      • Risk Factors for Early Recurrence of HBV-related Hepatocellular Carcinoma Meeting Milan Criteria after Curative Resection

        Background: The prognosis of patients with hepatocellular carcinoma (HCC) after curative resection varies greatly. Few studies had investigated the risk factors for early recurrence (recurrence-free time ${\leq}$ 1 year) of hepatitis B virus (HBV)-related HCCs meeting Milan criteria. Methods: A retrospective analysis was performed on the 224 patients with HCC meeting Milan criteria who underwent curative liver resection in our center between February 2007 and March 2012. The overall survival (OS) rate, recurrence-free survival (RFS) rate and risk factors for early recurrence were analyzed. Results: After a median follow-up of 33.3 months, HCC reoccurred in 105 of 224 patients and 32 died during the period. The 1-, 3- and 5-year OS rates were 97.3%, 81.6% and 75.6% respectively, and the 1-, 3- and 5-year RFS rates were 73.2%, 53.7% and 41.6%. Cox regression showed alpha-fetoprotein (AFP) > 800 ng/ml (HR 2.538, 95% CI 1.464-4.401, P=0.001), multiple tumors (HR 2.286, 95% CI 1.123-4.246, P=0.009) and microvascular invasion (HR 2.518, 95% CI 1.475-4.298, P=0.001) to be associated with early recurrence (recurrence-free time ${\leq}$ 1-year) of HCC meeting Milan criteria. Conclusions: AFP > 800 ng/ml, multiple tumors and microvascular invasion are independent risk factors affecting early postoperative recurrence of HCC. In addition resection appears capable of replacing liver transplantation in some situations with safety and a better outcome.

      • Expression Level of Valosin Containing Protein is Associated with Prognosis of Primary Orbital MALT Lymphoma

        Objective: To investigate whether the expression level of valosin-containing protein (VCP) is correlated with the prognosis of primary orbital mucosa-associated lymphoid tissue (MALT) lymphoma. Methods: VCP expression in 58 samples from primary orbital MALT lymphoma patients was determined by immunohistochemisty using monoclonal antibodies. Correlations between VCP expression level and prognosis were clarified by statistical analysis. Results: It was found that the percentage of VCP positive cells in samples of primary orbital MALT lymphoma ranged from 32% to 95%. The samples were divided into two groups (level 1 and level 2) according to the median value (45%) of the percentage of VCP positive cells. It was found that the expression level of VCP was significantly correlated with recurrence (P=0.003) and tumor size (P=0.008). At the same time, the 5-year disease-free and overall survival rate of patients of level 1 was significantly better than that of level 2 (P=0.001; P=0.032). There was no observed correlation between the expression level of VCP and other clinical features. Conclusion: VCP could be a useful marker for predicting the prognosis of primary orbital MALT lymphoma.

        A Fast Estimation Method of Soil Discharged by an Earth Pressure Balanced Shield Machine

        The soil discharged by an earth pressure balanced (EPB) shield machine reflects ground loss and controls surface settlement. In engineering, the discharge was mainly estimated by weighing the tunnelling spoil, however, human disturbance and information lag bring serious hidden security risks to tunnel construction. Through the analysis of conveying flow and force of screw conveyor, an energy model of interaction between screw conveyor and soil discharged was proposed and verified experimentally. Then, the energy model was combined with the discharge equation and the force balance equation, and a semi-analytical method to fast estimate the soil discharged of the EPB shield machine was proposed. Taking an EPB shield construction project as an example, according to the data of earth pressure, the torque and the rotation of the screw conveyor recorded by EPB shield machine, the real-time soil discharged of EPB shield machine can be accurately estimated. The mean absolute error (MAE) of this method is 3.177 × 103 kg compared with the measured weight of tunnelling spoil, which verifies the rationality of the method and provides a method for further effective evaluation and control of ground loss.

        Overexpression of rice phosphate transporter gene OsPT2 enhances nitrogen fixation and ammonium assimilation in transgenic soybean under phosphorus deficiency

        Phosphorus (P) deficiency is one of the major factors that limit legume nodulation and nitrogen (N) fixation, and thus legume productivity. In our previous study, we showed that three T2 transgenic soybean lines overexpressing rice phosphate transporter gene OsPT2 showed enhanced tolerance to low P stress. This study aimed to determine whether OsPT2 overexpression would increase N2 fixation and ammonium assimilation in three T3 homozygous transgenic lines (HTLs) under P deficiency in pot culture. Under low inorganic phosphate (Pi) conditions, the P accumulation, total N and total ureide concentrations were significantly higher in the T3 HTLs than in the wild type (WT) plants. Further, the T3 HTLs showed significantly better plant growth performance and nodule development than the WT plants under low-Pi conditions. Quantitative real-time PCR (qRT-PCR) analysis showed that the expression levels of GmENOD40-1, GmENOD40-2 (two early nodulin genes), and GmLba (one leghemoglobin gene) were significantly increased in T3 HTLs under P deficiency at 24 and 32 d after inoculation (DAI). The increased transcript levels of GmGS1β1 and GmGS1β2 (two cytosolic glutamine synthetase genes) in the T3 HTLs were consistent with the increase in glutamine synthetase (GS, EC 6.3.1.2) activity at 32 DAI. Our results indicated that the overexpression of OsPT2 in T3 HTLs enhances N2 fixation and ammonium assimilation activity under low P stress.

