        Low pH stress responsive transcriptome of seedling roots in wheat (Triticum aestivum L.)

        Soil acidification is one of major problems limiting crop growth and especially becoming increasingly serious in China owing to excessive use of nitrogen fertilizer. Only the STOP1 of Arabidopsis was identified clearly sensitive to proton rhizotoxicity and the molecular mechanism for proton toxicity tolerance of plants is still poorly understood. The main objective of this study was to investigate the transcriptomic change in plants under the low pH stress. The low pH as a single factor was employed to induce the response of the wheat seedling roots. Wheat cDNA microarray was used to identify differentially expressed genes (DEGs). A total of 1057 DEGs were identified, of which 761 genes were up-regulated and 296 were downregulated. The greater percentage of up-regulated genes involved in developmental processes, immune system processes, multi-organism processes, positive regulation of biological processes and metabolic processes of the biological processes. The more proportion of down-regulation genes belong to the molecular function category including transporter activity, antioxidant activity and molecular transducer activity and to the extracellular region of the cellular components category. Moreover, most genes among 41 genes involved in ion binding, 17 WAKY transcription factor genes and 17 genes related to transport activity were up-regulated. KEGG analysis showed that the jasmonate signal transduction and flavonoid biosynthesis might play important roles in response to the low pH stress in wheat seedling roots. Based on the data, it is can be deduced that WRKY transcription factors might play a critical role in the transcriptional regulation, and the alkalifying of the rhizosphere might be the earliest response process to low pH stress in wheat seedling roots. These results provide a basis to reveal the molecular mechanism of proton toxicity tolerance in plants.

        Temporal dynamics of Bactrocera (Zeugodacus) tau (Diptera: Tephritidae) adults in north Jiangxi, a subtropical area of China revealed by eight years of trapping with cuelure

        Bactrocera (Zeugodacus) tau (Walker) (Diptera: Tephritidae), a quarantine pest of international concern, has become widespread and economically important in South and Southeast Asia. It has been introduced into north Jiangxi from Fujian, China and become a threat to local fruits and vegetables. Field trapping with cuelure (CUE) as lure is needed, not only for monitoring population dynamics of B. tau but also for direct control of population. In this study, we collected weekly tephritids from 4 CUE-baited McPhail traps every year from 2008 to 2015, analysing seasonal and inter-year B. tau population fluctuations. The total number of captured tephritids reached 7811, including B. tau (5321), B. scutellata (Hendel) (2375) and B. cucurbitae (Coquillett) (1 1 5). Among them, B. tau was the dominant species. It had 3–4 generations a year in north Jiangxi with high generation overlapping. The average number of trapped B. tau males did not differ significantly in different years, while the average monthly number showed large variation from June to December. No B. tau males were trapped before June in any year. Adult males were present from mid-June to late December, with distinct peaks between late August and mid-October. This fruit fly can withstand below 13℃ as a monthly minimal temperature under field conditions, and adult capture rates were significantly related to monthly minimal temperature and monthly total precipitation. These results can be used to predict the occurrence time and population size of B. tau in different seasons, enabling growers and extension personnel to take efficient management measures before they damage the host crops.

        Behavior of three-tube buckling-restrained brace with circumference pre-stress in core tube

        The behavior of a new Three-Tube Buckling-Restrained Brace (TTBRB) with circumference pre-stress (σθ,pre) in core tube are investigated through a verified finite element model. The TTBRB is composed of one core tube and two restraining tubes. The core tube is in the middle to provide the axial stiffness, to carry the axial load and to dissipate the earthquake energy. The two restraining tubes are at inside and outside of the core tube, respectively, to restrain the global and local buckling of the core tube. Based on the yield criteria of fringe fiber, a design method for restraining tubes is proposed. The applicability of the proposed design equations are verified by TTBRBs with different radius-thickness ratios, with different gap widths between core tube and restraining tubs, and with different levels of σθ,pre. The outer and inner tubes will restrain the deformation of the core tube in radius direction, which causes circumference stress (σθ) in the core tube. Together with the σθ,pre in the core tube that is applied through interference fit of the three tubes, the yield strength of the core tube in the axial direction is improved from 160 MPa to 235 MPa. Effects of gap width between the core tube and restraining tubes, and σθ,pre on hysteretic behavior of TTBRBs are presented. Analysis results showed that the gap width and the σθ,pre can significantly affect the hysteretic behavior of a TTBRB.

