        Simple Synthesis of Multi-Halogen Pyrazino[1,2-a]indole-1,8(2H,5aH)-diones

        A concise and efficient one-pot synthesis of multi-halogen pyrazino[1,2-a]indole-1,8(2H,5aH)-dione (MHPID) derivatives by the reaction of an enamino ester with multi-halogen benzoquinone derivatives is described. MHPIDs 3a–3d were obtained with good yields (78–83%) by refluxing enamino esters 1a and 1b and tetrahalogen-1,4-benzoquinones 2a and 2b for 24 h without the use of catalysts. Compounds 3e–3p were also obtained with excellent yields (69–92%) via the reaction of the phenyl-substituted enamino esters 1c–1h with tetrahalogen-1,4-benzoquinones 2a and 2b in CH3CN catalyzed by Cs2CO3. These two protocols are efficient and effective for the synthesis of MHPIDs.

        EXISTENCE OF SOLUTION FOR A FRACTIONAL DIFFERENTIAL INCLUSION VIA NONSMOOTH CRITICAL POINT THEORY

        This paper is concerned with the existence of solutions to the following fractional differential inclusion $$\{-{\frac{d}{dx}}\(p_0D^{-{\beta}}_x(u^{\prime}(x)))+q_xD^{-{\beta}}_1(u^{\prime}(x))\){\in}{\partial}F_u(x,u),\;x{\in}(0,1),\\u(0)=u(1)=0,$$ where $_0D^{-{\beta}}_x$ and $_xD^{-{\beta}}_1$ are left and right Riemann-Liouville fractional integrals of order ${\beta}{\in}(0,1)$ respectively, 0 < p = 1 - q < 1 and $F:[0,1]{\times}{\mathbb{R}}{\rightarrow}{\mathbb{R}}$ is locally Lipschitz with respect to the second variable. Due to the general assumption on the constants p and q, the problem does not have a variational structure. Despite that, here we study it combining with an iterative technique and nonsmooth critical point theory, we obtain an existence result for the above problem under suitable assumptions. The result extends some corresponding results in the literatures.

        We formulate a stochastic control problem on proportional reinsurance that includes impulse and regular control strategies. For the first time we combine impulse control with regular control, and derive the expected total discount pay-out (return function) from present to bankruptcy. By relying on both stochastic calculus and the classical theory of impulse and regular controls, we state a set of sufficient conditions for its solution in terms of optimal return function. Moreover, we also derive its explicit form and corresponding impulse and regular control strategies.

      • Ten Year Literature on Psychological and Behavioral Interventions Against Cancer: a Terms Analysis

        We here performed a systematic review of PBIC literature using terms analysis in a hope of both identifying potential trends and patterns and exploring methods leveraging traditional literature reviews in this specific area. Articles meeting inclusion criteria were retrieved from PUBMED and translated into dichotomized article records representing presence or non-presence of MeSH terms and a metric consisting of numbers of times of co-occurrence between all pairs of terms identified using a self-designed program. The occurrence of and relations among the terms were calculated and visualized using Excel2007 and UCINET respectively. A total of 1,742 terms were identified from 997 articles retrieved. Put in a descending order, the lines representing the times of term occurrence formed a typical hyperbolic curve; when plotted along the x-axis of whole MESH terms, the lines clustered within four specific regions. Comparison of term occurrence between 2002 and 2011 revealed priority changes in population and subjects (from general groups to priority groups), intervention approaches (from medicine to exercise and psychotherapy), methodology and techniques (from cohort studies to randomized controlled trials) and outcomes (from health and mental health to quality of life, depression etc.). Networks of the terms featured a number of closely linked groups of topics including method and questionnaires, therapy and outcomes, survival management, psychological assessment and intervention, behavioral intervention (individual and community oriented). Terms analysis revealed interesting trends and patterns about PBIC publications and both the analysis methods and findings have implications for future research and literature reviews.

        Sequence analysis and expression profiles of TaABI5, a pre-harvest sprouting resistance gene in wheat

        Pre-harvest sprouting (PHS) is the germination of grains in the spike under wet conditions before harvest, which causes a severe reduction in crop yield and flour quality. The phytohormone abscisic acid plays a key role in regulating seed dormancy, which inhibits pre-harvest spouting or vivipary for many important crops. TaABI5, belonging to a family of basic leucine zipper transcription factors could regulate a subset of late embryogenesis abundant genes during seed development, was isolated and characterized. Phylogenetic analysis revealed that TaABI5 homologous gene belonged to the ABI5 subgroup of the ABF/AREB/ABI5 subfamily. TaABI5 consist of four conserved domains and one bZip domain. The expression pattern of TaABI5 indicated that it was seed-specific and accumulated at late stages of seed development. Accumulation of TaABI5 in synthetic hexaploid wheat SHW-L1 (PHS resistance) was 8.93-fold higher than that in wheat cultivar Chuanmai 32 (PHS susceptible) at 25 days post anthesis. Four expression quantitative trait loci (eQTL) of TaABI5 on chromosome 2DS, 4DS, 6DS, and 7DL were characterized from SHW-L1/Chuanmai 32 derived recombinant inbred line population.

