        돈육의 위생화와 품질보존을 위한 감마선 조사

        이상의 결과로 돼지고기는 비교적 미생물 오염도가 낮아 1 kGy조사로 저온성 미생물을 제외한 오염미생물의 생육을 검출한계 이하로 감소시킬 수 있었으며 모든 오염미생믈의 살균을 위해서는 3kGy의 선량이 요구되었다. 감마선조사와 저장기간에 따른 일반성분 변화의 경우 수분함량에 있어서만이 저장기간에 따라 다소 감소되었으며, pH 및 산도는 감마선 조사에 의한 영향은 없었으나 저장기간이 경과함에 따라 pH는 증가하고 산도는 감소하는 경향을 보였다. 총아미노산 조상에서는 가장 많이 검출된 것이 glutamic acid였고, 감마선 조사에 의한 영향도 매우 미비하였다. 무기질 함량의 경우 주요 무기질 성분으로 potassium, phosphorus, sodium, magnesium 순이었고 감마선 조사에 의한 조성변화는 없었다. Meat is a perishable product since it offers favorable conditions for microbial contamination and multiplication. Besides, undercooked and raw meat has been linked to outbreaks of hemorrhagic diarrhea due to the presence of microbial pathogens. Therefore, this study was conducted to find out the effect of the use of gamma-irradiation in order to improve preservation and eliminate this microorganisms. The initial level of microbial contamination in pork was 1.1×10⁴ CFU/g (aerobic bacteria), 5.3×10³ CFU/g (psychrophile), 3.1×10³ CFU/g (coliforms) and 3.2×10³ CFU/g (Listeria), All kinds of contaminated microorgnisms were sterilized by gamma irradiation at 3 kGy. The proximate composition of pork was not significantly changed by irradiation dose and storage period. The pH was slightly increased during storage period and titratable acidity decreased. However, no significant changes in pH and acidity were observed by gamma irradiation. There was no difference in total amino acid content regardless of irradiation dose and storage period. Glutamic acid was detected at high levels throughout the storage period. The major mineral compositions of pork were potassium, phosphorus, sodium, magnesium. No significant difference in the components of minerals were observed by gamma irradiation.

        Purposes : To investigate the viral etiology of community-acquired pneumonia in Korean adults, we have detected respiratory viruses (Respiatory syncytial virus, adenovirus, influenza virus and parainfluenza virus) in the way of prospective, multi-center study. Methods : From July 1997 to April 2000, nasal aspirates or sputum were obtained from adults patients with community pneumonia admitted to the participating hospitals and transferred immediately to the central laboratory at the Seoul National University Children's Hospital. The specimens were divided into three parts. One part was used for indirect immunofluorescent test for respiratory viruses, the other part for the culture of RSV and adenovirus in HEp-2 cell monolayer. The other part was used for the culture of influenza virus and parainfluenza virus in MDCK or LLC-MK2 cell monolayers. Results : Of 317 samples, 32 (10.1%) specimens were positive for viral agent by indirect IF staining or culture, including one dual-infected specimen (adenovirus and parainfluenza virus). Influenza virus was most commonly detected (16 specimens). Parainfluenza virus, adenovirus and RSV were detected in 10, 4 and 3 patients, respectively. All isolated influenza viruses were type A (H3N2 in 9 patients, HIN1 in 2 and unspecified in 5), and 8 out of 10 parainfluenza virus isolates were type 3. Conclusion : Similar to previous reports from other countries, a significant portion of community-acquired pneumonia in Korean adult is caused by respiratory viruses.         겸상적혈구 혈증에 의한 동통성 발작 1례

        The sickle cell disease are a group of hemoglobin disorders characterized by red cells that undergo sickle shape transformation when they are deoxygenated. Sickle cell disease is transmit-ted as an autosomal recessive trait. This unusual property, due to the polymerization of sickle hemoglobin results in anemia and vasoocclusive complication. 1) The most clinically significant of these disease are sickle cell anemia, sickle cell hemoglobin C disease, and sickle cell beta thalassemia. Symptoms of pallor, fever, abdominal and joint pain, enlargement of the liver and spleen, swelling of hands and feet first appear near the latter part of the first year of life. Intravascular sickling affects all organs. For clinical and therapeutic purposes, exacerbations may be classified as vasoocclusive or pain, aplastic, hemolytic or sequestration crisis. we experienced a 22 year old female patient who suffered severe multiple joint pain and back pain thought to be caused by vasoocclussive phenomena.

