        CD-ROM을 이용(利用)한 서지정보(書誌情報) 데이터베이스의 신뢰성향상(信賴性向上) 기법연구(技法硏究) - 다운로드와 업로드를 중심(中心)으로 -

        본연구(本硏究)는 레이저기술(技術)과 정보공학(情報工學)의 눈부신 발전과정(發展過程)에서 등장한 대용량 정보축적매체(情報蓄積媒體)인 CD-ROM Catalog Production Product 중에서 BiblioFile을 선택하고, CD-ROM BiblioFile과 과학기술대학도서관(料學技術大學圖書館)에서 자체 개발(開發)한 시스템인 KITLAS를 시스템구성(構成), 파일구조(構造), 특성(特性) 및 기능면(機能面)에서 비교분석(比較分析)한 후 CD-ROM에 수록(收錄)된 MARC 레코드의 변환(變換)과 온라인 대량, 동시 전송(傳送)하는 과정(過程)인 이른바 다운로드(download)와 업로드(up-load)하는 과정(過程)을 구현하여 서지정보(書誌情報) 데이터베이스의 신뢰성(信賴性) 향상(向上) 방법(方法)을 모색하였다. With regard to system configuration, file structure and function, this study has compared and analyzed KITLAS developed by KIT Library and CD-ROM BiblioFile, which is selected among CD-ROM Catalog Production Products, the mass storage media, produced by the fast-growing laser technology and information technology. Furthurmore, this study illuminates how to improve the reliability of bibliographic database, implementing downloading and uploading processes which make possible the conversion and online, mass transmission of CD-ROM MARC records at once.

      • 돼지 정액의 동결시 Taurine과 $\alpha$-Tocopherol 첨가가 동결$\cdot$융해 정자의 성상과 기능에 미치는 영향

        The present study evaluated whether an exogenous antioxidants, taurine and $\alpha$-tocopherol, could, when added to the freezing extender, improve the post-thaw sperm characteristics, function, the level of reactive oxygen species (ROS) generation, and the level of lipid peroxidation (LPO) in frozen-thawed porcine semen. CASA (computer-aided sperm analysis), HOST (hypoos-motic swelling test), chemiluminescence using luminol and lucigenin and the detection of malondialdehyde for LPO was performed in frozen-thawed porcine sper-matozoa. The results obtained in these studies are as follows. While no beneficial effects of taurine and $\alpha$-tocopherol supplementation were visible in motility, viability, acrosome reaction, tail swelling patterns, and the generation of $O^{2-}$ of frozen-thawed porcine sper-matozoa, $H_{2}O_{2}$ was decreased by all treatments except taurine 50mM treatment. In conclusion the taurine and $\alpha$-tocopherol treatments during freezing reduced generation of reactive oxygen species and production of malondialdehyde in frozen-thawed porcine semen, and the ROS savangers may minimize various damages of spermatozoa during freezing. 본 연구는 돼지 정자 동결시 taurine과 $\alpha$-tocopherol의 첨가가 융해후 정자 성상과 정자 기능 활성산소계(reactive oxygen species; ROS)의 발생 정도 및 지질 산화(lipid peroxidation, LPO)에 미치는 영향을 구명하기 위하여 시행하였다. 1차 및 2차 희석액내 taurine과 $\alpha$-tocopherol이 첨가된 돼지 동결정액의 융해 후 정자 운동성 양상, 정자 생존성, 정자 기법을 적용하여 다음과 같은 결과를 얻었다. Taurine(25mM, 50mM), $\alpha$-tocopherol($500{\mu}M,\;1,000{\mu}M$ 단일처리군 그리고 taurine과 $\alpha$-tocopherol의 혼합처리군($25mM\~500{\mu}M\~1,000{\mu}M$)은 동결$\cdot$융해 후 대조군과 비해 정자 운동성과 생존성은 유의적인 차이를 나타내지 않았다. Taurine과 $\alpha$-tocopherol의 혼합처리군 $50mM\~1,000{\mu}M$에서 HOST, 점체 반응이 대조군과 비교하여 유의차는 인정되지 않았으나 다소 증가하였다. 동결$\cdot$융해 정자의 ROS 발생 억제를 위한 taurine과 $\alpha$-tocopherol의 모든 처리군은 대조군과 비교하여$\cdot\{O_2}{^-}$ 발생을 유의적으로 완화시키지 못하였다. 그러나 taurine 단일처리군(25mM), $\alpha$-tocopherol 단일처리군($50mM,\;1,000{\mu}M$)과 혼합처리군($25mM\~500mM,\;50mM\~1,000{\mu}M$은 $H_{2}O_{2}$의 발생을 대조군과 비교하여 유의적으로 완화시켰다(P<0.05). 동결$\cdot$융해 정자의 malondialdehyde의 생산은 taurine과 $\alpha$-tocopherol의 모든 처리군에서 대조군과 비교하여 유의적인 감소를 보였다(P<0.05). 이상의 결과를 종합해 보면 돼지 정액의 동결보존시 taurine와 $\alpha$-tocopherol 같은 항산화제의 처리는 ROS 발생과 LPO을 효과적으로 완화시킴에 따라 돼지 정액의 동결보존 효율을 증진시킬 수 있는 유용한 방법이라 사료된다.

