        15-Deoxy-Δ<sup>12,14</sup>-prostaglandin J<sub>2</sub> activates PI3K-Akt signaling in human breast cancer cells through covalent modification of the tumor suppressor PTEN at cysteine 136

        <P><B>Abstract</B></P> <P>15-Deoxy-Δ<SUP>12,14</SUP>-prostaglandin J<SUB>2</SUB> (15d-PGJ<SUB>2</SUB>), one of the terminal products of cyclooxygenase-2-catalized arachidonic acid metabolism, has been shown to stimulate breast cancer cell proliferation and migration through Akt activation, but the underlying mechanisms remain poorly understood. In the present study, we investigated the effects of 15d-PGJ<SUB>2</SUB> on the activity of PTEN, the inhibitor of the phosphoinositide 3-kinase (PI3K)-Akt axis, in human breast cancer (MCF-7) cells. Since the α,β-unsaturated carbonyl moiety in the cyclopentenone ring of 15d-PGJ<SUB>2</SUB> is electrophilic, we hypothesized that 15d-PGJ<SUB>2</SUB>-induced Akt phosphorylation might result from the covalent modification and subsequent inactivation of PTEN that has several critical cysteine residues. When treated to MCF-7 cells, 15d-PGJ<SUB>2</SUB> bound to PTEN, and this was abolished in the presence of the thiol-reducing agent dithiothreitol. A mass spectrometric analysis by using recombinant and endogenous PTEN protein revealed that the cysteine 136 residue (Cys<SUP>136</SUP>) of PTEN is covalently modified upon treatment with 15d-PGJ<SUB>2</SUB>. Notably, the ability of 15d-PGJ<SUB>2</SUB> to covalently bind to PTEN as well as to induce Akt phosphorylation was abolished in the cells expressing a mutant form of PTEN in which Cys<SUP>136</SUP> was replaced by serine (C136S-PTEN). The present study demonstrates for the first time that electrophilic 15d-PGJ<SUB>2</SUB> directly binds to cysteine 136 of PTEN and provides new insight into PTEN loss in cancer progression associated with chronic inflammation. These observations suggest that 15d-PGJ<SUB>2</SUB> can undergo nucleophilic addition to PTEN, presumably at Cys<SUP>136</SUP>, thereby inactivating this tumor suppressor protein with concomitant Akt activation.</P> <P><B>Highlights</B></P> <P> <UL> <LI> PTEN is inactivated through covalent modification by 15-deoxy-Δ<SUP>12,14</SUP>-prostaglandin J<SUB>2</SUB> in breast cancer MCF-7 cells. </LI> <LI> 15-Deoxy-Δ<SUP>12,14</SUP>-prostaglandin J<SUB>2</SUB> directly binds to the cysteine 136 residue of the recombinant PTEN. </LI> <LI> PTEN interaction with 15-deoxy-Δ<SUP>12,14</SUP>-prostaglandin J<SUB>2</SUB> stimulates Akt phosphorylation. </LI> <LI> 15-Deoxy-Δ<SUP>12,14</SUP>-prostaglandin J<SUB>2</SUB> stimulates growth of MDA-MB-231 cells when inoculated to the athymic nude mice. </LI> </UL> </P>

        Nuclear Localization of Fibroblast Growth Factor Receptor 1 in Breast Cancer Cells Interacting with Cancer Associated Fibroblasts

        Cancer-associated fibroblasts (CAFs) represent a major component of the tumor microenvironment and interplay with cancer cells by secreting cytokines, growth factors and extracellular matrix proteins. When estrogen receptor-negative breast cancer MDAMB-231 cells were treated with the CAF-conditioned medium (CAF-CM), Akt and STAT3 involved in cell proliferation and survival were activated through phosphorylation. CAFs secrete fibroblast growth factor 2 (FGF2), thereby stimulating breast cancer cell progression. Akt activation induced by CAF-CM in MDA-MB-231 cells was abolished when FGF2-neutralizing antibody was added. Treatment of MDA-MB-231 cells directly with FGF2 enhanced the phosphorylation of Akt and the FGF receptor (FGFR) substrate, FRS2α. These events were abrogated by siRNA-mediated silencing of FGFR1. In a xenograft mouse model, co-injection of MDAMB-231 cells with activated fibroblasts expressing FGF2 dramatically enhanced activation of Akt. Stable knockdown of FGFR1 blunted Akt phosphorylation in xenograft tumors. MDA-MB-231 cells co-cultured with CAFs or directly stimulated with FGF2 exhibited enhanced nuclear localization of FGFR1. Notably, FGF2 stimulation produced reactive oxygen species (ROS) accumulation in MDA-MB-231 cells, and FGF2-induced nuclear accumulation of FGFR1 was abrogated by the ROS scavenging agent, N-acetylcysteine.

