        Targeted delivery of isoliquiritigenin by ultrasonic microbubbles attenuate myocardial injury via suppressing inflammation and oxidative stress and activating AMPK/SIRT1/eNOS signaling pathway in rats

        To investigate the protective efficacy of ultrasound targeted microbubble destruction (UTMD) combined with Isoliquiritigenin on myocardial injury in rats. The GK rat model of cardiomyopathy was successfully established by the induction of adriamycin. Then these rats with cardiomyopathy were randomly assigned into the model group, isoliquiritigenin microbubbles and ultrasound alone or combination group, using healthy ones as normal control. After 8-week consecutive treatment, the relevance indexes of diabetes, echocardiography as well as the hyperlipidemia, oxidative stress of model animals were examined. In addition, the fibrosis, morphological changes and inflammation response of myocardial tissues were also assessed. After further 4-week intervention, the blood biochemical indexes and the cardiac functions of model rats received the combined treatment were improved (all P < 0.05) compare to those received either monotherapy or saline. After chronic treatment, the heart/body weight ratio and serum cardiac index levels in model rats received combined treatment were significantly changed (all P < 0.05) compared with others. Furthermore, combination therapy could ameliorate excessive oxidation stress and inflammation response as well as up-regulate the expression levels of AMPK/SIRT1/eNOS signaling pathway. Targeted delivery of isoliquiritigenin by ultrasonic microbubbles can ameliorate the myocardial injury via activating AMPK/SIRT1/eNOS signaling pathways.

        A glassy carbon electrode modified with a composite consisting of electrodeposited chitosan and carboxylated multi‑walled carbon nanotubes for simultaneous voltammetric determination of dopamine, serotonin and melatonin

        A glassy carbon electrode modified with a composite consisting of electrodeposited chitosan and carboxylated multi-walled carbon nanotubes (e-CS/MWCNTs/GCE) was used as a working electrode for simultaneous determination of dopamine (DA), serotonin (5-HT) and melatonin (MT), which were related to circadian rhythms. The electrochemical characterizations of the working electrode were carried out via electrochemical impedance spectroscopy and chronocoulometry. It was found that electrochemical modification method, that was cyclic voltammetry, may can cause continuous CS polymerization on MWCNTs surface to form a dense membrane with more active sites on the electrode, and the electrochemically active surface area of e-CS/MWCNTs/GCE obtained was about 7 times that of GCE. The electrochemical behaviour of DA, 5-HT and MT on working electrode were carried out via differential pulse voltammetry and cyclic voltammetry. The results showed that e-CS/MWCNTs/GCE solved the problem that the bare electrode could not detect three substances simultaneously, and can catalyze oxidation potential difference as low as 0.17 V of two substances reaction at the same time, indicating very good electrocatalytic activity. By optimizing the detection conditions, the sensor showed a good linear response to DA, 5-HT and MT in the range of 20-1000 μmol/L, 9-1000 μmol/L and 20-1000 μmol/L, and the detection limits were 12 μmol/L, 10 μmol/L and 22 μmol/L (S/N = 3), respectively. In addition, the proposed sensor was successfully applied to the simultaneous detection of DA, 5-HT and MT in human saliva samples.

        High-Voltage Transmission Line Foreign Object and Power Component Defect Detection Based on Improved YOLOv5

        With the outstanding performance of deep learning in the feld of computer vision, the automatic visual detection of foreign bodies in transmission lines and electrical equipment defects by inspection robots and Unmanned Aerial Vehicles based on neural networks has become an attractive topic in smart grid. However, in practical application scenarios, small-sized target defects pose a great challenge to the detection accuracy of existing mainstream deep learning detection networks with limited perceptual felds. To solve the above problems, the paper proposes a detection model of YOLOv5 transmission line inspection image. Firstly, the key target images under diferent backgrounds and attitudes are collected and preprocessed. Specifcally, in order to improve the perception ability of networks for small-sized targets, the K-means clustering algorithm is used to optimize the size of the anchor box, which efectively improves the ftting ability of the key target features. Then, recursive gated convolution is used as the backbone network to improve the ability to extract key target features. Finally, considering the concealment of small-scale features, the space-to-depth convolution module is added to the neck network to realize down-sampling and retain all the information in the channel dimension. In addition, a feature prediction layer is added to optimize the scale of network detection, and a Simple Parameter-Free Attention Module is added to further optimize the characterization of network features. The experimental results show that the accuracy and recall of the proposed network are 96.8% and 93.3%, respectively. The average detection accuracy reaches 97.1%, which is 3.8% higher than that of YOLOv5 network, 5.2% higher than YOLOv6 and 1.0% higher than that of YOLOv7 network. The proposed method signifcantly improves the detection performance of critical targets and defects of high-voltage transmission lines.

