        천연 항균제처리를 병용한 과채류의 자연 저온저장기술 개발에 관한 연구

        과채류의 신선도 유지 및 병리적 장해를 감소시켜 저장성을 증진시키기 위하여, 천연 식물성 항균제언 grapefruit 종자추출물(Grapefruit seed extract : GFSE)로 처리하고 polyethylene film 포장하여 과채류의 품질 향상을 유도할 수 있었고 과채류 변패 미생물의 오염 및 생육을 억제할 수 있었다. GFSE는 세균, 곰팡이 등 광범위한 범위의 병리미생물에 대하여 250ppm~500ppm의 처리농도로 뚜렷한 항균효과를 나타내었다. 변패된 과채류에서 분리한 Bacillus subtilis와 같은 세균 균체세포에 대하여 100ppm 이상의 GFSE 용액처리로 균의 증식이 억제되는 뚜렷한 항균력을 관찰할 수 있었으며, 전자현미경 실험결과, 균체세포 및 내생포자의 세 포막기능이 파괴되어 세포내용물이 균체외부로 유출되어 균체의 생육이 억제되는 것을 관찰할 수 있었으며, 곰팡이 Fusarium sp.의 균사체도 1,000ppm의 용액처리로 세포벽기능이 파괴되었고, Aspergillus sp.의 포자 내용물도 GFSE 처리로 소실되어 정상적인 발아기능이 저해됨을 알 수 있었다. 한편, 농가에서 수확한 가지, 산나물, 오이, 양다래, 금귤, 포도 등을 GFSE용액에 침지처리하고 polyehtlyene film 포장하여 자연저온(10℃~15℃)에서 저장함으로서, 색택 및 조직면에서 우수한 저장효과를 얻을 수 있었고, 포도에 GFSE를 처리한 경우, Botrytis cinerea에 의한 부패율을 크게 감소시켰다. 아울러, GFSE 처리한 오이의 경도는 저장 4주 후에도 초기경도값의 65% 이상을 나타내어 대조구의 15%에 비하여 높은 경도를 유지하고 있었다. 실험결과를 고찰하여 볼 때, GFSE를 병용한 과채류의 자연 저온방법은 과채류의 선도연장 및 품질보존 효과에 우수함을 알 수 있었다. In order to prevent the postharvest decay and to promote the freshness retention of fruits and vegetables grapefruit seed extract(CFSE), natural microorganism control agent, was applied to the preservation of fresh fruits and vegetables. Fresh fruits and vegetables treated with GFSE and stored in polyethylene film (0.1㎜) at 10℃~15℃ of natural low temperature kept better qualities in color and texture than the GFSE-not-treated control. The treatment using GFSE in a 250ppm to 500ppm concentration seemed to be an effective one for the control of Botrytis cinerea isolated in red wine grapes. After 4 weeks of storage the firmness rate of cucumbers treated with the dilute GFSE was four times higher than that of non-treated ones. GFSE showed effective inhibitory action towards plant pathological bacteria and fungi which were involved in the decay of fruits and vegetables. Minimum inhibitory concentrations of GFSE towards them were in the range of 250ppm to 500ppm. Direct visualization of microbial cells and spores using electron microscopy showed microbial cells and fungal spores the function of which was destroyed by treating with the dilute solutions of GFSE. It was observed that GFSE would reduce disease damages and have bactericide & fungicide properties during the storage of such fruits and vegetables as egg plant, wild edible greens, kumquat, and kiwifruit.

