        Experimental Pathogenesis of Pullorum Disease with the Local Isolate of Salmonella enterica serovar. enterica subspecies Pullorum in Pullets in Bangladesh

        이 실험은 추백리의 병원성을 연구하고자 2006년 2월부터 12월까지 Salmonella enterica serovar. enterica subspecies (S.) Pullorum 야외주를 분리한 후, 이를 건강한 닭에 실험적으로 감염시킨 다음 임상증상, 여러 기관의 육안 및 조직병리학적 검색과 아울러 공격주의 재분리와 동정을 시도하였다. S. Pullorum에 혈청학적으로 음성인 12주령의 100수의 암탉을 $A{\sim}E$까지 20수씩 5그룹으로 구분하였다. $A{\sim}DS$. Pullorum을 $10^6\;CFU$, $10^7\;CFU$, $2{\times}10^7\;CFU$, $10^8\;CFU$로 각각 경구 감염시켰고, E는 비감염 대조군으로 삼았다. 실험 방법으로는 부검, 조직병리학적 검사, Salmonella에 대한 세균 배양, 염색, 생화학적 특성을 조사하고 그 결과를 기술하였다. The research work was carried out to study the pathogenesis covering the clinical signs, gross and histopathological lesions in different organs, and reisolation and identification of the organisms after experimental infection with the local isolate of Salmonella enterica serovar. enterica subspecies (S.) Pullorum at different time interval of the experiment during the period February 2006 to December 2006. One hundred pullets (seronegative to S. Pullorum of 12 weeks age were purchased and divided into 5 (A, B, C, D and E) groups and each group consisted of 20 birds. Four groups (A, B, C and D) were infected orally with a dose of $10^6\;CFU$, $10^7\;CFU$, $2{\times}10^7\;CFU$, $10^8\;CFU$ of S. Pullorum, respectively, and one group (E) was treated as uninfected control. The used methods were necropsy and histopathology, culture of bacteria, staining and biochemical test of Salmonella. Five birds from each group were randomly selected and sacrificed $1^{st}$ week, $2^{nd}$, $3^{rd}$ and $4^{th}$ weeks of post infection (PI). From all the groups, the bacteriological samples (crop, liver, lung, heart, spleen, bile duodenum, ceca and blood) were collected with pre enriched in buffered peptone water in sterile poly bags. Liver, lungs, heart, spleen, intestine, etc. were collected in 10% buffered-formalin for histopathological examination. No clinical signs, gross and histopathological lesions were found in control group and no S. Pullorum was reisolated. Clinical sign of experimentally infected with S. Pullorum in pullets were loss of appetite (100%), slight depression (75%), ruffled feathers (85%), diarrhea (60%) and loss of weight (100%) in chickens. The feed intake and body weight at different weeks after PI differed significantly (p<0.01) among the groups. Grossly, the highest recorded lesion was button-like ulcer in the ceca (80%) and the lowest was white nodules in lungs (1.25%). S. Pullorum were reisolated from crop (91.25%), liver (91.25%), lung (83.75%), heart (71.25%), spleen (87.75%), bile (33.25%), duodenum (92.50%), ceca (97.50%) and from different group of infection (61.25%). The highest microscopic findings were intestinal and cecal mucosa and submucosa exhibited infiltration of mononuclear cells and congestion (96.25%), and the lowest finding was nodule formation in the lungs (3.75%). The pattern of the disease production by local isolate of S. Pullorum in Bangladesh is almost similar with other isolates in different countries.

        Effects of Resveratrol and Essential Oils on Growth Performance, Immunity, Digestibility and Fecal Microbial Shedding in Challenged Piglets

