Adipogenesis related to energy consumption in DNAJC6- transfected 3T3-L1 cells

Resting metabolic rate accounts for 60-70% of total daily energy expenditure, is essential for establishing strategies for obesity prevention. In a previous GWAS obesity study, we found that the DNAJC6 gene was related to energy metabolism. Compared between DNAJC6-overexpressed 3T3-L1 cells (TgHsp), and the control (3T3-L1), we identified what mechanisms are involved, such as lipid metabolism, inflammatory cytokines, insulin resistance, and mitochondrial dysfunction. The adipogenesis (PPARγ, C/EBP α, aP2) and adipokines (leptin, adiponectin, leptin receptor) were significantly depressed in TgHsp cells complared to the control. The proteins involved in the insulin signals (IRS, AKT, GLUT4) were depressed in TgHsp cells, compared to the control. O₂ consumption, ATP production, and UCP1 in mitochondria were decreased in TgHsp cells. We found that the DNAJC6 gene was positively involved in inhibiting adipogenesis with the possibility of dysfunction of ATP production and insulin resistance.