        Preliminary study of genome-wide association identified novel susceptibility genes for thyroid-related hormones in Chinese population

        Background: Thyroid hormones are critical regulators of metabolism, development and growth in mammals. However, the genetic association of thyroid-related hormones in the Chinese Han population is not fully understood. Objective: We aimed to identify the genetic loci associated with circulating thyroid-related hormones concentrations in the healthy Chinese Han population. Methods: Genotyping was performed in 124 individuals using Applied Biosystems™ Axiom™ PMDA, and 796,288 single nucleotide polymorphisms (SNPs) were available for the GWAS analysis. For replication, eleven SNPs were selected as candidate loci for genotyping by Agena MassARRAY platform in additional samples (313 subjects). The values of p < 5 × 10- 6 suggest a suggestively significant genome-wide association with circulating thyroid-related hormones concentrations. Results: We identified that rs11178277 (PTPRB, p = 4.88 × 10- 07) and rs7320337 (LMO7DN-KCTD12, p = 1.22 × 10- 06) were associated with serum FT3 level. Three SNPs (rs4850041 in LOC105373394-LINC01249: p = 3.55 × 10- 06, rs6867291 in LINC02208: p = 2.40 × 10- 06 and rs79508321 in WWOX: p = 3.35 × 10- 06) were related to circulating T3 level. Rs12474167 (LOC105373394-LINC01249, p = 1.65 × 10- 06) and rs1864553 (IWS1, p = 2.00 × 10- 06) were associated with circulating T4 concentration. The association with TGA concentration was for rs17163542 in DISP1 (p = 3.46 × 10- 06) and rs12601151 in NOG-C17orf67 (p = 2.72 × 10- 07). Two genome-level significant SNPs (rs2114707 in LINC01314, p = 1.69 × 10- 06 and rs12601151, p = 1.41 × 10- 07) associated with serum TMA concentration were identified. Moreover, rs6083269 (CST1-CST2, p = 3.36 × 10- 06) was a significant locus for circulating TSH level. In replication, rs12601151 in NOG-C17orf67 was still associated with serum TGA level (p = 0.012). Conclusions: The GWAS reported 11 new suggestively significant loci associated with circulating thyroid-related hormones levels among the Chinese Han population. These findings represented suggestively biological candidates for circulating thyroid-related hormones levels and provided new insights into the mechanisms of regulating serum TGA concentration.

        Insulator Defects Detection for Aerial Photography of the Power Grid Using You Only Look Once Algorithm

        The safe operation of the power grid system depends partly on regular inspections of transmission lines, in which the insulator is one of the most important inspections objects. The manual inspection of transmission lines is a chaotic process that is both time and cost-consuming since it involves an accurate manual inspection by an expert. For insulator defect detection, an improved YOLOv4 algorithm is proposed. First, a new data augmentation method is proposed to solve the problem of insufficient sample size. Then, the size of the anchor boxes is redesigned base on the K-means algorithm to further improve the detection precision. Finally, an insulator defects detection network is constructed based on YOLOv4. Experimental results show that the detection precision of the improved network is 37.2% higher after data enhancement and anchor boxes redesign. In addition, the detection method proposed in this paper is superior to other popularity detection algorithms, including the single shot detector, region-convolutional neural networks (Faster-RCNN) and released version of you only look once (YOLO). The value of mean average precision is 99.08% and frame per second is 56. The robustness test results demonstrate that our proposed algorithm performs well under different light intensities and complex environmental backgrounds, and can accurately detect all targets, which is significantly better than other comparative algorithms. In terms of detection accuracy, test speed and robustness, our proposed algorithm meets the requirements of industrial field applications of insulator defect detection.

        Enhanced activity and stability of the monolithic Pt/SiO2–Al2O3 diesel oxidation catalyst promoted by suitable tungsten additive amount

        A series of the modified Pt/SiO2–Al2O3 diesel oxidation catalysts with different WO3 loading were prepared through co-impregnation method. With an optimum additive loading of 2 wt% WO3, the catalyst significantly improved the performance and its stability. Compared to the free-tungsten sample, the catalyst increased the maximum conversion of NO by 5–10% and decreased the reaction temperature for 30% NO conversion by 7–28 C. It was found that there exist the electron and structure interaction between platinum and tungsten species. The interactions can generate and maintain more Pt sites in metallic state for the catalytic oxidation with enhanced activity and stability.

