        Knockdown of Long Non-Coding RNA NEAT1 Inhibits Proliferation and Invasion and Induces Apoptosis of Osteosarcoma by Inhibiting miR-194 Expression

        Purpose: Long non-coding RNA (lncRNA) nuclear paraspeckle assembly transcript 1 (NEAT1) has been implicated as an oncogenein the development and progression of osteosarcoma. This study aims to explore the mechanism of NEAT1 in osteosarcoma. Materials and Methods: Expressions of NEAT1 and miR-194 in osteosarcoma tissues and cells were detected by quantitative real-time PCR. The effects of NEAT1 knockdown or miR-194 overexpression on cell proliferation, invasion, and apoptosis were determinedby 3-[4, 5-dimethylthiazol-2-yl]-2, 5 diphenyl tetrazolium bromide (MTT) assay, transwell invasive assay, and flow cytometry analysis, respectively. Luciferase reporter assay was performed to observe the possible interaction between NEAT1 and miR-194. Results: NEAT1 was upregulated and miR-194 was downregulated in osteosarcoma tissues and cells. Knockdown of NEAT1 or overexpression of miR-194 suppressed proliferation and invasion and induced apoptosis of osteosarcoma cells in vitro. Luciferase reporter assay validated that NEAT1 could interact with miR-194 and negatively modulated its expression. Furthermore, inhibitionof miR-194 reversed the suppression of proliferation and invasion and the promotion of apoptosis induced by NEAT1 depletionin osteosarcoma cells. Conclusion: Knockdown of NEAT1 suppressed proliferation and invasion and induced apoptosis in osteosarcoma cells by inhibitingmiR-194 expression.

        Isolation and Identification of Lactic Acid Bacteria from Traditional Dairy Products in Baotou and Bayannur of Midwestern Inner Mongolia and q-PCR Analysis of Predominant Species

        In this study, traditional culture method and 16S rRNA gene analysis were applied to reveal the composition and diversity of lactic acid bacteria (LAB) of fermented cow milk, huruud and urum from Baotou and Bayannur of midwestern Inner Mongolia. Also, the quantitative results of dominant LAB species in three different types of dairy products from Baotou and Bayannur were gained by quantitative polymerase chain reaction (q-PCR) technology. Two hundred and two LAB strains isolated from sixty-six samples were identified and classified into four genera, namely Enterococcus, Lactococcus, Lactobacillus, Leuconostoc, and twenty-one species and subspecies. From these isolates, Lactococcus lactis subsp. lactis (32.18%), Lactobacillus plantarum (12.38%) and Leuconosto mesenteroides (11.39%) were considered as the dominated LAB species under the condition of cultivating in MRS and M17 medium. And the q-PCR results revealed that the number of dominant species varied from samples to samples and from region to region. This study clearly shows the composition and diversity of LAB existing in fermented cow milk, huruud and urum, which could be considered as valuable resources for LAB isolation and further probiotic selection.

        MiR-371 promotes proliferation and metastasis in hepatocellular carcinoma by targeting PTEN

        Hepatocellular carcinoma (HCC) is the leading cause of cancer-related mortality worldwide. MiR-371 has recently emerged as an important regulator in tumorigenesis, and may serve as a biomarker for malignant tumors. We transfected miR-371 or its inhibitor in two human HCC cell lines, then used 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, soft agar colony formation, and transwell migration assays to evaluate the effects on cell proliferation, migration, and invasion. We found that miR-371 was positively correlated with HCC metastasis and poor prognosis in the inflicted patients, and the high expression of miR-371 was promoted, whereas a low level of miR-371 depressed cell proliferation and invasion. We found PTEN to be a direct target of miR-371. The overexpression or knockdown of PTEN exhibited the opposite effects from those of miR-371 on cell proliferation and migration. Our study demonstrates that miR-371 promotes proliferation and metastasis in HCC by targeting PTEN. [BMB Reports 2019; 52(5): 312-317]

        Construction engineering quality inspection is an important means to ensure the quality and safe use of construction engineering. On the base of analyzing the status and the main problems of Anhui provincial construction engineering quality inspection institutions, the five principles were proposed to put forward the total control, optimize the structure and increase and development ability, moderate competition and standard development. By optimizing the detection mechanism, enhancing the overall level of the testing agencies, and strengthening supervision, to achieve a basic balance and reasonable layout of supply and demand in various cities and regions. It provides reference for the research and practice of the supervision and management of construction engineering quality test institutions.