      • Flowering time is repressed by sumoylation of FLC

        The transition from vegetative growth to flowering is a major developmental switch in the plant cycle and the timing of flowering is very critical for reproduction of plant species. In transition to flowering in plants, Flowering locus C (FLC) is one of the crucial factors. Here, we showed How the stability and activity of FLC are regulated by sumoylation mechanism. By pull-down assay, we showed that FLC interact with E3 SUMO ligase in vitro and vivo. And we showed that FLC is sumoylated in vitro condition with AtSUMO1 protein. In transgenic plants with overexpression of FLC and inducible expression of AtSIZ1, sumo E3 ligase led to increase of FLC protein level and delayed the post-translation degradation of FLC indicating that Arabidopsis E3 sumo ligase AtSIZ1 stabilizes FLC. Also, the plants with overexpression of mutant FLC (K154R, a mutation of the sumoylation site on FLC) flowered considerably earlier than plants with overexpression of FLC but comparable with wild type indicating that sumoylation is a important part for function of FLC. Our data indicate that the sumoylation of FLC is critical for its role in the control of flowering time.

        Nitric oxide {NO} laduces apoptotic cell death In murine RAW 264.7 macrephages. To elucidate the inhlabitory effects of protein kinmac C (PKC) on NO-lmdwced apoptoles, we generated clones of RAW 264.7 cells that overexpress one of the PKC looforma and explered the powble Interactions between PKC and three stractarally related metegen-activated preblem (MAP)kiness In NO actiens. Treatment of RAW 264.7 cells with acdium mkrepruside (SNP), a NO tenerating agent , activated both c-Jun N-terminal kinesseltress-activated pretein kinase (JNK/SAPK) and p38 kinase, but did not activate extracellular signal-regulated kinase (ERK)-1 and ERK-2 In additlen, SNP-imduced apopteels was mightly blocked by the selective p-38 kinesse Inhtblter (SB203580)but not by the MAP?ERK1 kiness Inbibitor (PD090059). PKC tranerrectants (PKC-βⅡ, -&, and -n) showed substantial pretection from cell deeth Induced by the expreture to NO downs such as SNP and S-nitroglutathiese (GSNO). In contract, in RAW 264.7 parent or in empty vecter-tracformed cells, these NO demmors imdneed imternucleosomal DNA cleavage. Mereover, overexpression of PKC isofornas signiflcammtly auppressed SNP-induced JNK/SAPK and p38 kinase activation, but did not affect ERK-1 and -2. We also explored the invelvement of CPP32-like preteose In the NO-INduced apeptoceds. Inhibithion of CPP32-like proteose prevented apoptions in RAWW 267.7 pareced cells In addition, SNP dramntically activated CPP32 in the parent or in empty vector-transformed cells, white slightly activated CPP32 in PKC transfectants. Therefore, we conclude that PKC protects NO-induced apoptotic cell death, preammably maMKying the NO-mediated activation of JNK/SAPK, p38 kinese, and CPP32-like protease in RAW 267.7 macrephages.

        Inhibitors Targeting ABA Biosynthesis and Catabolism Can Be Used to Accurately Discriminate between Haploid and Diploid Maize Kernels during Germination

        There is a growing preference for using doubled haploids (DHs) in maize breeding programs because they reduce the time required to generate and evaluate new lines to 2 years or less. However, there is an urgent need for efficient techniques that accurately discriminate between haploid and diploid maize kernels. Here, we investigate the effects of several hormones and chemicals on the germination of haploid and diploid maize kernels, including auxin, cytokinin, ethylene, abscisic acid (ABA) biosynthesis inhibitor (fluridone), ABA catabolism inhibitor (diniconazole), methyl jasmonate (MeJA), and NaCl. Ethylene effectively stimulated the germination of both haploid and diploid maize kernels. The ABA biosynthesis inhibitor fluridone, the ABA catabolism inhibitor diniconazole, and MeJA selectively stimulated the germination of haploid maize kernels. By contrast, gibberellin, 1-naphthaleneacetic acid (NAA), kinetin, and NaCl inhibited the germination of both haploid and diploid maize kernels. These results indicate that the germination of haploid maize kernels is selectively stimulated by fluridone and diniconazole, and suggest that ABA-mediated germination of haploid maize kernels differs from that of diploid maize kernels and other plant seeds.

