        Soluble Epoxide Hydrolase Inhibitory and Anti-inflammatory Components from the Leaves of <i>Eucommia ulmoides</i> Oliver (Duzhong)

        <P><I>Eucommia ulmoides</I> leaves have been used as a functional food and drink in China. The purpose of this study was to identify the bioactive constituents with soluble epoxide hydrolase (sEH) inhibitory activity and anti-inflammatory properties. Twenty-seven known compounds (<B>1</B>–<B>27</B>) were isolated from the leaves of <I>E. ulmoides</I> Oliver, and their structures were identified by NMR and ESIMS analysis; three of these, 2,5-dimethoxy-3-glucopyranosyl cinnamic alcohol (<B>11</B>), foliasalacioside E<SUB>2</SUB> (<B>26</B>), and icariside F<SUB>2</SUB> (<B>27</B>), were obtained from this plant for the first time. Compounds <B>1</B>–<B>7</B> exhibited soluble epoxide hydrolase (sEH) inhibitory activity at 100 μM; among them, quercetin (<B>1</B>) and kaempferol (<B>5</B>) displayed potential activities with IC<SUB>50</SUB> values of 22.5 ± 0.9 and 31.3 ± 2.6 μM, respectively, with noncompetitive inhibition mode. Nuclear factor kappa B (NF-κB) inhibitory activity of the isolated compounds was evaluated by the NF-κB liciferase assay in HepG2 cells. Compounds <B>1</B>, <B>9</B>, <B>20</B>, and <B>27</B> displayed potent NF-κB inhibitory effects, with IC<SUB>50</SUB> values of 15.14 ± 2.29, 15.23 ± 2.34, 16.88 ± 2.17, and 16.25 ± 2.19 μM, respectively, whereas other compounds showed weak inhibition of NF-κB transcriptional activity ranging from 17.54 to 92.6 μM. A structure–activity relationship of flavonoids <B>1</B>–<B>9</B> was also discussed.         Genetic variations in the bitter taste receptor gene TAS2R38 are related to cigarette smoking behavior in Han Chinese smokers

        Background: Smoking behavior is influenced by multiple genes, including the bitter taste gene TAS2R38. It has been reported that the correlation between TAS2R38 and smoking behavior has ethnicity-based differences. However, the TAS2R38 status in Chinese smokers is still unclear. Objective: This study aims to investigate the possible relationship between genetic variations in TAS2R38 (A49P, V262A and I296V) and smoking behaviors in the Han Chinese population. Methods: The haplotype analyses were performed and smoking behavior questionnaire was completed by 1271 individuals. Genetic association analyses for smoking behavior were analyzed using chi-square test. Further, for investigating the molecular mechanism of TAS2R38 variants effect on smoking behavior, we conducted TAS2R38-PAV and TAS2R38-AVI expression plasmids and tested the cellular calcium assay by cigarette smoke compounds stimulus in HEK293. Results: Significant associations of genetic variants within TAS2R38 were identified with smoking behavior. We found a higher PAV/PAV frequency than AVI/AVI in moderate and high nicotine dependence (FTND ≥ 4; X2 = 4.611, 1 df, p = 0.032) and strong cigarette smoke flavor intensity preference (X2 = 4.5383, 1 df, p = 0.033) in participants. Furthermore, in the in vitro cellular calcium assay, total particle matter (TPM), N-formylnornicotine and cotinine, existing in cigarette smoke, activated TAS2R38-PAV but not TAS2R38-AVI-transfected cells. Conclusion: Our data highlights that genetic variations in TAS2R38 are related to smoking behavior, especially nicotine dependence and cigarette smoke flavor intensity preference. Our findings may encourage further consideration of the taste process to identify individuals susceptible to nicotine dependence, particularly Han Chinese smokers.