        Aerodynamic Numerical Analysis of the Low Reynolds Number Diamond Joined-Wing Configuration Unmanned Aerial Vehicle

        Aerodynamic numerical simulation of low Reynolds number long-endurance Diamond Joined-Wing configuration unmanned aerial vehicle was performed. First, low Reynolds number aerodynamic characteristics of a typical wing are numerically simulated by solving the Reynolds averaged Navier–Stokes (RANS) equations based on the structural grid technology and the k–kL–ω transition model. To prove the accuracy of this method, experimental data and numerical simulation results were compared. Second, the performance of low Reynolds number Diamond Joined-Wing configuration UAV (unmanned aerial vehicle) aerodynamic characteristics and control surfaces was studied by numerical simulation and theoretical analysis. The analysis of the interference between the Frt-wing (front wing) and the Aft-wing (after wing) of the Diamond Joined-Wing configuration UAV and the reason for the change in the pitching moment in the lateral-directional motion is carried out. The results show that the Diamond Joined-Wing configuration UAV has a typical low Reynolds number surface flow characteristics, Frt-wing and Aft-wing flow interference are relatively minor at a small angle of attack, which has a high basic aerodynamic performance in addition to good stall characteristics, with longitudinal and lateral-directional static stability. The performance of control surfaces and flaps is higher to meet the design requirements. Finally, the accuracy of the numerical simulation results was verified by vehicle aerodynamic testing.

        A quantitative analysis of the wing outlines of owlflies (Insecta: Neuroptera: Ascalaphidae)

        We quantitatively described and analyzed variation in the wing outlines of owlflies using 135 forewings and 135 hindwings from eight species in five genera. We captured and digitized images of the wing outlines from 135 individual specimens and subjected them to Elliptic Fourier analysis to produce matrices of Fourier coefficients based on 20 harmonics. The first five principal components extracted from the Fourier coefficient matrices described the wing outlines, explaining 94.87% of the total forewing variation and 95.06% of the total hindwing variation. Relationships among species were estimated using multivariate analysis of variance, principal component analysis, and cluster analysis. The results indicated highly significant differences in the wing outlines among the eight owlfly species. Pair-wise comparisons indicated significant differences between 96% of the species pairs based on the forewing outlines and between 100% of the species pairs based on the hindwing outlines. The results of the cluster analysis were partially consistent with the existing classification system of Ascalaphidae. Our results show that quantitative data of wing outlines can effectively differentiate Libelloides from the other four owlfly genera.

        The influences of process parameters on the preliminary roll-forging process of the AISI-1045 automobile front axle beam

        Roll-forging process is widely used to fabricate the automobile front axle beams. The reasonable selection of the process parameters is significant to ensure the quality and improve the property of the automobile front axle beam owing to the complexity and importance of the preliminary roll-forging process. This paper developed a 3D Finite element (FE) model of the preliminary roll-forging process of the AISI-1045 automobile front axle beam under the Deform-3D platform. A corresponding experiment is subsequently conducted. Good agreement between the experimental and simulative results validates the employed model. Based on this dependable FE model, the influences of initial temperature of billet T 0 , rotational speed of rolls n and friction factor m on the geometric dimension, temperature inhomogeneity, deformation inhomogeneity, and roll-forging load are investigated comprehensively. The results can provide reliable guidelines for the selection of process parameters.

        MicroRNA-590-5p Regulates Proliferation and Invasion in Human Hepatocellular Carcinoma Cells by Targeting TGF-beta RII

        MicroRNAs (miRNAs) are regulatory small non-coding RNAs that can regulate gene expression by binding to gene elements, such as the gene promotor 5UTR, mainly in the 3UTR of mRNA. One miRNA targets many mRNAs, which can be regulated by many miRNAs, leading to a complex metabolic network. In our study, we found that the expression level of miR-590-5p is higher in the human hepatocellular carcinoma cell line HepG2 than in the normal hepatocellular cell line L02. Downregulation of miR-590-5p inhibited proliferation and invasion of hepatocellular carcinoma cells (HCCs). We also showed that expression of TGF-beta RII, which has been regarded as a regulator of tumor proliferation, invasion, and migration in hepatocellular carcinoma, is regulated by miRNA-590-5p. In addition, miR-590-5p downregulated the expression of TGF- beta RII by targeting the 3UTR of mRNA. We also found that downregulation of miR-590-5p was associated with an elevation of TGF-beta RII and inhibition of proliferation and invasion in HepG2 cells. Furthermore, overex-pression of miR-590-5p was associated with upregulation of TGF-beta RII and could promote proliferation and invasion in L02 cells. In conclusion, we determined that TGF-beta RII is a novel target of miRNA-590-5p. Thus, the role of TGF-beta RII in regulating proliferation and invasion of human HCCs is controlled by miR-590-5p. In other words, miR-590-5p promotes proliferation and invasion in human HCCs by directly targeting TGF-beta RII.
