        Investigation of Germicide and Growth Enhancer Effects on Bean Sprout using NMR-based Metabolomics

        Bean sprouts are often cultivated in the circumstances prevailing in the improper using of germicide and growth enhancer. The influence of ingestion those bean sprouts are unknown. The components of the bean sprouts are needed to evaluate for food safety. The extracts of the control, 0.5 g/L germicide, 1 g/L germicide, 12.5 mL/L growth enhancer and 25 mL/L growth enhancer were used to compare the components in the experiment. Nuclear Magnetic Resonance spectroscopy (NMR) was used to analyze the extracts. Statistical analysis of metabolomics showed significant changes between the control and head and the stem of the bean sprouts. Significant changes in metabolites were identified with the bean sprouts cultivated with germicide and growth enhancer by applying qualitative and quantitative analysis. Similar changes in the area of the bean sprouts were observed after treated to germicide and growth enhancer. Although treating germicide and growth enhancer showed no particular harmful metabolites changes to human, it made significant changes in the morphological and the metabolites of the bean sprouts. These changes indicate that the germicide and growth enhancer has substantially potential to influence the growth of the bean sprouts.

        HR-MAS NMR Technique for Metabolic Profiling of Powdery Ginseng

        Ginseng is used as a medicinal ingredient. The quality control of species, age, origin and manufacturing process is important. The metabolome of ginseng about quality was studied in many reports. Almost studies carried out the extract of ginseng, however, the reproducibility cannot be obtained using extracted sample. In this study, powdery ginseng samples were analyzed using high resolution-magic angle spinning nuclear magnetic resonance (HR-MAS NMR)-based metabolomics except extraction step. Sample was measured three times using 600 MHz NMR spectrometer equipped with nano probe. Reproducibility can be enhanced using this method and the metabolic profiles of ginseng were identified and quantified.

        1H-NMR-based Metabolomic Study of Miamiensis avidus-infected Olive Flounder

        The scuticociliate Miamiensis avidus (M. avidus) is an aetiological agent of mass mortalities in cultured fish worldwide. The external signs of infection by this histiophagous parasite are hemorrhage, muscle ulceration, and exophthalmia. In this study, we infected olive flounder (Paralichthys olivaceus) with M. avidus under laboratory conditions, and used1H-nuclear magnetic resonance (NMR) spectroscopy to detect the metabolomic alterations during the infection. Multivariate data analysis showed that there were distinct differences in metabolomes between the control and the infected groups. Analysis of the metabolomic changes indicated that the olive flounder experienced severe metabolic shifts to anaerobic respiration and hypoxia.

        The difference of metabolic profile between male and female zebrafish

        Various experiments using zebrafish have been highlighted recently in the scientific community. Because it is possible to conduct practical experiment from various neurological research to area of genetic study or toxicity experiment. However, gender difference effects are nearly not considered. If the gender differences of zebrafish are considered it is possible to obtain more accurate data. In this study, zebrafish which have different genders were compared each other with NMR-based metabolomics. The extracts of male and female zebrafish were measured by 600 MHz NMR spectrometer. Statistical analysis and target profiling were conducted. As a result, muscle related metabolites were observed in male zebrafish and nerve related metabolites were observed in female zebrafish.

        Toxicological Assessment of Microcystin-LR to Zebrafish (Danio rerio) Using Metabolomics

        Microcystins are one of the most frequently found hepatotoxic cyclic peptides in fresh- and brackish-water. Zebrafish (Danio rerio) were exposed to 20 ppb of microcystin-LR (MCLR) for 72 h in order to investigate the metabolic responses. Proton nuclear magnetic resonance (1H-NMR) spectroscopy was used to analyze the metabolic profiles. As a result, male and female zebrafish showed different patterns of metabolic perturbation in relation to their detoxification profiles. Glutathione was reduced for the MCLR excretion, and fumarate and creatine were increased for the energy demands required for detoxification in the male zebrafish. The correlation of metabolites in the metabolic pathway network was changed due to MCLR exposure. The correlations connected with myo-inositol, betaine, and NADPH considerably altered in the MCLR-exposed group.