        UPLC-QTOF-MS/MS Analysis for Steaming Times-dependent Profiling of Steamed Panax quinquefolius and its Ginsenosides Transfromations induced by Repetitious Steaming

        The metabolic profiles of Panax quinquefolius and its associated therapeutic values are critically affected by the repetitious steaming times. The times-dependent steaming effect of P. quinquefolius is not well-characterized and there is also no official guidelilne on its times of steaming, In this paper, a UPLC-Q-TOF-MS/MS method was developed for the qualitative profiling of multi-parametric metabolic changes of raw P. quinquefolius durilng the repetitious steaming process. Our method was successful in discriminating the differentially multi-steamed herbs. Meantime, the repetitious steaming-inducing chemical transformations in the preparation of black American ginseng (American ginseng that was subjected to 9 cycles of steaming treatment) were evaluated by this UPLC-Q-TOF-MS/MS based chemical profiling method. Under the optimized UPLC-Q-TOF-MS/MS conditions, 29 major ginsenosides were unambiguously identified and/or tentatively assigned in both raw and multi-steamed P. quinquefolius within 19 min, among them 18 ginsenosides were detected to bo newly generated during the preparatory process of black American ginseng. The mechanisms involved were further deduced to be hydrolysis, dehydration, decarboxylation and addition reactions of the original ginsenosiders in raw P. quinquefolius through analyzing 9 cycles of steaming extracts of 14 pure reference ginsenosides. Our novel steaming times-dependent metabolic profiling represents the paradigm shift in the global quality control of multi-steamed P. quinquefolius products.

        A finite-element analysis of the connecting bolts of slewing bearings based on the orthogonal method

        This paper presents an analysis of the main factors affecting the performance of connecting bolts for slewing bearings. For double row four-point contact ball bearings, major factors include the preload, friction coefficient and height of the bushes. First, bolt connection performances with different levels of each factor are calculated with the finite element method (FEM). Then, the calculation results are analyzed with the orthogonal method. The analysis indicates that the preload is the most important factor affecting the bolt connection performance, followed, to a lesser degree, by the height of the bushes. The friction coefficient is the least important factor.

        Determination of 11 Ginsenosides in Black Ginseng Developed from Panax ginseng by High Performance Liquid Chromatography

        A high performance liquid chromatography (HPLC) method has been developed for determination of 11 ginsenosides in black ginseng (BG, white ginseng that is subjected to 9 cycles of 95℃ for 3 hr). After eluted by gradient elution of water-acetonitrile without buffer in 70 min, 11 ginsenosides in BG were identified. The proposed method provided good linearity (R²>0.9995), accuracy (92.2-106.6%), and intra- and interday precision (RSD<2.6%). In addition, ginsenosides compositions in white, red, and black ginsengs were investigated using this method, respectively. Interestingly, in BG, the content of ginsenoside Rg3 which does not existed in white ginseng was 7.51 ㎎/g, approximately 20 times than that in red ginseng.

        Characteristics of registered studies for Coronavirus disease 2019 (COVID-19): a systematic review

        Background: The World Health Organization characterized the Coronavirus disease 2019 (COVID-19) as a pandemic on March 11th. Many clinical trials on COVID-19 have been registered, and we aim to review the study characteristics and provide guidance for future trials to avoid duplicated effort. Methods: Studies on COVID-19 registered before March 3rd, 2020 on eight registry platforms worldwide were searched and the data of design, participants, interventions, and outcomes were extracted and analyzed. Results: Three hundred and ninety-three studies were identified and 380 (96.7%) were from mainland China, while 3 in Japan, 3 in France, 2 in the US, and 3 were international collaborative studies. Two hundred and sixty-six (67.7%) aimed at therapeutic effect, others were for prevention, diagnosis, prognosis, etc. Two hundred and two studies (51.4%) were randomized controlled trials. Two third of therapeutic studies tested Western medicines including antiviral drugs (17.7%), stem cell and cord blood therapy (10.2%), chloroquine and derivatives (8.3%), 16 (6.0%) on Chinese medicines, and 73 (27.4%) on integrated therapy of Western and Chinese medicines. Thirty-one studies among 266 therapeutic studies (11.7%) used mortality as primary outcome, while the most designed secondary outcomes were symptoms and signs (47.0%). Half of the studies (45.5%) had not started recruiting till March 3rd. Conclusion: Inappropriate outcome setting, delayed recruitment and insufficient numbers of new cases in China implied many studies may fail to complete. Strategies and protocols of the studies with robust and rapid data sharing are warranted for emergency public health events, helping the timely evidence-based decision-making.