        소 미성숙 난포란의 체외성숙시 ${\beta}-Mercaptoethanol$의 첨가가 체외성숙, 체외수정 및 Glutathione 수준에 미치는 영향

        본 연구는 소의 미성숙 난포란의 체외성숙시 ${\beta}-mercaptoethanol({\beta}-ME)$의 첨가가 체외성숙, 체외수정 후 웅성전핵의 형성 및 세포질 내의 GSH 수준에 미치는 영향을 알아보고자 실시하였다. 체외성숙시 $25\;{\mu}M$과 $50\;{\mu}M$의 ${\beta}-ME$를 첨가한 경우 대조구에 비하여 성숙율이 증가하는 것으로 나타났으며(p<0.05), 모든 실험구에 있어서 12시간 체외성숙보다 24시간 체외성숙에서 높은 성숙율을 나타냈다(p<0.05). 체외수정 후 웅성전핵 형성에 있어서는 $25\;{\mu}M$와 $50\;{\mu}M$ 농도의 ${\beta}-ME$ 첨가구에서 대조구에 비하여 높게 나타났으나(p<0.05), $25\;{\mu}M$과 $50\;{\mu}M$ 농도구와의 유의적인 차이는 없었다. GSH의 수준은 체외성숙 후 $50\;{\mu}M$의 ${\beta}-ME$ 첨가구가 다른 처리구에 비교하여 높게 나타났으며(p<0.05), 체외수정 후 웅성전핵이 형성된 다음 세포질 내 GSH 수준 역시 $50\;{\mu}M$의 ${\beta}-ME$ 첨가구에서 가장 높은 결과를 나타냈다(p<0.05). Experiments were conducted to determine the effects of beta-mercaptoethanol(${\beta}-ME$) supplements to the maturation medium on in vitro fertilization(IVF) and intracellular glutathione(GSH) concentration. Bovine cumulus-intact oocytes were matured in TCM-199 medium containing FBS, hormonal supplements, and ${\beta}-ME$(0, 25 and $50\;{\mu}M$) for 12h and 24 h. After culture, cumulus-free matured oocytes were co-incubated with frozen-thawed spermatozoa for 24h. Maturation rate increased(p<0.05) in ${\beta}-ME$ treatment group, but no significant differences among treatment groups. Also, increases(p<0.05) in intracellular GSH concentration before and after fertilization were observed in $50\;{\mu}M\;{\beta}-ME$ supplements to the maturation medium. Male pronuclear formations after IVF was increased(p<0.05) in ${\beta}-ME$ treatment group, but no significant difference among treatment groups. In conclusion, supplementing ${\beta}-ME$ into the maturation medium increased maturation rates, fertilization rates, and intracellular GSH concentrations.

