        牧楡湯이 人體 肺癌細胞의 變化에 미치는 影響에 關한 分子生物學的 硏究

        This experimental study was performed to investigate the effects of Mokyou-Tang on cell cycle and gene expressions of human lung cancer cell. I examined the cell cycle progression and cell cycle-related gene expression in A549 using a flow cytometry and a quantitative PT-PCR analysis. The treatment groups, in accordance with concentration of Mokyou-Tang were classified as 3 group(MYT 100㎕/㎖, MYT 500㎕/㎖, MYT 1,000㎕/㎖), and according to treatment time, were classified as 2 group(24hours, 48hours). The results were summarized as follows; 1. In cell cycle stage, MYT 100㎕/㎖(48hours) and 1000㎕/㎖(48hours) groups were increased as compared with control group, especially 1000㎕/㎖(48hours) group showed eminent increase. 2. In p53 gene, MYT 500㎕/㎖(48hours) and 1000㎕/㎖(48hours) groups showed eminent suppressive effect to cancer cell. 4. CDKNI gene was decreased in all treatment groups, this mean that MYT has suppressive effect to proliferation of cancer cell. 4. In GADD45 gene was decreased in all treatment groups, this mean that MYT has suppressive effect to proliferation of cancer cell. The results suggest that MYT promotes G1 cell cycle arrest and suppresses the proliferation of cancer cell. Thus, it seems likely that MYT has suppressive effect to proliferation of lung cancer cell.

      • 都市街路의 遲滯度推定에 관한 硏究

        This study suggested input variables whose effects have been considered by adjusting various input variables in the TRANSYT-7F and NETSIM, the Simulation programs employed in calculating the delay time at street sections. The results can be summarized as follow. 1. Among the input variables in the TRANSYT-7F needed to calculate the delay time at street sections, the excluded are those which effect the most on time delay, such as bus stops, pedestrian crossings and on/off ramps. Among the input variables the most affecting factors are Saturation flow rate and time delay whose suggested quotients are well summarized in the text of this study. 2. There are variables which affect the change of time delay in the NETSIM, such as bus stop, pedestrian crossing, bus appearance period, and departure time delay. This study did the Simulation program using those variables except pedestrian crossing because, as the result of the correlation analysis suggests, there is a strong correlation between bus stop and pedestrian crossing. In this study, the survey had been done in the morning and afternoon except the Peak time, so heavily congested sections were excluded. And both sides of three street sections were surveyed. Therefore, there should be further studies on heavily congested situations and many other street sections.

      • 고립성 폐결절의 조직학적 진단에 있어서 경기관지폐생검의 진단적 가치

        Transbronchial lung biopsy(TBLB) is a relatively simple and convenient diagnostic tool in solitary pulmonary nodule(SPN) on chest radiograph, which is cannot be diagnosed through routine tests. To investigate the diagnostic value and diagnostic rate of TBLB according to the variable factor, we analyzed 69 patients with radiologically SPN lesion on chest radiograph which was confirmed histologically. A histologic diagnosis of TBLB was made in 47 of 69 patients(68.1%), in which 26 of 37(70.3%) with malignancy and 21 of 32(65.6%) with benign lesion. There were no significant differences in diagnostic yield according to age, sex and location of the lesion. The diagnostic rate tended to increase with the more than 5 pieces to acquired by TBLB. When the size of SPN was less than 3 ㎝, it cannot be diagnosed. In conclusion, TBLB is a diagnostic procedure with good yield and safety, but percutaneous needle aspiration, transbronchial needle aspiration, and bronchial washing fluid cytology may be a complementary procedure to overcome the limitation of TBLB and get higher diagnostic yield for SPN.

        Development of Microstrain in Aged Lithium Transition Metal Oxides

        <P>Cathode materials with high energy density for lithium-ion batteries are highly desired in emerging applications in automobiles and stationary energy storage for the grid. Lithium transition metal oxide with concentration gradient of metal elements inside single particles was investigated as a promising high-energy-density cathode material. Electrochemical characterization demonstrated that a full cell with this cathode can be continuously operated for 2500 cycles with a capacity retention of 83.3%. Electron microscopy and high-resolution X-ray diffraction were employed to investigate the structural change of the cathode material after this extensive electrochemical testing. It was found that microstrain developed during the continuous charge/discharge cycling, resulting in cracking of nanoplates. This finding suggests that the performance of the cathode material can be further improved by optimizing the concentration gradient to minimize the microstrain and to reduce the lattice mismatch during cycling.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/nalefd/2014/nalefd.2014.14.issue-8/nl5022859/production/images/medium/nl-2014-022859_0012.gif'></P>

        Influence of Anion and Mole Ratio on the Coordination Behavior of an NO<sub>2</sub>S<sub>3</sub>-Macrocycle: The Formation of a Dumbbell-Shaped Macrocyclic Cadmium(II) Iodide Complex

