      • Characterization of endoplasmic reticulum oxidoreductase (ERO1) gene from Bombyx mori

        Disulfide bond formation, reduction and isomerization are important posttranslational modification in proteins that occur in most, if not all, living organisms. In eukatoyes, disulfide bond in substrate proteins are primarily formes by ERO1 and PDI. ERO1, oxidized by molecular oxygen, acts as a specific oxidant of PDI, which then makes disulfide bonds in folding proteins oxidized directly. It means that ERO1 plays an essential role in setting the redox potential in the ER, and the regulation of Ero1p activity is critical to maintain redox homeostasis and proper ER folding activity. We have isolated and analysed a endoplasmic reticulum oxidoreductase (ERO1) from Bombye mori. It apperas that both an N-terminal CxxxxC motif and a C-terminl CxxCxxC motif are necessary for Ero1p fuction. In vivo, the result of the 5day of 5th instar larvae by RT-PCR and Real-Time PCR shows that posterior silkgland, skin and mid silkgland are revealed more than rhose of other tissues. The same result for tissue distribution of transcripts is appeared about ERO1 and PDI. In Bombyx mori, ERO1 is also supposed to correlate with PDI. Afterwards, more experiments are needed to figure out accurate interrelation between ERO1 and PDI.

      • Molecular characterization of heat shock protein 70 promoter from the silkworm, Bombyx mori

        Recently Transgenesis was achieved in Bombix mori. For stable and effective transgenesis in B.mori, B.mori cytoplasmic actin gene (BmA3) promoter was used to expression of marker gene, the green fluorescent protein(GFP). Green fluorescent protein expression for selection of transformants was visible in all larval, pupal, and adult tissues but, unexpectdly, was not detectable in embryos. So, it spend times and money on rearing of silkworm. Furthermore, the BmA3 promoter is predominantly active in the midgut, which makes it difficult to reliably identify transformants since autofluorescence of many insect foods can mask low-level fluorescence and only allows the detection of strongly expressing individuals with potentially multiple insertions. Therefore, we need more intensely promoter than BmA3 promoter for selected by expression of GFP in embryos and selected by reliable expression of GFP in larvae. We performed dot blot hybridization to develop strong promoter. Nine differentially expressed clones were isolated and we focused one clone of them which has high similarity with heat shock protein 70 gene from D.melanogaster. We named it as bHSP70 (Bombyx mori heat shock protein 70). Expression from the hsp70 promoter was strong and heat shock-dependent. And Drosophila hsp70 promoter appears useful for regulating expression of Exogenous DNA. So, we analyzed transcriptional activity of promoter with bHSP70 gene by using dual luciferase assay system. bHSP70 promoter has about 264 folds more intensely than BmA3 promoter. Also, when bHSP70 promoter treated heat shock(42℃), transcriptional activity incresed 2 times more than normal condition. Therefore, we suggest that bHSP70 promoter is more effective candidate for stable transformation and selection of transformants.

      • Large area multi-stacked lithium-ion batteries for flexible and rollable applications

        <P>The demand for lithium ion batteries (LIBs) in various flexible mobile electronic devices is continuously increasing. With this in mind, a vast number of smart approaches, such as implementation of conductive nanomaterials onto paper and textiles, have been recently demonstrated. Most of them were, however, limited to the single-cell level. In the present study, large area flexible battery modules were developed in an attempt to expand the knowledge and design accumulated from the single-cell level approaches to larger-scale applications. A multi-stacked configuration was adopted to produce a high areal energy density in each single-cell. Meanwhile textile-based electrodes on both sides grant mechanical stability, even on the module level, by efficiently releasing the stress generated during aggressive folding and rolling motions. Moreover, the connection between and stacking of the single-cells allow the wide tuning of the overall voltage and capacity of the module. This battery design should be immediately applicable to a broad range of outdoor, building, and military items.</P>

        말산클레보프리드 서방성 제제의 제조 및 약물동태학적 평가

        Clebopride malate(Cm) is a new benzamide drug which has a potent central antidopaminergic activity possessing antiemetic and anxiolytic properties. A purpose of this study was to assess the feasibility of formulating sustained release preparation by dispersing a drug in hydrophilic polymeric matrices and double layered tablets(DLT), using HPMC, carbopol, PEO, PVP/VA and other polymers as a rate controlling barrier. The matrix and DLT showed optimum dissolution pattern up to 8 hours and the contents of polymer were optimized at 30% level in this preparation. After an oral administration in beagle dog, Cm concentration was determined by use of GC-ECD and pharmacokinetic parameters were calculated by Vallner`s method. The AUC of DLT showed similar results and the duration of action was increased 55% compared to that of regular release dosage form. The calculated absorption rate effectiveness(ARE) and controlled release effectiveness(CRE) for DLT increased 50% compared to that of matrix, the overall effectiveness(E) of this product appears to be about 70%. in vivo effectiveness test, DLT showed significant differences from control on antiemetic action of Cm. In consequence, it was possible to conclude that double layered tablet might be a good candidate for the sustained release dosage forms.

      • A Study on the Malicious Process Detection System Model based on Security Agent

        As recently use of Internet gets increased rapidly, infection path through the network becomes diverse. In particular, attacks by Bot mainly work at C&C (command-andcontrol) server but it can be secured just by blocking IP because C&C server runs in form of IP. However, these attackers too get gradually intelligent as they try to connect periphrastically in order to avoid server blocking. Once these malicious codes infiltrate user’s system, it is not easy to detect it through general detection method while it is running. In this paper, so, we propose malicious process detection system based on security agent in order to prevent damage caused by malicious code infection from being spread.