      • 공감각적 후각체험을 위한 향기와 음악 매칭 연구

        시각, 청각, 후각, 미각, 촉각은 인간의 5대 감각으로 인간의 생존과 직결되어 있다. 인간은 5가지 감각을 총동원하여 세상을 경험하고 위험으로부터 자신을 보호하며 상대방과 소통한다. 하나의 감각만 자극을 받는 것보다 동시다발적으로 주어지는 감각은 인식의 대상을 더욱 명확하고 깊게 경험할 수 있다. 기존의 크로스 모달 연관관계 연구는 하나의 감각이 다른 감각을 연상시키는 것에 초점을 맞춘 반면1), 본 연구에서는 접근방식을 바꿔 향기에서 속도를 의미하는 증기압과 음악에서 속도를 의미하는 진동수를 수식으로 연결하여 향기와 음악을 함께 경험할 수 있는 초기연구를 진행하였다. 그 결과 136개의 화학 향료성분과 피아노 음정을 매칭하였으며, 음악으로부터 향기를 조향하는 새로운 조향 방법을 개발하였다. The five basic human senses: sight, hearing, smell, taste, and touch are directly related to human survival. Humans use all five senses to perceive the world, protect themselves from danger, and communicate with each other. As the sensory systems function simultaneously, not one at a time, humans are able to perceive an object more clearly and deeply. While traditional cross-modal association studies focus on the associations between sensory systems, we took a different approach. In this study, an initial study was conducted to cross-experience scent and music by connecting the vapor pressure, determining speed in scent, and the frequency, determining speed of sound, with a simple formula. As a result of this study, 136 chemical fragrance components and piano notes were matched, and a new perfumery method was developed to create scent from music.

        Resveratrol suppresses migration, invasion and stemness of human breast cancer cells by interfering with tumor-stromal cross-talk

        <P><B>Abstract</B></P> <P>Cancer-associated fibroblasts (CAFs) constitute a major compartment of the tumor microenvironment. CAFs produce a variety of cytokines, growth factors and extracellular matrix proteins, thereby stimulating tumor progression. CAFs are distinct from normal fibroblasts for their overexpression of α-smooth muscle actin. Recent studies suggest that CAFs play an important role in proliferation and migration of cancer cells through cross-talk with them. Resveratrol (<I>trans</I>-3,4′5,-trihydroxystilbene), a phytoalexin present in grapes, has been reported to possess chemopreventive and chemotherapeutic activities. In the present study, we examined the effects of resveratrol on CAF-induced migration, invasion and self-renewal activity of breast cancer cells. Resveratrol inhibited proliferation, migration and invasion of human breast cancer cells treated with CAF-conditioned media (CAF-CM). Resveratrol treatment suppressed the CAF-CM-induced expression of Cyclin D1, c-Myc, MMP-2 and MMP-9. In addition, resveratrol inhibited Sox2 expression as well as activation of Akt and STAT3 induced by CAF-CM in breast cancer cells. Further, resveratrol abrogated stemness properties and reduced the expression of self-renewal signaling molecules in stem-like breast cancer cells. Taken together, the present study provides insights into the role of resveratrol in tumor microenvironment with focus on interaction between cancer cells and the hosting niche.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Resveratrol inhibited proliferation, migration and invasion of human breast cancer cells treated with CAF conditioned media. </LI> <LI> Resveratrol suppressed the expression of cyclin D1, c-Myc, MMP-2, MMP-9 and Sox-2 in breast cancer cells stimulated with CAFs. </LI> <LI> Resveratrol inhibited activation of Akt and STAT3 induced in human breast cancer cells stimulated with CAF conditioned media. </LI> <LI> Resveratrol abrogated the expression of self-renewal signaling molecules in stem-like breast cancer cells. </LI> </UL> </P>

        A Study on the Method and System for Organization’s Name Authorization of Korean Science and Technology Contents

        Science and technology contents (research papers, patents, reports) are the most common reference material for researchers involved in research and development in the fields of science and technology. Based on various search elements (title, abstract, keyword, year of publication, name of journal, name of author, publisher, etc.), many services are available for users to search science and technology contents and bibliographic information owned by libraries. Authority data on organization name can be useful as an element for author identification and as an element to search for results produced by specific organizations. However, organization name is not taken into account by current search services for domestic academic information and bibliographic records. This study analyzed organization name data contained in the metadata of science and technology contents, which are the basis of the establishment of authority data, and proposes a method and system based on string containment and exact string matching.

