      • Combined Genotype Analyses of Precursor miRNA-196a2 and -499a Variants with Hepatic and Renal Cancer Susceptibility- a Preliminary Study

        MicroRNAs, a novel class of small non-coding RNAs, are key players in many cellular processes, including cell proliferation, differentiation, invasion and regeneration. Tissue and circulatory microRNAs could serve as useful clinical biomarkers and deregulated expression levels have been observed in various cancers. Gene variants may alter microRNA processing and maturation. Thus, we aimed to investigate the association of MIR-196a2 rs11614913 (C/T), MIR-499a rs3746444 (A/G) polymorphisms and their combination with cancer susceptibility in an Egyptian population. Sixty five renal cell carcinoma (RCC) and 60 hepatocellular carcinoma (HCC) patients and 150 controls were enrolled in the study. They were genotyped using real-time polymerase chain reaction technology. Both $miR-196a2^*T$ and $miR-499a*G$ were associated with RCC risk, but only $miR-196a^*T$ was associated with HCC development. Carriage of the homozygote combinations ($MIR196a2^*TT+MIR499a^*AA$) and ($MIR196a2^*CC+MIR499a^*GG$) was associated with 25 and 48 fold elevation of likelhood to develop RCC, respectively. The miR-196a2 SNP was also linked with larger tumor size in RCC and advanced tumor stage in HCC. miR-196a2 and miR-499a combined genotypes were associated with RCC and HCC. Further functional analysis of SNPs is required to confirm relationships between genotypes and phenotypes.

        Insect Cell Surface Expression of Hemagglutinin (HA) of Egyptian H5N1 Avian Influenza Virus Under Transcriptional Control of Whispovirus Immediate Early-1 Promoter

        In the present study, whispovirus immediate early 1 promoter (ie-1) was used to initiate surface expression of the hemagglutinin (HA) protein of Egyptian H5N1 avian influenza virus (AIV) by using the baculovirus expression vector system. The HA gene and whispovirus ie-1 promoter sequence were synthesized as a fused expression cassette (ie1-HA) and successfully cloned into the pFastBac-1 transfer vector. The recombinant vector was transformed into DH10Bac competent cells, and the recombinant bacmid was generated via site-specific transposition. The recombinant bacmid was used for transfection of Spodoptera frugiperda (Sf-9) insect cells to construct the recombinant baculovirus and to induce expression of the HA protein of H5N1 AIV. The recombinant glycoprotein expressed in Sf-9 cells showed hemadsorption activity. Hemagglutination activity was also detected in both extra- and intracellular recombinant HAs. Both the HA and hemadsorption activities were inhibited by reference polyclonal anti-H5 sera. Significant expression of the recombinant protein was observed on the surface of infected insect cells by using immunofluorescence. SDS-PAGE analysis of the expressed protein revealed the presence of a visually distinguishable band of ~63 kDa in size, which was absent in the non-infected cell control. Western blot analysis confirmed that the distinct 63 kDa band corresponded to the recombinant HA glycoprotein of H5N1 AIV. This study reports the successful expression of the HA protein of H5N1 AIV. The expressed protein was displayed on the plasma membrane of infected insect cells under the control of whispovirus ie-1 promoter by using the baculovirus expression vector system.

        Synthesis of polyoxometalates supported on HZSM-5 for the photocatalytic purification of crude terephthalic acid under mild conditions