      • CFD Modeling of a Thermally Efficient Modular Reactor for Fischer–Tropsch Synthesis: Determination of the Optimal Size for Each Module

        <P>A modular multichannel reaction module with microchannel-based coolant channels was considered, and a computational fluid dynamics (CFD) model was developed to describe the hydrodynamic behavior of the module. Reaction rates for the lumped chain length distribution of hydrocarbon products generated by the Fischer-Tropsch synthesis reaction were proposed, and the developed kinetic and CFD models were shown to satisfactorily fit the experimental data under different production rates. High heat transfer rates resulting from the use of microchannel-based cooling channel maintained the temperature peak below 10 degrees C, and simulation results with increased size of the catalytic bed and absence of inert materials showed that the high heat of reactions could be efficiently removed over entire catalytic beds, preventing the creation of local hot spots, which are usually observed in conventional fixed bed reactors. In addition, the efficient use of thermal energy could guarantee that methane selectivity, which needs to be maintained as low as possible, was close to approximately 10% under all conditions, while the selectivity of the desired hydrocarbons (C5+) slightly increased with increasing feed flow rates.</P>

        한국형 노인우울검사 보호자용(Collateral Source Version of the Korean Form of Geriatric Depression Scale:CS-KGDS)의 타당도 연구

        In order to examine elderly depression in the elderly that too weakened cognitively, and physically to check depressive symptoms reliably, the authors deviced collateral source version of elderly depression scale and tested it's validity. Sixty elderly depression patients and forty-eight old person in normal group completed the KGDS (Korean Form of Geriatric Depression Scale), and their collateral source (CS) completed a CS version of the KGDS (CS-KGDS). The differences of CS-KGDS means was signified at α =.001 level (patients group mean=18.33 (4.71), control group mean=7.60 (5.25), t=11.18). The values of Cronbach's α and Split-half reliability were .88 and .81 respectively, and correlational coefficent with KGDS was .68. On factor analysis, 5 factors were extracted. They were labeled ‘Physical weakening and decreased vitality’(factor 1), ‘Emotional discomfort’(factor 2), ‘Negative thinking and Unhappiness feeling’(factor 3), ‘Cognitive dysfunction’(factor 4), ‘Decreased social interest and activity’(factor 5), which congruous with internal structure of KGDS. The result of discrimination analysis showed 87.09% of hit ratio, and suggested the score of 13 as optimal cut-off score. This cut-off score was identical to that of KGDS. Conclusively, CS-KGDS could be a useful tool for evaluaion of elderly depression instead of KGDS in such case that KGDS can not be administered reliably. 치매가 의심될 정도로 인지적, 신체적으로 약화된 노인들에서 우울증을 효율적으로 검사하고자 보호자용을 만들어 신뢰도와 타당도를 검증하고자 하였다. 연구대상은 노인우울증 환자 60명, 신체적으로 건강하고 정신과적 질환이 없는 정상노인 48명과 그 보호자들이었다. 노인들에게는 KGDS(Korean form of Geriatric Depression Scale)을 실시하였고 보호자들에게는 KG DS의 보호자용(CS-KGDS;Collateral Source Version of KGDS)을 실시하였다. 연구 결과, CS-KG DS는 KGDS와 마찬가지로 우울증 환자집단[Mean(SD)=18.33(4.71)]과 정상집단[Mean(SD)=7.60(5.25)]에서 유의하게 점수의 차이를 보였다(t=11.18, p<.001). KGDS와 높은 상관을 보였고(r=.68, p<.001) 내적일치도(α=.88), 반분신뢰도(.81) 모두 만족스러운 수준이었다. 요인분석결과'신체적 약화와 활력감소','정서적 불편감','부정적 사고 및 불행감','인지적 기능저하','사회적 관심과 활동저하'의 5개 요인이 밝혀져 KGDS의 5개 요인과 동일한 내적 구조를 보였다. 판별분석결과 KGDS와 전체 판별률은 87.09%였고 최적절단점수는 KGDS와 같은 13점으로 밝혀졌다. 이상에서 볼 때, CS-KGDS는 기능이 좋지않아 검사를 실시하기 어려운 노인들의 우울증을 평가할 때 대체용 검사로 신뢰롭게 쓰일 수 있을 것으로 생각된다.
