<P>A sensitive and specific high-performance liquid chromatography–quadrupole time-of-flight–mass spectrometry (LC–QTOF–MS) method combined with liquid chromatography–tandem mass spectrometry (LC–MS/MS) was developed for the determination of grayanotoxins I and III in dietary supplements and homemade wine. Grayanotoxins I and III were successfully extracted using solid-phase extraction cartridges, characterized by LC–QTOF–MS, and quantitated by LC–MS/MS. The LC–MS/MS calibration curves were linear over concentrations of 10–100 ng/mL (grayanotoxin I) and 20–400 ng/mL (grayanotoxin III). Grayanotoxins I and III were found in 51 foodstuffs, with quantitative determinations revealing total toxin concentrations of 18.4–101 000 ng/mL (grayanotoxin I) and 15.3–56 000 ng/mL (grayanotoxin III). 본 연구에서는 식육의 중금속 기준규격 설정을 위하여 다소비 식품인 소, 돼지, 닭, 오리, 햄 및 소시지에 대하여 중금속 함량을 조사하였다. 국내 유통 중 식육 및 식육 가공품 총 466건을 2008년 5월부터 11월까지 구입하고 납, 카드뮴, 비소 및 수은의 함량을 분석하였다. 시료의 분해를 위해 관련 전처리 방법을 비교하여 검증된 중금속 분석법을 찾고자 하였으며, 이를 근거로 소, 돼지, 닭, 오리, 햄 및 소시지 6종류의 시료에 대하여 납, 카드뮴, 비소 및 수은을 모니터링하고 안전성 평가를 위해, 수집된 자료와 식이섭취량으로 JECFA의 주간잠정섭취허용량와 비교 분석하고자 하였다. 전처리법으로 비교한 습식 분해법과 마이크로웨이브법은 회수율이 모두 80% 이상이며 재현성(RSD)은 10% 미만으로 나타났으나 전처리 시간과 조작의 편리성에서 마이크로웨이브법이 나은 것으로 판단되었다. 따라서 납, 카드뮴, 비소의 경우 산 혼합액을 사용한 마이크로웨이브법으로 시료를 전처리하여 ICP-MS로 분석하였고, 수은은 수은분석기를 사용하여 직접 분석하였다. 전반적으로 낮게 검출된 원소는 카드뮴과 수은이었으며, 납과 비소는 상대적으로 높게 나타났다. 납의 경우 평균함량이 식육은 0.006-0.010 mg/kg이었고, 식육가공품은 0.005-0.009 mg/kg이었다. 카드뮴의 경우 평균함량이 식육은 0.000-0.001 mg/kg이었고, 식육가공품은 0.001-0.002 mg/kg이었다. 비소의 경우 평균함량이 식육은 0.004-0.021 mg/kg이었고, 식육가공품은 0.014-0.018 mg/kg이었다. 그리고 수은 함량은 식육에서 0.710-0.902 μg/kg이었고, 식육가공품은 1.052-1.141 μg/kg이었다. 안전성 평가를 위해 본 연구에서 측정된 모니터링 자료를 이용하여 JECFA(Joint FAO/WHO Expert Committee on Food Additives)의 주간잠정섭취허용량(Provisional Tolerable Weekly Intake, PTWI)와 비교하여 노출량을 평가한 결과, 본 사업에서 검출된 중금속 함량은 비교적 안전한 수준으로 확인할 수 있었다. This study was conducted to examine the contents of lead (Pb), cadmium (Cd), arsenic (As), and mercury (Hg) in meat and meat products in Korea. The contents of Pb, Cd, As, and Hg in 466 samples of beef, pork, chicken, duck, ham, and sausage were measured using inductively coupled plasma mass spectrometry or a mercury analyzer. Wet ashing and microwave method were compared, and the recovery and reproducibility of the microwave method were better than those of wet ashing for meat and meat products. The recovery of the microwave method was 98.1% for Pb, 104.6% for Cd, and 103.4% for As, respectively. The best result was obtained through digestion using an acid mixture (HNO<sub>3</sub>/H<sub>2</sub>O<sub>2</sub>, 6:2). Hg content was measured using a mercury analyzer. As a result, the contents of Hg and Cd in samples were lower than those of Pb and As. The average contents of Pb were 0.009 mg/kg in beef, 0.010 mg/kg in pork, 0.006 mg/kg in chicken, 0.007 mg/kg in duck, 0.005 mg/kg in ham, and 0.009 mg/kg in sausage. The average Cd contents were 0.0004 mg/ kg in beef, 0.0004 mg/kg in pork, 0.0005 mg/kg in chicken, 0.0012 mg/kg in duck, 0.0015 mg/kg in ham, and 0.0019 mg/kg in sausage. The average As contents were 0.016 mg/kg in beef, 0.004 mg/kg in pork, 0.021 mg/kg in chicken, 0.010 mg/kg in duck, 0.014 mg/kg in ham, and 0.018 mg/kg in sausage. The average Hg contents were 0.713 μg/kg in beef, 0.902 μg/kg in pork, 0.710 μg/kg in chicken, 0.796 μg/kg in duck, 1.141 μg/kg in ham, and 1.052 μg/kg in sausage. Based on the results of the National Health and Nutrition Survey 2005, the levels of dietary exposure to heavy metal contaminants in meat and meat products were compared with the provisional tolerable weekly intake(PTWI) established by the Joint FAO/WHO Expert Committee on Food Additives. The average dietary exposure of the general population from meat and meat products was 0.03-0.2% of PTWI for Pb, Cd, As, and Hg, which indicates a safe level for public health at present.