        <P><B>Abstract</B></P> <P>Phosphomolybdic (PMo) and vanadium-containing phosphomolybdic (PV<SUB>2</SUB>Mo) acids were synthesized and supported on HZSM-5 zeolite via a wet impregnation technique and used as photocatalysts for the purification of terephthalic acid (TA) via an economic process. Nowadays, purified TA can be obtained from crude TA due to their great commercial importance by separating 4-carboxybenzaldehyde (4-CBA), which represents high environmental toxicity and negatively affects the applications of TA. For these reasons, conversion of 4-CBA to TA is highly desirable for both enhancing the efficiency of purified TA production and environmental reasons. The synthesized catalysts were characterized by XRD, FTIR, Raman spectroscopy, BET, SEM, and TEM. The purification process was carried out by under different parameters as VUV irradiation, persulfate (PS), time, and different types of catalysts (PMo or PV<SUB>2</SUB>Mo)/HZSM-5. This kind of combination showed complete removal of the 4-CBA with a high yield of PTA in 2 h. The high efficiency attributed to the acidity of the polyoxometalates/HZSM-5, which increase the selectivity to oxidize 4-CBA at room temperature and much shorter reaction time compared to other common methods.</P> <P><B>Highlights</B></P> <P> <UL> <LI> PMo or PV<SUB>2</SUB>Mo / HZSM-5 used as a photocatalyst for the economic purification of CTA. </LI> <LI> Catalyst achieve complete purification for CTA at 2 h and at room temperature. </LI> <LI> PV<SUB>2</SUB>Mo/HZSM-5 show no significant loss of catalytic activity for four consecutive. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        THE HAEMATOLOGY OF GROWING CAMELS (Camelus dromedarius) DURING THE FIRST YEAR OF LIFE

        The haematological profiles of camel calves of either sex were monitored at monthly intervals between 1-12 months of age. RBC, Hb and PCV increased with age for several months, with corresponding increases in MCH and MCHC, and a relative reduction in MCV. WBC and lymphocyte counts were highest at the age of 1 month, fell progressively during the following 6 months, and stabilized thereafter. Neutrophils were also highest at the age of one month and decreased slowly with advancing age. Eosinophils, monocytes and basophils fluctuated only from month to month. Age effect was significant for WBC, lymphocytes and neutrophils, and all the erythrocytic parameters, except MCV. Sex effect was significant for Hb and eosinophils.

        Experimental and FE Study on RC One-Way Slabs Upgraded with FRP Composites

        The use of externally bonded Fiber Reinforced Polymer (FRP) composites as a means of upgrading the flexural capacity of Reinforced Concrete (RC) one-way slabs is experimentally and numerically investigated in this study. A total of four groups of eight slabs were tested under four-point bending. The two slabs of the first group were left unstrengthened to be used as control specimens. The two slabs of the second group were externally strengthened with adhesively bonded pultruded, pre-cured CFRP plates. The four slabs of the last two groups were externally upgraded with unidirectional carbon (or E-glass) fiber fabric impregnated with an epoxy resin. In addition to the experimental program, a numerical investigation utilizing nonlinear Finite Element (FE) analysis was conducted using LS-DYNA software. Besides the eight slabs tested in this study, another eleven slabs were collected from the literature for the purpose of finite element validation. A comparison was made between the experimental and numerical results and good agreement was achieved. Based on FE validation, the numerical analysis was extended to include additional cases to study the effect of axial FRP stiffness and FRP-to-concrete width ratio on the flexural performance of upgraded slabs. As a result of the numerical study, new stiffness and reinforcement parameters were introduced in this research. These parameters were employed in the development of two new formulas for predicting the FRP debonding strain and percent gain in flexural capacity of FRPstrengthened slabs.

        Effect of Supplemental Chromium Levels on Performance, Digestibility and Carcass Characteristics of Transport-stressed Lambs

        A trial was conducted to study the effect of supplemental chromium (Cr) levels from a Cr-yeast source on performance, digestibility and carcass characteristics of transport-stressed lambs. Forty-eight Naemi lambs (avg. BW 31.7 kg) were transported by truck for a distance of 1,450 km. On arrival day, the lambs were randomly allocated to four groups receiving diets supplemented with 0.0, 0.3, 0.6 or 0.9 ppm Cr. Each group consisted of four separately housed replicates of three lambs each. The lambs were fed their respective diets ad libitum for 84 d (21 d stress period, followed by 63 d growing period). Road transit of lambs resulted in a decreased (p<0.001) live body weight of 8.5%. Supplementation of Cr-yeast did not alter the performance of lambs during the stress period. Linear and quadratic increases (p<0.05) were observed in DMI and ADG, respectively, with increasing supplemental Cr levels in the diets during the growing period. Values were greater (p<0.05) by 14.7% and 20.8%, respectively, for lambs fed 0.3 ppm Cr compared to control, while those fed on the other two levels were intermediate. Over the 84-d feeding period, a trend was noted towards a slight increase in loin eye area and a decrease in body wall fat thickness for lambs fed Cr supplementation compared to the control group. This study suggests that the supplementation of Cr-yeast, especially at 0.3 ppm level, is beneficial for improving the performance of growing lambs whether the animals are stressed or not.

