To develop a natural re-seeding technology, the optimum soil incorporation time of Chinese milk vetch (CMV) plant residues, seed persistence in soil, biomass production and subsequent rice yield were determined from 2005 to 2007 in rice with CMV green manure in southern Korea. Generally, insufficient seedling stand was regenerated with CMV incorporation to the soil at 20 and 25 days after flowering (DAF), while high regeneration of greater than 400 seedlings per m2 was observed at 35 and 40 DAF. High re-seeding stand of CMV incorporated at 35 DAF or later was related with high seed viability and heavy seed weight. Appreciable number of CMV seeds remained 4 month after burial in soil and a good number of CMV seedlings regenerated from this seed bank at rice harvest time in the fall. Based on the relationships among re-seeding number of CMV plants, seed weight and seed viability, CMV plants should be incorporated into the soil 35 DAF (May 30) or later when CMV seeds were sufficiently matured. The natural re-seeding stand for the 3-year trials was stable ranging from 437 to 700 plants per m2 and the biomass production was sufficient to supply nitrogen for rice growth. The use of re-seeding CMV plant can produce similar rice yield like that of rice without CMV green manure. 본 연구의 목적은 휠체어컬링 국가대표선수와 팀의 요구에 기초하여 현장 활용도와 시합 및 훈련 기여도가 높은 스포츠심리기술훈련 프로그램을 개발하고 그 적용 가능성을 평가해 보는데 있다. 본 연구의 대상자는 현재 휠체어컬링 국 가대표선수 4명을 대상으로 하였다. 상담은 내담자와 편안한 분위기에서 일주일에 1회씩 매 60-90분 총 12회를 실시하였다. 측정도구는 스포츠심리기술 질문지, 운동선수 자기관리행동 질문지, 한국판수행전략검사지, 기분상태프로파일질문지, 경쟁 상태불안 검사지, 경기 참여 휠체어컬링 수행결과 변화, 심리기술훈련 평가질문지, EEG 대뇌반구 비대칭 차이지표 분석 등을 측정하였다. 본 연구결과는 다음과 같다. 첫째, 심리기술훈련은 사전단계에서 연구대상자의 문제점으로 지적되었던 3개의 하위 요인인 각성조절, 걱정, 주의집중을 긍정적으로 변화시킨 것으로 나타났다. 둘째, 심리기술훈련은 휠체어컬링 대표선수들의 수행변화에도 긍정적인 영향을 미치는 것으로 나타났다. 셋째, 개방형질문지 분석결과 연구대상자는 심리기 술훈련 프로그램에 대한 필요성을 인식하고 경기력에도 도움이 된다고 인식하고 있는 것으로 나타났다. 넷째, 스포츠심리 기술훈련 처치 전에는 EEG대뇌반구비대칭 차이지표가 부정적 정서 상태를 보였지만 처치 후 긍정적 정서 상태를 보였다. 이것은 뇌 과학적 접근인 EEG 뇌파분석을 통해 심리기술훈련이 휠체어컬링 대표선수들의 심리적 상태에 긍정적인 영향을 미친다는 것을 과학적이고 객관적으로 검증하였다. The present study aimed to develop a sports psychological skill training program and to test its applicability to improve the performance of national wheelchair curling players. A total of 4 participants completed the study. Subjects have been to 12 sessions of 6 different psychological skills training programs (routine training, attentional focus training, writing practice training in diary, relation training, self-control training, and positive self-talk training). The effectiveness of psychological skills training has been evaluated with 6 questionnaires and EEG measurement. The result was that psychological skills training had improved coping with adversity, peaking under pressure, and concentration, self-management and reduced anxiety of Korean national wheelchair curling player. The result was that the EEG inter-hemispheric asymmetry index showed negative emotional states before psychological skill training but positive emotional states after. The result implies that psychological skill training plays an important mediating role in bringing about positive effects in the psychological elements and competitiveness in national wheelchair curling players.

