      • Induction of heme oxygenase-1 protects against podocyte apoptosis under diabetic conditions

        Heme oxygenase-1 (HO-1) is an anti-oxidant enzyme normally upregulated in response to oxidant injury. Here we determined the role of HO-1 in podocyte apoptosis in glomeruli of streptozotocin-treated rats and in immortalized mouse podocytes cultured in media containing normal or high glucose. HO-1 expression, its activity, the ratio of Bax/Bcl-2 protein, and active caspase-3 fragments were all significantly higher in isolated glomeruli of diabetic rats and in high glucose–treated podocytes. These increases were inhibited by zinc protoporphyrin treatment of the rats or by HO-1 siRNA treatment of the podocytes in culture. The number of apoptotic cells was also significantly increased in the glomeruli of diabetic rats and in high glucose–treated podocytes. Inhibition of HO-1 accentuated the increase in apoptotic cells both in vivo and in vitro. Our findings suggest that HO-1 expression protects against podocyte apoptosis under diabetic conditions.

        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.       • Curcumin protects retinal pigment epithelial cells against oxidative stress via induction of heme oxygenase-1 expression and reduction of reactive oxygen

        <P><B>Purpose</B></P><P>To determine whether curcumin induces expression of the defensive enzyme heme oxygenase-1 (HO-1) and protects cells against oxidative stress in cultured human retinal pigment epithelial cells.</P><P><B>Methods</B></P><P>Effective concentrations and toxicities of curcumin were determined after 3 h of curcumin treatment with the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Confluent human retinal pigment epithelium cell lines (ARPE-19) were preincubated with curcumin and oxidatively challenged with H<SUB>2</SUB>O<SUB>2</SUB>. HO-1 expression was determined with western blot analysis. To confirm the protective role of HO-1 in oxidative stress, small interfering RNA (siRNA) against HO-1 or inhibitor of HO-1 was treated with curcumin in retinal pigment epithelium cells. Intracellular levels of reactive oxygen species (ROS) were measured with flow cytometry and confocal microscopy. Apoptosis was evaluated with Annexin V-fluoroscein isothiocyanate staining.</P><P><B>Results</B></P><P>Curcumin had little cytotoxicity at concentrations less than 30 μM, and HO-1 expression was the highest at the 15 μM concentration. At this concentration, curcumin also increased the cytoprotective effect against the oxidative stress of H<SUB>2</SUB>O<SUB>2</SUB> through the reduction of ROS levels in human retinal pigment epithelial cells. Curcumin’s effect on the reduction of ROS was mediated by the increase in HO-1 expression.</P><P><B>Conclusions</B></P><P>Curcumin upregulated the oxidative stress defense enzyme HO-1 and may protect human retinal pigment epithelial cells against oxidative stress by reducing ROS levels.</P>

        RIZOMA SAURURI (RS) has been known to use antiinflammatory drug. To investigated the mechinism of RS-induced Peritoneal Macrophage activation, I examineded the ability of morphological change of Peritoneal Macrophages by RS. RS alone had only a small effect, whereas in combination with rIFN-γ markedly increased NO synthesis in a dose dependent manner. RS potently increased TNF-α secretion in Peritoneal Macrophages. In conclusion, RS induced the morphological change of the Peritoneal Macrophages through NO.

      • Underactuated 유영하는 우주 매니퓰레이터에 대한 강인한 적응 직교 제어

        A free-floating space robot manipulator system is a robotic system that has a robot manipulator mounted on a space station or satellite which freely moves in reaction to the robot manipulator due to the dynamic coupling. In usual, a robot manipulator that has fewer number of joint actuators than the number of its total joints is so-called an underactuated manipulator. For the control in Cartesian space in which tasks of robot manipulators are planned, we assume that the number of active joints is greater than or equal to the number of configurational dimension of the manipulator's end-effector in Cartesian space. In this paper, a robust adaptive Cartesian control scheme of an underactuated free-floating space manipulator is proposed in the presence of the parametric uncertainty and disturbances. The proposed scheme does not need a prior knowledge of the exact dynamic parameters and uncertainty bounds. To show the feasibility and robustness of the proposed control scheme, simulation results for a six-link PUMA560 arm with three passive joints mounted on a free-floating space vehicle are presented.

