        홍삼 추출물 첨가 혼합 발효유의 특성

        우유와 두유를 2:1로 혼합하고 홍삼 추출물을 첨가하여 발효유를 제조하였고 2종류의 스타터를 사용하여 발효 특성을 시험하였다. 2종류의 스타터를 사용한 모든 처리구에서 배양 15시간 후 pH는 $3.90{\sim}3.94$ 사이로 나타나 산 생성 촉진 효과가 있었고, 산 생성은 홍삼 추출물 첨가량이 증가함에 따라 조금씩 증가되는 것으로 나타났다. 홍삼 추출물 첨가량이 증가할수록 생균수는 증가하여 Lactobacillus acidophilus KCTC 3150를 스타터로 사용한 균에서 1.0% 홍삼 추출물 첨가량은 $6.26{\times}10^8\;cfu/mL$로서 최대 균수를 나타내었으며, 배양 12시간 이후에 대조구에서는 감소하는 반면 홍삼 추출물 첨가구에서는 젖산균수는 계속해서 최대균수를 유지하고 있었다. 유기산 생성은 lactic acid인 경우 Lactobacillus acidophilus KCTC3150에서 1.0% 홍삼 추출물 첨가구에서 332.22 mM로 가장 높았으며 홍삼 추출물 첨가량이 증가됨에 따라 lactic acid의 생성량도 증가되었다. 당 분해율은 두 균주 모두 홍삼 추출물 첨가량이 증가함에 따라 stachylose의 분해율이 증가하였고, Lactobacillus acidophilus KCTC3150 균주의 경우 stachylose의 분해율이 최대 19%인 반면, Lactobacillus salivarius subsp. salivarius CNU27의 경우 stachylose의 최대 분해율은 54%로서 분해율이 월등히 높았다. 점도는 홍삼추출물 첨가량이 1.0% 첨가구에서 Lactobacillus acidophilus KCTC3150, Lactobacillus salivarius subsp. salivarius CNU27 각각 780 cP로 가장 높게 나타났다. 관능검사 결과는 0.2% 홍삼 추출물을 첨가하고 Lactobacillus salivarius subsp. salivarius CNU27를 스타터로 사용한 발효유가 $4.14{\pm}0.64$로 기호도가 가장 좋게 나타났다. This experiment was carried out to examine the fermentation properties of yogurt with bovine milk and soybean milk at the mixed ratio of 2:1 and added 0.1, 0.2, 0.4 and 1.0% red ginseng extract. The effect on promoting the fermentation by additives 0.1, 0.2, 0.4 and 1.0% red ginseng extracts were higher and pH was $3.90{\sim}3.94$ when Lactobacillus acidophilus KCTC3150 and Lactobacillus salivarius ssp. salivarius CNU27 were used. Titratable acidity showed a little inhibiting due to increasing red ginseng extract content. The average viable counts of lactic acid bacteria after 15 hour culture was the highest level of $6.26{\times}10^8cfu/mL$ when Lactobacillus acidophilus KCTC3150 was used, and the additives content of red ginseng extract was 1.0% The production of lactic acid was the highest and the concentration was 332.22 mM when Lactobacillus acidophilus KCTC3150 was used, and the additives content of red ginseng extracts was 1.0% Lactose hydrolysis was completely hydrolyzed when Lactobacillus acidophilus KCTC3150 and Lactobacillus salivarius ssp. salivarius CNU27 were used. The highest viscosity of yogurt was 780 cP when Lactobacillus acidophilus KCTC3150 and Lactobacillus salivarius subsp. salivarius CNU27 were used and red ginseng extract was added 1.0% The overall acceptability, $4.17{\pm}0.64$, was the highest when Lactobacillus salivarius subsp. salivarius CNU27 was used and the additives content of red ginseng extract was 0.2%.

        Enterococcus faecium KHM-11를 이용한 요구르트 급여가 자돈의 성장에 미치는 영향

        A total of 70 colonies were isolated from the Korean human milk samples on the BCP plate count agar. These LAB isolates were subcultured in 10% reconstituted skim milk, and two strain thereof were finally selected for their highest acid productions. These strains were identified as Enterococcusfaecium based on 16S rDNA sequencing data, named as Enterococcus faecium KHM-11. Sugar utilization of E. faecium KHM-11 was investigated by API 50CH kit, and 19 different sugars including D-arabinose, L-arabinose, galactose, D-glucose, D-fructose, and D-mannose were utilized. For fermentation profiles, a yogurt inoculated by E. faecium KHM-11 after 15 hour, reached at pH 4.09, titratable acidity at 1.10% and average viable counts $1.30{\times}10^9\;CFU/mL$. Effects of the administration of yogurt 0.5% of piglet diet to piglets were investigated for growth rate, analysis of blood and incidence of diarrhea. 24 heads of piglets were divided into two groups: the experimental and the control of 12 animals each. The average growth rate in the yogurt-fed group was higher for 21.67%, compared with control (p<0.05). There were no differences in the concentrations of blood glucose, cholesterol, albumin and globulin between the two treatments. Incidence of diarrhea was no in pigs fed yogurt as compare to control.

