        Assessment of In Vitro Assay System for Thyroid HormoneDisruptors Using Rat Pituitary GH3 Cells

        The development of in vitro assays has been recommended to screening and test-ing the potential endocrine disruptors (EDs). These assay systems focus only on identifying thethe thyroid hormone (TH) disruptors. The aim of this study was to evaluate a test system to detectTH disruptors using rat pituitary tumor GH3 cells. The test system is based on the TH-dependentincrease in growth rate. As expected, L-3,5,3-triiodothyronine (T3) markedly induced a morphologicalchange in GH 3 cells from flattened fibroblastic types to rounded or spindle-shaped types. T3 stimu-lated GH3 cell growth in a dose-dependent manner with the maximum growth-stimulating effect9 M. In addition, T3 increased the release of growth hor-mone and prolactin into the medium of the GH3 cells culture. Using this assay system, the TH-dis-rupting activities of bisphenol A (BPA) and its related compounds were examined. BPA,dimethylbisphenol A (DMBPA), and TCI-EP significantly enhanced the growth of GH3 cells in therange of 1 × 10-5M to 1 × 10-6M concentrations. In conclusion, this in vitro assay system might bestandardization before it can be used as a broad-based screening tool.

        Qigong is on the developmental stage and is expanding to the entire world, while it acknowsedges to the West as a universial method of human healthy-longevity. The conception of health known to thr west is based on the providence of modem civilization and science focusing on the physical practices, however, which usually limit the projected effective execution itself. Recently the eastern method of more effective physical-mental training was explored and by which is replaced for the human-healthy longevity in human life. The western standard form and its criterion was established through researches done by the scheme of biological-medical science, effectively it is revolutionizing forward the the eastern medical science as a formal integration of psychology, physiology and the social environmental elements. According to the eastern thought human life is a unified form of mental and physical integration. In China, the government actually supports in official for the development of qigong so that it is activated, practically suggested and progressed lively throughout the nation. Qigong in Korea is activated today. It will be expanded further from now on. Qigong is considered as a method of natural healing, physical theraphy and healthy longevity while giving attention to a lot of people in our country. As they demand qigong in daily lives, there increased the amount of service institute In our community. However, they are not qualified enough both of the knowledge and practical technique. In a future it may cause social problems and there need countermeasure. Therefore, the government must consider about certain problems such as Qigong clinics and its practicioners, which need to be examined before approval, and should enact the national policy and system so that Qigong in korea can be identifie as physical activity for all, as well as systematically established for a way of national health.

      • 초음속 터빈 익렬 앞전 형상 및 노즐-익렬 간격에 따른 유동 특성에 대한 실험적 연구

        본 연구에서는 초음속 충동형 터빈의 유통특성을 알아보기 위해 소형 초음속 풍동을 설계 하였으며 Single pass Schlieren system을 이용하여 유통을 가시화하였다. 실험은 2차원 초음속 노즐과 익렬을 조합하여 블레이드 앞전 형상과 노즐-익렬 간극에 따라 실시하였다. 실험을 통해 충격파를 포함한 복잡한 유동 형태와 노즐-익렬, 충격파-경계층 상호작용 등을 관찰할 수 있었다. In this paper, a small supersonic wind tunnel is designed and built to study the flow characteristics of a supersonic impulse turbine cascade. The flow is visualized by means of a single pass Schlieren system. The supersonic cascade with 2-dimensional supersonic nozzle was tested for various blade leading edge shapes and gaps between the nozzle and cascade. Highly complicated flow patterns including shocks, nozzle-cascade interaction and shock boundary layer interactions are observed.

        본 연구에서는 초음속 충동형 터빈의 유동특성을 알아보기 위해 소형 초음속 풍동을 설계 하였으며 Single pass Schlieren system을 이용하여 유통을 가시화하였다. 실험은 2차원 초음속 노즐과 익렬을 조합하여 압력비에 따라 실시하였다. 실험을 통해 충격파를 포함한 복잡한 유동 형태와 노즐-익렬, 충격파-경계층 상호작용 등을 관찰할 수 있었다. In this paper, a small supersonic wind tunnel is designed and built to study the flow characteristics of a supersonic impulse turbine. The flow is visualized by means of a single pass Schlieren system. The supersonic cascade with 2-dimensional supersonic nozzle was tested over a wide range of pressure ratio.       • 선천성 사지기형 발생 기전 연구(Ⅲ) : Hox유전자 발현에 관한 연구 Studies on the Expression of Hox gene