        Prediction of the Amount of Soil Discharged by an Earth Pressure Balanced Shield Machine Based on Feature Engineering

        The ground loss caused by earth pressure balance (EPB) shield tunneling is a serious threat to the safety of ground and underground buildings. The amount of soil discharged is a direct factor causing the ground loss, but the feedback of the amount of soil discharged has the problems of human subjectivity and information lag. Based on the theory of soil discharged and shield construction data, a data model for predicting the amount of soil discharged is constructed, and a predictive method of soil discharged based on adaptive adjustment strategy and parameters to achieve optimal regression is proposed. Taking Changzhou shield tunnel construction project as an example, through comparative analysis, the main influencing factors of soil discharge data model are screw-conveyor torque, screw-conveyor revolutions, total thrust, screw-conveyor entrance earth pressure, the revised standard penetration test blow-count, and compression modulus. The results show that the mean absolute error (MAE) of the model is 0.911 m3, and the mean absolute percentage error (MAPE) is 2.78%, which verifies the effectiveness of the prediction of soil discharged by feature engineering.

        CFD based simulations of flutter characteristics of ideal thin plates with and without central slot

        In this paper, the airflow around an ideal thin plate (hereafter referred to as ITP) with various ratios of central slot is simulated by using the finite-difference-method (FDM)-based Arbitrary-Lagrangian-Eulerian descriptions for the rigid oscillating body. The numerical procedure employs the second-order projection scheme to decouple the governing equations, and the multigrid algorithm with three levels to improve the computational efficiency in evaluating of the pressure equation. The present CFD method is validated through comparing the computed flutter derivatives of the ITP without slot to Theodorsen analytical solutions. Then, the unsteady aerodynamics of the ITP with and without central slot is investigated. It is found that even a smaller ratio of central slot of the ITP has notable effects on pressure distributions of the downstream section, and the pressure distributions on the downstream section will further be significantly affected by the slot ratio and the reduced wind speeds. Continuous increase of $A_2^*$ with the increase of central slot may be the key feature of the slotted ITP. Finally, flutter analyses based on the flutter derivatives of the slotted ITP are performed, and moreover, flutter instabilities of a scaled sectional model of a twin-deck bridge with various ratios of deck slot are investigated. The results confirm that the central slot is effective to improve bridge flutter stabilities, and that the flutter critical wind speeds increase with the increase of slot ratio.

      • miR-9 Modulates Osteosarcoma Cell Growth by Targeting the GCIP Tumor Suppressor

        Osteosarcoma is the most common primary bone tumor in humans, especially in childhood. However, the genetic etiology for its pathogenesis remains elusive. It is known that microRNAs (miRNAs) are involved in the development of tumor progression. Here we show that microRNA-9 (miR-9) is a potential oncogene upregulated in osteosarcoma cells. Knockdown of miR-9 in osteosarcoma resulted in suppressed colony formation and cell proliferation. Further study identified GCIP, a Grap2 and cyclin D interacting protein, as a direct target of miR-9. In addition, GCIP overexpression activated retinoblastoma 1 (Rb) and suppressed E2F transcriptional target expression in osteosarcoma cells. Moreover, GCIP depletion reversed miR-9 knockdown induced colony formation and cell proliferation suppression. In sum, these results highlight the importance of miR-9 as an oncogene in regulating the proliferation of osteosarcoma by directly targeting GCIP and may provide new insights into the pathogenesis of osteosarcoma.

      • Expression and Role of ICAM-1 in the Occurrence and Development of Hepatocellular Carcinoma

        Intercellular adhesion molecule-1 (ICAM-1) is a member of the immunoglobulin superfamily, its main function being to participate in recognition and adhesion between cells. ICAM-1 is considered closely related to occurrence, development, metastasis and invasion process of hepatocellular carcinoma (HCC). A variety of inflammatory cytokines and stimulus affect its expression through the nuclear factor-kappa B (NF-${\kappa}B$) signal transduction pathway. In the initial stage of inflammation, hepatocirrhosis and tumor development, ICAM-1 is expressed differently, and has varied effects on different cells to promote occurrence of malignancy and metastasis. ICAM-1 has diagnostic significance for AFP-negative or suspected HCC, and may be a prognositic significance. It is thus widely used in studies as a biomarker which reflects cancer cells metastasis as well as curative effect of drugs. Many new treatments of HCC may be based on the effects of ICAM-1 on different levels of function.