        A study was conducted to evaluate the effects of resveratrol and essential oils from medicinal plants on the growth performance, immunity, digestibility, and fecal microbial shedding of weaned piglets. A total of 48 weaned piglets (8 kg initial weight, 28-d-old) were randomly allotted to four dietary treatments with 3 replications of 4 piglets each. The dietary treatments were NC (negative control; basal diet), PC (positive control; basal diet+0.002% apramycin), T1 (basal diet+0.2% resveratrol), and T2 (basal diet+0.0125% essential oil blend). All piglets were orally challenged with 5 ml culture fluid containing $2.3{\times}10^8$ cfu/ml of Escherichia coli KCTC 2571 and $5.9{\times}10^8$ cfu/ml Salmonella enterica serover Typhimurium. The PC group (p<0.05) showed the highest average daily gain (ADG) and average daily feed intake (ADFI) throughout the experimental period, although feed conversion ratio (FCR) was improved in the T1 group (p>0.05). Serum IgG level was increased in the T1 group, whereas TNF-${\alpha}$ levels was reduced in the supplemented groups compared to control (p<0.05). The PC diet improved the dry matter (DM) digestibility, whereas PC and T2 diets improved nitrogen (N) digestibility compared to NC and T1 diets (p<0.05). Fecal Salmonella and E. coli counts were reduced in all treatment groups compared to control (p<0.05). Fecal Lactobacillus spp. count was increased in the T2 group compared to others (p<0.05). Dietary treatments had no significant effect on fecal Bacillus spp. count throughout the entire experimental period. Based on these results, resveratrol showed strong potential as antibiotic alternatives for reversing the adverse effects of weaning stress on growth performance, immunity and microbial environment in E. coli and Salmonella-challenged piglets.

        Evaluation of Physicochemical Deterioration and Lipid Oxidation of Beef Muscle Affected by Freeze-thaw Cycles

        This study was performed to explore the deterioration of physicochemical quality of beef hind limb during frozen storage at -20ºC, affected by repeated freeze-thaw cycles. The effects of three successive freeze-thaw cycles on beef hind limb were investigated comparing with unfrozen beef muscle for 80 d by keeping at -20±1°C. The freeze-thaw cycles were subjected to three thawing methods and carried out to select the best one on the basis of deterioration of physicochemical properties of beef. As the number of repeated freeze- thaw cycles increased, drip loss decreased and water holding capacity (WHC) increased (p<0.05) till two cycles and then decreased. Cooking loss increased in cycle one and three but decreased in cycle two. Moreover, drip loss, WHC and cooking loss affected (p<0.05) by thawing methods within the cycles. However, pH value decreased (p<0.05), but peroxide value (p<0.05), free fatty acids value (p<0.05) and TBARS value increased (p<0.05) significantly as the number of repeated freeze-thaw cycles increased. Moreover, significant (p<0.05) interactive effects were found among the thawing methods and repeated cycles. As a result, freeze-thaw cycles affected the physicochemical quality of beef muscle, causing the degradation of its quality.

        Integrated Management of Foot Rot of Lentil Using Biocontrol Agents under Field Condition

        The efficacy of cowdung, Bangladesh Institute of Nuclear Agriculture (BINA)-biofertilizer, and Bangladesh Agricultural University (BAU)-biofungicide, alone or in combination, was evaluated for controlling foot rot disease of lentil. The results exhibited that BINA-biofertilizer and BAUbiofungicide (peat soil-based Rhizobium leguminosarum and black gram bran-based Trichoderma harzianum) are compatible and have combined effects in controlling the pathogenic fungi Fusarium oxysporum and Sclerotium rolfsii, which cause the root rot of lentil. Cowdung mixing with soil (at 5 t/ha) during final land preparation and seed coating with BINA-biofertilizer and BAU-biofungicide (at 2.5% of seed weight) before sowing recorded 81.50% field emergence of lentil, which showed up to 19.85% higher field emergence over the control. Post-emergence deaths of plants due to foot rot disease were significantly reduced after combined seed treatment with BINA-biofertilizer and BAU-biofungicide. Among the treatments used, only BAU-biofungicide as the seed treating agent resulted in higher plant stand (84.82%). Use of BINA-biofertilizer and BAU-biofungicide as seed treating biocontrol agents and application of cowdung in the soil as an organic source of nutrient resulted in higher shoot and root lengths, and dry shoot and root weights of lentil. BINA-biofertilizer significantly increased the number of nodules per plant and nodules weight of lentil. Seeds treating with BAUbiofungicide and BINA-biofertilizer and soil amendment with cowdung increased the biomass production of lentil up to 75.56% over the control.