        한국인 모유로부터 분리한 Enterococcus faecium KHM-11의 Amylase 활성과 쌀 첨가 요구르트의 특성

        This studies were carried out to assess fermentation properties of amylase activity and added rice yogurt of Enterococcus faecium KHM-11 isolated from Korean human milk. The amylase activity of Enterococcus faecium KHM-11 was above 23 units. Characterization of carbohydrate fermentation of Enterococcus faecium KHM-11 has D-ribose, D-lactose, L-arabinose and starch. Titratable acidity and viable count of lactic acid bacteria of 4% rice yogurt was higher compared to the 0%. Therefore we were discussed Enterococcus faecium KHM-11 is suitable microorganism for fermented milk added rice powder. Hydrolysates of sugars of fermented milk with 4% rice powder cultured Enterococcus faecium KHM-11 were analyzed by TLC and HPLC. Hydrolysates of lactose and galactose were revealed and hydrolysates of glucose was not revealed in results of TLC and HPLC.

        녹색 홀그레인 보리, 밀, 찹쌀, 멥쌀 분말을 첨가한 발효유의 특성

        Yogurt was prepared from skim milk added with 2, 4 or 6% of green whole grain of barley, wheat, glutinous rice and common rice, respectively. Changes in pH, titratable acidity, viable cell counts and viscosity during fermentation were monitored and its sensory evaluation was also performed. The optimum level of additives such as green whole grain of barley, wheat, glutinous rice and common rice for yogurt manufacture was selected to 2%. The samples added with green whole grain of barley and wheat powders reached pH 4.5 in 12 hours. After 4 hours of fermentation, pH, the viable cell counts and viscosity in samples added with 2% green whole grain of barley, wheat, glutinous rice and common rice powders were the highest. Especially, in overall sensory evaluation using 5 parameters, the sensory scores of glutinous rice and common rice 2% yogurts were significantly high. From this experiment, the additives optimum level of glutinous rice and common rice powders was selected to be 2%.

        Holstein 초유 중 Lactoferrin, IgA, IgG<sub>1</sub>, IgG<sub>2</sub> 정량과 미생물의 성장에 미치는 영향

        본 실험은 홀스타인 젖소의 초유에 함유된 lactoferrin, IgA, $IgG_1,\;IgG_2$의 함량과 병원성미생물의 항균효과, 유산균증식효과를 알아보기 위하여 수행하였다. 홀스타인종 젖소 초유의 lactoferrin, IgA, $IgG_1,\;IgG_2$의 함량은 여름철의 경우 분만 후 첫째 날에 각각 0.30 mg/mL, 0.37 mg/mL, 4.00 mg/mL, 0.37 mg/mL, 겨울철의 경우 분만 후 첫째 날에 각각 1.16 mg/mL, 2.60 mg/mL, 13.35 mg/mL, 1.30 mg/mL로 측정되었다. 무열처리한 초유와 $65^{\circ}C$에서 30분간 열처리한 초유에서 배양한 병원성 미생물의 변화는 E. coli 균주에 대하여 초유의 항균활성이 나타났다. 초유 첫째 날, 둘째 날, 세째 날의 유산균수 성장촉진 효과는 B. longum, L. acidophilus S. themophilus의 상업용 혼합균주와 L. acidophilus, L. casei, L. bulgaricus, L. lactis subsp. cremoris에서 모두 정상유보다 균수가 증가하였다. 초유 첫째 날, 둘째 날, 세째 날의 pH는 배양 15시간 후 B. longum, L. acidophilus S. themophilus의 상업용 혼합균주와 L. acidophilus, L. casei, L. bulgaricus, L. lactis subsp. cremoris에서 각각 4.97-5.22 및 5.96-6.47로 나타났으나, 정상유에서는 각각 4.89, 6.50-6.81이었다. 초유 첫째 날, 둘째 날, 세째 날의 산도는 B. longum, L. acidophilus S. themophilus의 상업용 혼합균주와 L. acidophilus, L. casei, L. bulgaricus, L. lactis subsp. cremoris에서 각각 0.75-0.88% 및 0.29-0.48%로 나타났으나, 정상유에서는 각각 0.70%, 0.20-0.25% 이었다. This experiment was carried out to measure the content of lactoferrin, IgA, $IgG_1,\;IgG_2$, in Holstein colostrum, and to test the effect of it's colostrum on the antibacterial activity to pathogenic bacteria and the growth stimulation of lactic acid bacteria. Colostrum was collected at the first, second, and third day after parturition in summer and winter season. The levels of lactoferrin, IgA, $IgG_1,\;and\;IgG_2$ in Holstein cow colostrum were 0.30 mg/mL, 0.37 mg/mL, 4.00 mg/mL, 0.37 mg/mL, respectively, on the first day of the summer season whereas they were 1.16 mg/mL, 2.60 mg/mL, 13.35 mg/mL, 1.30 mg/mL on the first day of the winter season, postpartum. Heat treated ($65^{\circ}C$ for 30 min) or non-treated colostrum showed antibacterial activity toward Escherichia coli. The growth of commercial mixed strains (Bifidobacterium longum, Lactobacillus acidophilus, Streptococcus themophilus), L. acidophilus, L. casei, L. bulgaricus, and L. lactis subsp. cremoris were improved in first, second and third day colostrum compared to normal milk. Commercial miked strains (B. longum, L. acidophilus S. themophilus) lowered the pH to 4.97-5.22 and 4.89 while increasing the titratable acidity to 0.75-0.88% and 0.70% in colostrum and normal milk, respectively. However, L. casei, L. bulgaricus, L. lactis subsp. cremoris lowered the pH to 5.96-6.47 and 6.5-6.8 while increasing the titratable acidity to 0.29-0.48% and 0.20-0.25% in colostrum and normal milk, respectively.