        Transcriptional profiling of MED Bemisia tabaci exposed to thermal stress and verification of HSP70 expression

        Temperature changes impact agricultural production and also influence the behavior and community structure of many insect species. Bemisia tabaci, is an invasive and destructive pest with at least 44 cryptic species and occurring in over 100 countries and regions. In this study, transcriptional profiling was used to evaluate the response of the Mediterranean (MED) cryptic species B. tabaci exposed to cold and hot temperature stress. In response to cold stress, 3,024 unigenes were differentially expressed and mapped to 39 Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment pathways. A total of 4,437 unigenes were differentially expressed during heat stress, and these represented 18 KEGG enrichment pathways. Quantitative RT-PCR was used to determine the relative expression level of the 11 HSP70 fragments; these were significantly up-regulated during high temperature stress relative to the control group that was maintained at 26°C. In conclusion, the transcriptome data indicated that B. tabaci responds to thermal stress via multiple genes and networks. Furthermore, HSP70 plays an important role in stress tolerance of the MED B. tabaci, especially during heat stress.

      • Association of a Pre-miR-27a Polymorphism with Cancer Risk: an Updated Meta-analysis

        MicroRNA-27a is highly expressed in cancers and has been identified as an oncogenic microRNA. A genetic variant in pre-miR-27a (rs895819) with a transition of A to G has been demonstrated to be associated with cancer risk; however, the results of these studies remain conflicting rather than conclusive. Therefore, we performed a meta-analysis to derive a more precise estimation. Through searching PubMed or other databases up to March 2014 using the following MeSH terms and keywords, "miR-27a", "polymorphism" and "cancer", seventeen case-control studies were identified in this meta-analysis, including 7,813 cases and 9,602. Crude odds ratios (ORs) and corresponding 95% confidence intervals (CIs) were calculated to investigate the association strength between rs895819 and the susceptibility of cancer. The results of the overall meta-analysis did not suggest any association between rs895819 polymorphism and cancer susceptibility, and this remained in Asians as a subgroup. In Caucasians, however, the rs895819 was associated with a reduced cancer risk in heterozygous (OR, 0.83; 95%CI, 0.75-0.93) and dominant models (OR, 0.84; 95%CI, 0.76-0.93), and the [G] allele of rs895819 showed a protective effect (OR, 0.90, 95%CI, 0.84-0.97). Further studies showed a significant association between the [G] allele of rs895819 and decreased risk of breast cancer (0.91; 95%CI, 0.85-0.98), and stratified analyses indicated a protective effect of the [G] allele in Caucasians (OR, 0.89; 95%CI, 0.82-0.98), younger breast cancer cases (OR, 0.87; 95%CI, 0.79-0.96), and in the group of unilateral breast cancer patients (OR, 0.90; 95%CI, 0.83-0.97). These findings suggest an association between pre-miR-27a polymorphism rs895819 and cancer risk in Caucasians. The protective effect of rs895819 [G] allele in younger breast cancer and in the group of unilateral breast cancer patients await further confirmation since the included studies in this meta-analysis were limited.

        An efficient Automatic Modulation Classification method based on the Convolution Adaptive Noise Reduction network

        Due to the influence of noise in the received signal in non-cooperative communication, it is difficult for existing Automatic modulation classification methods to balance classification accuracy and model complexity. This paper proposes a novel Convolutional Adaptive Noise Reduction (CANR) network, which consists of an Adaptive Noise Reduction (ANR) module and a Convolutional Feature Extraction (CFE) module. The ANR and CFE modules denoise the combined input and capture the spatiotemporal features in the time series. Experiments on benchmark datasets show that the proposed network has the fewest training parameters and state-of-the-art recognition accuracy under the same conditions.
