        Induction of the second reproductive cycle and ovarian maturation, and changes of protein were investigated in the black blowfly, Phormia regina. A height in between hardboard and bottom of the experimental device based on the length of mouthpart and ovipositor was 2.5cm. Ovipositional rate of the first reproductive cycle in the experimental device was 81.2±6.1% without liver juice in the crop and midgut. During the second reproductive cycle terminal oocyte was 385, 977, 1121㎛ and ovary was 1732, 3819, 3946 ㎛ in length at 24, 48, and 60h after the second liver-feeding, respectively. Haemolymph protein increased 36h after the second liver-feeding and decreased thereafter while ovarian protein increased continuously until 60h in the second reproductive cycle. This result should be applicable as reference data for the embryo-physiological research of the second reproductive cycle in other Dipteran insects.

        본 연구의 목적은 바로끄 건축이 이룬 개념의 하나인 동적 표현의 관점을 통해서 그 건축에서 표현하거나 체험할 수 있는 동적의사들 속에서 그 표징을 밝히고, 그 표현의 개념과 원리를 체득함으로서 작금의 시대의사 속에 새로이 시사하는 바를 밝히고자 함에 있다. 따라서 이 논문은 두가지 접근으로 그 목적을 이룬다. 먼저 건축에 있어 동적표현을 개념하는 것이고, 다른 하나는 그 개념이 전개한 조형의 표징을 바로끄 건축 속에서 추적하는 것이다. 건축에 있어 동적 표현을 이루는 조형의사로 전개된 바로끄 건축에서 성과로 파동의 입면, 상호관입, 수축이완의 세가지 관점을 통해서 체득한다. The purpose of this research has tried to find the signs of dynamic intention which was expressed or experienced with a point of view of dynamic expression in Baroque architecture and has redefined dynamic intention of current thoughts in contemporary architecture by realizing the conceptions and principles of the expression. This thesis accomplishes its purpose with two points of view. One is conceptualizing the dynamic expression in architecture, the other is tracing the sign of formative arts which has developed with its concept in Baroque architecture. As a result, you may comprehend three points of view in architecture-wave of elevation, mutual infiltration, and oontraction/relaxation through Baroque architecture that has developed with the intention of formative arts on dynamic expression.

        Porphyromonas endodontalis의 침투에 따른 혈관 내피세포의 유전자 발현

        During the last two decades, there has been an increasing interest in the impact of oral health on atherosclerosis and subsequent cardiovascular disease (CVD). To date, some periodontal pathogens including Porphyromonas gingivalis (P. gingivalis) have been reported to be relevant to CVD. Porphyromonas endodontalis (P. endodontalis), which shares approximately 87% sequence homology with P. gingivalis, is mostly found within infected root canals. However, recent studies reveal that this pathogen also resides in the dental plaque or periodontal pocket in patients with periodontitis. It has been shown that P. endodontalis invades human coronary artery endothelial cells (HCAEC) and coronary artery smooth muscle cells (CASMC). To evaluate whether P. endodontalis can participate in the progression of atherosclerosis and CVD, we examined the changes in transcriptional gene expression profiles of HCAEC responding to invaion by P. endodontalis in this study. The following results were obtained. 1. Porphyromonas endodontalis was invasive of HCAEC. 2. According to the microarray analysis, there were 625 genes upregulated more than two-folds, while there were 154 genes downregulated by half. 3. Upregulated genes were relevant to inflammatory cytokines, apoptosis, coagulation and immune response. Enhanced expression of MMP-1 was also noticeable. 4. The transcription profiles of the 10 selected genes examined by real-time PCR agreed well with those observed in the microarray analysis. Thus, these results show that P. endodontalis presents the potential to trigger and augment atherosclerosis leading to CVD. 본 연구에서는 Porphyromonas endodontalis와 죽상경화증 및 심혈관질환과의 관계를 알아보기 위해, P. endodontalis가 사람의 관상동맥 내피세포에 침투했을 때 나타나는 유전자 발현의 변화를 microarray와 real-time PCR로 측정하였고, 발현이 증가된 유전자 중에서 죽상경화증과 연관된 유전자들이 관련 KEGG pathway 상에서 유의성 있는 영향을 미치는지를 분석하여 다음과 같은 결과를 얻었다. 1. Porphyromonas endodontalis는 사람의 관상동맥 내피세포에 침투하였다. 2. Microarray 분석결과, 대조군보다 발현이 2배 이상 증가된 유전자는 625개였고, 1/2이하로 감소된 유전자는 154개였다. 3. 발현이 2배 이상 증가된 유전자 중에는 염증성 cytokine 및 chemokine, 세포자멸사, 혈액응고와 면역반응 관련 유전자들이 포함되었다. 4. Microarray 분석결과를 확인하기 위해 발현이 2배 이상 증가된 유전자 중에서 10개의 유전자를 선택하여 real-time PCR을 시행하였고, 그 결과 microarray에서와 마찬가지로 발현 정도가 대조군보다 모두 높게 나타났다. 따라서 P. endodontalis가 사람의 관상동맥 내피세포에 만성적으로 작용했을 때, 심혈관질환에서 중요한 부분을 차지하는 죽상경화증을 유발하는 위험 요소 중의 하나로 작용할 수 있을 것으로 판단된다. 이와 관련된 자세한 기전을 이해하기 위해서는 앞으로 더 많은 연구가 필요할 것으로 보인다.

