Chitin의 탈아세틸화로 얻어지는 화합물인 chitosan과 chitooligosaccharide는 다양한 생리활성 기능을 나타내므로 생물의학적인 응용에 많은 관심이 집중되고 있다. 본 연구는 수용성 chitooligosaccharide가 생쥐의 간 기능에 미치는 영향을 조사하기 위하여 수행되었다. 6주 동안 3% cholesterol를 식이한 생쥐에서는 혈장 cholesterol 함량이 증가하였으나, 1% chitooligosaccharide와 3% cholesterol을 함께 식이한 생쥐에서는 혈장 cholesterol 함량이 23% 정도 감소하였다. 그러나 혈장내 HDL-cholesterol과 간내 cholesterol 함량에는 유의한 차이가 없었다. 한편, 15% ethanol을 8주간 투여한 생쥐에서는 inflammation, necrosis, 지방구의 축적, GPT의 활성 증가 등 간 상해가 관찰되었다. 그와 반면에 0.5% chitooligosaccharide와 15% ethanol을 함께 투여한 생쥐에서는 ethanol에 의해 유발된 간 상해가 현저히 억제되어 지방구의 축적이 관찰되지 않았고, 혈장의 GPT 활성이 25%정도 감소하였다. 따라서, chitooligosaccharide가 생쥐에서 혈장 cholesterol 함량을 감소시키고, 알코올성 지방간을 예방하는 등 부분적인 기능이 있음을 보여주었다. Chitosan is the deacetylated product of chitin. Chitosan and its derivatives have many properties that make them attractive for a wide variety of health applications. This study was performed to investigate some effects of water soluble chitooligosaccharides on liver function in the mouse. The animals given the sterol diet containing 3% cholesterol for 6 weeks showed increase in plasma cholesterol level, which were lowered by 23% when they were fed on 1% chotooligosaccharide. While there was no significant change in liver cholesterol and plasma HDL-cholesterol levels. Continuous administration of 15% ethanol via drinking water to mice for 8 weeks elicited pathological alterations such as inflammation, necrosis, accumulation of lipid droplets in the liver and increase in GPT activity, while simultaneous administration of ethanol and chitooligosaccharide prevented remarkedly ethanol-induced liver injury; there was no observable lipid droplet and GPT activity was decreased by 25% in the liver. These results suggest that chitooligosaccharide play some roles in liver function, such as reducing the plasma cholesterol level and preventing alcoholic liver disease.

본 연구에서는 제주도 자생 감귤 17종에 대하여 탈색소효과(depigment effect)를 연구하였다. B16 멜라닌 생성세포(B16 melanoma cell)를 이용하여 tyrosinase 및 멜라닌 생성억제 효과를 연구한 결과 대조군 대비하여 농도 의존적으로 감소하는 3종의 미성숙 온주밀감(immature Citrus unshue), 팔삭(Citrus hassaku), 진지향(Citrus sinensis x reticulata)을 도출하였다. 또한, 이와 같이 도출된 감귤 추출물을 이용하여 멜라닌 세포 생성(melanogenesis)에 영향을 미치는 tyrosinase, DOPAchrome tauto-merase (TRP-2)와 DHICA oxidase (TRP-1) 단백질 발현을 실험한 결과 tyrosinase, TRP-1, TRP-2 단백질 발현이 감소함을 나타내었다. 특히 이들 중 미성숙 온주밀감은 tyrosinase와 TRP-1 단백질 발현에서 유의적으로 감소하는 결과를 보였다. 본 연구 결과로써 미성숙 온주밀감은 모든 실험에서 우수한 효과를 나타내어 UV에 의해서 야기되는 피부 색소 침착을 방지할 수 있는 물질로서 기대할 수 있다. We examined the depigmentation effect on Korean traditional citrus 17 species. With B16 melanoma cells, we have seen inhibition of the tyrosinase and melanin formation, which eventually were dose dependently decreased by three citrus fruits, immature Citrus unshiu, Citrus hassaku, and Citrus sinensis × reticulata as compared with positive control. Also, we examined expression of tyrosinase, DOPAchrome tautomerase (TRP-2), and DHICA oxidase (TRP-1) which affect melanin synthesis. Especially, immature Citrus unshiu decreased the protein levels of tyrosinase and TRP-1. In conclusion, immature Citrus unshue showed the strongest activity in all the experiments mentioned above and we expect that it can be used for preventing UV-induced pigmentation.