        Macrophage and Adipocyte Mitochondrial Dysfunction in Obesity-Induced Metabolic Diseases

        Obesity is one of major health burdens of modern society as it contributes to the growing prevalence of its related comorbidities, such as diabetes, cardiovascular diseases, and some cancers. A series of innate immune cells, especially macrophages, and adipocytes have been implicated in the pathogenesis of obesity. Mitochondrial dysfunction, which is induced by obesity, are critical mediators in initiating inflammation in macrophages and adipocytes, and subsequent systemic insulin resistance. In this review, we discuss new findings on how obesity impairs mitochondrial function in macrophages and adipocytes and how this dysfunction contributes to obesity and its comorbidities. We also summarize drugs that treat metabolic diseases by targeting mitochondrial dysfunction.

        Isolation and characterization of heat-responsive gene TaGASR1 from wheat (Triticum aestivum L.)

        GA-stimulated transcript (GAST) family genes have been identified in numerous plant species. In this paper, we isolated and characterized a heat-responsive gene, TaGASR1, from heat tolerant variety TAM107. The complete ORF of TaGASR1 was cloned, which encoded a 98-kDa protein, and the sequence shared 51.52% similarity to OsGASR1. Analysis of the TaGASR1 promoter region showed that it contained a heat shock element (HSE) and several cis-elements involved in abiotic stress response and hormone signal transduction. Expression patterns of TaGASR1 revealed that it was strongly induced by stress factors, such as high temperature, drought, high salinity and oxidation, as well as the phytohormones, including MeJA, ACC and ABA, which suggested the TaGASR1 gene might participate in these stress and hormone signal transduction pathways. Transient expression of TaGASR1-GFP fusion proteins in onion epidermal cells indicated that TaGASR1 protein was localized to the cell membrane or cytosol. Further analysis showed that ectopic expression of TaGASR1 in Arabidopsis enhanced thermotolerance and reduced the accumulation of reactive oxygen species (ROS) after heat stress. Moreover, we also found that TaGASR1-overexpressing wheat improved tolerance to heat stress and oxidative stress.

        Direct Synthesis of H2O2 over Ti-Containing Molecular Sieves Supported Gold Catalysts: A Comparative Study for In-situ-H2O2-ODS of Fuel

        Direct synthesis of H2O2 and in situ oxidative desulfurization of model fuel over Au/Ti-HMS and Au/TS-1 catalysts has been comparatively investigated in water or methanol. Maximum amount (82%) of active Au0 species for H2O2 synthesis was obtained. Au/Ti-HMS and Au/TS-1 exhibited the contrary performances in H2O2 synthesis as CH3OH/H2O ratio of solvent changed. H2O2 decomposition and hydrogenation in water was inhibited by the introduction of methanol. Effect of O2/H2 ratio on H2O2 concentration, H2 conversion and H2O2 selectivity revealed a relationship between H2O2 generation and H2 consumption. The highest dibenzothiophene removal rate (83.2%) was obtained over Au/Ti-HMS in methanol at 1.5 of O2/H2 ratio and 60 oC. But removal of thiophene over Au/TS-1 should be performed in water without heating to obtain a high removal rate (61.3%). Meanwhile, H2 conversion and oxidative desulfurization selectivity of H2 were presented.

