LHC signals of theSO(5)×U(1)gauge-Higgs unification

Signatures of the SO(5) x U(1) gauge-Higgs unification at LHC and future colliders are explored. The Kaluza-Klein (KK) mass spectra of.; Z; Z(R) and the Higgs self-couplings obey universality relations with the Aharonov-Bohm phase theta(H) in the fifth dimension. The current data at low energies and at LHC indicate theta(H) < 0.2. Couplings of quarks and leptons to KK gauge bosons are determined. Three neutral gauge bosons, the first KK modes Z(R)((1)), Z((1)), and gamma((1)), appear as Z' bosons in dilepton events at LHC. For theta(H) = 0.114, the mass and decay width of Z(R)((1)), Z((1)), and gamma((1))are (5.73, 482), (6.07, 342), and (6.08 TeV, 886 GeV), respectively. For theta(H) = 0.073 their masses are 8.00 similar to 8.61 TeV. An excess of events in the dilepton invariant mass should be observed in the Z' search at the upgraded LHC at 14 TeV.

Arthrobacter luteus가 생산하는 AL - Protease의 효모세포벽 용해 촉진작용

효모세포벽 용해효소의 일종인 Zymolyase(endo-β-1, 3-glucanase)와 더불어 Arthrobacter luteus로 부터 생산되었고, 또한 Zymolyase의 조효소중에서 발견된 바 있는 염기성 AL-protease의 효모세포벽 용해작용을 S. sake의 생세포와 그 세포벽 標品을 사용하여 조사하였다.<br/> AL-protease의 효모 생세포에 대한 용해활성은 그 단독작용만으로는 지극히 미약하였으나, Zymolyase와의 복합작용에 의해서 용해활성은 고도로 상승하였다. 효모생세포를 AL-protease와 Zymolyase로써 단계적인 처리를 할 경우, 효모세포는 AL-protease로 전처리된 뒤에 Zymolyase로 처리되는 처리 순서에 한하여 효과적으로 용해되었다. 이러한 AL-protease의 촉진적 작용은 AL-protease처럼 염기성이고 serine protease로 알려진 몇가지 시판의 효소들 중에서는 발견되지 않았으며, 또한 AL-protease의 이러한 작용은 실험에 사용된 효모들의 배양조건 및 균종에 따라서 커다란 영향을 받는 것으로 밝혀졌다. AL-protease는 효모세포벽 標品으로부터 일정량의 peptide와 상당량의 당을 유리시키고 있으나, 그 세포벽을 66% 이상은 용해시키지 못하였다. 반면에, Zymolyase는 그 단독작용으로도 효모세포벽을 거의 완전히 용해시킬 수 있었다.<br/> 이상의 실험결과를 기초로 하여, S. sake 세포벽의 용해에 있어서 AL-protease의 효소적 작용은, 먼저 AL-protease가 mannan과 단백질로 구성되는 세포벽 표층부에 결합하고, 이어서 그들의 구조를 변화시킴으로써, Zymolyase를 세포벽의 외부로 부터 알카리 불용성 glucan으로 구축되고 있는 세포벽 내부의 골격구조로까지의 침투를 촉진시키는 것으로 추론되었다. The yeast cell wall lytic action of the alkaline AL-protease, which was found out of the crude Zymolyase that a kind of yeast cell wall lytic endo-β-1, 3-glucanase produced from Arthrobacter luteus, was investigated with the viable cells of S. sake and it's cell wall preparation.<br/> AL-protease on the lysis of the viable yeast cells showed very low activities with the alone, but the lytic activities were highly increased with the combination of AL-protease and Zymolyase. On the stepwise treatment of the viable yeast cells with AL-protease and Zymolyase, the cells were lysed highly only by the course having a treatment with Zymolyase after pretreatment with AL-protease. Thus synergistic action of AL-protease was not observed with any some commercial enzymes, known as a type of alkaline and serine protease such as AL-protease, and was also found to be affected greatly by the culture conditions and species of the yeast tested. AL-protease caused the release of some peptide and a lot of sugar from the cell wall preparation, but could not lysed the cell wall more than 66%. Whereas Zymolyase could lysed the cell walls almost completely with alone.<br/> On the basis of these results, the synergistic action of AL-protease on the lysis of S. sake cells is hypothesized that at first AL-protease bind to the yeast cell surface layer consisting of mannan and protein, and then changes their conformation to facilitate the penetration of Zymolyase from the outside to the inside framework layer constituted of alkali insoluble β-1, 3-glucan.
Arthrobacter luteus로부터 유래한 염기성 AL - Protease의 효소학적 성질 및 활성 아미노산 잔기의 검색

Zymolyase 조효소로부터 분리, 정제되었고, 효모 세포벽 용해 촉진물질로 밝혀진 바있는 Arthrobacter luteus 로부터 유래한 염기성 protease(AL-protease)의 효소학적 성질 및 활성 아미노산 잔기를 검색한 결과는 다음과 같다.<br/> 1. AL-protease 는 저해제 DFP 및 PMSF 에 의해서 그 protease 활성 및 용해 촉진활성이 동시에 완전히 소멸되었으며, 그 저해 반응속도는 chymotrypsin 에 대한 것에 비하여 대단히 완만하였다. 그 반응에서 AL-protease와 DFP의 결합 mole 비는 1:1 로 추정되었다.<br/> 2. 정제된 AL-protease의 동결건조품 중에는 종래 효모세포벽 용해반응에 관여하는 것으로 알려진 yeast phosphomannase를 비롯한 다당류 가수분해 효소들의 활성은 그 어느 것도 인정되지 않았다.<br/> 3. AL-protease의 casein에 대한 최적 pH 및 최적온도는 pH 10.5와 65℃이었고, 그 활성은 pH 5~11 사이와 65℃이하에서 안정하였다. 또한, AL-protease의 활성에 미치는 여러가지 금속이온의 영향은 인정되지 않았다.<br/> 4. [(32)^P]-DFP에 의하여 화학수식된 [(32)^P]-DIP-AL-protease에 대한 활성부위의 아미노산 잔기를 검색, 동정하기 위하여 조제용 PAG-전기영동, SDS-PAG-전기영동, Dowex 이온교환 크로마토그래피 및 고압 여지 전기영동을 실시하였고, 그 결과, AL-protease는 활성 부위에 1 분자당 1 mole의 serine 잔기를 가지 는 염기성 protease로 밝혀졌다. The enzymatic properties of the alkaline AL-protease, which had been prepared from the crude zymolyase of Arthrobacter luteus, was investigated together with its active amino acid residue.<br/> Complete inactivation of the proteolytic activity of AL-protease by either DFP or PMSF was simultaneously accompanied by the loss of its lytic effect on the lysis of yeast cell wall. In the reaction, AL-protease showed the pattern of inactivation to decrease very slowly, as compared to that of chymotrypsin, and that enzyme and DFP were found to react with a molar ratio of 1 : 1.<br/> The preparation of AL-protease exhibited no hydrolytic activity in any substrates of polysaccharases, playing a significant role in the lysis of yeast cell wall. The optimum pH and temperature of AL-protease was pH 10.5 and 65℃, respectively. It also showed stability in the pH range from 5 to 11 and at the temperature below 65℃.<br/> Through the identification of the amino acid residue in the active site of the (32)^P-diisopropylph-osphorylated(DIP) AL-protease modified specifically with (32)^P-labeled DFP, AL-protease was found to be a DFP-sensitive enzyme which has a mole of active serine residue involved in its proteolytic activity per mole of the enzyme.
