Pectobacterium carotovorum subsp. carotovorum (PCC)는 세균성 무름병균으로 주로 감자, 양배추 등의 식물에서 질병을 일으킨다. 본 연구에서는 현장에서 신속하게 진단하기 위해 loop-mediated isothermal amplification법을 이용하여 1시간 내에 등온에서 검출 가능한 진단법을 개발하였으며, 이를 PCC-LAMP법이라 명명하였다. PCC의 lytic murein transglycolase 유전자를 특이적으로 증폭시키는 4개의 프라이머를 제작하였으며 최적 온도가 $61^{\circ}C$임을 확인하고 최적 조건을 확립하였다. 최적 조건을 바탕으로 4개의 프라이머가 $1{\times}10^3$ copies까지 검출하는 민감성을 확인할 수 있었다. 본 연구에서 개발된 PCC-LAMP법은 특이성 검사를 통해 PCC만이 특이적으로 검출됨을 확인하였으며, 이는 실제 시료에서도 적용 가능함을 확인하였다. PCC-LAMP법을 통하여 PCC를 신속하고 정확하게 검출함으로써 현장에서 유용하게 적용될 수 있을 것으로 사료된다. Pectobacterium carotovorum subsp. carotovorum is the causative agent of soft rot in crops such as potato and cabbages. Loop-mediated isothermal amplification (LAMP) is a simple DNA amplification method, as well as isothermal PCR technique. In this study, a new method for the rapid detection of Pectobacterium carotovorum subsp. carotovorum was developed using LAMP that named PCC-LAMP. Based on lytic murein transglycolase gene of Pectobacterium carotovorum subsp. carotovorum, a set of four primers for LAMP was designed. The optimal PCC-LAMP reaction temperature was established at $61^{\circ}C$. Under standard conditions, PCC-LAMP amplified $1{\times}10^3$ copies of clone PCC-pBX437 per reaction. Further, this method can also assay directly by SYBR Green I without electrophoresis. Amplification was not detected for five other bacterial species. In conclusion, PCC-LAMP may be a useful method for the detection Pectobacterium carotovorum subsp. carotovorum in the field.

노송당(老松堂) 송희경(宋希璟)의 사행시(使行詩) 연구(硏究)

본 논문은 老松堂宋希璟의 생애와 그의 使行詩를 다루었다. 그는 回禮使로 일본을 使行하면서 일본 사회의 생활과 풍속, 문화 등 여러 모습을 詩로 표현하였으며, 주로 객관적 묘사를 통한 사실 전달에 치중하여 보다 진솔하고 현실적인 느낌을 갖게 하는 등 후대의 일본 기행문과는 또 다른 면모를 보였다. 宋希璟은 使行중 일왕의 부당한 요구와 회유에도 交隣의 道를 강조하고 절의를 굽히지 않는 등 강직한 성품을 보였다. 일본인 高僧을 비롯하여 그가 교유했던 지인들과 나눈많은 詩작품, 그리고 일본 견문을 기록한 使行詩에서도 진솔한 인간적 모습을 보여주고 있다. 宋希璟의 使行詩는 그의 소박한 성품처럼 꾸밈과 화려함이 없는 간결하고 淡淡한 표현으로 그 여운이 깊다. 특히 儒者로서의 편견을 露呈하지 않고, 있는 그대로의 현실을 사실감 있게 표현하고 있는 점에서 그의 使行詩가 갖는 의미는 더욱 크다 할 수 있다. This a paper has been considered about Nosongdang, Song Hui-gyeong (1376-1446)’s life and Envoy poetry. 『Ilbonhaengrok』, there were also records of reporting schedule to the ex-king, King Taejong on the next day. Through this record of visit to Japan as an envoy, we can see the rising gentries` recognition of Japan and all the circumstance related to such visits directly after establishment of the Chosun. 『Ilbonhaengrok』 have on envoy poetry. Song Hui-gyeong’s visit to Japan as an envoy was made under an anxious and critical situation. There was fear of ‘political retaliation’ because it was the next year of an expedition to the Daemado and there were also anxious elements such as the ‘rough and difficult sea’ and ‘pirates’. However, in expressing his experience of visit to Japan in poems, he showed an attitude to ‘record all things he could contact by means of his ears and eyes without thinking of vulgarness’. While visiting to Japan on relations with neighboring countries. he found a strange quirk Japanese society and suggested countermeasures thereof, but he didn’t despise Japan. he recognized Japanese as an existence having same ‘disposition’ as the Chosun people. Because of such reasons, he tried to keep an objective view as the 3rd party of Japanese society and customs being different from Chosun’s. his poetry is characterized by abundant use of sensuous literature flavor.