        유청단백질의 분리 및 단백질 분해 효소에 의한 유청단백질의 가수분해 양상

        The aim of this study was to introduce a simple method for isolation of α-lactalbumin, β-lactoglobulin and bovine serum albumin from cow’s milk, and peptides produced by enzymatic hydrolysis of α-lactalbumin, β-lactoglobulin and bovine serum albumin with alcalase. Whey protein were precipitated from whey by ammonium sulfate and, α-lactalbumin and β-lactoglobulin were isolated using Hi Prep 26/60 Sephacryl S-100 column gel filtration chromatography. Bovine serum albumin and β-lactoglobulin were isolated by Mono-Q 5/50 GL column anion exchange chromatography of the 50% Ammonium Sulfate-supernatant. Isolated whey proteins were hydrolyzed by proteolytic alcalase. Tricine SDS-PAGE and reverse-phase HPLC analyses revealed that almost hydrolyzed all the α-lactalbumin, β-lactoglobulin and bovine serum albumin with alcalase. Molecular weight of various peptides derived from alcalase hydrolysate were small molecular weight than 3.5 kDa.

        발효초유사료 급여가 자돈의 성장에 미치는 영향

        This study was carried out to assess the fermentation properties of lactic acid bacteria (LAB) isolated from bovine colostrum and effects of feeding fermented colostrum feed on the growth to piglet. A total of 427 colonies were isolated from bovine colostrum on the BCP plate count agar. These LAB isolated were subcultured in 10% reconstituted skim milk, and seven strain thereof were selected for their highest acid productions, and two strain thereof were finally selected for their excellent sugar utilization. These strains were identified as Streptococcus thermophilus and Streptococcus macedonicus based on l6S rDNA sequencing data, named S. thermophilus CNB-11 and S. macedonicus CNB-11 respectively. For fermentation profiles, sugar utilization, acid production and viable cell counts were excellent in S. thermophilus CNB-11 as compared with S. macedoniclts CNB-11 after 48 hour. The effect of feeding fermented colostrum feed 0.5% using S. thermophilus CNB-11 was investigated for growth rate, analysis of blood and incidence of diarrhea. 24 heads of piglets were divided into two groups: the experimental and the control of 12 animals each. The average growth rate in the pigs fed fermented colostrum feed was higher 16.73% compared with control diet (p<0.05). There were no differences in the concentrations of blood glucose, cholesterol, albumin and globulin in pigs fed fermented colostrum feed as compared with control piglets. Incidence of diarrhea was no in pigs fed fermented colostrum feed as compared with control piglets.

