In this study, nutrition and hygiene education textbooks and teaching and learning curriculum were developed to improve the nutrition and hygiene level of children attending Community Children’s Center. The nutrition and hygiene education workbook consisted of 4 sessions by selecting related topics for each season. Seasonal topics were selected as follows, spring: importance of hygiene to prevent fine dust, summer: prevention of food poisoning, autumn: in-between season and seasonal foods, and winter: cold prevention. In comparison with the effects before and after nutrition and hygiene education, the level of nutrition knowledge increased significantly in the prevention of food poisoning, the in-between season changes and seasonal food, and the prevention of cold after education. However, in the education of the importance of hygiene for fine dust and prevention, there was no significant effect before and after education. After nutrition education using the nutrition and hygiene workbook developed in this study, the students answered 'very satisfied' to the question of whether the education helped to form the correct eating habits. Also, they would like to continue to participate in education, and more than half of the children were positive about the content of the education. Therefore, the workbook developed in this study contributes to the improvement of nutritional and hygiene knowledge level of the children attending the community children’s center and contributes to correct eating behavior in life, thus might helping children prepare for a healthy future.

<P>Adipose tissue is the key organ coordinating whole-body energy homeostasis. Although it has been reported that ring finger protein 20 (RNF20) regulates lipid metabolism in the liver and kidney, the roles of RNF20 in adipose tissue have not been explored. Here, we demonstrate that RNF20 promotes adipogenesis by potentiating the transcriptional activity of peroxisome proliferator–activated receptor-γ (PPARγ). Under normal chow diet feeding, <I>Rnf20</I> defective (<I>Rnf20</I><SUP><I>+/−</I></SUP>) mice exhibited reduced fat mass with smaller adipocytes compared with wild-type littermates. In addition, high-fat diet–fed <I>Rnf20</I><SUP><I>+/−</I></SUP> mice alleviated systemic insulin resistance accompanied by a reduced expansion of fat tissue. Quantitative proteomic analyses revealed significantly decreased levels of PPARγ target proteins in adipose tissue of <I>Rnf20</I><SUP><I>+/−</I></SUP> mice. Mechanistically, RNF20 promoted proteasomal degradation of nuclear corepressor 1 (NCoR1), which led to stimulation of the transcriptional activity of PPARγ. Collectively, these data suggest that RNF20-NCoR1 is a novel axis in adipocyte biology through fine-tuning the transcriptional activity of PPARγ.</P>

Background and Objectives: Human induced pluripotent stem cell (hiPSC)-derived cardiomyocyte (CM) hold great promise as a cellular source of CM for cardiac function restoration in ischemic heart disease. However, the use of animal-derived xenogeneic substances during the biomanufacturing of hiPSC-CM can induce inadvertent immune responses or chronic inflammation, followed by tumorigenicity. In this study, we aimed to reveal the effects of xenogeneic substances on the functional properties and potential immunogenicity of hiPSC-CM during differentiation, demonstrating the quality and safety of hiPSC-based cell therapy. Methods and Results: We successfully generated hiPSC-CM in the presence and absence of xenogeneic substances (xeno-containing (XC) and xeno-free (XF) conditions, respectively), and compared their characteristics, including the contractile functions and glycan profiles. Compared to XC-hiPSC-CM, XF-hiPSC-CM showed early onset of myocyte contractile beating and maturation, with a high expression of cardiac lineage-specific genes (ACTC1, TNNT2, and RYR2) by using MEA and RT-qPCR. We quantified N-glycolylneuraminic acid (Neu5Gc), a xenogeneic sialic acid, in hiPSC-CM using an indirect enzyme-linked immunosorbent assay and liquid chromatography-multiple reaction monitoring-mass spectrometry. Neu5Gc was incorporated into the glycans of hiPSC-CM during xeno-containing differentiation, whereas it was barely detected in XF-hiPSC-CM. Conclusions: To the best of our knowledge, this is the first study to show that the electrophysiological function and glycan profiles of hiPSC-CM can be affected by the presence of xenogeneic substances during their differentiation and maturation. To ensure quality control and safety in hiPSC-based cell therapy, xenogeneic substances should be excluded from the biomanufacturing process.