본고는 향후 한국의 자치 단체장 선거에서 어떠한 경력의 소유자가 당선되는 것이 바람직할 것인가를 전망해 보는데 있어 일본의 사례를 살펴보고자 하였다. 특히 광역단체장인 지사의 경력과 정치성향에 초점을 맞추어, 정치 환경의 변화와 시대의 흐름 속에서 지사 선거 입후보자와 당선자들의 출신경력을 검토해 봄으로써, 금후 한국의 지방자치 발전과정에서 중추적 역할을 담당할 지사상(知事像)을 그려보고자 하였다. 전후 일본에서 지방자치가 실시된 이래 보수계 자민당 정권이 유지되어온 1990년대 초까지의 지사후보자와 당선자들의 경력을 분석해 보면, 국정 레벨에서 ``자유당과 민주당의 경쟁시기``였던 1950년대 중반까지는 과거 특권적 지위를 누렸던 관료출신이 급감하고, 그 대신에 정치가와 공직미경험자의 진출이 뚜렷하였다. 그 후 자민당의 결당으로 1960년대 중반까지의 ``자민당1당우위의 시기``에는 보수계 후보의 단일화가 가능해지면서 ``보수·혁신 연합형 후보``는 감소하고, 보수정당 단일후보가 증가하였다. 이 시기에 자민당은 국회의원을 지사후보로 적극 공천하여 당선시켰다. 1960년대 후반부터 1970년대 말까지의 ``보수·혁신 백중의 시기``에는 고도경제성장이 둔화되면서 지방재정의 건전화와 자치체의 내부관리가 중요시되어, 지역에서의 근무경험이 있는 지방관료 출신 후보가 지사 선거전에 본격적으로 진출한다. 동시에 이시기에는 보수·혁신 백중의 중앙정국 구도 때문에 국회의석수를 고려하여 여·야 모두가 현직 국회의원을 지사후보로 공천하기가 쉽지 않았다. 1980년대와 1990년대 초는 ``보수·중도 연합``의 중앙정국구도 아래 국가재정의 재건과 환경, 복지문제가 최우선 과제로 등장한 시기이다. 이 시기의 단체장들은 지방정치에 중앙정치의 ``보수·혁신 대립`` 구도를 끌어들이는 것은 옳지 않다고 주장하였다. 보수·혁신간의 정책대결이 이완되자 ``여·야 공동추천후보`` 형태가 많아졌고, 후보로는 관료 출신자, 특히 지방 관료가 선호되었는데, 여기에는 지방화시대가 주창되면서 지사후보의 선발과정에 있어 지방의 영향력이 크게 증대하고, 지사후보자의 기준으로 ``지역에서의 평판``을 중요시 하는 사회적 분위기가 존재하였다. 지사후보의 경력이 지역주민의 요구와 평판을 중시하는 쪽으로 변화하게 되었다는 것은 지사후보의 공천에 있어 지역의 주도권이 그만큼 강화되었다는 점을 의미한다. 특히 각 정당의 지방조직의 정당화가 진행되면서 지방정당조직을 중심으로 지역이익의 대변자로서의 지사의 역할에 대한 기대는 한층 높아지고 있다. 이와 같은 추세는 1990년대 후반은 물론, 최근의 지사 선거에서도 계속되는 현상으로 ``지사 선거에 있어서의 지방화 시대``는 당분간 유지 될 것으로 보인다. The paper is to analyze Japan`s cases with respect to predicting what kind of political background of local politicians is more desirable in the Korea`s local autonomy election of local governments in the future. The paper focuses on the desirable characteristics of Korea`s provincial governors who will play an important role in the process of Korea`s local autonomy development in the future by especially investigating the career and background of local autonomy candidates and elected candidates of provincial governors who face the changes of political environment and time_series trend. First of all, the most prominent characteristics of Japan`s prefectural governors` election is that the influential power of regions has increased gradually with respect to the selection of local autonomy candidates. It seems that central parties decide prefectural governor`s candidates regardless of ruling party and opposition party. But in fact, the federation of prefectural branches playes a key role in deciding candidate. Especially the expectation for the role of prefectural governors as a representative for the interest of regions has risen considerably in the local organizations of central parties, as the normalization of the local organizations of each of the central parties has proceeded.

This study examined how online gamers were divided into different segments in the Korean online game industry. In addition, this study attempted to understand their wants in order to get some ideas on how a game can be developed. Online gamers were found to be divided into three major market segments, and each represented different lifestyles and was related to hardcore vs. casual players and male vs. female players. We found that compared to non-gamers, gamers are more outgoing, active and fashion- oriented, which is contrary to our common-knowledge. We also found that gamers were divided into `family-oriented type`. `social-leader type` and `active-outgoing type`, according to their lifestyle characteristics. In general, gamers` loyalty toward a game(i.e., game loyalty) were influenced by three factors such as flow factor, social interaction factors, and promotion factor. One of the interesting findings was that `family-oriented type considered sevice factor as the most important while `active-outgoing type` regarded factors such as price, service, and pioneering are more important than others. We provided some managerial implications for online game developers and marketers on how to make games to be successful.