        It is important to interpret the soil chemical properties in order to effectively manage the farmland. This study was conducted to investigate the soil chemical properties of upland soybean field according to the soil series at 32 upland fields located in the parts of Gangwon-do. Soil sampling sites were selected by using the National Statistics DB and KSIS DB that is a soil environment information system provided by the Rural Development Administration. Soil samples were collected from 0~15 cm of top soil before seeding or transplanting, air-dried, passed through a 2 mm sieve, and analyzed for soil pH, EC, organic matter, available phosphorus (P2O5), exchangeable potassium, exchangeable calcium and exchangeable magnesium. The average chemical properties have exceeded the each nutrient optimum range level of RDA, except the organic matter in experimental soil 2015. And despite application of conventional fertilization, each soil series nutrient contents were change respectively. The results of this study suggest that conventional fertilization should be applied based on the analysis about the amount of accumulated nutrient contents in the soil.

        Clinical and molecular characterizations of novel POU3F4 mutations reveal that DFN3 is due to null function of POU3F4 protein.

        <P>X-linked deafness type 3 (DFN3), the most prevalent X-linked form of hereditary deafness, is caused by mutations in the POU3F4 locus, which encodes a member of the POU family of transcription factors. Despite numerous reports on clinical evaluations and genetic analyses describing novel POU3F4 mutations, little is known about how such mutations affect normal functions of the POU3F4 protein and cause inner ear malformations and deafness. Here we describe three novel mutations of the POU3F4 gene and their clinical characterizations in three Korean families carrying deafness segregating at the DFN3 locus. The three mutations cause a substitution (p.Arg329Pro) or a deletion (p.Ser310del) of highly conserved amino acid residues in the POU homeodomain or a truncation that eliminates both DNA-binding domains (p.Ala116fs). In an attempt to better understand the molecular mechanisms underlying their inner ear defects, we examined the behavior of the normal and mutant forms of the POU3F4 protein in C3H/10T1/2 mesodermal cells. Protein modeling as well as in vitro assays demonstrated that these mutations are detrimental to the tertiary structure of the POU3F4 protein and severely affect its ability to bind DNA. All three mutated POU3F4 proteins failed to transactivate expression of a reporter gene. In addition, all three failed to inhibit the transcriptional activity of wild-type proteins when both wild-type and mutant proteins were coexpressed. Since most of the mutations reported for DFN3 thus far are associated with regions that encode the DNA binding domains of POU3F4, our results strongly suggest that the deafness in DFN3 patients is largely due to the null function of POU3F4.</P>

      • Resistant Mechanism of Escherichia coli to HK3140-A New Fluoroquinolone

        In our study, the resistant mechanism of Escherichia coli to a new fluoroquinolone-HK3140 (Kim et al., 1991) was characterized. In the presence of carbonylcyanide m-chlorophenyl hydrozone (CCCP), HK3140 uptake was increased and in vivo DNA synthesis was inhibited. This result suggested that both susceptible and resistant strains have efflux systems which are proton gradient dependent. The resistant strain had another efflux system which was ATPase dependent. DNA synthesis of the permeabilized resistant cells was less inhibited by HK3140 than that of the permeabilized susceptible cells. Results suggested that the resistant mechanism against HK3140 came from an ATPase dependent efflux system and changes in DNA gyrase and porin proteins (Lee et al., 1992).

      • ROSPath : An Intergrated Database for Reactive Oxygen Species-mediated Signaling Pathway

        Progress in understanding complex signaling pathways and networks has been hampered by the current lack of a formal and structured collation of the available information, in a format suitable for analysis via software tools. In order to facilitate the organization. and understanding of such complex network of information, it is essential to have a formal means to represent and analyze cellular pathways. We have defined a formal ontology for cell-signaling events that allows us to describe these cellular pathways at various levels of abstractions. Using this formal representation, ROSPath(Reactive Oxygen Species related signaling Pathway) database system has been implemented and made available on the web(http://rospath.ewha.ac.kr). ROSPath is a database system for reactive oxygen species(ROS)-mediated cell signaling pathways and signaling processes in molecular detail, that facilitates a comprehensive understanding of the regulatory mechanisms in signaling pathways. ROSPath includes growth factor-, stress- and cytokine-induced signaling. It is a web-based structured repository of information on the signaling pathways of interest and provides a means for managing data produced by high-throughput tools such as proteomics and genomics. It also provides effective and flexible tools for querying, displaying and analyzing pathways, thus providing an integrated web environment for visualizing and manipulating ROS-mediated cell-signaling events.

        Comparison of Two Apheresis Systems of COBE and Optia for Autologous Peripheral Blood Stem Cell Collection

        <P>Peripheral blood stem cell (PBSC) transplantation following myeloablative therapy is a mainstay of treatment for various types of malignancies. This study aimed to evaluate the differences between the Optia MNC and COBE Spectra MNC systems (Terumo BCT, Japan) according to apheresis procedures and the parameters of apheresis, products, and collection. The clinical data of 74 patients who underwent autologous PBSC collection from July 2012 to July 2015 were reviewed retrospectively. The patients comprised 48 (65%) men and 26 (35%) women with a median age of 56 yr (range, 23–66 yr). Of 216 procedures, 111 (51%) and 105 (49%) were processed by using COBE and Optia MNC, respectively. PBSC collection rates, throughput, numbers of stem cells retrieved, collection efficacy, and platelet loss were compared. There were no significant differences in the median CD34+ cell counts of collected products (0.61×10<SUP>8</SUP> vs 0.94×10<SUP>8</SUP>), CD34 collection efficiency (43.5% vs 42.1%), and loss of platelets (40.1% vs 44.7%). The Spectra Optia MNC apheresis system was comparable to the COBE Spectra system in collecting autologous CD34+ hematopoietic stem cells and retention of platelets.</P>
