Interferon preparations, in addition to their antiviral properties, may inhibit cell growth and multiplication, enhance the expression of cell surface antigens and influence some functions of T-1ymphocytes, macrophages and natural killer cells. The study was performed in order to investigate the cytolytic effect, antiproli ferative effect and HLA antigen expression effect of the gamma-interferon(IFN- r ) on human melanoma cell line A-375. the cytolytic activities were checked by 'Cr release assay, the antiproliferative activities were analyzed by the 'H-thymidine uptake test and HLA antigen expressions were observed by the indirect imrnuno fluor esent method. The results were as follows : 1. The cytolytic effect of the peripheral rnononuclea.r cells treated with INF- on A 375 human melanoma cell line was increased to 31.4%, 36.5%, 33.9%, 53.9 %, 13.9% respectively in the experiment 1, 2, 3, 4, 5 as compared to control group (-11.4%, 3.6%, 18.5%, 35.5%, 6.7% respectively). 2. 1ri the dose response check of the peripheral mononuclear cells treated with INF r to A 375, the cytolytic effect was definitely observed at 200U/ml concentration of the INF r and maximal effect was observed at 500U/ml concentration. 3. The IIUF r treated natural killer cells did not show any significant increase in cytolytie activity as compared to that of untreated natural killer cell and the same results were obtained with monocytes. This cytolytic activity was significantly increased when INF-r treated natural killer cells were co-culturd with monoeytes. 4. The cyaolytjc activity of natural killer cells was increased at the same degree when using culture supernant of INF- treated monocytes as in the case of using

N/A Recently murine monoclonal antibodies have studied actively for radioimmuno-scintigraphy and radioinamunotherapy, especially on patients with leukemia and lymphoma. In this research, we studied radiolabeling and immunologic characteristics of two in-house anti-leukemic monoclonal antibodies(anti-CALLA & anti-J1-l antibodies) to make the basis tor their clinical application. Each antibody was radiolabeled successfully with 99mTc by pretargeting transchelation method and with 125I by Iodogen method. We also studied cell binding assay, Scatchard analysis and modulation phenomenon. 125I showed 90% labeling efficiency for each anti-body which was satisfactory, but 99mTc showed labeling efficiency below 70%, for which we need better labeling method, In cell binding assay, the immunoreactivity(IR) was low for 99mTc-labeled antibodies. Scatchard analysis showed satisfactory data for both binding affinity. The affinity constant and antibody binding sites per cell are around 109M-1 and 104, respectively. There was no modulation phenomenon in cases of 125I or 99mTc labeled antibodies. We expect that two anti-leukemic monoclonal antibodies may be useful in diagnosis and therapy for leukemia and lymphoma patients.

A clinicopathological study was made on 17 case with leukemia cutis diagnosed in a period of 10 years frorn 1980 to 1989 in Seoul Xational University I-lospital. The results were summarized as follows : 1. There were 4 cases with acute lyrnphocytic leukernia, 7 with acute myelocytir. leukemia (AML), 5 with chronic myelocytic leukemia (CML), 1 with eosinophilic leukemia. 2. The clinical appearance of leukemia cutis included nodules (88.2%), papules, macules, plaques and ulcerative lesions. Leukemia cutis dis not show any discernible clinical qppearance with each different type of leukemia. 3. Leukemia cutis showed a wide spectrum of histopathologic features, diffuse infiltration, patchy infiltration with linear infiltration between collagen bundles, perivascular and peridnexal involvement, perivascular involvement, and mainly subcutaneous tissue involvement. There was no distinctive histopathologic pattern with each different type of leukemia. 4. The demonstration of intracytoplasmic chloroacetate esterase and lysozyme was helpful to refine the diagnosis of leukemia cutis. (Kor J Dermatol 28(3): 321 330, 1990)