      • Adenovirus-mediated Expression of Both Antisense Ornithine Decarboxylase and S-adenosylmethionine Decarboxylase Induces G<sub>1</sub> Arrest in HT-29 Cells

        To evaluated the effect of recombinant adenovirus Ad-ODC-AdoMetDCas which can simultaneously express both antisense ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (AdoMetDC) on cell cycle distribution in colorectal cancer cell and investigated underlying regulatory responses, human colorectal cancer cells HT-29 were cultured in RPMI 1640 medium and infected with Ad-ODC-AdoMetDCas. Cell cycle progression was detected by flow cytometry analysis. The expression levels of cell cycle regulated proteins were measured by Western blot analysis. The mRNA level of cyclin D1 was measured by RT-PCR. And a luciferase reporter plasmid of cyclin D1 promoter was constructed to observe the effect of Ad-ODC-AdoMetDCas on cyclin D1 promoter activity. The results showed that recombinant adenovirus Ad-ODC-AdoMetDCas significantly induced $G_1$ arrest, decreased levels of cyclin D1 protein and mRNA and suppressed the promoter activity. Ad-ODC-AdoMetDCas also inhibited nuclear translocation of $\beta$-catenin. In conclusion, downregulation of ODC and AdoMetDC mediated by Ad-ODC-AdoMetDCas transfection induces $G_1$ arrest in HT-29 cells and the arrest was associated with suppression of cyclin D1 expression and inhibition of $\beta$-catenin nuclear translocation. As a new anticancer reagent, the recombinant adenovirus Ad-ODC-AdoMetDCas holds promising hope for the therapy of colorectal cancers.

        RIS Selection and Energy Efficiency Optimization for Irregular Distributed RIS-assisted Communication Systems

        In order to improve spectral efficiency and reduce power consumption for reconfigurable intelligent surface (RIS) assisted wireless communication systems, a joint design considering irregular RIS topology, RIS on-off switch, power allocation and phase adjustment is investigated in this paper. Firstly, a multi-dimensional variable joint optimization problem is established under multiple constraints, such as the minimum data requirement and power constraints, with the goal of maximizing the system energy efficiency. However, the proposed optimization problem is hard to be resolved due to its property of nonlinear nonconvex integer programming. Then, to tackle this issue, the problem is decomposed into four sub-problems: topology design, phase shift adjustment, power allocation and switch selection. In terms of topology design, Tabu search algorithm is introduced to select the components that play the main role. For RIS switch selection, greedy algorithm is used to turn off the RISs that play the secondary role. Finally, an iterative optimization algorithm with high data-rate and low power consumption is proposed. The simulation results show that the performance of the irregular RIS aided system with topology design and RIS selection is better than that of the fixed topology and the fix number of RISs. In addition, the proposed joint optimization algorithm can effectively improve the data rate and energy efficiency by changing the propagation environment.

        Resource Allocation for D2D Communication in Cellular Networks Based on Stochastic Geometry and Graph-coloring Theory

        In a device-to-device (D2D) underlaid cellular network, there exist two types of co-channel interference. One type is inter-layer interference caused by spectrum reuse between D2D transmitters and cellular users (CUEs). Another type is intra-layer interference caused by spectrum sharing among D2D pairs. To mitigate the inter-layer interference, we first derive the interference limited area (ILA) to protect the coverage probability of cellular users by modeling D2D users’ location as a Poisson point process, where a D2D transmitter is allowed to reuse the spectrum of the CUE only if the D2D transmitter is outside the ILA of the CUE. To coordinate the intra-layer interference, the spectrum sharing criterion of D2D pairs is derived based on the (signal-to-interference ratio) SIR requirement of D2D communication. Based on this criterion, D2D pairs are allowed to share the spectrum when one D2D pair is far from another sufficiently. Furthermore, to maximize the energy efficiency of the system, a resource allocation scheme is proposed according to weighted graph coloring theory and the proposed ILA restriction. Simulation results show that our proposed scheme provides significant performance gains over the conventional scheme and the random allocation scheme.
