The development of in vitro assays has been recommended to screening and test-ing the potential endocrine disruptors (EDs). These assay systems focus only on identifying thethe thyroid hormone (TH) disruptors. The aim of this study was to evaluate a test system to detectTH disruptors using rat pituitary tumor GH3 cells. The test system is based on the TH-dependentincrease in growth rate. As expected, L-3,5,3-triiodothyronine (T3) markedly induced a morphologicalchange in GH 3 cells from flattened fibroblastic types to rounded or spindle-shaped types. T3 stimu-lated GH3 cell growth in a dose-dependent manner with the maximum growth-stimulating effect9 M. In addition, T3 increased the release of growth hor-mone and prolactin into the medium of the GH3 cells culture. Using this assay system, the TH-dis-rupting activities of bisphenol A (BPA) and its related compounds were examined. BPA,dimethylbisphenol A (DMBPA), and TCI-EP significantly enhanced the growth of GH3 cells in therange of 1 × 10-5M to 1 × 10-6M concentrations. In conclusion, this in vitro assay system might bestandardization before it can be used as a broad-based screening tool.

The catalytic dehydrocoupling of bis(1-sila-3-butyl)benzene 1 and 2-phenyl-1,3-disilapropane 2 by Cp2ZrCl2/Red-Al and Cp2ZrCl2/n-BuLi was reported to compare their catalytic efficiency. The dehydrocoupling of monomeric silanes 1 with the Cp2ZrC12/Red-Al and Cp2ZrCl2/n-BuLi combination catalysts produced two phases of polymers: one is a highly crosslinked insoluble solid, and the other is noncross-linked or slightly cross-linked soluble oil and could be a precursor for the solid polymer. The dehydrocoupling of 2 with the Cp2ZrCl2/n-BuLi combination catalyst similarly produced two phases of polymers. By contrast, the catalytic reaction of 2 with the Cp2ZrCl2/Red-Al combination catalyst produced a soluble polymer via redistribution/dehydrocoupling process.

Two diastereomeric chiral stationary phases (CSPs) were applied to the liquid chromatographic resolution of various racemic α-amino methyl esters, α-amino N,N-diethylamides and α-amino N-propylamides. The CSP incorporating (R)-3,3’-diphenyl-1,1’-binaphtyl and (R,R)-tartaric acid unit as chiral barriers did not show any chiral recognition. In contrast,the CSP incorporating (R)-3,3’-diphenyl-1,1’-binaphtyl and (S,S)-tartaric acid unit as chiral barriers was found to show excellent chiral recognition especially for the two enantiomers of α-amino N-propylamides. Some of α-amino methyl esters and α-amino N,N-diethylamides were also resolved on the CSP incorporating (R)-3,3’-diphenyl-1,1’-binaphtyl and (S,S)-tartaric acid unit. From these results it was concluded that the two chiral units composing the diastereomeric CSPs can show “matched” or “mismatched” effect on the chiral recognition according to their absolute stereochemistry.