The marine microalga Chlorella sp. was cultivated under mixotrophic conditions using methanol as an organic carbon source, which may also act to maintain the sterility of the medium for long-term outdoor cultivation. The optimal methanol concentration was determined to be 1% (v/v) for both cell growth and lipid production when supplying 5% CO_2 with 450 μE/m^2/sec of continuous illumination. Under these conditions, the maximal cell biomass and total lipid production were 4.2 g dry wt/L and 17.5% (w/w), respectively, compared to 2.2 g dry wt/L and 12.5% (w/w) from autotrophic growth. Cell growth was inhibited at methanol concentrations above 1% (v/v) due to increased toxicity, whereas 1% methanol alone sustained 1.0 g dry wt/L and 4.8% total lipid production. We found that methanol was preferentially consumed during the initial period of cultivation, and carbon dioxide was consumed when the methanol was depleted. A 12:12 h (light:dark) cyclic illumination period produced favorable cell growth (3.6 g dry wt/L). Higher lipid production was observed with cyclic illumination than with continuous illumination (18.6% (w/w) vs 17.5% (w/w)), and better lipid production was also obtained under mixotrophic rather than autotrophic conditions. Interestingly, under mixotrophic conditions with 12:12 (h) cyclic illumination,high proportions of C_(16:0), C_(18:0), and C_(18:1) were observed,which are beneficial for biodiesel production. These results strongly indicate that the carbon source is important for controlling both lipid composition and cell growth under mixotrophic conditions, and they suggest that methanol could be utilized to scale up production to an open pond type system for outdoor cultivation where light illumination changes periodically.
Immobilization of Yeast Pichia stipitis for Ethanol Production

Cotton을 효모 세포(Pichia stipitis)의 고정화 담체로 사용하기 위하여 2-(diethylamino)ethyl chloride hydrochloride (DEAE·HCl)로 derivatization 시켰다. 0.5 M DEAE·HCl로 처리하였을 때, 효모 세포가 완전히 흡착하였으며, 이것은 DEAE-cotton g 당 101.8 mg의 효모 세포가 흡착하는 것이고, DEAE-cellulose에 효모 세포가 흡착하는 양의 약 6배 이상인 것으로 확인되었다. DEAE-cotton을 이용하여 효모 세포를 고정화하고, 이것을 ethanol 생산에 이용하였을 경우, glucose와 xylose가 포함된 배지에서 단당류에 대한 ethanol 수율로 0.33 정도로 ethanol을 생산 할 수 있다는 것을 실험적으로 확인하였다. 이를 이용하여 lignocellulosic bomass의 가수분해물로부터 bioethanol 생산에 이용될 수 있을 것으로 기대되어진다. DEAE-cotton에서 얻어진 결과는 DEAE-cellulose에서 같은 실험을 실시하여 서로 비교 분석하였다. In this study, DEAE-cotton [derivatized by 2-(diethylamino)ethyl chloride hydrochloride (DEAE·HCl)] was prepared as a carrier for immobilized Pichia stipitis for ethanol production. When cotton was derivatized with 0.5 M DEAE·HCl, the yeast cell suspension was adsorbed at 100% of the initial cell OD600. The adsorbed yeast cells were estimated to be 101.8 mg-dry cells/g-DEAE-cotton. In particular, when a flask culture using the immobilized yeast cells was conducted in a glucose and xylose-containing medium, the yeast cells on the DEAE-cotton gradually produced ethanol, according to glucose and xylose consumption; the ethanol yield was approximately 0.33 g-ethanol/g-monosaccharide. Because DEAE-cotton was successfully used as a carrier for ethanol production from a glucose and xylose-containing medium, we expect that this bioethanol production process may be used for the bioethanol production process from the hydrolysate of lignocellulosic biomass. All the results of DEAE-cotton were compared with those of DEAE-cellulose as a carrier for immobilization.