      • Expression of the Proto-oncogene Pokemon in Colorectal Cancer - Inhibitory Effects of an siRNA

        Objective: This study aimed to investigate expression of the proto-oncogene POK erythroid myeloid ontogenic factor (Pokemon) in colorectal cancer (CRC), and assess inhibitory effects of a small interference RNA (siRNA) expression vector in SW480 and SW620 cells. Methods: Semi-quantitative reverse transcription-polymerase chain reaction (PCR) and immunohistochemistry were performed to determine mRNA and protein expression levels of Pokemon in CRC tissues. Indirect immunofluorescence staining was applied to investigate the location of Pokemon in SW480 and SW620 cells. The siRNA expression vectors that were constructed to express a short hairpin RNA against Pokemon were transfected to the SW480 and SW620 cells with a liposome. Expression levels of Pokemon mRNA and protein were examined by real-time quantitative-fluorescent PCR and western blot analysis. The effects of Pokemon silencing on proliferation of SW480 and SW620 cells were evaluated with reference to growth curves with MTT assays. Results: The mRNA expression level of Pokemon in tumor tissues ($0.845{\pm}0.344$) was significantly higher than that in adjacent tumor specimens ($0.321{\pm}0.197$). The positive expression ratio of Pokemon protein in CRC (87.0%) was significantly higher than that in the adjacent tissues (19.6%). Strong fluorescence staining of Pokemon protein was observed in the cytoplasm of the SW480 and SW620 cells. The inhibition ratios of Pokemon mRNA and protein in the SW480 cells were 83.1% and 73.5% at 48 and 72 h, respectively, compared with those of the negative control cells with the siRNA. In the SW620 cells, the inhibition ratios of Pokemon mRNA and protein were 76.3% and 68.7% at 48 and 72 h, respectively. MTT showed that Pokemon gene silencing inhibited the proliferation of SW480 and SW620 cells. Conclusion: Overexpression of Pokemon in CRC may have a function in carcinogenesis and progression. siRNA expression vectors could effectively inhibit mRNA and protein expression of Pokemon in SW480 and SW620 cells, thereby reducing malignant cell proliferation.

        Promoting effect of the Maillard reaction products produced during the stir-frying process of Hordei Fructus Germinatus on the intestinal absorption of active ingredients in Hordei Fructus Germinatus

        This study was designed to evaluate theabsorption promoting capacity of Maillard Reaction Products(MRPs) produced during the stir-frying process ofHordei Fructus Germinatus on catechin, ferulic acid,quercetin and kaempferol by the ex vivo rat everted gut sacmodel, in situ single-pass intestinal perfusion model andthe whole animal model. Moreover, verapamil, EDTA andmannitol were used for determining the transport mechanismof catechin, ferulic acid, quercetin and kaempferol. The tight junction (TJ) proteins including zonula occudens-1(ZO-1) and claudin-1 were chosen to investigate thepromoting mechanism of MRPs by quantitative real-timePCR (qRT-PCR) and western blot analyses. The resultsshowed that the MRPs produced during the stir-fryingprocess of Hordei Fructus Germinatus could improve theintestinal absorption of catechin, ferulic acid, quercetin andkaempferol. And the absorption-promoting effect of MRPswas related to chelating effect and the reduced expressionof claudin-1 and ZO-1. Our results suggested that MRPscould be promising oral absorption promoters, which mightbe another processing mechanism of Hordei FructusGerminatus.

        Structural-based Screening of L-phenylglycine Aminotransferase using L-phenylalanine as the Optimal Amino Donor: Recycling of L-phenylalanine to Produce L-phenylglycine

        The aproteinogenic amino acid, L-phenylglycine, is an important side chain building block for some drugs. It would be of great commercial and environmental value to biocatalyse L-phenylalanine to L-phenylglycine, and thus replace the organic synthesis method. To produce Lphenylglycine from L-phenylalanine, an L-phenylglycine aminotransferase was screened and characterized. HpgTAO showed high homology to α-aminoadipate aminotransferase. The L-phenylalanine binding site was near the residues S26, R401, N201, and G46 in HpgTAO, and L-phenylalanine formed a hydrogen bond with Asn20, which was similar to the substrate binding mechanism of α-aminoadipate aminotransferase. HpgTAO showed increased activity in alkalescent environment below 40°C. The kinetic analysis showed that L-phenylalanine had the highest affinity to HpgTAO, which ensured the recycle biosynthesis of Lphenylglycine from L-phenylalanine. To date, it was the only aminotransferase using L-phenylalanine as an optimal amino donor. The L-phenylglycine biocatalysis operon was also constructed by co-expressing the hmaS, hmo and hpgT by a single plasmid. The first in vitro conversion of Lphenylalanine to L-phenylglycine was achieved by directly using the L-phenylalanine fermentation broth as the raw material.