        요양소에 수용중인 정신장애자의 입소전 치료방법에 관한 연구

        In Korea, a large number of the mentally ill are still institutionalized in asylums where they can not receive modern psychiatric treatments. This survey was designed to investigate previous treatment methods of these psychiatric patients and to offer basic data in order to develop modern psychiatric delivery system for patients institutionalized in asylums. The 694 subjects who could give cooperative informations wee selrected form 6 asylums which are located in the Kwang-ju and Chonnam area. The data on this survey was collected by investigator's interview from Mar. 30, 1987 to Apr. 10, 1987. The previous treatment methods were divided into 8 groups(modern psychiatric treatment, asylym, Korean shamanic rite, religious healing, herb medicine, drug store, others and no treatment) and multiple responses were allowed. The methods were studied relating to the following psychiatric and sociodemographic factors : psychiatric diagnosis, age at the time of institutionalization, morbid duration before institutionalization, sex, occupation, residence, religion, economic status, marital status, educational level, and guardian. 49% of the patients had no experienced any previous treatment for their mental illness. 35% of the patients had been given modern psychiatric treatment and 23% had been previously institutionalized in asylums. The other treatment methods were reported in less than 10% of the patients. There were significant relationships between the choice of treatment methods and the psychiatric and sociodemographic factors.

        As information processing recently increases over Internet. there have been many researches about designing an efficient storage system to process a huge amount of information, which is based upon DTD in XML However, most of them have some difficulties in meaningful and structural presentation of XML documents and some restrictions in integration of many heterogeneous documents. In other word, we need a storage system which well-defines the structure of heterogeneous XML documents and integrates them. In this paper, we proposed a technique of an XML documentation storage system using DTD integration graphs based on CSB+-tree. This technique retrieves structural information from elements of XML documents, creates a CSB+-tree based DTD integrated graphs of the man storage system, and generates an integrated storage system by using element mappings. The proposed technique utilizes the man storage system more efficiently in space and provides more faster response time.

      • 18S-srRNA 분석에 의한 Acantbamoeba 한국 분리주들의 분류 검토

        Acanthamoeba spp. spread in soil, ponds, air and swimming pool, cause a granulomatous amoebic meningitis and amoebic keratitis in human and experimental animals. Their classification had depended upon morphological characteristics and physiological conditions such as thermophilic status and generation times. However, because amoeba species showed the morphological and physiological diversity in isolates, many attempts have been made on identification of Acanthamoeba spp. Restriction fragments length polymorphism (RFLP) analysis of IBS-small subunit ribosomal RNA (srRNA) was an useful tool for classification among morphologically and genetically closely-related species. In this study, 18S-srDNAs of amoebae were amplified by PCR with primer encoding 18S-srRNA gene and digested with restriction endonudeases, and RFLP analysis was applied on classification of Acanthamoeba Korean isolates (YM-4, YM-5 and YM-7), comparing with reference amoebae, A. culbertsoni, A. polyphaga and A. royreba. Trophozoites of six Acanthamoeba spp. had typical acanthopoda, but did not show any morphological differences. Cyst of Acanthamoeba sp. YM-7 was similar to that of A. polyphaga which was designated as group Ⅱ Acanthamoeba. Morphologically other amoebae belonged to group Ⅲ Acanthamoeba. Digestion of 18S-srDNA with six enzymes, Dde Ⅰ, Hae Ⅲ, Hind Ⅲ, EcoR Ⅰ, Rsa Ⅰ and Sph Ⅰ, resulted in various DNA fragments. Using the method of Nei and U (1979) for RFLP analysis, genetic divergence was not observed between A culbertsoni and Acanthamoeba sp. YM-4. Between A. culbertsoni and Acanthamoeba sp. YM-5, genetic distance was 0.070, 0.364 between Acanthamoeba sp. YM-4 and A. polyphaga, and 0.277 between Acanthamoeba sp. YM-7 and A. polyphaga. In comparison with A. culbertsoni and A. polyphaga, Acanthamoeba sp. YM-7 showed genetic distance of 0.330 and 0.154, respectively. Thus, Acanthamoeba sp. YM-7 is a similar species to A. polyphaga. Acanthamoeba sp. YM-5 is a different species from A. polyphaga, but closely-related to A. culbertsoni. Acanthamoeba sp. YM-4 can be classified as a subspecies or a strain of A. culbertsoni.