Purpose: Vascular endothelial growth factor (VEGF) is a vascular endothelial cell specific mitogen and a major regulator of angiogenesis. VEGF secretion is activated in some thyroid cancers and that VEGF secretion is stimulated by TSH. So we postulated that TSH may promote growth and invasion in some thyroid cancers by stimulating VEGF secretion and angiogenesis. Methods and Results: We investigated the TSH effect for the VEGF secretion, endothelial cell proliferation and invasion in vitro with the primary cultured normal thyroid cell (NT-1) and thyroid cancer cell line (TPC-1). And to evaluate the relationship between TSH and VEGF, angiogenesis and tumor growth in vivo, we xenografted human dermal matrix inoculated with thyroid cells into nude mice or directly injected subcutaneously. For the study, mice were made hypothyroid (Group 1) by antithyroid hormone p.o, hyperthyroid (Group 2) by L-thyroxine injection and euthyroid (Group 3). One week after the treatment, significant difference were noted in T3, T4 and TSH level between each group, but the VEGF level showed significant difference in group 1 only compared with group 2 and 3. NT-1 or TPC-1 were seeded in the upper chamber of Transwell and HUVEC were cultured in lower chamber, and added different concentration of TSH. NT-1 and TPC-1 secreted VEGF under basal condition, but the level were similar. TPC-1 cells secreted significantly more VEGF than NT-1 after TSH (1, 10, 100 mIU/dl) stimulation, which were also parallel with the concentration of TSH. In low concentration of TSH (0, 1 mIU/dl), there were no difference of HUVEC proliferation between NT-1 and TPC 1. In high concentration of TSH (10, 100 mIU/dl), however, TPC-1 enhanced HUVEC proliferation than NT-1 significantly (p<0.05). Similar findings were noted in thyroid cell invasion. Invasion was higher in TPC-1 than in NT-1 in high concentration of TSH (10, 100 mIU/dl). In vivo study using the dermal matrix showed that number of blood vessels ingrowth were higher in Group 1 (25/HPF) than Group 2 (16/HPF) or Group 3 (17/HPF). But there was no difference between Group 2 and Group 3. Level of TSH and VEGF were also increased significantly in Group 1 compared with in Group 2 and Group 3. The size of tumor did not showed significant difference between each group during observation. The tumor from Group 1 (6.2 gm) were larger compared with Group 2 (5.1 gm) or Group 3 (5.6 gm), but this difference was not significant statistically (p>0.05). The number of blood vessels in tumor were also more increased in Group 1 and were commonly located in the peripheral portion of tumor. Conclusion: We conclude that thyroid cancer cell line secrete the VEGF and TSH secretion is more enhanced by the stimulation of TSH. And increased VEGF promote the vascular endothelial cell proliferation, invasion and angiogenesis in thyroid cancer. (Korean J Endocrine Surg 2001; 1:51-60)

목적: To compare the effect of long-acting recombinant FSH (rFSH), corifollitropin alfa with daily rFSH administered during 1st 7days of controlled ovarian stimulation (COS) using GnRH antagonist multiple dose protocol (MDP) in poor responders undergoing IVF/intracytoplasmic sperm injection (ICSI). 방법: In this prospective randomized study total of 90 poor responders who were scheduled for IVF/ICSI were enrolled and divided into the corifollitropin alfa group (study group) or the daily rFSH group (control group). The diagnosis of poor responder was based on the Bologna criteria of the 2011 ESHRE consensus. In all subjects, GnRH antagonist MDP was used for COS. 결과: There were no differences in patients` characteristics between the two groups. Duration of COS was comparable between the study and control groups. The numbers of oocytes retrieved and grade I or II embryos were also similar in the two groups. However, the number of mature oocytes was higher in the study group with a borderline significance (p< .07) and the ratio of mature oocytes among total oocytes retrieved was significantly higher in the study group of 78.6 ± 13.0%, compared with 61.2 ± 16.0% in control group (p< .001). There were no differences in the clinical pregnancy rate per cycle initiated, embryo implantation rate and miscarriage rate between the two groups. 결론: Corifollitropin alfa is at least as effective as daily rFSH treatment for COS using GnRH antagonist MDP, and may improve the synchrony of follicular development in poor responders undergoing IVF/ICSI. Therefore, corifollitropin alfa may be a feasible, patient-friendly alternative for poor responders.

Angiosarcomas are rare malignant neoplasms, which mainly occur on the skin and superficial tissue. Angiosarcoma in the small intestine after radiation therapy is very rare and there are less than 20 cases of small-intestine angiosarcoma after pelvic irradiation. We report a case of double cancer with gastric adenocarcinoma and postradiation angiosarcoma of the small intestine which has been diagnosed 20 years after pelvic irradiation therapy for uterine cervical cancer.