Effects of hot-water extract from Sasa quelpaertensis leaf (HWES) on melanogenesis were investigated in B16/F10 mouse melanoma cells. HWES inhibited cellular tyrosinase activity and melanin biosynthesis in a dose-dependent manner. Western blotting analysis showed that HWES dose-dependently inhibited tyrosinase and tyrosinase related protein-1 expression. Also, HWES suppressed sustained ERK activation in a concentration-dependent manner, suggesting that HWES inhibits the melanin biosynthesis through the suppressive effect against pathway involving sustained ERK activation.
제주도, 한반도 및 대만 내 팔색조의 생태적 서식특성 분석을 위한 유전적 접근

A Fairy Pitta is a bird known to breed only in mainland China, Taiwan, Japan and Korea and is listed as Vulnerable in the IUCN Red List. We carried out a DNA analysis to contribute to conserve the genetic diversity of Fairy Pitta. 32 samples were collected at Jeju Island, the Korean Peninsula and Taiwan from 2004 to 2013 and DNA was extracted from them and several sequences were amplified it through PCR. And then we performed the population genetic analysis. We found there was a transversion between nucleotide sequences at CO1 gene, while there was no changes at Cyt-b gene. And we confirmed the polymorphism from two genes was caused from genetic drift not from selection. Through this analysis, the group within the Peninsula was found bigger than other two groups based on the analysis of CO1 gene, and the group from Taiwan was found bigger than other two groups through the analysis of Cyt-b gene. The population genetic structure of mitochondria gene of three group was showing CO1 gene had 5 haplotypes and Cyt-b gene had 6 haplotypes. Haplotype 2 in CO1 gene was found in three group and many individuals of samples had this haplotype. Like CO1 gene, haplotype 2 in Cyt-b gene was found in three group and was included in plenty of individuals. Other haplotypes were not overlaped and broke off among the three groups. To prevent from the extinction of Fairy Pitta and to obtain the genetic diversity, we need to compare with other regional group such as Japan, China and perform additional research in the non-breeding area.
김치에서 분리한 Lactococcus lactis W-44에 의한 감귤발효물의 양식 넙치 성장에 미치는 영향

젖산균을 접종하여 감귤 분쇄액을 발효시켜 얻은 감귤발효물의 어류양식 사료 첨가제로의 가능성을 넙치(Paralichthys olivaceus) 양식에서 검토하였다. 김치에서 항균활성이 우수한 젖산균 W-44를 분리한 후 16S rDNA 염기서열 분식을 통하여 Lactococcus lactis W-44로 동정하였다. L. lactis W-44를 이용하여 감귤 분쇄액 발효를 수행한 결과, 생리활성이 우수한 무배당체 flavonoids인 naringenin, hesperitin의 함량이 각각 약 10과 6배 증가함을 확인하였다. L. lactis W-44를 이용하여 발효된 감귤발효액을 양식넙치의 사료첨가제로 투여한 결과, 감귤발효액을 투여하지 않은 대조구의 평균 전장 및 체중 증가율과 현격한 차이가 있었다. 또한 0.2% (v/v)의 감귤발효액을 투여한 실험군에서 넙치의 성장이 가장 우수한 것으로 나타났다. 이러한 결과로부터 젖산균의 감귤발효산물을 넙치 양식에서 기능성 사료첨가제로 사용할 수 있는 가능성을 확인하였다. We evaluated the use of citrus fruit fermented by lactic acid bacteria, as a feed supplement for flounder (Paralichthys olivaceus) cultivation. For the fermentation, a lactic acid bacterial strain W-44 showing antibacterial activity was isolated from kimchi. From the phylogenetic analysis based on, 16S rDNA sequence, the strain W-44 was identified as Lactococcus lactis. After the fermentation of citrus fruit with L. lactis W-44, the contents of naringenin and hesperetin, bioactive flavonoid aglycones, were increased about ten-fold and six-fold, respectively. The effects of fermented citrus fruit-based feed additives (CFBFA) were tested on the growth of flounder, Paralichthys olivaceus. There were significant differences in average total length and body weight between the experimental and control group. The growth rate of the experimental group fed with the 0.2% CFBFA-supplemented diet was increased 4.5% and 20.9% more than the control group in total length and body weight, respectively. These results suggest that the fermented citrus fruit could be used as a functional feed additive for flounder cultivation.
DNA 분석에 의한 팔색조의 암수 구분 및 암수별 피해 현황 그리고 크기 차이에 관한 연구