        도라지의 휘발성분에 관한 연구

        To identify the volatiles of Platycodon roots, the three-year-old and the 24-year-old Platycodon roots cultivated in a local farm were extracted with ethyl ether by SDE method. 54.71㎎% and 80.01㎎% of volatile components were collected in the three-year-old and in 24-year-old Platycodon roots, respectively. As a result of GC/MS(TIC) analysis, 24 and 28 compounds were identified, respectively. The amounts of volatile components such as hexanol, trans-2-hexenol and nonanal in 24-year-old roots were detected above ten times than those in 3-year-old roots. The Volatiles of 24-year-old roots were fractionated by SCC and identified by GC/MS to be 10 hydrocarbons, 8 alcohols, 10 aldehydes, 5 esters, 6 acids and 1 unknown compound as the major components. The species and total amounts of volatile component in 24-year-old Platycodon roots were considered to be higher than those in 3-year-old Platycodon roots. * Abbreviations 1.         Platycodon root contains abundant pharmaceutical substances and is widely used as a food and a medicinal herb. The three-year-old and the 24-year-old Platycodon roots cultivated in a local farm were analyzed and compared with their pharmaceutical substances to identify the major components. Reverse column HPLC analysis of ethanol extracts from the 24-year-old roots showed some distintive peaks after the retention time of 50 min which were absent in the extracts from the three-year-old roots. This indicates that several pharmaceutical substances are present in the older roots. There were no differences in sterol composition and saponin content between the 24-year and three-year-old roots. The 15-year-old and the 23-year-old Platycodon roots, the 6-year-old Ginseng root, and the three-year-old Condonopsis root all did not show significant anticomplement activity.

        Platycodon root contains abundant pharmaceutical substances and is widely used as a food and a medicinal herb. To identify the major components of the three-year-old and the 24-year-old platycodon roots cultivated in a local farm were analyzed and compared with their chemical compositions prior to their pharmaceutical substances, which were used as the fundamental data. There were no differences in chemical composition, inorganic compositions and fatty acid compositions between the 24-year-old and three-year-old roots. Total amino acid contents were 375.8 ㎎% and 891.6 ㎎% in the 24-year-old and three-year-old platycodcon roots, respectively. Sixteen species of amino acid were identified by amino acid autolyzer except cysteine both in the 24-year-old and three-year-old roots. Linoleic acid, which was the highest fatty acid contained both in the 24-year-old and three-year-old roots, was estimated to be 48.7% and 39.1% of total fatty acids, respectively.

        District Heating System(DH) is one of the most excellent system in terms of energy saving and environmental protection. In order to comply with the global energy crisis and environment problem , it is necessary to research and develop the energy utilization technology with the reduction of the primary energy usage. Although the incresing rate of energy consumption started to attenuate , the entire growth in Korea. The energy demand in Korea increses by annual 3.7% during the period from 2000 to 2006. And it is expected that we should take a charge of the greenhouse gas reduction obligation by the Climatic Charge Convention(Kyoto Protocol) during the 2nd pledge period(2013~2017). This thesis reports something about comparisons between Braze & Gasket heat exchanger.

        도라지 조직배양의 최적배지와 배양조건

        도라지의 약리성 성분을 많이 함유하고 있어 생약재료로서의 가치가 높다. 도라지의 잎, 줄기, 뿌리의 조직부위별 기내배양을 통해 캘러스 유기, 체세포배 형성및 식물체 재분화방법을 조사하였다. 줄기나 뿌리의 절편체보다도 잎 절편체를 치상시 캘러스가 더 잘 유기되었다. 캘러스가 유기배지는 1.0㎎/L2,4-D 와 1.0㎎/L kinetin이 첨가된 MS 기본배지에서 가장 양호하였다. 고체배지 상태에서 유기된 캘러스를 6-7일간 현탁배양하는 과정을 거쳐 활력있는 캘러스 덩어리를 만들고 MS 기본배지에 1.0㎎/L BA와 1.0㎎/L NAA가 혼합된 배지에 치상함으로 75일만에 완전한 도라지 재분화 개체를 얻을 수 있었다. Platycodon grandiflorus DC contains abundant parmaceutical substances and is widely used .is a medicinal herb. Int this study, through tissue cultures of different organelles such as leaves, stems and roots of Platycodon, callus and somatic embryos were induced and whole plants were regenerated. Whole plautlets could be regenerated in 75 days after initiaton of of culture of callus on the MS medium containing 1㎎/L BA and 1㎎/L NAA To facilitate the regeneration, the callus induced from the suspension culture for 6-7 days was transferred to the solid medium. The leaf explant was better than stem or root explants in callus induction and regeneration of whole plantlets.
