The feasibility of laccase production by immobilization of Pleurotus ostreatus 1804 on polyurethane foam (PUF) cubes with respect to media composition was studied in both batch and reactor systems. Enhanced laccase yield was evidenced due to immobilization. A relatively high maximum laccase activity of 312.6 U was observed with immobilized mycelia in shake flasks compared to the maximum laccase activity of free mycelia (272.2 U). It is evident from this study that the culture conditions studied, i.e. biomass level, pH, substrate concentration, yeast extract concentration, $Cu^{2+}$ concentration, and alcohol nature, showed significant influence on the laccase yield. Gel electrophoretic analysis showed the molecular weight of the laccase produced by immobilized P. ostreatus to be 66 kDa. The laccase yield was significantly higher and more rapid in the packed bed reactor than in the shake flask experiments. A maximum laccase yield of 392.9 U was observed within 144 h of the fermentation period with complete glucose depletion.

<P><B>Abstract</B></P> <P>In the present study, carbon nanotube (CNT), thermally reduced graphite oxide (TRGO), and CNT-TRGO were functionalized with p-sulfonatocalix[4]arene (SC4) and then investigated for fluorescent detection of cholesterol in the presence of rhodamine 6G (R6G). All of the composite samples show effective quenching of R6G fluorescent intensity without cholesterol, but the fluorescence intensity significantly increases with the addition of cholesterol. The results indicate a strong interaction between SC4 composites and cholesterol rather than between the SC4 composites and R6G, increasing R6G fluorescence intensity in the presence of cholesterol due to the release of R6G from the SC4 composites. Formation of the complex, made of the SC4 composite plus the cholesterol, enables the detection of cholesterol in the wide range of cholesterol concentration up to 109 µM. Purpose: Diagnosis of extrapulmonary tuberculosis (EPTB) poses serious challenges. A careful selection of appropriate gene targetsis essential for designing a multiplex-polymerase chain reaction (M-PCR) assay. Materials and Methods: We compared several gene targets of Mycobacterium tuberculosis, including IS6110, devR, and genes encodingMPB-64 (mpb64), 38kDa (pstS1), 65kDa (hsp65), 30kDa (fbpB), ESAT-6 (esat6), and CFP-10 (cfp10) proteins, using PCR assayson 105 EPTB specimens. From these data, we chose the two best gene targets to design an M-PCR. Results: Among all gene targets tested, mpb64 showed the highest sensitivity (84% in confirmed cases and 77.5% in clinically suspectedcases), followed by IS6110, hsp65, 38kDa, 30kDa, esat6, cfp10, and devR. We used mpb64+IS6110 for designing an M-PCR assay. Our M-PCR assay demonstrated a high sensitivity of 96% in confirmed EPTB cases and 88.75% in clinically suspected EPTB cases with a high specificity of 100%, taking clinical diagnosis as the gold standard. Conclusion: These M-PCR results along with the clinical findings may facilitate an early diagnosis of EPTB patients and clinical management of disease.
An Efficient Semantic Ranked Keyword Search of Big Data Using Map Reduce

Information retrieval is fast becoming the prevailing form of information access, surpassing traditional database style searching. Ontologies have become the tool of choice employed in many information retrieval systems and more prominently in semantic information retrieval. In order to overcome the disadvantages in key word based information retrieval systems, which transfer irrelevant information, ontology has been designed. A system with ontology mimics the real world, where every task is laced with certain meaning as this is basic idea behind knowledge processing. Hadoop, which is an open source frame work for storing and processing large datasets, is used for pre-processing the text documents. First, a set of text documents are considered. Pre-processing is performed on a large domain of data using Hadoop MapReduce. This includes the removal of the stop words along with stemming and excluding less frequency words. Despite this pre-processing, owing to the colossal number of index terms still floating in the considered domain data, the problem of high dimensionality is encountered. Therefore the dimensionality of such a group of terms is reduced by identifying it as a concept and those concepts can be viewed as a single dimension in a ontology based information retrieval system. Now ontology is constructed by assigning synonym set to each concept in this structure using tools like word net. Thus constructed ontology can be mapped on to the processed query which gives us the relevant information from the data pool considered.

Helicoverpa armigera is an important pest infesting a number of crops of high commercial value leading to huge economic losses globally. RNA interference (RNAi) is a potent tool for the control of insect pests and towards this objective, transgenic tomato expressing dsRNA of Juvenile hormone acid methyl transferase (JHAMT) gene of H. armigera was developed. The H. armigera larvae fed on HaJHAMT tomato leaves led to a severe (90%) down regulation of the cognate gene expression thus adversely affected the feeding and metamorphosis. Reduction in larval and pupal weight and inability to undergo complete metamorphosis were observed that led to larval-pupal intermediates and subsequently affected the eclosion. Thus, JH down regulation is an attractive target due to specificity to insects and its important role in insect growth and development to engineer insect resistance in crops using RNAi.
