Enhanced adipogenic differentiation and reduced collagen synthesis induced by human periodontal ligament stem cells might underlie the negative effect of recombinant human bone morphogenetic protein‐2 on periodontal regeneration. J Periodont Res 2011; 46: 193–203. © 2010 John Wiley & Sons A/S</I> </P><P><B>Background and Objective: </B> Recombinant human bone morphogenetic protein‐2 (rhBMP‐2) is a potent inducer for the regeneration of mineralized tissue, but has a limited effect on the regeneration of cementum and periodontal ligament (PDL). The aim of the present study was to determine the effects of rhBMP‐2 on the <I>in vitro</I> and <I>in vivo</I> biologic activity of well‐characterized human PDL stem cells (hPDLSCs) and to elucidate the underlying mechanism of minimal periodontal regeneration by rhBMP‐2.</P><P><B>Material and Methods: </B> hPDLSCs were isolated and cultured, and then transplanted into an ectopic subcutaneous mouse model using a carrier treated either with or without rhBMP‐2. Comprehensive histologic, histometric and immunohistochemical analyses were performed after an 8‐wk healing period. The effects of rhBMP‐2 on the adipogenic and osteogenic/cementogenic differentiation of hPDLSCs were also evaluated. The effect of rhBMP‐2 on both soluble and insoluble collagen synthesis was analyzed, and the expression of mRNA and protein for collagen types I, II, III and V was assessed.</P><P><B>Results: </B> In the present study, rhBMP‐2 promoted both adipogenic and osteogenic/cementogenic differentiation of hPDLSCs <I>in vitro</I>, and the <I>in vivo</I> potential of hPDLSCs to form mineralized cementum and organized PDL tissue was down‐regulated following treatment with rhBMP‐2. Collagen synthesis, which plays a crucial role in the regeneration of cementum and the periodontal attachment, was significantly reduced, with associated modification of the relevant mRNA and protein expression profiles.</P><P><B>Conclusion: </B> In summary, the findings of the present study suggest that enhanced adipogenic differentiation and inhibition of collagen synthesis by hPDLSCs appear to be partly responsible for the minimal effect of rhBMP‐2 on cementum and PDL tissue regeneration by hPDLSCs.</P>

<P>A highly pathogenic avian influenza (HPAI) H5N8 virus was first detected in poultry and wild birds in South Korea in January 2014. Here, we determined the pathogenicity and transmissibility of three different clades of 1-15 viruses in mandarin ducks to examine the potential for wild bird infection. H5N8 (Glade 2.3.4.4) replicated more efficiently in the upper and lower respiratory tract of mandarin ducks than two previously identified H5N1 virus clades (clades 2.2 and 2.3.2.1). However, none of the mandarin ducks infected with H5N8 and H5N1 viruses showed severe clinical signs or mortality, and gross lesions were only observed in a few tissues. Viral replication and shedding were greater in H5N8-infected ducks than in H5N1-infected ducks. Recovery of all viruses from control duck in contact with infected ducks indicated that the highly pathogenic H5 viruses spread horizontally through contact. Taken together, these results suggest that H5N8 viruses spread efficiently in mandarin ducks. Further studies of pathogenicity in wild birds are required to examine possible long-distance dissemination via migration routes. (C) 2016 Elsevier B.V. All rights reserved.</P>

<P>The deposition of high-quality SiO2 films has been achieved through the use of both plasma-enhanced chemical vapor deposition (PE-CVD) and plasma-enhanced atomic layer deposition (PE-ALD) methods using H2Si[N(C2H5)(2)](2) as a Si precursor. We systematically investigated growth characteristics, chemical compositions, and electrical properties of PE-CVD SiO2 prepared under various deposition conditions. The SiO2 films prepared using PE-CVD showed high purity and good stoichiometry with a dielectric constant of similar to 4. In addition, the PE-ALD process of the SiO2 films exhibited well-saturated and almost linear growth characteristics of similar to 1.3 cycle(-1) without notable incubation cycles, producing pure SiO2 films. Electrical characterization of metal-oxide silicon capacitor structures prepared with each SiO2 film showed that PE-ALD SiO2 films had relatively lower leakage currents than PE-CVD SiO2 films. This might be a result of the saturated surface reaction mechanism of PE-ALD, which allows a smooth surface in comparison with PE-CVD method. In addition, the dielectric properties of both SiO2 films were further evaluated in the structures of In-Ga-Zn-O thin-film transistors, and they both showed good device performances in terms of high I (on) - I (off) ratios (> 10(8)) and low off-currents (< 10(-11) A). However, based on the negative bias stress reliability test, it was found that PE-ALD SiO2 showed better reliability against a negative V (th) shift than PE-CVD SiO2, which might also be understood from its smoother channel/insulator interface generation at the interface.</P>

The effects of CO2 addition on the downstream interactions of H2-air and CO-air premixed flames were investigated. Flame stability diagrams mapping the extinction limits for various fuel concentrations as functions of the strain rate were developed for the downstream interactions between H2-air and CO-air premixed flames with 0-20% CO2 addition. Of particular concern was the impact of the chemical reactions of the additional CO2 on the flame extinction characteristics. The results showed that CO2 addition suppressed the flammable regions appreciably and decreased the critical strain rate, opening up the lean extinction boundary. For highly stretched interacting flames, two flammable island regions on the maps were combined into one when 10% or 20% CO2 was added to either the H2-air or CO-air premixed mixtures. The chemical reactions of the additional CO2 were significant, especially for high strain rate flames, while the effect of the radiative heat loss of the additional CO2 was minor, even for low strain rate flames. Such chemical reactions were determined by competition between the reaction steps for the consumption and production of HCO and the reverse reaction of CO+OH=CO2+H with the reaction step of H+O2→O+OH, an indicator of the overall reaction rate for the H atoms. This resulted in a weakened interacting flame. Such chemical reactions of the additional CO2 were examined in detail to describe the flame characteristics.