The differentiation of sex is important for species preservation. However, Fairy Pitta is sexually monomorphic and sex of an individual is indistinguishable with its external characteristics. We determined the sex of Fairy Pitta through DNA analysis and investigated the causes and time of injury and mortality and the size based on sex. We collected 21 samples at Jeju Island, Korean Peninsula from 2004 to 2013 and extracted DNA from them and amplified chromo helicase DNA-binding gene from Z and W chromosomes through Polymerase Chain Reaction (PCR). We confirmed their sex with the banding pattern through Agarose gel electrophoresis, i.e. male (ZZ): one banded and female (ZW) two banded. We distinguished the sex of 17 of 21 samples resulting in 9 males and 8 females. Most casualties were recorded in adult of both sexes. Causes of injury and mortality proved that female casualties occurred from window strikes, dehydration, car accident, predation by natural enemies, and male occurred from window strikes, car accident and dehydration. The time of injury and mortality in adults differ by sex. There was no difference between sexes in any of the six size parameters. As the time of injury and mortality differ by sex, the survey on the role and ecological nature by sex in breeding season must be carried out in the future. External measurements may not be reliable for sexing of Fairy Pitta and other traits such as vocal or characteristics are required to identify the sex of individuals in the field.

누리장나무의 잎과 줄기는 예로부터 항고혈압, 관절염, 류마티스 관절염, 항염증 등을 치료하는 민간약재로 사용되어 왔다. 본 연구에서는 누리장나무의 잎 추출물과 그 분획물의 항산화 및 항염증 소재로서의 활성가능성을 확인하기 위하여 수행되었다. 누리장나무 잎의 ethanol (EtOH)로 추출물과 이 추출물을 분획하여 얻은 hexane, chloroform (CHCl3), ethyl acetate (EtOAc), n-butanol (BuOH) 분획물들의 총 폴리페놀 함량을 비교한 결과, 총 폴리페놀 함량은 EtOAc 분획물(78.08 μg/mg)과 BuOH 분획물(77.54 μg/mg)에서 높게 측정되었다. 각 분획물의 항산화 활성을 비교한 결과, 폴리페놀 함량이 높은 EtOAc 분획물과 BuOH 분획물에서 높은 DPPH (1,1-diphenyl-2-picrylhydrazyl) 및 ABTS (2,2’-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid)) radical 소거활성을 나타내었다. 그리고 xanthine oxidase 저해활성은 CHCl3 분획물(IC50 = 4.43 μg/ml)과 EtOAc 분획물(IC50 = 5.69 μg/ml)에서 높게 나타났다. EtOAc 분획물은 RAW 264.7 세포에서 lipopolysaccharide (LPS)로 유도된 nitric oxide (NO)의 생성을 효과적으로 억제하였다(IC50 = 18.87 μg/ml). 또한, EtOAc 분획물(100 μg/ml)은 LPS로 지극한 RAW 264.7 세포에서 tumor necrosis factor α (TNF-α)와 interleukin-6 (IL-6)의 생성과 inducible nitric oxide synthase (iNOS)와 cyclooxygenase-2 (COX-2) 단백질의 발현을 효과적으로 억제하였다. 본 연구결과는 누리장나무 잎 추출물과 분획물이 항산화 및 항염증 소재로서의 활용가능성이 있음을 제시해 준다. Clerodendrum trichotomum (CT) leaves and stems have been used in folk medicine for their anti-hypertension, arthritis, rheumatism, and anti-inflammatory properties. This study was performed to evaluate the potential of CT as an anti-oxidant and anti-inflammatory agent. CT leaves were extracted using 70% ethanol (EtOH). Then, using this extract, a hexane, chloroform (CHCl3), ethyl acetate (EtOAc), and n-butanol (BuOH) fraction was prepared. The polyphenol contents were higher in the EtOAc fraction (78.08 μg/mg) and BuOH fraction (77.54 μg/mg) compared to the other fractions. Also, these two fractions exhibited strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2’-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activities. Xanthine oxidase inhibitory activities were higher in the CHCl3 fraction (IC50 = 4.43 μg/ml) and EtOAc fraction (IC50 = 5.69 μg/ml). Moreover, the EtOAc fraction effectively inhibited nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells (IC50 = 18.87 μg/ml). Thus, we investigated the effects of the EtOAc fraction on the expression of pro-inflammatory cytokines, inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) in LPS-stimulated RAW 264.7 cells. The treatment of the EtOAc fraction (100 μg/ml) effectively decreased the levels of the tumor necrosis factor α (TNF-α) and interleukin-6 (IL-6), and the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). These results suggest the potential for CT extract and fractions as promising anti-oxidant and anti-inflammatory agents.
버섯균사체 배양기질로서의 손바닥선인장의 활용과 그 배양추출물의 기능적 특성