        한우의 조정기능발달에 관한 연구 1 . 한우와 교잡종의 전반기 고환발육과 조정기능

        In order to study on the development of the spermatogenic function and the sexual maturity between the Korean purebred and three-way crossbred ((Angus × Korean purebred) × Holstein), this experiment was conducted to investigate the testicular development and the spermatogenetic function by means of repeating biopsy at one month intervals from one to 9 months of age with 6 Korean purebreds and 3 three-way crossbreds. 1. In the testicular development, the scrotum circumference of the Korean purebred and three-way crossbred increased steadily with increasing age. The scrotum circumferences of the Korean purebred and three-way crossbred were 19.4 ㎝ and 28.9 ㎝ at 9 months of age, respectively, and doubled to one month of age. The testicular width and length increased rapidly from 6-7 months of age, but the differences between two groups were not statistically significant. 2. The seminiferous tubules in the testis of two groups, solid at 3 months of age, increased appearently in diameter at 5 to 6 months of age, but were not reached mature size. 3. Only gonocytes and indifferent supporting cells were present in the tubules at 3 months of age and spermatogonia appeared from 5 months of age, spermatocytes increased rapidly from 6 months of age and spermatid were seen for the first time in the 7 months old Korean purebred and three-way crossbred. On the basis of the above results it is concluded that sexual maturity of three-way cross bulls was somewhat earlier than the Korean bulls, but the difference of two groups was not significant.

        Chlorpromazine activates p21Waf1/Cip1 gene transcription via early growth response-1 (Egr-1) in C6 glioma cells

        2-Chloro-10-[3(-dimethylamino)propyl]phenothiazinemonohydrochloride (chlorpromazine) is a phenothiazine derivative used clinically to control psychotic disorders. It also exhibits an anticancer activity. Treatment with chlorpromazine (CPZ) results in cell-cycle arrest at the G2/M phase in rat C6 glioma cells. CPZ reduces the expression of cell cycle-related proteins, such as cyclin D1, cyclin A, and cyclin B1, but causes an increase in the p21Waf1/Cip1 level. The molecular mechanism by which CPZ regulates p21Waf1/Cip1 expression is unknown. Here, we provide evidence that CPZ activates the p21Waf1/Cip1 gene promoter via induction of the transcription factor early growth response-1 (Egr-1) independently of p53 in C6 cells. A point mutation in the Egr-1-binding motif within the p21Waf1/Cip1 promoter abrogated promoter inducibility due to CPZ. Forced expression of Egr-1 enhanced p21Waf1/Cip1 promoter activity. In contrast, knockdown of endogenous Egr-1 by small interference RNA attenuated CPZ-induced p21Waf1/Cip1 promoter activity. A chromatin immunoprecipitation assay demonstrated that Egr-1 binds to the p21Waf1/Cip1 gene promoter. Further analysis showed that the ERK and JNK MAP kinases are required for induction of Egr-1 by CPZ. Finally, stable silencing of Egr-1 expression lead to attenuated CPZ-inducible p21Waf1/Cip1 expression and inhibited G2/M phase cell-cycle arrest. These results demonstrate that a functional link between ERK and JNK MAP kinase pathways and p21Waf1/Cip1 induction via Egr-1 contributes to CPZ-induced anticancer activity in C6 glioma cells.

        소 정자에 있어서 활성산소계가 정자 기능과 지방산화 및 DNA 절편화에 미치는 영향

        Objective : To evaluate the effects of the reactive oxygen species (ROS) generated with a xanthine (X) and xanthine oxidase (XO) system on sperm function, the change of sperm characteristics, lipid peroxidation, and DNA fragmentation in bovine spermatozoa. Materials and Methods: ROS were produced using a combination of 1000 uM X and 50 mU/ml XO. The ROS scavengers: superoxide dismu tase (SOD) (200 U/ml) and catalase (500 U/ml) were also tested. Spermatozoa were incubated for 2 hours in BWW medium with a combination of X-XO supplemented with or without ROS scavengers at $37^{circ}C$ under 5% $CO_2$ incubator. Sperm movement characteristics by CASA (computer-aided sperm analysis), HOST (hypoosmotic swelling test), Caionophore induced acrosome reaction, malondialdehyde formation for the analysis of lipid peroxidation, the percentage of DNA fragmentation using the method of TdT-mediated nick end labelling (TUNEL) by flow cytometry were determined after 2 hours incubation. Results: The action of ROS on bovine spermatozoa resulted in a decreased in capacity for sperm motility, Ca-ionophore induced acrosome reaction and membrane integrity, an increased in malondialdehyde formation and the percentage of sperm with DNA fragmentation. In the effects of antioxidant, catalase completely alleviated the toxic effects induced by the ROS in terms of sperm function and characteristics, however SOD exhibited no capacity to reduce the toxic effects. Conclusion: The ROS can induce significant damages to sperm functions and characteristics. The useful ROS scavengers can minimized the defects of sperm function and various damages of spermatozoa.