        Quality Determination of Different Wholesale Cuts of Goat Carcass at Different Ages

        본 연구는 도매상에서 판매하고 있는 염소육의 영양학적 및 미생물학적 품질을 평가하기 위하여 수행하였다. 시료는 1년생, 2년생, 3년생 염소육을 대상으로 수행하였으며, 각 연령별에서 3마리씩 4부분의 다른 부위(어깨, 목, 허리, 다리)를 선정하여 총 36점을 대상으로 본 실험을 수행하였다. 염소육의 품질을 평가하기 위하여 각 시료의 외형, 색, 냄새, 육즙, 일반조성분, pH, 총균수, 대장균군과 효모수를 측정하였다. 각 부위별 염소육의 pH는 평균적으로 5.647-5.692로 나타내어 각 부위별 염소육의 pH는 유의적 차이가 나타나지 않은 반면 연령별 염소육의 pH는 5.585-5.743로 유의적 차이(p < 0.01)를 나타냈다. 각 연령별 염소육의 육즙함량은 32.24-42.10%로 유의적인 차이(p < 0.01)를 나타내었으며, 마블링은 다른 부위보다 목살부위에서 더 명확하게 확인할 수 있었다. 염소육의 단백질 함량은 20.78-27.71%의 결과로 유의적 차이(p < 0.01)를 나타내었고, 다리부위는 다른 부위에 비하여 높은 단백질 함량을 나타내었으며 각 부위별 염소육의 지방 함량은 2.657-11.469%로 유의적인 차이(P < 0.01)를 나타냈다. 염소육의 수분함량은 69.20-73.31%, 1년생 염소육의 회분함량은 0.989 $\pm$ 0.129%로 다른 연령대의 염소육보다 높은 것으로 나타났으며 이 결과 또한 유의적인 차이(P < 0.01)를 나타내었다. 칼슘의 함량은 유의적인 차이를 나타내지 않은 반면 1년생 염소육의 인의 함량은 0.149 $\pm$ 0.0251%로 다른 년생의 염소고기에 비하여 높은 것으로 나타냈다. 연령별 염소육의 총균수 수준은 5.05-5.15 log cfu/g으로써 유의적 차이를 나타내지 않은 반면 각 부위별 염소육의 대장균군의 오염수준은 2.56-3.05 log cfu/g으로 유의적인 차이(P < 0.01)를 나타냈다. 연령별 염소육 또한 대장균군 오염수준은 2.79-2.84 log cfu/g으로써 유의적인 차이(P < 0.05)를 나타냈으며 1년생 염소육의 대장균 오염 수준은 다른 연령의 염소육에 비해 높은 것으로 나타났다. 효모의 오염수준 또한 각 부위별(P < 0.01), 연령별(P < 0.05)로 유의적인 차이를 나타내었다. 결론적으로 염소육의 영양학적, 미생물학적 품질은 염소의 연령과 판매 부위에직접적인 영향을 받는 것으로 사료된다. The experiment was conducted to find out the nutritive value and microbial status of different wholesale cuts of goat carcass. The meat sample (4 cuts from each animal $\times$ 3 different age groups $\times$ 3 animals in each group = 36 samples) was obtained from 1-, 2-, and 3-year aged goats. The whole sale cuts were shoulder, rack, loin and leg of each goat carcass. To assess the quality of meat sample, the general appearance, color, smell, juiciness, proximate composition, pH, total bacteria, coliform bacteria, and yeast were studied. The mean pH value of different cuts ranges from 5.65-5.69 didn't differ significantly, but due to age differences the pH values (5.59-5.74) differed significantly (p < 0.01). The values of juiciness in different ages ranged from 32.24-42.10% which differed significantly (p < 0.01). The marbling of the cuts of rack portion was more pronounced than that of other cuts. The ranges of crude protein (CP) content of goat carcass (20.78-27.71%) differed significantly (p < 0.01) and leg portion contained higher CP than other portion. Fat contents of different cuts ranged from 2.66-11.47% differed significantly (P < 0.01). The moisture content of the carcass differed significantly which ranged from 69.20-73.31%. The ash content of the cuts of 1-year aged groups (0.99 $\pm$ 0.13%) was higher than that of other age groups and differed significantly (P < 0.01). The calcium (Ca) content did not differ significantly. The phosphorus (P) content was higher in one year old goat (0.15 $\pm$ 0.03%) than that of the goats of other ages. The total viable count (TVC) content of microorganisms ranging from 5.05-5.15 log cfu/g at different ages did not differ significantly. The coliform count (CC) of different cuts differed significantly (P < 0.01) which ranged from 2.56-3.05 log cfu/g; it also differed significantly (P < 0.05) in different ages (2.79-2.84 log cfu/g) and was higher in 1 year old goat carcass. The yeast count differed significantly in different cuts (P < 0.01) and ages (P < 0.05). From the study it is concluded that the age and different wholesale cuts have direct influence on quality of goat carcass.
