>2</SUB> nanopores on the surface of clinically utilized micro-grooved titanium implants. Primary macrophages, osteoblasts and fibroblasts were cultured on the nano-engineered implants, and it was demonstrated that the modified surfaces selectively reduced the proliferation of macrophages (immunomodulation), while augmenting the activity of osteoblasts (osseo-integration) and fibroblasts (soft-tissue integration). Additionally, the mechanically robust nanopores also stimulated osteoblast and fibroblast adhesion, attachment and alignment along the direction of the pores/grooves, while macrophages remained oval-shaped and sparsely distributed. This study for the first time reports the use of cost-effectively prepared nano-engineered titanium surface via anodization, with aligned multi-scale micro/nano features for selective cellular bioactivity, without the use of any therapeutics.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Single-step fabrication of dual micro-rough and nanoporous titanium surfaces </LI> <LI> Mechanically robust aligned nanopores with preserved underlying micro-features </LI> <LI> Upregulated attachment and adhesion of osteoblasts and fibroblasts </LI> <LI> Reduced macrophage proliferation and adherence </LI> <LI> Osteoblasts and fibroblasts align parallel to the nanopore arrangement. </LI> </UL> </P>

        Genetically-engineered <i>Salmonella typhimurium</i> expressing TIMP-2 as a therapeutic intervention in an orthotopic glioma mouse model

        <P><B>Abstract</B></P> <P>Glioma is one of the most devastating and refractory cancers. The main factors underlying therapeutic failure include extremely invasive characteristics and lack of effective methods for drug delivery. Attenuated <I>Salmonella</I> strains presented a high concentration of tumor targets in various types of cancer models, suggesting a role as potential vectors for drug delivery. In this study, we genetically engineered an attenuated strain of <I>Salmonella</I> as an anti-invasive vector for the targeted delivery and expression of tissue inhibitor of metalloproteinases 2 (TIMP-2) in an orthotopic nude mouse model of glioma. The bioluminescence signals related to tumor size significantly declined in the TIMP-2-expressing <I>Salmonella</I> (SLpTIMP-2)-treated group compared with the control group. Compared with the control group with a survival rate of an average of 33 days, the SLpTIMP-2 group showed an extended survival rate by nearly 60% and lasted an average period of 53 days with TIMP-2 induction. These results indicated the promising therapeutic potential of <I>S. typhimurium</I> for targeted delivery and secretion of TIMP-2 in glioma.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Intracranial injection of <I>Salmonella</I> has been demonstrated to be a more effective than tail vein injection. </LI> <LI> Treatment with TIMP-2-expressing bacteria showed down regulation of MMP-2 in orthotopic glioma. </LI> <LI> TIMP-2-expressing bacteria significantly inhibited tumor growth and elongated animal survive. </LI> </UL> </P>

        Bacitracin Inhibits the Migration of U87-MG Glioma Cells via Interferences of the Integrin Outside-in Signaling Pathway

        Objective : Protein disulfide isomerase (PDI) acts as a chaperone on the cell surface, and it has been reported that PDI is associated with the tumor cell migration and invasion. The aims of this study are to investigate the anti-migration effect of bacitracin, which is an inhibitor of PDI, and the associated factor in this process. Methods : U87-MG glioma cells were treated with bacitracin in 1.25, 2.5, 3.75, and 5.0 mM concentrations. Western blot with caspase-3 was applied to evaluate the cytotoxicity of bacitracin. Adhesion, morphology, migration assays, and organotypic brain-slice culture were performed to evaluate the effect of bacitracin to the tumor cell. Western blot, PCR, and gelatin zymography were performed to investigate the associated factors. Thirty glioma tissues were collected following immunohistochemistry and Western blot. Results : Bacitracin showed a cytotoxicity in 3rd (p<0.05) and 4th (p<0.001) days, in 5.0 Mm concentration. The cell adhesion significantly decreased and the cells became a round shape after treated with bacitracin. The migration ability, the expression of phosphorylated focal adhesion kinase (p-FAK) and matrix metalloproteinase-2 (MMP-2) decreased in a bacitracin dose- and time-dependent manner. The U87-MG cells exhibited low-invasiveness in the 2.5 mM, compared with the untreated in organotypic brain-slice culture. PDI was expressed in the tumor margin, and significantly increased with histological glioma grades (p<0.001). Conclusion : Bacitracin, as a functional inhibitor of PDI, decreased the phosphorylated FAK and the secreted MMP-2, which are the downstream of integrin and play a major role in cell migration and invasion, might become one of the feasible therapeutic strategies for glioblastoma.

