      • Application of Improved Grid Search Algorithm on SVM for Classification of Tumor Gene

        According to the merits and shortcomings of the traditional gridsearch algorithm in parameters optimization of support vector machine (SVM), an improved grid search algorithm is proposed. Dichotomous search algorithm is used to reduce target searching range. First, searching range is determined roughly, and a set of parameters are obtained. Then fine search is applied in reduction the range for searching, and searching the optimum parameters.Three kinds of famous tumor gene data set are used in the comparison experiments to validate the classification accuracy of principal component analysis (PCA)-SVM and kernel principal component analysis (KPCA)-SVM. Experiment results and data analysis shows that, comparing with traditional gridsearch algorithm, the proposed method has higher classification accuracy and less search time.

        AtFes1A is essential for highly efficient molecular chaperone function in Arabidopsis

        AtFes1A is a co-chaperone of heat shock protein 70 (Hsp70), which is a key component of plant molecular chaperone systems. Plants without AtFes1A generally present defective thermotolerance. However, the underlying mechanism of this molecule has remained elusive. In the present study, we used thermolabile firefly luciferase as a molecular reporter to evaluate the efficiency of the plant molecular chaperone system. Renaturation of firefly luciferase in atfes1a mutants lagged far behind that of wild-type (WT) plants. In contrast, in vitro AtFes1A did not affect renaturation of firefly luciferase, whereas Hsp70 did significantly. We propose that AtFes1A is indirectly involved in plant molecular chaperone systems.

        Association of the Porcine Cluster of Differentiation 4 Gene with T Lymphocyte Subpopulations and Its Expression in Immune Tissues

        Cluster of differentiation 4 (CD4) is mainly expressed on $CD4^+$ T cells, which plays an important role in immune response. The aim of this study was to detect the association between polymorphisms of the CD4 gene and T lymphocyte subpopulations in pigs, and to investigate the effects of genetic variation on the CD4 gene expression level in immune tissues. Five missense mutations in the CD4 gene were identified using DNA pooling sequencing assays, and two main haplotypes (CCTCC and AGCTG) in strong linkage disequilibrium (with frequencies of 50.26% and 46.34%, respectively) were detected in the population of Large White pigs. Our results indicated that the five SNPs and the two haplotypes were significantly associated with the proportions of $CD4^-CD8^-$, $CD4^+CD8^+$, $CD4^+CD8^-$, $CD4^+$ and $CD4^+/CD8^+$ in peripheral blood (p<0.05). Gene expression analysis showed the mRNA level of the CD4 gene in thymus was significantly higher than that in lymph node and spleen (p<0.05). However, no significant difference was observed between animals with CCTCC/CCTCC genotype and animals with AGCTG/AGCTG genotype in the three immune tissues (p>0.05). These results indicate that the CD4 gene may influence T lymphocyte subpopulations and can be considered as a candidate gene affecting immunity in pigs.

        Drying kinetics and physicochemical properties of kumquat under hot air and air-impingement jet dryings

        Kumquat is famous for its unique flavor and nutritional value. In this study, the drying kinetics, moisture effective diffusivity, phytochemical properties, and antioxidant capacities of kumquat dried by hot air drying (HAD) and air-impingement jet drying (AIJD) were comparatively investigated. The results showed that drying rate, moisture effective diffusivity, and nutrient retention under AIJD were better than those under HAD. Fourteen polyphenols were identified by UPLC-QqQ-MS/MS in kumquat slices. The content of limonoid was significantly increased after AIJD. It was also found that high temperature contributed to a higher drying rate. However, most of the polyphenol components decreased at high drying temperatures. Accordingly, AIJD 60 °C was regarded as the optimum condition for kumquat drying. This work contributed to a better understanding of the drying character of kumquat under AIJD and showed the bioactive compounds and antioxidant activities are affected by drying methods.

        在全球能源转型与环境保护的大背景下,新能源专业的人才培养显得尤为重要。工程教育认证视角下新能源专业劳动教育协同育人模式,以储能原理与技术课程为例,提出了一个创新的教学方案。本研究围绕工程教育认证的核心要求,探索了将劳动教育融入专业学习的具体实践路径,旨在提升学生的实践能力、创新能力和社会责任感。通过实验、项目驱动学习、企业实习等方式,强化理论与实践的结合,促进学生能力的全面发展。研究结果表明,该协同育人模式能有效地提升学生的专业技能和工程素养,为新能源领域培养出更具实践经验和创新能力的高质量人才。本研究不仅对新能源专业教育具有重要的指导意义,也为其他工程专业的人才培养提供了可借鉴的经验。 In the context of global energy transformation and environmental protection, talent cultivation in the field of new energy is particularly important. A collaborative education model for labor education in the field of new energy from the perspective of engineering education professional certification, taking the Energy Storage Principles and Technology course as an example, proposes an innovative teaching plan. This study focuses on the core requirements of engineering education professional certification and explores specific practical paths for integrating labor education into professional learning, aiming to enhance students’ practical ability, innovation ability, and social responsibility. By conducting experiments, project-driven learning, and enterprise internships, we aim to strengthen the integration of theory and practice and promote the comprehensive development of student abilities. The research results indicate that this collaborative education model can effectively enhance students' professional skills and engineering literacy and cultivate high-quality talent with more practical experience and innovative abilities in the field of new energy. This study not only has important guiding significance for the education of new energy majors but also provides valuable experience for talent cultivation in other engineering majors.

        Tissues Expression, Polymorphisms Identification of FcRn Gene and Its Relationship with Serum Classical Swine Fever Virus Antibody Level in Pigs

        Neonatal Fc receptor (FcRn) gene encodes a receptor that binds the Fc region of monomeric immunoglobulin G (IgG) and is responsible for IgG transport and stabilization. In this report, the 8,900 bp porcine FcRn genomic DNA structure was identified and putative FcRn protein included 356 amino acids. Alignment and phylogenetic analysis of the porcine FcRn amino acid sequences with their homologies of other species showed high identity. Tissues expression of FcRn mRNA was detected by real time quantitative polymerase chain reaction (Q-PCR), the results revealed FcRn expressed widely in ten analyzed tissues. One single nucleotide polymorphism (SNP) (HQ026019:g.8526 C>T) in exon6 region of porcine FcRn gene was demonstrated by DNA sequencing analysis. A further analysis of SNP genotypes associated with serum Classical Swine Fever Virus antibody (anti-CSFV) concentration was performed in three pig populations including Large White, Landrace and Songliao Black pig (a Chinese indigenous breed). Our results of statistical analysis showed that the SNP had a highly significant association with the level of anti-CSFV antibody (At d 20; At d 35) in serum (p = 0.008; p = 0.0001). Investigation of expression and polymorphisms of the porcine FcRn gene will help us in further understanding the molecular basis of the antibody regulation pathway in the porcine immune response. All these results indicate that FcRn gene might be regarded as a molecular marker for genetic selection of anti-CSFV antibody level in pig disease resistance breeding programmes.