        Homeobox-containir을 (Hox) 운전자는 발생단케, 위치 및 조직 특이적인 발현에 따라 배자의 발생 과정동안 형태형성에 관여하는 것으로 알려져 있다. 딘ok 유진자들(또oxa-1, Hoxa-2, Hoxb-6 Hoxa-7, Hoxc-8) 중 특히 Hoxa-7 유전자는 ectoderm에서 유래하는 neural tube 및 neoral crest ceIBs과 mesoderm에사 유래하는 prevertebratae에서 전 ·후축 형성 시에 발현하는 것으로 잘 알려진 유전자이다. 그러나 Hol(a-? 유전자가 배자의 사지 및 꼬리 발생에 직접적으로 관여하는지에 대해서는 아직까지 잘 알려지지 않았다. 따라서 본 연구에서는 최기형성 물질로 알려진 all-trans retinoic acid (RA)하 cyclophosphandde (CP)를 임신 9.5일의 마우스에 투여한 후 임신 10.5, 11.5, 12.5, 13.5 및 14.5꿀에 배자를 적출 f':4였을 때 나타나는 형태학적 이상과 Hoxa·-7 유전자 발현의 위치와의 상관성을 비교하기 위하여 whoBe mount in situ hybridization(WISH)을 실시하였다. 그 결과, 템oxa-7의 antisense RNA probe를 사용하였을 경우, 용매대조군과 비교하여 RA 투여 시 나타난 형태학적 이상인 꼬리단락의 위fl됐 「loxa-7이 강하게 발현되었고, CP 투여 시에는 용매대조군과 비교하여 배자꼭 크기가 현저히 적어서, 전체적인 발달이 저해된 것으로 나타났으며, 특히 발달이 저해된 foreBimb과 hindlimb의 위치에 Hoxa-7이 강하게 발현되었다 한편, antisense RNA probe의 음성대조군인 Hoxa-7의 sense RNA 1)robe로 WISH를 실시한 결과 antisense probe를 사용한 결과와는 달리 색깔 반응이 현저히 약하게 나타났다. 파라서 본 연구결과, 최기형성 물질에 노출시 나타나는 마우스 배자의 형태학적이상은 Hoxa-7 유전자의 발현 증가와 상관성이 있는 것으로 추정된다. However, it is not clearwhether Hoxa-7 gene if; direcfBy involved in mouse embryonic timb and tail development.0order to establish the correlation between Hoxa-7 gene expression and the effect of teratagensduring mouse embryogenesis, we have analyfed Hoxa-7 gene expression in the developing limbbuds and tails of ICR mouse embryes (gestation day 10,5, 11,5, 12.5, 12.5, 14.5) after maternaladministration of teratogens, atl-trans retineic actd (RA) and cycBophosphahEde (CP), ungestation day 9.5 using whole mount in situ hybridization (WISH). We f4nd that RA causesshortening and crookedness of tail and CP causes develoPmental retardation of embryo,especia31y Ihfb buds. Rerutts from IfnSH show that expression of Hoxa-7 is increased in thetail of embryos which were treated with RA. Whereas Hoxa-7 gene expression is increased inthe limb buds of embryos, wich were treated with CP compared to vehicle control group.Taken together, these data suggest that the pattern of Hoxa-7 erpression correlates with theabnormality caused by ter;3togens during the mouse embryogenesis.

These assay systems focus only on identifying the estrogenic or antiestrogenic activity of EDs, whereas a few studies have been carried out to screen the thyroid hormone (TH) disruptors. The aim of this study was to evaluate a test system to detect TH disruptors using rat pituitary tumor $GH_3$ cells. The test system is based on the TH-dependent increase in growth rate. As expected, L-3,5,3-triiodothyronine ($(T_3)$ markedly induced a morphological change in $GH_3$ cells from flattened fibroblastic types to rounded or spindle-shaped types. $T_3$ stimulated $GH_3$ cell growth in a dose-dependent manner with the maximum growth-stimulating effect being observed at a concentration $1{\times}10^9M$. In addition, $T_3$ increased the release of growth hormone and prolactin into the medium of the $GH_3$ cells culture. Using this assay system, the TH-disrupting activities of bisphenol A (BPA) and its related compounds were examined. BPA, dimethy/bisphenol A (DMBPA), and TCI-EP significantly enhanced the growth of $GH_3$ cells in the range of $1{\times}10^{-5}M\;to\;1{\times}10^{-6}M$ concentrations. In conclusion, this in vitro assay system might be useful for identifying potential TH disruptors. However, this method will require further evaluation and standardization before it can be used as a broad-based screening tool.

        A Preliminary Design of Flight Test Conditions for a Sub-scale RBCC Engine using a Sounding Rocket

        Various R&D programs for rocket-based combined cycle (RBCC) engines have progressed worldwide for the space development and the defense applications. As a way toward indigenous domestic RBCC program, a preliminary design of flight test conditions was studied in this study for a sub-scale RBCC engine using a sounding rocket. Launch and flight profiles were calculated for several booster options and compared with that of HyShot II program. The result shows that the Korea Sounding Rocket-II (KSR-II) is a proper candidate to perform the flight test available in Korea. The recommend flight test conditions with KSR-II are Mach 6.0 with a test vehicle of 230 kg and Mach 7.4 with 50 kg. Present study will soon be followed by a design of sub-scale RBCC for a flight test using a sounding rocket.
