Red Blood Cell Velocity Field in Rat Mesenteric Arterioles Using Micro PIV Technique

As endothelial cells are subject to flow shear stress, it is important to determine the detailed velocity distribution in microvessels in the study of mechanical interactions between blood and endothelium. This paper describes a velocity field of the arteriole in the rat mesentery using an intravital microscope and high-speed digital video system obtained by a highly accurate PIV technique. Red blood cells (RBCs) velocity distributions with spatial resolutions of $0.8{\times}0.8{\mu}m$ were obtained even near the wall in the center plane of the arteriole. By making ensemble-averaged time-series of velocity distributions, velocity profiles over different cross-sections were calculated for comparison. The shear rate at the vascular wall also evaluated on the basis of the ensemble-averaged profiles. It was shown that the velocity profiles were blunt in the center region of the vessel cross-section while they were steep in the near wall region. The wall shear rates were significantly small, compared with those estimated from the Poiseuille profiles.
Performance Evaluation of Two e-mail Filtering Systems with Different Machine Learning Techniques

We have tried to compare the accuracy rates of classification methods in two spam mail filter systems with different machine learning techniques. One of these systems employs BONSAI and the other employs support vector machine (in short SVM) as a machine learning techniques. These two systems can classify e-mails into the sets of spam mail or non-spam mail according to the appearing rate of words in combination with the sequence analysis based on its appearing rate. The mail filtering system using BONSAI could classify e-mails into the sets of spam mail or non-spam mail with more than 90% accuracy. The other system using SVM could also classify e-mails with more than 90% accuracy. Although two systems could classify e-mails into two categories with high accuracy rate, the system using BOSAI was characterized by an efficient classification on the condition that the less learning examples were given.
Extension of Artificial Genetic Circuits with Mathematical Analysis

Synthetic biology is a study which aims at constructing artificial genetic circuits with the support of computational methods to understand biosystems deeply. We proposed a new procedure to design a computational model of a genetic circuit based on logical and dynamical formalisms. In this report, we investigated the mathematical behavior of the dynamic model for the artificial genetic oscillation circuit consisted of three genes by our proposed procedure, and compered with the one of the 3-state genetic toggle circuit by defining the differential equations for these genetic circuits and analyzing their nullclines. Each gene which composes the genetic oscillation circuit is activated cyclically by regulating each other, and the 3-state genetic toggle circuit is finally transformed into any one of three stable states.
Visualization of micro-interfacial conditions using Micro PIV

A new micro-resolution PIV (Particle Image Velocimetry) has been developed. To investigate transient phenomena in a microfluidic device, Dynamic micro-PIV system was realized by combining a high-speed camera and a CW (Continuous Wave) laser. The technique was applied to a micro-counter-current flow, consisting of water and butyl acetate. The velocity fields of water in the micro counter-current flow were visualized for a time resolution of 500 ㎲ and a spatial resolution of 2.2 × 2.2 ㎛. Using the Dynamic micro-PIV technique, the vortex-like motions of fluorescent particles at the water-butyl acetate interface were captured clearly.

For the purpose of mapping linguistic features of the world’s languages, it is necessary to identify the languages in Yamamoto-Data and SilGIS-Data. In this paper, firstly we point out the problems in the primary method that uses language name(s) for language identification. Secondly, noticing that the world’s languages are classified and grouped into languages family trees, we propose an improved method for the language classification. Finally, we give our experimental results.
Evaluation of N-myristoylation Prediction Tool using Machine Learning

Protein sequences constitute molecular complex in an organism. However it is difficult to find a sequence rule such as cascade reaction signals, post translational modification signals and so on. These sequence signals perform an essential role in regulating cellular structure and function. In previous study, we could find sequence rules of Nmyristoylated proteins easily with computational approach. Subsequently, we have developed a CGI tool to predict Nmyristoylated proteins with their sequence rules. In this study, we performed accuracy evaluation of our developed CGI tool. As a result, we show that developed CGI tool predict Nmyristoylated proteins effectively with characteristics of Nmyristoylated protein sequences.
Arp2/3 complex regulates adipogenesis by controlling cortical actin remodelling.

<P>Extensive actin cytoskeleton remodelling occurs during adipocyte development. We have previously shown that disruption of stress fibres by the actin-severing protein cofilin is a requisite step in adipogenesis. However, it remains unclear whether actin nucleation and assembly into the cortical structure are essential for adipocyte development. In the present study we investigated the role of cortical actin assembly and of actin nucleation by the actin-related protein 2/3 (Arp2/3) complex in adipogenesis. Cortical actin structure formation started with accumulation of filamentous actin (F-actin) patches near the plasma membrane during adipogenesis. Depletion of Arp2/3 by knockdown of its subunits Arp3 or ARPC3 strongly impaired adipocyte differentiation, although adipogenesis-initiating factors were unaffected. Moreover, the assembly of F-actin-rich structures at the plasma membrane was suppressed and the cortical actin structure poorly developed after adipogenic induction in Arp2/3-deficient cells. Finally, we provide evidence that the cortical actin cytoskeleton is essential for efficient glucose transporter 4 (GLUT4) vesicle exocytosis and insulin signal transduction. These results show that the Arp2/3 complex is an essential regulator of adipocyte development through control of the formation of cortical actin structures, which may facilitate nutrient uptake and signalling events.</P>