<P>In the development of three-dimensional printable materials for high-speed and high-resolution printing, UV-curing polymers can guarantee fast and precise printing of high performance load-bearing structures, but the injected drops of the monomers tend to spread over the substrates due to their low viscosity. In this study, we imposed the self-standing and shape-memorable capability of an epoxy acrylate (EA) monomer to ensure continuous filamentary 3D printing while maintaining its low viscosity nature. Using octadecanamide (ODA) with EA, strong hydrogen-bond networks (−N−H⋯OC−, −N−CO⋯H-O-, -N-H⋯N-) were additionally achieved in the material system and the developed material distinctively exhibited rheological duality at different processing stages: a low-viscosity liquid-like behavior (viscosity of ∼50 Pa) while passing through the nozzle and a self-standing solid-like behavior (static yield stress of ∼364 Pa) right after being printed. This reversible liquid-to-solid transitional capability was quantified by viscoelastic complex moduli provided a dynamic yield stress (<I>τ</I>y,G) of 210 Pa corresponding to the upright stacking up to ∼3.2 cm (3 wt% of ODA). The time (<I>t</I>y,G) required for conformational rearrangement was evaluated to be as fast as ∼10−2 s. After UV curing, the 3D printed layers exhibited no air pockets or weld lines at the stacked interfaces, which could guarantee excellent mechanical performance and structural integrity.</P>

<P>It is a challenge to develop methodologies involving multiple transformations for preparing new materials that cannot be obtained via direct synthesis. Herein, we report an anion exchange process accompanying cation reduction and dimerisation via a single-crystal-to-single-crystal transformation. First, a direct reaction of the flexible tripyridyl dithioether ligand L with CuI afforded a mixture of four bis(ligand) complexes (1a-1d). To avoid the formation of undesired mixed products, a copper(II) nitrate complex-mediated approach involving anion exchange and cation reduction was employed to generate a monomeric complex, [Cu-II(L)NO3]NO3 center dot toluene (2). When the dark blue crystals of 2 were immersed in an aqueous NaI solution, the crystals were transformed to a pale yellow dimeric copper(I) iodide complex, [(mu-CuI2+I2)(L)(2)] (3). The observed anion exchange promotes the reduction of copper(II) to copper(I) at the expense of I-/I-3(-) oxidation as well as dimerisation via the formation of a Cu2I2 cluster. This result corresponds to the synthesis of a compound that otherwise was not able to be prepared via a direct synthetic procedure.</P>

<P>Summary</P><P>Enterotoxin produced by enterotoxigenic <I>Bacteroides fragilis</I> (BFT) has been associated with mucosal inflammation and diarrhoeal diseases. In this study, the anti-inflammatory molecular mechanism of 5,7-dihydroxy-3,4,6-trimethoxyflavone (eupatilin) was characterized in an HT-29 intestinal epithelial cell line stimulated with BFT. Pre-treatment of HT-29 cells with eupatilin decreased the production significantly of both interleukin (IL)-8 and prostaglandin E2 induced by BFT in a dose-dependent manner. BFT-activated nuclear factor-kappaB (NF-&kgr;B) signals in HT-29 cells and pretreatment with eupatilin suppressed NF-&kgr;B activation that resulted in the significant inhibition of IL-8 and cyclo-oxygenase-2 expression. BFT-induced phosphorylation of both I&kgr;B&agr; and I&kgr;B kinase (IKK) signals was prevented in eupatilin-pretreated HT-29 cells. Transfection of siRNA for IKK-&agr; and IKK-&bgr; decreased the production of IL-8 and prostaglandin E2; however, the transfection of IKK-&bgr; siRNA showed a more significant reduction of BFT-induced I&kgr;B&agr; phosphorylation compared with that of IKK-&agr; siRNA. In addition, herbimycin A, a specific inhibitor of heat shock protein 90 (Hsp90), decreased the BFT-induced activation of IKK and NF-&kgr;B, suggesting that Hsp90 is associated with a pathway of IKK-NF-&kgr;B-IL-8/cyclo-oxygenase-2 gene signalling. Furthermore, eupatilin dissociated the complex between Hsp90 and IKK-&ggr; in BFT-stimulated HT-29 cells. These results suggest that eupatilin can suppress the NF-&kgr;B signalling pathway by targeting the Hsp90-IKK-&ggr; complex in intestinal epithelial cells and may attenuate BFT-induced inflammatory responses.</P>