본 연구는 버섯균사체의 배양 기질로서 손바닥선인장 열매의 활용 가능성을 타진하기 위하여 수행하였다. 5 종류(신령, 잎새, 노루궁뎅이, 차가, 상황)의 버섯균사체를 MEB배지와 손바닥선인장 열매 액체배지(CB)에서 각각 배양하여 에틸아세테이트로 추출한 후, 각 추출물의 수율, 총 폴리페놀 함량, 그리고 몇몇 생리활성을 분석하여 비교하였다. CB 배지에서 배양한 버섯 균사체 중에 잎새버섯 균사체가 추출 수율(0.4 g/l)과 총 폴리페놀 함량(22.7%)에서 가장 양호하였다. CB 배지에서 배양한 버섯 균사체 추출물들은 항산화 활성(DPPH 자유기 소거활성, superoxide의 소거활성, xanthine oxidase 억제 활성), LPS로 활성화시킨 RAW264.7세포에서의 NO 생성 억제 활성, 항균활성, 그리고 HL-60에서의 세포증식 억제 활성 모두가 MEB 배지에서 배양한 버섯균사체 추출물의 것과 비교해 현저하게 증진되어 있음을 확인할 수 있었다. 특히, 잎새버섯 발효추출물에서 DPPH 자유기 소거활성($IC_{50}=362.9{\mu}g/ml$), xanthine oxidase 저해활성(80% at $500{\mu}g/ml$), superoxide 소거활성(80% at $500{\mu}g/ml$), NO 생성 억제 활성($IC_{50}=43.1{\mu}g/ml$), 항균활성, 그리고 HL-60 세포 증식 억제 활성 등 분석된 모든 생리활성이 가장 높게 나타났다. 본 연구 결과는 손바닥선인장 열매가 버섯균사체의 기능성 증진을 위한 배양기질로서 활용될 수 있음을 시사하였다. This study was performed to know the potentialities of the fruits of Opuntia ficus-indica, as a medium for mushroom mycelial culture. Five mushroom mycelial (Agrocus blazei, Grifola frondosa, Hericium erinaceum, Innonotus obliquus, Phellinus linteus) were frown on the malt extract broth (MEB) and the cactus broth medium (CB). The submerged culture mixtures were extracted using equal volume of ethyl acetate, and their extract yields, total polyphenol contents, and some physiological activities were compared with each other Each extract from mycelial culture grown on CB medium showed remarkable enhancement in physiological activities compared with each counterpart grown on MEB. Among five mycelial cultures grown on CB medium, the extract yield and polyphenyl content were highest in the extract from Grifola frondosa (extract yield, 0.4 g/L and polyphenol content, 22.7%). Also, the extracts from Grifola frondosa showed the highest physiological activities, such as DPPH radical scavenging ($IC_{50}=362.9{\mu}g/ml$), xanthine oxidase inhibition (about 80% at $500{\mu}g/ml$), and superoxide radical scavenging (about 80% at $500{\mu}g/ml$), and NO production inhibition ($IC_{50}=43.1{\mu}g/ml$) in LPS-stimulated RAW 264.7 cells. This result suggests that the fruit of Opuntia ficus-indica can be used as a culture medium for improving the functional properties of various mushroom mycelia.