        냉장 및 실온 보존에 따른 모유와 환원조제분유 중의 총균수와 대장균수의 변화

        본 연구는 손착유 모유와 breast-pump에 의한 기계착유 모유 중의 총균수와 대장균수를 검사하고 또한 냉장 및 실온 보존에 따른 모유(초유, 성숙유)와 환원조제분유의 저장성을 파악하고자 시험하였다. 시험 결과 breast-pump로 수집한 모유 중의 총균수는 2.02×10⁴ CFU/mL로 손착유에 의한 총균수 1.06×10⁴ CFU/mL보다 약 2배 가량 많이 나타났으며 특히 대장균의 경우, 손착유에 의한 감염율이 12.9%인 반면 breast-pump로 수집한 모유의 감염율은 19.5%로 높게 나타났다. 또한 모유와 환원조제분유를 냉장(4℃) 및 실온(20℃, 30℃) 상태에서 보존하면서 총균수와 대장균수의 변화를 시험한 결과, 4℃에서 대장균수의 변화는 10일 동안 뚜렷한 변화를 보이지 않았으며 총균수는 지속적으로 약간 감소하는 경향을 나타냈다. 20℃에서 총균수의 변화는 모유 성숙유와 모유 초유에서 뚜렷한 증가 추세를 보이지 않았으나 대장균의 경우 지속적인 증가 추세를 보였으며, 특히 환원조제분유에서 대장균수는 12시간 이후 급속히 증가하는 경향을 나타냈다. 30℃에서 총균수의 변화는 모유 성숙유의 경우 12시간 이후 급속히 증가하는 경향을 보였으나 모유 초유의 경우 뚜렷한 증가 추세를 보여주지 않았다. 반면에 환원조제분유는 6시간 이후 급속히 증가하는 추세를 나타냈다. Human milk is normally contaminated with various microorganisms, which seem to produce no ill effects. A contamination of hand-expressed and pump-expressed human milk is a major concern in the collection of storage milk. In this study we compared milks collected by two methods, hand expression and suction breast pump, to quantify differences in the degree of bacterial contamination. Thirty-one samples had been manually expressed. The mean of total bacterial counts was 10,600 CFU/mL (range: 360~59,200 CFU/mL) and coliform counts was 43 CFU/mL (range: 20~1,060 CFU/mL) in these samples. Whereas in the 118 breast pump-expressed samples, the mean of total bacterial counts was 20,200 CFU/mL (range: 240~492,000 CFU/mL) and coliform counts was 158 CFU/mL (range: 4~10,600 CFU/mL). There was no bacterial growth when the samples were incubated for 10 days at 4℃. We also compared total bacterial growth in colostrum and in matured human milk for 24 hr at 20℃ and 30℃. Although bacterial growth had not shown for 24 hr at 20℃, but shown slight growth in colostrum and rapidly increase in matured human milk for 24 hr at 30℃. The coliform bacteria in all samples, particulary in formula milk, had grown at 20℃ and 30℃.

        젖소초유 급여가 마우스의 성장과 사료섭취에 미치는 효과

        본 연구는 젖소 초유를 마우스에 급여후 성장과 사료섭취에 관하여 연구한 것이다. 초유구, 시유구, 대조구로 나누어 각 시험구당 10마리씩 나누어 실험하였다. 마우스의 사료섭취는 시험기간 2주 동안 대조구는 4.73 g, 시유구는 3.95 g, 초유구는 3.4.1g 이었다. 마우스의 시유 및 초유섭취는 시험기간 2주 동안 대조구는 0 g, 시유구는 9.93 g 섭취, 초유구는 10.17 g을 섭취하였다. 물섭취는 2주간 대조구가 6.28 g, 시유구가 4.74 g, 초유구가 4.67 g을 섭취하였다. 마우스의 증체율은 초유구가 대조구에 비해서 16.73%(P<0.05) 높았다. 마우스 혈액으로부터 조사한 엘러지와 관련된 사이토카인 IL-4는 초유에서 낮게 나타났다. 따라서 젖소 초유가 마우스의 증체율 및 사료 섭취에 큰 영향을 미치는 것으로 사료된다. This studies were carried out to assess the effects of feeding bovine colostrum on growth and feed intake of mouse. A total of 30 mice were divided into three groups(10 mice for each groups), and treatments(colostrum, city milk, or no milk as the control) were randomly assigned. Average daily intake of solid feed during the experimental period were 4.73, 3.95 and 3.41g for control, normal milk, and colostrum, respectively. Average daily intake of milk were 0, 9.93, and 10.17g for control, normal milk, and colostrum, respectively. Average daily water intake were 6.28, 4.743, and 4.67g for control, normal milk, and colostrum, respectively. The growth rate of the colostrum group was 16.7% higher compared to the control group (P<0.05). There were differences in the concentrations of IL-4 and IL-13 associated with allergy in mouse blood at colostrum and city milk feeding as compared with control.