        Assessment of supervision monitoring for radiation environment around the typical research reactors in China

        The supervision mode, monitoring basis and monitoring scheme of radiation environment monitoringconcerning typical research reactors in China were investigated in this study. Summary and analysis wereconcluded of the present situation of supervised monitoring of radiation environment, such as monitoring objects, points, frequency and so on, based on the relevant data of monitoring points of fourtypical research reactors in China. Some experiences and existing problems were analyzed concerningthe supervised monitoring of China's research reactors. Tips on topics related to strengthen the monitoring of radiation environment around the research reactors has noted.

        현대시 교과서본 확정의 문제 연구 - 김영랑의 <돌담에 속삭이는 햇발>의 국어 교과서 표기를 중심으로 -

        이 연구는 교과서본 표기 확정의 중요성에 대한 인식을 고취하여 문학교육적 목적을 고려하여 현대시 교과서본을 확정할 필요가 있음을 말하고자 하였다. 이에 교과서에서 출처로 제시하고 있는 현대시의 원본과 정본, 교과서본 표기를 검토하고, 검정 체제 아래 동일한 현대시 작품이 각기 다른 방식으로 표기되는 것을 문제시하였다. 현대시의 교과서본 확정에는 현행 한글 맞춤법 규정을 적용하는 것보다 해석의 가능역을 규정하는 시 표기의 중요성을 인식하고, 시마다의 특수한 미감을 온전하게 살려내는 방향으로의 문학교육적 논의가 필요하다. 또한 교과서본 확정에 적용된 원칙과 논의의 과정은 공유하여 교육 주체 간 교육 목적에 대한 합의를 바탕으로 작품 선택의 다양성뿐만 아니라 감상의 가능역 또한 넓혀가야 할 것이다. This study aims to promote awareness regarding the importance of textbook notation finalizations and it makes suggestions regarding the need for modern poetry textbooks to be finalized with considerations made to literary education purposes. At this, examinations were made of notations in the original, formal, and textbook versions of modern poems presented as sources in textbooks and the problem was raised that identical modern poems within the authorization system are being noted using respectively different methods. In textbook finalizations of modern poetry, rather than applying the current rules of Korean orthography, it is necessary to recognize the importance of poetry notations that define possible areas of interpretation and to conduct discussions of literary education that wholly vitalize the unique esthetic senses of each poem. Also, principles applied to textbook finalizations and discussion processes should be shared to expand the possible areas of appreciation in addition to the diversity of work selection based on agreements made regarding educational purposes among educational agents.

        ‘문학의 생활화’를 위한 시 암송 교육 연구 - D중학교의 사례를 중심으로-

        본고에서는 ‘문학의 생활화’의 방법으로 시 암송에 주목하였다. 암송은 전통적인 시 교육의 방법인 구술 연행으로, 학습자의 내면 변화를 관찰 가능한 범주 내에서 평가해야 하는 설계 방식 아래 기능적 훈련으로 간주되어 경시되고 있다. 간과되고 있는 암송의 교육적 가치를 되살리고자 시 교육의 방법으로서 암송의 위상과 시 암송 교육 프로그램에 참여한 학습자의 내면 변화의 양상을 검토하였다. 이를 바탕으로 시 암송 교육에서 활용 가능한 시 읽기의 방법을 ‘익숙하게 읽기’, ‘음미하며 읽기’, ‘체인하며 읽기’로 제안하였다. 시는 학습자에게 투입되어야 할 고정적 실체가 아니라 학습자에 의해 발제된 역동적 실체로, 암송은 산출되어야 하는 교육적 결과가 아니라 문학의 생활화를 위한 시 교육의 방법이다. 자율적 체화 구조를 갖춘 학습자가 일상의 삶에서 시를 발제할 때 문학의 생활화가 이루어질 수 있다. 시 암송은 학습자 누구나 할 수 있는 문학 활동으로 학습자에게 체인된 시는 평생토록 문학을 생활화하게 하는 원동력이 되어 줄 것이다. This study focused on poetry recitation as a method of 'living of literature'. Recitation is oral performance, a traditional method of poetry education. Recital is regarded as a functional training under the design method that should be evaluated within the observable category of learner's inner change. In order to revive the educational value of recitation, the educational status of recitation was reviewed and the internal changes of the learners who participated in the recitation education program were examined. Based on the results, the methods of reading available poems in poetry recitation education were proposed as 'Recognitive reading', 'Aesthetic reading' and 'Embodied cognitive reading'. Poetry is not a static entity to be put into the learner, but a dynamic entity presented by the learner. Recitation is not an educational result to be produced but a poetry education method for 'living of literature'. When a learner with an autonomous embodied structure enacts poetry in their daily life, 'living of literature' can be achieved. Poetry recitation is a literary activity that any learner can do, and the embodied cognitive poetry to the learners will be the driving force for the life of literature for a lifetime.