      • A Low-Cost Portable Real-Time EEG Signal Acquisition System Based on DSP

        EEG apparatus are expensive and bulky. Their real-time performance is weak, and EEG signals are easy to be distorted. In this paper, a low-cost portable EEG signal acquisition system based on DSP is developed. By a noninvasive method with bipolar leads, weak EEG signals are induced to the pre-processing circuits, where they will undergo multi-level amplifying and filtering. Then, the analog signals are converted into digital signals using ADS8320. These digitized signals are filtered in DSP (TMS320VC5509) so that the power interference and physiological artifacts are removed with LMS (Last Mean Square) algorithm and ICA (Independent Component Analysis) algorithm. Experimental results demonstrate that the system can acquire weak EEG signals in real time, display and save the processing results. The acquisition system has the advantages of usableness and portability, and helpful to the popularity of community-based and family-based EEG diagnostic equipments.

      • Establishment of Full Genomic Length Resistance-Associated Variant Genotype 2 Hepatitis C Viruses and Applications for Future Therapeutic Strategies

        Aims: Several directly acting antiviral agents (DAA) were currently approvedfor the treatment of chronic hepatitis C (CHC). AlthoughDAA therapies are associated with better tolerability and improvedresponse rates, occurrence of drug resistance has been the drawback.The aim of the present study is to develop full-length resistance associatedvariants (RAV) HCV culture systems to evaluate the efficacyand the cross resistance of current antiviral drugs for future therapeuticstrategies.Methods: Resistance associated substitutions on NS3 (A156T, D168V),NS5A (L31V, Y93H, L31V+Y93H), and NS5B (S282T) domains weregenerated by site directed mutagenesis and cloned into genotype2a J6/JFH1 HCV plasmid with or without luciferase gene. After In vitro RNA transcription, RNAs of RAV were transfected into Huh 7.5.1cells. HCVcc in the supernatants were collected and used for thereinfection and treatment experiment to confirm drug susceptibility.We performed HCV core staining and Renilla luciferease assays toassess treatment response to multiple DAAs and other antiviral drugswith different mechanism of action after transfection or infectionof RAVs.Results: DAAs in the same classes shared cross resistance to correspondingRAVs: boceprevir, telaprevir, simeprevir, and asunaprevirto NS3 RAVs; daclatasvir and ledipasvir to NS5A RAVs; sofosbuvirto NS5B RAV. However, DAAs of the other classes effectively suppressedRAVs as well as wild type. All the RAVs were sensitive todrugs with different action of mechanism including interferon alfa,ribavirin, cyclosporine (cyclophilin inhibitor) or chlorcyclizine (entry inhibitors).Conclusions: We developed full-length RAV HCV culture systemsbased on genotype 2a strain. This system will be useful to assessantiviral response of drugs with different action of mechanisms.Combination of different classes of DAA or new drugs with differentaction mechanisms (e.g. cyclophilin inhibitor or entry inhibitors) shouldbe a future therapeutic strategy for overcoming drug resistance inthe treatment of CHC.

        Characterization and identification of OsFTL8 gene in rice

        The rice genome contains 13 Flowering Locus T-Like (FT-Like) genes, among which, OsFTL2/Hd3a and OsFTL3/RFT1 are characterized as forigen-encoding genes, promoting rice fowering under short-day and long-day conditions, respectively. However, the functions of the other FT-Like members are still unknown in rice. In this study, we characterized and identifed a member of FT-Like family, OsFTL8 gene. The investigation of photoperiodic expression pattern showed that OsFTL8 transcription was mainly induced under short-day conditions. The qRT-PCR and GUS staining assays indicated that OsFTL8 was broadly expressed in various organs, including stems, leaves and panicles. OsFTL8 overexpression transgenic lines and its knockout mutants displayed no obvious change in their heading date, compared with the wild type. However, the pollen viability and seed setting rate were decreased in the overexpression transgenic lines. OsFTL8 localizes in both nucleus and cytoplasm. And we found that OsFTL8 does not interact with OsFD1 in vivo, suggesting that OsFTL8 may not be the component of forigen activation complex (FAC). In summary, our results indicate that OsFTL8 is associated with pollen viability and not involved in regulating rice fowering time, implying functional divergence of FT-Like members.

        <P>UV-induced cell pigmentation represents an important mechanism against skin cancers. Sun-exposed skin secretes alpha-MSH, which induces the lineage-specific transcriptional factor MITF and activates melanogenesis in melanocytes. Here, we show that the autophagic tumor suppressor UVRAG plays an integral role in melanogenesis by interaction with the biogenesis of lysosome-related organelles complex 1 (BLOC-1). This interaction is required for BLOC-1 stability and for BLOC-1-mediated cargo sorting and delivery to melanosomes. Absence of UVRAG dispersed BLOC-1 distribution and activity, resulting in impaired melanogenesis in vitro and defective melanocyte development in zebrafish in vivo. Furthermore, our results establish UVRAG as an important effector for melanocytes' response to alpha-MSH signaling as a direct target of MITF and reveal the molecular basis underlying the association between oncogenic BRAF and compromised UV protection in melanoma.</P>