        성장중인 수흰쥐의 거세가 혈청LH 와 FSH수준 , 체중 및 정낭중량에 미치는 영향

        The purpose of this experiment was to investigate the effects of castration on serum LH and FSH level, body weight and seminal vesicle weight in growing male rats. The animals used in this experiment were 60 male rats of Wister Imamichi strain weighing 93.03±4.02g at 42 days of age. Five rats from each group were randomly sacrificed on the day of castration and at 1st, 2nd, 3rd, 4th and 5th week after castration. Seurm eras separated from the blood which was obtained by cardiac puncture and the serum was assayed for LH and FSH level by radioimmunoassay. Changes of the body weights were measured before sacrificing. Seminal vesicles were removed after obtaining blood and the weights were measured. The results which were obtained in this experiment were as follows: 1. Serum LH levels in castrated groups were increased gradually after castration and after 2 weeks of castration the levels were increased significantly (P$lt;0.05) as compared with the intact control groups. After 3 weeks of castration the serum LH level began to decrease and then maintained the same level to 5 weeks after castration. However, the serum LH level in the castrated groups were higher than those of the intact control groups at all periods. 2. Serum FSH levels in castrated groups were greatly increased after 1 week of castration. But the level began to decrease after 2 weeks of castration and got back to the level of the day of castration after 5 weeks of castration. The serum FSH levels in the castrated groups were higher than those of the intact control groups at all periods. 3. There were no differences in the both groups at 2 weeks of castration in body weights of growing male rats. From 3 weeks to 5 weeks after castration the body weights of the castrated groups were increased less than those of the intact control groups. 4. The weights of seminal vesicles in the castrated groups began to decreased from the first week of castration. After 4 weeks of castration the weights of seminal vesicles were significantly decreased (P$lt;0.05) and at 5th week after castration the weight of seminal vesicles were significantly decreased in the castrated group (P$lt;0.01) as compared with intact control group.

        돼지의 조정기능발달에 관한 연구

        In order to study on the spermatogenic function and the sexual maturity in male three-way crossbred ((Berkshire × Hampshire) × Landrace), this experiment was conducted to investigate the testicular development and the spermatogenic function by means of repeating biopsy testes at one month internals from one to 4 months of age and 15 days intervals from 4 to 6 months of age and the quality of semen collected after 27 weeks of age. 1. In the testicular development, the scrotum circumference increased steadily with increasing age up to 3 months of age, but increased rapidly after 4 months of age. The correlation between body weights, testicular lengths, testicular widths, and scrotum circumferences were statistically highly significant (p$lt;0.01). 2. The seminiferous tubules increased steadily at 3 months of age, increased apparently in diameter at 4 to 6 months of age. The percentage of intratubular tissue in the testis increased apparently at 4 to 6 months of age. 3. Only indifferent supporting cells, spermatogonia and spermatocytes were present in the seminiferous tubules at 3 months of age. Spermatids were appeared from 4 months of age, the percentage of which increased rapidly up to 5.5 months of age. 4. The first ejaculation of spermatozoa were observed in 1 of the 4 boars at 27 weeks of age, in 2 of the 4 boars at 29 weeks and in all of the 4 boars at 30 weeks. 5. The total volume ejaculated, sperm concentration and percentage of alive spermatozoa per ejaculate averaged 196.7㎖, 284.77x 10^6/㎖, and 80.0, respectively, at 35 weeks of age. On the basis of the above results it is concluded that three-way crossbred boars reached puberty at 27 seeks of age and can be used for breeding at about 35 weeks of age.