        As the number of scientific and technical contents increases, services that support efficient search of scientific and technical contents are required. When an author’s affiliation is used as a keyword, not only the contents produced by the affiliation can be searched, but also the identification rate of the search result using the author and the term as keyword can be improved. Because of the ambiguity and vagueness of the data used as a search keyword, the search result may include false negative or false positive. However, the previous research on the control through identification of the search keyword is mainly focused on the author data and terminology data. In this paper, we propose the algorithm to identify affiliations and experiment with show the experiment with scientific and technological contents held by the Korea Institute of Science and Technology Information.

        Gold-layered calcium phosphate plasmonic resonants for localized photothermal treatment of human epithelial cancer

        <P>We fabricated <I>de novo</I> biocompatible mineral plasmon resonants for localized and systemic treatment of cancer. Biodegradable calcium-phosphate gold nanocomposites were synthesized for inducing a superior surface plasmon resonance effect.       • Development of an Inter-Organizational Collaboration Model based on CoP: Project Management Perspective

        The importance of collaboration has long been emphasized not only within an organization but also among organizations. Without collaborations, access to partner’s knowledge or skill would probably be restricted and innovation among organizations could not be realized. Collaboration is also an important success factor for organizations in the project management area. Many companies collaborate on a project with partners for the accomplishment of strategic goals. However, to date, a research on project management has tended to focus on the tools of management. For example, the Gantt chart, Risk classification and Prototyping, instead of making an effort to increase collaboration between the owners and contractor organizations during the project. The main objective of this paper is to present an inter-organizational collaboration model based on CoP during the period of project management. Thus, the model demonstrates that project management through collaboration is critically essential for not only simple achieving the success of the project, but also reaching strategic goals that eventually enrich the organization’s overall performance and capability. And then a case study was performed with a real company to apply this model.

        노인 류마티스 관절염 환자에서의 요양급여심사기준 변경에 따른 TNF-α 억제제 약물사용양상

        2014년 1월부터 류마티스 관절염 환자의 TNF-α 억제제 사용에 관한 급여기준이 기존의 다양한 검사 결과를 요구하는 방법에서 DAS28이라는 지표를 활용하는 방향으로 완화되었다. 본 연구에서는 이 급여기준의 변경 전후 1년간 류마티스 관절염(한국표준질병사인분류 M05, M06)노인 환자들의 TNF-α 억제제 이용 변화를 분석비교하였다. 건강보험심사평가원의 2013, 2014년 고령환자데이터셋을 활용하여 분석하였고, 구간별 회귀분석을 이용한 개입 시계열 분석방법을 적용하여 급여기준의 변경 전후 1년간 월별 TNF-α 억제제의 처방건과 사용량을 분석하였다. 사용량은 천명의 류마티스 관절염 환자의 일당 정립 용량(Daily Defined Dose)를 사용하였다. 분석결과, 급여기준 변경 후 월별 TNF-α 억제제의 처방건과 사용량은 유의한 변화를 보이지 않았다. 그러나 의료기관 종별에 따라 하위분석을 실시한 결과, 급여기준 변경 후 상급종합병원에서의 월별 처방과 사용량은 증가하였고, 종합병원, 병원/의원에서의 월별 처방과 사용량은 감소한 것으로 나타났다. 전체적으로 급여기준의 변경이 TNF-α 억제제의 처방과 사용에 영향을 미치지 않은 것으로 나타났으나 의료기관 종별에 따라 유의한 변화가 있었다. 급여기준의 개선으로 인해 장기간으로 보았을 때 류마티스 관절염 환자들의 TNF-α 억제제에 대한 접근성이 증가했는지에 대한 추가적인 연구가 필요할 것으로 보인다. In January 2014, there was a change in the reimbursement criteria of TNF-α inhibitor (TNFi) for RA patients. The new reimbursement criteria require DAS28 of a patient to assess disease activity, while previous reimbursement criteria required laboratory data and several subjective assessments. We examined the effect of a change in reimbursement criteria on the utilization of TNFi in RA patients. Data sources were 2013-2014 Aged Patient Sample data of Health Insurance Review and Assessment. Monthly trends in prescription and consumption of TNFi were estimated using segmented regression of interrupted time series. The consumption of TNFi was measured in terms of Daily Defined Dose per 1,000 RA patients per day. Subgroup analysis was conducted by the types of institution. There were increased baseline trends in prescription and consumption of TNFi, which were not significant. In subgroup analysis, tertiary hospital showed a significant increase in prescription and consumption immediately after the change in reimbursement criteria, while general hospital, hospital, and clinic showed a significant decrease in prescription and consumption. These trends were consistent throughout one year after the change in reimbursement criteria. This study shows that the change in reimbursement criteria of TNFi in RA patients had little impact on the overall utilization of TNFi. However, the change in reimbursement criteria significantly affected the pattern of TNFi utilization by the types of institution. Further research is needed to examine whether this change improved the accessibility to TNFi in RA patients.