        국소적으로 진행된 자궁경부암에서 동시 항암화학방사선치료의 효과

        목적 : 국소적으로 진행된 자궁경부암 환자들에 대한 치료로서 동시 항암화학방사선치료의 효과를 확인하고 그 부작용을 비교하고자 하였다. 연구 방법 : 1994년 9월부터 2000년 8월까지 아주대학교병원 산부인과를 방문하여 임상병기 Ⅱb에서 Ⅵa의 국소적으로 진행된 자궁경부암으로 진단 받은 환자들과, 병기 Ib나 Ⅱa로 진단된 환자 중 병소의 최대직경이 5 cm 이상인 거대 종괴 환자들 및 자기공명영상촬영상 직경 1 cm 크기 이상의 골반내 림프절 비대 소견을 보여 림프절 전이가 있는 것으로 추정되는 환자를 대상으로 하였다. 총 242명의 방사선치료를 받은 환자들 중 재발 고위험군을 대상으로 동시 항암화학방사선요법을 시행한 24명과 방사선치료만을 받은 62명의 치료 효과 와 부작용을 후향적으로 분석하였다. 골반 방사선조사는 45 Gy 시행 후 (중앙차폐 36-40 Gy에 시행) 골반의 국소 전이와 림프절 전이를 보인 경우 자궁 주위 조직에 총 5,800 cGy까지 추가 조사하였다. 통상적으로 하루에 1.8 Gy씩 주 당 5회 실시하였으며 고선량 근접치료로 A 점에 4 Gy 또는 5 Gy씩 6-7회 조사하였다. 항암화학치료는 방사선치료 제 1일과 29일에 cisplatin 75 ㎎/m2, 제 2일부터 5일과 제 30일부터 33일까지 5-FU 1000 ㎎/m2/day를 주사하였다. 부작용을 GOG 정의에 따라 평가하고 생존율과 재발율을 통계적으로 분석하였다. 결과 : 2년 생존율은 동시항암화학방사선치료를 받은 전체 18명의 대상 환자 중 3명이 사망하여 15명 (83.3%), 방사선치료군은 61명 중 15명이 사망하고 46명 (75.4%)이 생존함으로써 방사선치료에 비해 상대적 사망위험이 66% 감소하였다 (RR 0.34, p=0.092). 치료 종료 후 2년째에 동시항암화학방사선치료군 18명 중 2명 (11.1%)이 재발하고, 방사선치료군은 61명 중 19명 (32.3%)이 재발함으로써 방사선치료군의 재발위험에 비하여 동시항암화학방사선치료군은 32% (RR 0.68, p=0.5) 감소하는 효과를 보였다. 동시항암화학방사선치료 기간 중 나타난 부작용은 grade 3의 장염이 3명 (12.5%), grade 3의 백혈구 감소는 2명 (8.3%), grade 3의 빈혈은 1명 (4.1%), grade 4의 빈혈은 2명 (8.3%)에서 발생하였다. 단독 방사선치료시 급성 독작용은 grade 3의 장염이 7명 (11.3%), grade 3의 직장 출혈이 1명 (1%)으로써 동시항암화학방사선치료군에서 급성 부작용의 발생이 증가되는 경향을 보였다. 결론 : 국소적으로 진행된 자궁경부암 환자에서 동시항암화학방사선치료를 함으로써 재발율을 감소시키고, 무병생존율은 향상시킬 수 있었다. 그러나 동시항암화학방사선치료 환자군에서 급성 부작용이 증가하는 경향을 보였으나 적절한 처치가 가능하였다. Objective : The purpose of this study was to evaluate the efficacy and toxicity of concurrent chemoradiotherapy (CCRT) in loco-regionally advanced cervical cancer patients. Methods : The medical records of 24 patients were retrospectively reviewed who underwent CCRT' and 62 patients who had underwent radiation therapy alone at the Department of Obstetrics and Gynecology, Ajou University Hospital from September, 1994 to August 2000 for loco-regionally advanced cervical cancer (stage Ⅱb-Ⅳa and Ib-Ⅱa with bulky tumor mass or pelvic lymph node involvement detected by magnetic resonance imaging). Radiotherapy was performed for 7 weeks during which the patients underwent radiotherapy and chemotherapy concomitantly. Chemotherapeutic regimen consisted of cisplatin (75 ㎎/m2 on day 1, 4) and 5-FU (1000 ㎎/m2 on day 2-5, 30-33). The toxicity was assessed according to the GOG toxicity criteria. Results : In the CCRT group, 2 of 18 patients (11.1%) had recurrent disease. Of the 22 patients who were followed up, the overall survival rate was 83.3%, and the relapse-free survival rate was 89.9%. In the radiotherapy alone group, 19 of 61 patients (32.3%) had recurrent disease. Of the 62 patients who were followed up, the overall survival rate was 75.4% and the relapsed-free survival rate was 67.7%. Relative risk reduction rate of the death rate in the CCRT group was 66% (RR 0.34), and 32% (RR 0.68) in the recurrence rate. Acute toxicities in the CCRT group were enteritis grade 3:3 (12.5%), leukopenia grade 3:2 (8.3%), anemia grade 3:1 (4.1%) and grade 4:2 (8.3%). The acute toxicities in the radiation therapy alone group were enteritis grade 3:7 (11.3%), rectal bleeding grade 3:1 (1%). Increased acute toxicities developed in the CCRT group. Conclusion : This study shows improved overall survival rates and relapse-free survival rates but some increase in acute toxicity. It is suggested that CCRT be the standard treatment of loco-regionally advanced cervical cancer. Due to small size sample and short duration of follow up, further study of a large group of patients and the long term survival rate are necessary.