        <P>Anion and mole ratio dependent formations of cadmium(II) complexes with an NO2S3-macrocycle (L) incorporating a pyridine subunit are reported. When the cadmium(II) salts (1-10 equiv) with different halide ions (Br- or I-) were reacted with L, CdBr2 afforded a monomer complex, [Cd(L)Br](2)[Cd2Br6]center dot CH2Cl2 (1), with three separated parts in the whole mole ratio range: two 1:1 stoichiometric complex cation parts and one Cd2Br6 cluster anion part. After separation of 1 by filtration, [Cd(L)Br](2)[CdBr4]center dot CH2Cl2 (2) with similar composition was afforded, except the cluster was isolated from the filtrate. Unlike the CdBr2 complexation, CdI2 afforded the mole ratio dependent products (3-5). Below 2.5 equiv of CdI2, [Cd(L)I](2)[CdI4]center dot CH2Cl2 (3) and [Cd(L)I](2)[Cd2I6].0.5CH(2)Cl(2) (4) with different cadmium(II) iodide clusters were isolated as kinetic (3) and a thermodynamic (4) products. Notably, the use of 3 equiv or above amount of CdI2 gave a dumbbell-shaped complex, {[Cd(L)](2)(mu-Cd4I12)} (5), in which two mononuclear macrocyclic complex units are linked by a (mu-Cd4I12)(4-) cluster. To monitor the mole ratio dependency as well as their reactivities, the systematic powder X-ray diffraction (PXRD) analysis has also been applied.</P>

        Genetic Analyses of the Chimeric <i>CYP11B1</i> / <i>CYP11B2</i> Gene in a Korean Family with Glucocorticoid-Remediable Aldosteronism

        <P>Glucocorticoid-remediable aldosteronism (GRA) is an autosomal-dominant inheritable form of hyperaldosteronism with early onset hypertension. GRA is caused by unequal crossing-over of the steroid 11β-hydroxylase (<I>CYP11B1</I>) and aldosterone synthase (<I>CYP11B2</I>) genes. As a result of chimeric gene duplication, aldosterone is ectopically synthesized in the adrenal zona fasciculata under the control of adrenocorticotropin. Here, we describe three cases of GRA in a Korean family. The proband-a 21-yr-old female-was incidentally found to have high blood pressure (170/108 mmHg). Her 46-yr-old father had been treated twice for cerebral hemorrhage at the ages of 29 and 39 yr. Her 15-yr-old brother had a 2-yr history of hypertension; however, he was never treated. Their laboratory test results showed normokalemia, hyporeninemia, hyperaldosteronism, and a high plasma aldosterone concentration-to-plasma renin activity ratio. Normal saline loading failed to suppress aldosterone secretion. However, dexamethasone administration effectively suppressed their plasma aldosterone concentrations. Following genetic analyses with PCR and direct sequencing to document the chimeric gene and crossover site, respectively, we identified <I>CYP11B1</I>/<I>CYP11B2</I> and determined the breakpoint of unequal crossover to be located between intron 2 of <I>CYP11B1</I> and exon 3 of <I>CYP11B2</I>.</P>

      • Bone Mineral Density in Prediabetic Men

        <P><B>Background</B></P><P>There are many studies regarding the effects of insulin on bone metabolism and changes in bone mineral density (BMD) in the setting of diabetes. The effect of prediabetes on BMD is not known.</P><P><B>Methods</B></P><P>A total of 802 men participated in the Korea Rural Genomic Cohort Study (in Geumsan County). According to the results of an oral glucose tolerance test, subjects were classified into normal, prediabetic, and diabetic categories. One hundred twenty-four subjects diagnosed with type 2 diabetes were excluded, leaving 678 subjects for the study inclusion. BMD was estimated with a quantitative ultrasonometer.</P><P><B>Results</B></P><P>The average BMD T scores of normal and prediabetic subjects were -1.34 ± 1.42 and -1.33 ± 1.30, respectively; there was no significant difference in the BMD T scores between these groups. The BMD T score was inversely associated with age and positively correlated with body weight, body mass index, total cholesterol, low density lipoprotein cholesterol, and HbA1c. On multiple linear regression analysis, low density lipoprotein cholesterol was the only statistically significant variable for prediabetes (β = 0.007, <I>P</I> = 0.005). On the stepwise regression analysis, age (β = -0.026, <I>P</I> < 0.001), the body mass index (β = 0.079, <I>P</I> < 0.001), and low density lipoprotein cholesterol (β = 0.004, <I>P</I> = 0.016) were significant variables for prediabetes.</P><P><B>Conclusions</B></P><P>There was no significant difference in the BMD T score between the normal and prediabetic subjects. Further studies are needed regarding the association of fracture risk and changes in BMD with the development of overt diabetes.</P>