        Cerebral fat embolism syndrome: diagnostic challenges and catastrophic outcomes: a case series

        Fat embolism syndrome is a rare but alarming, life-threatening clinical condition attributed to fat emboli entering the circulation. It usually occurs as a complication of long-bone fractures and joint reconstruction surgery. Neurological manifestations usually occur 12 to 72 hours after the initial insult. These neurological complications include cerebral infarction, spinal cord ischemia, hemorrhagic stroke, seizures, and coma. Other features include an acute confusional state, autonomic dysfunction, and retinal ischemia. In this case series, we describe three patients with fat embolism syndrome who presented with atypical symptoms and signs and with unusual neuroimaging findings. Cerebral fat embolism may occur without any respiratory or dermatological signs. In these cases, diagnosis is established after excluding other differential diagnoses. Neuroimaging using brain magnetic resonance imaging is of paramount importance in establishing a diagnosis. Aggressive hemodynamic and respiratory support from the beginning and consideration of orthopedic surgical intervention within the first 24 hours after trauma are critical to decreased morbidity and mortality.

        Performance and Carcass Composition of Broilers under Heat Stress : II. The Effects of Dietary Lysine

        An experiment was conducted to determine the effect of lysine on performance and carcass composition of broilers under heat stress during the grower period (3-6 weeks). A factorial arrangement of three levels of dietary protein (18, 20, and 22%), three levels of dietary lysine (1.26, 1.39, and 1.52%), and two rearing temperature regimens were used in this study. Birds were kept under either moderate temperature ($24{\pm}1^{\circ}C/24h$) or hot cycling temperature ($26-34^{\circ}C/6h$, $34{\pm}1^{\circ}C/12h$, and $34-26^{\circ}C/6h$). Body weight (BW), weight gain (WG), feed intake (FI), feed conversion (FE), carcass weight (CW), carcass yield (CY), and percentages of breast meat (BM), abdominal fat (AF), drumsticks (DS), and thighs (TH) were determined at the end of experiment. Exposure to high ambient temperature significantly (p<0.05) decreased BW, WG, FI, FE, CW, BM, AF, and increased CY, DS, and TH. High dietary protein significantly (p<0.05) decreased AF and TH, and improved CW only under moderate temperature, resulting in significant (p<0.05) protein by temperature interaction. High dietary lysine significantly (p<0.05) decreased BW, WG, FI, CW, CY and AF, while BM was reduced only when high dietary protein was fed, resulting in significant (p<0.05) protein by lysine interaction. It is concluded that increasing dietary lysine adversely affected broilers' performance and carcass composition irrespective of rearing temperature.

        Synthesis and Antimicrobial Activity of Certain New 1,2,4-Triazolo[1,5-a]Pyrimidine Derivatives

        Certain new derivatives of 1,2,4-triazolo[1,5-a]pyrimidines were synthesized through the reaction of 1,2,4-triazolo[1,5-a]pyrimidine-7-ol with ethyl bromoacetate to afford the ethyl acetate ester, which upon hydrazinolysis gives the corresponding hydrazide. The hydrazide is the key intermediate which was used for the synthesis of the target compounds. The structures of the new compounds were assigned by spectral and elemental methods of analyses. The synthesized compounds were tested for their in vitro antibacterial and antifungal activities. Most of the tested compounds showed comparable results with those of ampicillin and fluconazole reference drugs.
