The effects of high relative humidity (RH) on the physicochemical properties and microbial activity of mature green tomatoes ('Dombito') during refrigerated storage were determined at three temperatures (5, 10, and $15^{\circ}C$) and four different RH levels (91, 94, 97, and 99%). At each temperature, the weight loss rates of tomatoes at different levels of RH were significantly (p<0.05) different from each other. For the samples stored at $10^{\circ}C$, the weight losses were generally higher than those for the samples at $15^{\circ}C$ within the same RH level (i.e., greater vapor pressure deficit). The color change rates ('a' value) showed positive slopes, indicating that the tomato color was changing from green to red. Neither bacteria nor fungi caused visible damages to the samples, and the microbial counts were below 650 colony forming units/$cm^2$ during the test period.

The optimum condition for rat hepatocyte primary cell culture as a model system for studying the role of stearate on cholesterol metabolism was determined. Up to the cell concentration of 1.5x10^6 cells/well, there was a linear relationship (r = 0.96) between the cell number and the quantity of cellular protein recovered after the incubation. Stearate bound to BSA at high levels in the medium (>97%) at all stearate-BSA ratios. Therefore, it is concluded that the cellular protein (mg) can be used as the base unit to express the results and stearate-BSA ratio of 5.6 was determined to be the optimum concentration when considering the difficulty of handling the medium at higher ratios.

The release of soluble peptides, methionine(Met) and cysteine(Cys) from cooked dry beans- Phaseolus vulgaris cv. Great Northern, Navy, nd Pinto beans- by in vitro proteolysis using an enzyme mixture of trypsin, a-chymotrypsin, peptidse, and protease were investigted. After in vitro enzymatic hydrolysis, more than 90% of the proteins were available as soluble peptides and free amino acids for all dry bean smaples. Greater than 80% of total Met and Cys were available either a soluble peptides of free form. More than 90% of available Met was released during first 4 hr of hydrolysis while the rate fo Cys release increased during the hydrolysis period.

The effects of storage temperature and high relative humidity (RH) on the weight loss, color change, and microbial activity of a mushroom ('Sylvan' hybrid white) were investigated. The experiment was performed at three temperature (5, 10 and $15^{\circ}C$) and four different relative humidity levels (91, 94, 97 and 99%). The weight loss of the tested samples had a highly correlated linear relationship with storage time at each RH level during storage. Both the storage temperature and RH levels in the experiment had signigicant effects (p<0.05) on the weight-loss rate of the tested samples. The loss whiteness of mushrooms was not significantly affected (p>0.05) by RH ranges at the same temperature. No visible damage was caused by either bacteria or fungi in all samples during storage.
상추(Lactuca sativa L.) 유래 항동맥경화물질 규명 및 가공적성 실험을 통한 건강기능성 식품 시제품 개발

우리나라 전역에서 전통적으로 식용 및 약용으로 사용되어온 상추에서 생리활성물질을 탐색한 결과 항동맥경화성이 발견되어 상추로부터 항동맥경화성을 갖는 물질을 분리·정제하여 물질의 구조규명을 한 결과 phytol로 밝혀졌다. 또한 이 상추유래 항동맥경화물질의 열안정성, 광안정성 등 가공적성을 연구하여 이를 토대로 최적의 건강기능성 식품의 모델에 대한 연구를 하여 항동맥경화물질을 함유한 건강기능성 시제품을 개발하였다. The anti-atherosclerotic substance originated from lettuce(Lactuca sativa L.) was extracted with MeOH and further partitioned by using EtOAc, n-BuOH and H_(2)O. The substance was determined as phytol by NMR and MS. The lettuce extract with anti-atherosclerotic activity was tested for processing characters such as heat and light stability. Food model was developed with the processing parameters of the lettuce extract. The extracts was finally processed as a functional food in a tablet form with other biologically active ingredients.
β-Cyclodextrin Reduces Obesity in C57BL/6J Mice Induced by High Fat Diet

Anti-obesity effects of β-cyclodextrin in obese C57BL/6J mice induced by a high fat diet (HD) were observed. The administration of β-cyclodextrin reduced the gain of body weight, abdominal fat, liver weight, the lipid deposits of hepatocytes and the size of adipocytes in the HD group. In serum analysis, the total and low-density lipoprotein-cholesterols were significantly decreased in the β-cyclodextrin-supplemented HD group than in the HD group. However, high-density lipoprotein-cholesterol was not changed in these groups. In hypothalamic homogenates, the decrease of neuropeptide Y and increase of α-melanocyte stimulating hormone were detected in the β-cyclodextrin-supplemented HD group compared to that in the HD group. These effects of β-cyclodextrin were similar to those of Garcinia cambogia, which is widely used as a natural anti-obesity product. These results suggest that β-cyclodextrin has anti-obesity effects through the lowering of the abdominal fat pad and inhibits the central effects of hunger.

Diospyros kaki (Persimmon) fruits and leaves have been used as a traditional medicine and include a number of therapeutic components. The present study investigated neuroprotective effect of the extract of Diospyros kaki stem (EDKS) against ischemic damage in the gerbil hippocampal CA1 region after 5 min of transient cerebral ischemia. To elucidate the neuroprotective effect of EDKS on ischemic damage, 50 mg/kg EDKS was administered orally to gerbils for 7 days before the ischemic surgery, and we used cresyl violet and Fluoro-Jade C (F-J C) staining. In the EDKS-treated group, many survived pyramidal neurons in the CA1 region was stained with cresyl violet and a few degenerative pyramidal neurons was stained with F-J C. The survival rate of the CA1 pyramidal neurons was about 62% compared to that in the vehicletreated group. In the EDKS-treated group, isolectin B4 (a marker for activated microglia) immunoreactivity was significantly lower than that in the vehicle-treated group. These results suggest that repeated supplement of EKDS can protect neurons from ischemic damage.