        Applications of NMR spectroscopy based metabolomics: a review

        Metabolomics is the study which detects the changes of metabolites level. Metabolomics is a terminal view of the biological system. The end products of the metabolism, metabolites, reflect the responses to external environment. Therefore metabolomics gives the additional information about understanding the metabolic pathways. These metabolites can be used as biomarkers that indicate the disease or external stresses such as exposure to toxicant. Many kinds of biological samples are used in metabolomics, for example, cell, tissue, and bio fluids. NMR spectroscopy is one of the tools of metabolomics. NMR data are analyzed by multivariate statistical analysis and target profiling technique. Recently, NMR-based metabolomics is a growing field in various studies such as disease diagnosis, forensic science, and toxicity assessment.

        Liver abscess due to Klebsiella pneumoniae in a healthy 12-year-old boy

        Pyogenic liver abscess (PLA) is rare in healthy children. We report a case of PLA in an immunocompetent 12-year-old boy. Percutaneous catheter drainage was performed for the abscess. In addition,parenteral antibiotics were administered for 3 weeks. Klebsiella pneumoniae was detected in the culture of blood and drained fluid. Here, we present this case and a brief review of the literature on this subject.

      • 암모니아, 이산화탄소 센서기반 퇴비 부숙도 측정법 개발

        이 연구는 선행 연구에서 전자센서를 이용한 암모니와 이산화탄소 측정과 측정 범위 설정을 통해 센서기반 부숙도 측정가능을 확인하였으나 고농도 시료에 측정시 센서 안정화에 대한 문제점을 보안하기 위해 수행되었다. 센싱기반 장치 및 부숙도 판별기준(안)을 활용한 부숙도 분석값과 기존 비료공정규격 부숙도 측정 방법인 콤백과 솔비타의 분석값간의 매우 유의한(P<0.001) 상관관계를 통해 기존의 기계적 측정법을 기준으로 적합하게 기준이 설정되었음을 확인하였다. 판별기준 중 특히 암모니아 발생량 기준값이 상관관계가 0.82 (P<0.001) 매우 높았다. 이는 기계적 측정법인 솔비타와 콤백 판정값이 일치하는 값을 기준(총 시료 773점 중 645점 일치)으로 발생량 측정법의 상관관계를 분석했다. 신규 기체발생량측정법의 비료공정규격상 추가는 2022년 본 과제를 통해 만들어진 휴대용 장치의 현장 실증 결과를 통한 2차 검증 후 비료관리법 비료공정규격상 신규 부숙도 측정 방법을 추가할 예정이다. 또한, 1차년도에 EC센서와 PID센서를 통한 부숙도 판정 기준에 관한 검증을 실시했으며, 2차년도에 NDIR센서 측정을 통해 3가지 센서중 현장 시료 측정을 위한 센서를 최종 선택함으로써 본 과제를 통한 축산농가현장에서 부숙도 측정을 신속하게 할 수는 장치를 개발할 수 있을 것이다. 2차년도에 3가지 센서에 대한 분석값 값의 신뢰도 및 상관관계 분석을 진행해 신규부숙도 측정법의 적용 범위 또한 확대할 계획이다.

      • 대학생의 장애체험 교육을 통한 장애에 대한 태도 및 인식 변화

        The purpose of the study was to observe the change in attitude and awareness towards disability through experience-based education of university students. The data for the analysis was collected through questionnaire survey before and after disability experience from July 25 to August 23, 2016, and the sample consisted of 30 participants who were university students. The experience included blindness and physical disability and the participants were given several activities to try out in either blindfolds or wheelchairs. The results were as follows : 1) When we compare the change in attitude before and after the experience, there were significant change in items such as ‘being near a disabled person doesn’t make me uncomfortable’, ‘they aren’t that different from abled people’ and ‘it wouldn"t make me uncomfortable if a friend introduced me to a disabled person.’ 2) The difference in all the items before and after the experience were small, which shows that change in attitude cannot be achieved in a short amount of time. Therefore, such factor should be considered when developing a future experience-based educational programs for accurate results. 3) Change in awareness after the experience were big in items related to spatial problems, such as ‘toilet for the disabled’, ‘raised blocks for the blind’, ‘the surroundings’, etc. which shows that awareness towards the space designed for the disabled was small.
